Molecular Cloning and Expression of a Sodium-Driven Flagellar Motor Component Gene(motX) from Vibrio fluvialis

  • The bacterial flagellar motor is a molecular machine that couples proton or sodium influx to force generation, mostly for driving rotation of the helical flagellar filament. In this study, we cloned a gene (motX) encoding a component of the sodium-driven flagellar motor from Vibrio fluvialis. The nucleotide sequence of the motX gene, composed of 633 bp and 211 amino acid residues, was determined. Overexpression of the motX gene in Escherichia coli using a strong promoter induced growth inhibition and cell lysis. The lethal effect of E. coli was suppressed by adding amiloride, as a potent inhibitor for the sodium channel. Electron microscopic observation of the expressed protein indicated that MotX protein induced by isopropyl ${\beta}$-D-thiogalactopyranoside caused the lysis of host cell.

  • The rotation of the flagellar motor is powered by the electrochemical gradient of specific ions across the cytoplasmic membrane. Recently, the gents of the Na'-driven motor have been cloned from marine bacterium of Vibrio sp. and some of the motor proteins have been purified and characterized. Also, motx gene encoding a channel component of the sodium type flagellar motor was identified from Vibrio Huuiaiis (KTCC 2473). The amino acid sequence of MotX protein from V, Huvialis shared 90, 85, $85\%$ identity with V, cholerae, V. alginolyticus, V parahaemolyticus, respectively. We have studied the localization of the expressed MotX protein in Escherichia coli by immune-gold labeling of ultra-thin frozen section. Our observation of the expressed protein indicated that MotX protein could be existed as attachment to inner membrane in E. coli.

소성 조건과 Zr/Ti 몰비에 따른 졸겔 $Pb(Zr_{1-x}Ti_x)O_3$ 박막의 구조 및 강유전 특성 (Structural and ferroelectric characteristics of sol-gel $Pb(Zr_{1-x}Ti_x)O_3$ thin films according to the sintering conditions and Zr/Ti mol%)

  • In this study, we have analyzed structural analysis and measured ferroelectric characteristics of PZT thin films prepared by sol gel process with different sintering conditions and different Zr/Ti mot%. When the Zr mot% of PZT thin film was increased, it was found that the remanent. polarization and coercive field were decreased and increased, respectively. Also, the maxium dielectric constant of PZT(50/50) thin film was 786.8. We got double hysteresis(anti-fcrroelectric) curve from PbZrO$_{3}$ thin film. As heating rate goes up, pyrochlore phase of PZT thin film was decreased and dielectric and ferroelectric characteristics were improved. As a result of variation of sintering temperature and time 500.deg. C-800.deg. C and 5 sec.-8 hours, respectively, we got optimal sintering temperature and time. The optimium sintering temperature and time of conventional furnace method and rapid thermal processing method were 650.deg. C-700.deg. C for 30-60 minutes and 700.deg. C/20 seconds-2 minutes, respectively.

Annuloaortic Ectasia 의 치험 1례 보고 (Annuloaortic Ectasia Associated with Aortic Regurgitation (One case report))

  • The incidence of annuloaortic ectasia has known rare, and approximately 5-10% of aortic regurgitation. The patient was 44 years old male who complained exertional dyspnea and left anterior chest pain. He had done Lt. side 2 stage thoracoplasty for pulmonary tuberculosis about 20 years ago at Dept.of Chest surgery of National Medical Center. At that time, there was no abnormal findings in cardiovascular system. The preoperative aortic cineangiogram showed pear shaped dilatation [7.3 cm x 6.8 cm] of aortic mot with aortic valve regurgitation but left ventricular ejection function was fair. Preop. ventilatory function test showed mixed type pulmonary insufficiency. Recently, we corrected surgically, by AVR with Carpentier-Edwards Bioprosthesis [29mm] & supracoronary Woven Dacron graft [29mm x 5cm] replacement, with good clinical result for follow up 6 months.

가변 시간 뉴톤-랍손 부동소수점 역수 제곱근 계산기 (A Variable Latency Newton-Raphson's Floating Point Number Reciprocal Square Root Computation)

  • 부동소수점 제곱근 계산에 많이 사용하는 뉴톤-랍손 부동소수점 역수 제곱근 알고리즘은 일정한 횟수의 곱셈을 반복하여 역수 제곱근을 계산한다. 본 논문에서는 뉴톤-랍손 역수 제곱근 알고리즘의 반복 과정의 오차를 예측하여 오차가 정해진 값보다 작아지는 시점까지 반복 연산하는 알고리즘을 제안한다. `F`의 역수 제곱근 계산은 초기값 '$X_0={\frac{1}{\sqrt{F}}}{\pm}e_0$'에 대하여, '$X_{i+1}=\frac{{X_i}(3-e_r-{FX_i}^2)}{2}$, $i\in{0,1,2,{\ldots}n-1}$'을 반복한다. 중간 곱셈 결과는 소수점 이하 p 비트 미만을 절삭하며, 절삭 오차는 '$e_r=2^{-p}$' 보다 작다. p는 단정도실수에서 28, 배정도실수에서 58이다. '$X_i={\frac{1}{\sqrt{F}}}{\pm}e_i$'라고 하면 '$X_{i+1}={\frac{1}{\sqrt{F}}}-e_{i+1}$, $e_{i+1}{<}{\frac{3{\sqrt{F}}{{e_i}^2}}{2}}{\mp}{\frac{{Fe_i}^3}{2}}+2e_r$이 된다. '$|{\frac{\sqrt{3-e_r-{FX_i}^2}}{2}}-1|<2^{\frac{\sqrt{-p}{2}}}$'이면,'$e_{i+1}<8e_r$이 부동소수점으로 표현 가능한 최소값보다 작아지며, '$X_{i+1}\fallingdotseq{\frac{1}{\sqrt{F}}}$'이다. 본 논문에서 제안한 알고리즘은 입력 값에 따라서 곱셈 횟수가 다르므로, 평균 곱셈 횟수를 계산하는 방식을 도출하고, 여러 크기의 근사 역수 제곱근 테이블($X_0={\frac{1}{\sqrt{F}}}{\pm}e_0$)에서 단정도실수 및 배정도실수의 역수 제곱근 계산에 필요한 평균 곱셈 횟수를 계산한다 이들 평균 곱셈 횟수를 종래 알고리즘과 비교하여 본 논문에서 제안한 알고리즘의 우수성을 증명한다. 본 논문에서 제안한 알고리즘은 오차가 일정한 값보다 작아질 때까지만 반복하므로 역수 제곱근 계산기의 성능을 높일 수 있다. 또한 최적의 근사 역수 제곱근 테이블을 구성할 수 있다. 본 논문의 연구 결과는 디지털 신호처리, 컴퓨터 그라픽스, 멀티미디어, 과학 기술 연산 등 부동소수점 계산기가 사용되는 분야에서 폭 넓게 사용될 수 있다.

치과용(齒科用) 은(銀)-파라디움합금(合金)의 합금원소(合金元素)가 제성질(諸性質)에 미치는 영향(影響)에 관(關)한 연구(硏究) (EFFECTS OF ALLOYING ELEMENTS ON VARIOUS PROPERTIES OF DENTAL SILVER-PALLADIUM ALLOYS)

  • Even though the tarnishing and corrosion problems characteristic with dental silver-palladium alloy are not yet fully solved, it is recently widely used because of its low cost. However the effects of major alloying elements on the various properties of this system are not fully understood. The object of this research is to clarify the effects of In and Zn additives on the corrosion and tarnishing resistances and precipitation hardening behavior of this sytem, using electrodynamic polarization, immersion, and Vicker's microhardness test and X-ray diffraction and electron probe micro analysis methods. The obtained results were as follows: I. As indium content is increased, both the corrosion resistance in Cl-solution and microhardness are also increased while the tarnishing resistance is decreased. 2. As Zinc content is increased, the corrosion resistance is decreased, but tarnishing resistance is increased 3. At 70Ag-25Pd-2.5Zn-2.5In composition, the precipitation harding behavior was mot significant. The optimum aging temperature was $450^{\circ}C$ and the time was 2 hrs. The resulting specimen of this work carried 180VHN. 4. Under the heat treatment, the changes in the mechanical property are due to the changes in the shape and composition of dendrite matrix, namely, it is because of the precipitation hardening behavior which has been proved by electron probe micro analysis and optical microscopic finding.

진도개 동결정액 제조를 위한 정액성상과 동결정액의 운동성에 관한 연구 (Study on the Motility of Frozen Semen and Characteristics for Frozen Semen Processing of Jindo-Dog)

  • 본 연구는 진도개 동결정액 제조기술을 정립하기 위하여 진도개 정액성상과 동결 전후 정액의 활력과 생존율 및 CASAs를 이용한 운동성 등에 대하여 조사하였고, 백구와 황구간, 개체간의 정액성상과 내동성을 비교, 조사하였다. 이상의 연구결과는 다음과 같다. 1. 총 63회 정액채취 후 신선정액의 평균 정액량 3.8 $m\ell$, 농도 145.6$\times$$10^{6}$$m\ell$, 총정자수 396.2$\times$08/$m\ell$, 전진운동율 79.7% 및 생존율 89.5% 였다. 황구와 백구간에는 황구가, 개체간에는 황구 2호가 정자농도, 총정자수, 전진운동정자율 및 생존율 등의 정액성상에서 유의적으로 우수하였다(P<0.05). 2. 동결전.후 정자의 전진운동율과 생존율을 46회 조사한 결과 동결전 73.5%와 82.3%를, 동결후 51.1%와 64.9%를 나타내 동결과정이 전진운동율과 생존율에 영향이 있었으며, 역시 황구와 백구간에는 황구가, 개체간에는 황구 2호가 동결전후 전진운동율과 생존율에서 유의적인 차이가 인정되었다 (P<0.05). 3. 동결.융해정자의 보다 객관적인 평가를 위하여 CASA system을 이용한 총 44회 평가한 결과, 생존율 65.6%, 전진운동율 54.8%, VAP 75.3 $\mu\textrm{m}$/sec, VCL 90.0 $\mu\textrm{m}$/sec, VSL 69.4 $\mu\textrm{m}$/sec 및 ALH 4.4 $\mu\textrm{m}$로 동결 융해 정액의 운동성은 양호하였고, 황구와 백구간의 운동성에는 유의적인 차이가 없었으나, 개체간에는 역시 황구 2호가 운동성이 우수하여 유의적인 차이가 인정되었다 (P<0.05). 4. 채취된 정액중 백구는 46%(13/28), 황구는 94%(33/35)가 동결정액 제조가 가능해 황구가 내동성이 좋았으며, 전체적으로 73%(46/63) 동결정액 제조가 가능하였다. 결론적으로 진도개의 동결정액을 제조하기 위하여 정액성상 및 동결 전후 운동성을 조사한 결과 동결정액 제조와 생산체계의 구축이 가능하였으며, 황구와 백구간, 개체간의 정액성상과 동결전후의 운동성에 차이가 인정되므로 정액성상과 내동성을 고려한 종견선발 체계가 필요하다고 사려되었다.

$PbWO_{4}-TiO_{2}-CuO-B_{2}O_{3}$ 세라믹의 고주파 유전특성 (Microwave Dielectric Properties of $PbWO_{4}-TiO_{2}-CuO-B_{2}O_{3}$ Ceramics)

  • PbWO$_4$ can be densified at 85$0^{\circ}C$ and it shows fairy good microwave dielectric properties; dielectric constant($\varepsilon$$_{r}$) of 21.5, quality factor(Q $\times$f$_{0}$) of 37,224 GHz, and temperature coefficient of resonant frequency($\tau$/suf f/) of -31ppm/$^{\circ}C$. Due to its low sintering temperature, PbWO$_4$ can be used as a multilayered chip component at microwave frequency with high electrical performance by using high conductive electrode metals such as Ag and Cu. However, in order to use this material for microwave communication devices, the $\tau$$_{f}$ of PbWO$_4$ must be stabilized to near zero with high Q$\times$f$_{0}$. In present study, PbWO$_4$ was modified by adding TiO$_2$, B$_2$O$_3$, and CuO in order to improve the microwave dielectric properties without increasing the sintering temperature. The addition of TiO$_2$ increased the $\tau$$_{f}$ and $\varepsilon$$_{r}$, due to its high rr(200ppm/$^{\circ}C$) and $\varepsilon$$_{r}$(100). However, the addition of TiO$_2$ reduced the Q$\times$f$_{0}$ value. When the mot ratio of PbWO$_4$ and TiO$_2$ was 0.913:7.087, near zero $\tau$$_{f}$(0.2ppm/$^{\circ}C$) was obtaibed with $\varepsilon$$_{r}$=22.3, and Q$\times$f/$_{0}$=21,443GHz. With this composition, various amount of B$_2$O$_3$ and CuO were added in order to improve the quality factor. The addition, of B$_2$O$_3$ decreased the $\varepsilon$$_{r}$. However, increased Q$\times$f$_{0}$ and $\tau$$_{f}$. When 2.5 wt% of B$_2$O$_3$ was added to the 0.913PbWO$_4$-0.087TiO$_2$ ceramic, $\tau$$_{f}$ =8.2, $\varepsilon$$_{r}$=20.3, Q$\times$f$_{0}$=54784 GHz. When CuO added to the 0.913PbWO$_4$-0.087TiO$_2$ ceramic, $\tau$$_{f}$ was continuously decreased. And $\varepsilon$$_{r}$ . and Q$\times$f$_{0}$ were increased up to 1.0 wt% then decreased. At 0.1 wt% of CuO addition, the 0.913PbWO$_4$-7.087Ti0$_2$ Ceramic Showed $\varepsilon$$_{r}$=23.5, $\tau$$_{f}$=4.4ppm/$^{\circ}C$, and Q$\times$f$_{0}$=32,932 GHz.> 0/=32,932 GHz.X>=32,932 GHz.> 0/=32,932 GHz.
