The Fertility beween Morus nigra L. from Turkey and Indigenous or Cultivated Mulberry in Korea (터-키산 흑상(Morus nigra L.)화분과 한국 자생뽕 및 재배뽕간의 임성)

  • The fertilities of indigenous and cultivated mulberry species(varieties) which were polli nated with Morus nigra L. brought from Turkey were investigated. The percentage of ripening sorosis was very high in indigenous species, Morus tiliaefolia Makino and cultivated variety, Keomseolppong and it was around 50% in both Daeryukppong and Cheongilppong However, the flowers of Morus bombycis Koidz. (BOM5-8) and Yongcheonppong fell after pollination. Looking at the sizes of seeds, they are also various. Generally, it was extremely small in Morus tiliaefolia, but it was medium in Keomseolppong, Cheongilppong and Daeryukppong. The fertilization percentage was generally low with 32-42% in the varieties, but it was comparatively high in Keomseolppong and Morus tiliaefolia. The seed germination percentages of Daeryukppong and Morus tiliaefolia are 88.0% and 23.6%, respectively, but it was below 2% in Cheongilppong and Keomseolppong. The cross success percentages are 14.2% and 8.0% in Daeryukppong and Morus tiliaefolia, respectively, but it was below 0.3% in Cheongilppong and Keomseolppong.

Antioxidative Activities and Tyrosinase Inhibitory Effects of Korean Medicinal Plants

  • To discover the sources with antioxidant and tyrosinase inhibitory activities in Korean traditional medicines, 10 extract of medicinal plants were screened for their potential to scavenge stable 1,1-diphenyl-2-picryhydrazyl (DPPH) free radical, inhibit hydroxyl radical (${\cdot}OH$), total phenolic content, and inhibition of tyrosinase. The potency of DPPH radical scavenging activity was shown in the extract of Ulmus davidiana var. japonica Nakai that has a greater effect with $IC_{50}$ values of $6.49{\pm}5.43{\mu}g/mL$, than BHA ($IC_{50}=20.99{\pm}0.74{\mu}g/mL$), L-ascorbic acid $(IC_{50}=20.59{\pm}1.06{\mu}g/mL),\;and\;{\alpha}-tocopherol\;(IC_{50}=25.55{\pm}0.26{\mu}g/mL)$ as a positive control. The ${\cdot}OH$ scavenging activities were observed in the plants tested. Acanthopanax senticosus, Cirsium setiders, U. davidiana exhibited scavenging activity of more than 60% at $500{\mu}g/mL$. The scavenging activity(%) of BHA and a-tocopherol were 64.32 and 55.87% at $500{\mu}g/mL$, respectively. The total phenolic content was determined, in order to assess its effect on the extract's antioxidant activity. The total phenoic content of $33.37{\pm}0.52mg/g$ was conformed by methanolic extract of U. davidiana. The U. davidiana and Morus bombycis exhibited tyrosinase inhibitory activity with a $34.28{\pm}1.32\;and\;75.57{\pm}1.10%$, respectively. In particular, M. bombycis has stronger tyrosinase inhibitory activity than albutin with $36.48{\pm}3.56%$ as a positive control. This work showed that the inhibitory abilities of Korean medicinal plants, such as U. davidiana and M. bombycis, on DPPH free radical, inhibit hydroxyl radical (${\cdot}OH$), and inhibition of tyrosinase and total phenolic content, can be useful in the prevention and treatment of free radical-relate disease. Investigations into further isolation of inhibitory principles of U. davidiana and M. bombycis are now in progress.

Cryopreservation of Mulberry Germplasm Core Collection and Assessment of Genetic Stability through ISSR Markers

  • A simple and reliable cryo technique using desiccation and slow freezing of winter dormant buds was employed for 238 core collection of mulberry germplasm collected from diverse geographical regions and maintained under tropical conditions in the ex situ field gene bank to develop long-term biodiversity conservation for ensuring sustainable utilization of these valuable resources. Desiccation and freezing tolerance of bud grafts and excised shoot apices in the axillary buds of different Morus species under in vivo and in vitro condition indicated species-specific variation and most of the wild Morus species were found sensitive. In vitro regeneration and cryopreservation($-196^{\circ}C$) protocols using differentiated bud meristem like axillary winter dormant buds were worked out for a wide range of Morus species, land races, wild and cultivated varieties. Successful cryopreservation of mulberry winter dormant buds of different accessions belonging to M. indica, M. alba, M. latifolia, M. cathayana, M. laevigata, M. nigra, M. australis, M. bombycis, M. sinensis, M multicaulis and M. rotundiloba was achieved. Among wild species Morus tiliaefolia, and M. serrata showed moderate recovery after cryopreservation. Survival rates did not alter after three years of cryopreservation of different Morus species. ISSR markers were used to ascertain the genetic stability of cryopreserved mulberry, which showed no difference detected among the plantlets regenerated from frozen apices in comparison to the non-frozen material.

Antioxidant Capacity Analysis of Water-Soluble Substances according to Maturity Stages in Yield-type Mulberry Leaves and Fruits Collected from Kang-Won Province (강원도 일대에서 채취한 야생뽕의 시기별 뽕잎과 오디의 항산화능 분석)

  • The antioxidant capacity of yield-type mulberry leaves and fruits (Morus alba L., M. bombycis Koidz, and M. Lhou (Ser). Koidz) collected from In-je, Won-ju and Yang-yang regions, Kang-won province, Korea, was investigated with minium L-100 device and ARAW-KIT (anti-radical ability of water-soluble substance), in comparison to the ascorbic acid. The results indicated that total antioxidant capacity of yield-type mulberry leaves was 2711.2 nmol (ascorbic acid equivalents). The highest stage of antioxidant capacity (3587.6 nmol) was opening stage of five leaves in spring. Especially, antioxidant capacity of mulberry leave collected from In-je region was high than that of other regions. No. 7 mulberry leave showed the strongest antioxidant capacity (6184.9 nmol) at opening stage of five leaves in spring. Also, fluit growing condition including fruit size and yield was good, too. Therefore, No. 7 mulberry three from In-je region, possessed not only high antioxidant capacity but also considerable fruit quality, was selected as potent resource for mulberry breeding and functional material development.

  • We conducted this study to investigate the morphological characteristics and physiological effects of mulberry leaves and fruits with wild varieties. Morphologically sampling materials were showed Morus bombycis type and Morus alba type characteristics. Water contents of mulberry fruits were higher than that of mulberry leaves. Sugar degree of sampling materials was 5.1∼22.7 Brix %. The yield of 85% MeOH extract from freezing dried Mulberry Fruits was 60.2% and Hexane, BuOH and H$_2$O fraction from MeOH extract were 2.8%, 4.6%, 92.3% respectively. In the cholesterol-induced hyperlipidemia rats administered with subfraction of mulberry fruit extract, total cholesterol and serum triglyceride were decreased in the MeOH extract group and H$_2$O soluble fraction group. Also H$_2$O soluble fraction group decreased GPT, GOT and LDH level. Therefore, the above results suggested that mulberry fruit with wild varieties can help to maintain normal liver functions and to protect hyperlipidemia.

A Study on Vegetation Structure and Soil condition of Leontice microrhyncha Population (한계령풀(Leontice microrhyncha) 개체군의 식생과 토양특성)

  • This study was carried out to analyze the vegetation properties, soil characteristics and ordination of Leontice microrhyncha population in South Korea. The Leontice microrhyncha population was classified into Quercus mongolica dominant population, Morus bombycis dominant population, Fraxinus mandshurica population and Leontice microrhyncha typical dominant. The Leontice microrhyncha population was located at an elevation of 633m to 1,336m in Korea. In the study sites, soil organic matter, total nitrogen, available phosphate, exchangeable potassium, exchangeable calcium, exchangeable magnesium, cation exchange capacity and soil pH were 8.40~10.58%, 0.42~0.61%, 14.15~25.07mg/kg, 0.44~0.59cmol$^+$/kg, 2.35~6.33cmol$^+$/kg, 0.35~0.98cmol$^+$/kg, 26.04~33.48cmol$^+$/kg and 4.69~5.32 respectively. Morus bombycis dominant population was found in the low elevation and gentle sloped area that has high percentage of phosphoric acid and less percentage of total nitrogen, exchangeable calcium, exchangeable magnesium than other dominant population. Fraxinus mandshurica dominant population and Leontice microrhyncha typical population were found in the comparatively high elevation area that has low percentage of phosphoric acid and high percentage of total nitrogen, exchangeable calcium, exchangeable magnesium. Quercus mongolica dominant population was found in the medium elevation area that has medium percentage of total nitrogen, exchangeable calcium, exchangeable magnesium.

Hardwood Cutting with Callusing in the Mulberry(Morus bombycis Koidz.) II. Effect of Callusing Temperature on Root Formation and Growth (뽕나무 유합촉진 고조삽목에 관한 연구 II. 삽목온도가 발근생장에 미치는 영향)

  • Mulberry cuttings from shoots of Shinkwangppong(Morus bombycis koidz.) had been callused in vermiculite separately at 15, 20, 25 and 30$^{\circ}C$ for 15 days before transplanting them in greenhouse to make clear the effect of temperature on root formation and growth is as follow. The buds of cuttings started sprouting in 4 and 6 days of callusing at 30 and 25$^{\circ}C$, respectively, reaching 100% budding in 10 and 15 days of callusing. Budding was delayed, however, at low temperature, showing 86% and 92% at 15 and 20$^{\circ}C$, respectively, in 15 days. Rooting from the cuttings was also accelerated at high temperature, showing 97-100% rooting at 25$^{\circ}C$ and 30$^{\circ}C$, in 15 days of callusing but no more than 93% at low temperature even in 35 days. Although high temperature increased root number and length after 15 days in callusing, no differences showed in the number and the weight at more than 20$^{\circ}C$ in 35 days of cuttings.

  • Various concentrations of ${\alpha}$-naphthalence acetic acid(NAA) as a root-promoting substance were tested in hardwood cutting of the mulberry(Morus bombycis Koidz., cultivar : Shinkwangppong) to make clear the callusing effect on the budding and root growth. Budding and shoot growth of cuttings were delayed at high concentrations of NAA within 10 days of callusing. Especially more severe is it at higher than 50ppm. More than 93% of them, however, budded in two weeks when callused at less than 100ppm NAA. Although rooting was accelerated at high concentration of NAA from the bigining of cutting, after that, rooting percentage increased to reach 100% in 35 days of cutting in any concentration except 150ppm with relatively low rooting. Root growth was utmostly accelerated at 50ppm NAA to show the highest amount in number, length and weight of roots per cutting although high concentration of it decreased mean root length.

  • This study was conducted to analyze character of distribution of plants by landforms and altitude in Mt. myeonbong. The chemical properties of soil in this study sites were as follows; pH 4.86, organic 52.53 g/kg, $P_2O_5$ 7.57 mg/kg, potassium $0.18cmol^+/kg$, calcium $2.45cmol^+/kg$ and magnesium $0.49cmol^+/kg$. Generalists in the upper layer were analyzed as follows; Quercus mongolica, Pinus densiflora, Quercus variabilis, and specialists; Carpinus cordata, Tilia amurensis, Morus bombycis etc. In the middle layer, generalists were Betula schmidtii, Quercus mongolica, Sorbus alnifolia and specialists; Lindera erythrocarpa, Quercus serrata, Staphylea bumalda etc. Generalist of shrubs; Lindera obtusiloba, Rhododendron mucronulatum, Sorbus alnifolia, and specialists; Morus bombycis, Pinus densiflora, Deutzia glabrata etc. Generalists of herbaceous layer; Rhus tricocarpa, Lespedeza maximowiczii, Polygonatum odoratum var. pluriflorum and specialists; Liparis kumokiri, Davallia mariesii and Chrysosplenium flagelliferum etc.

  • In our country there are five species of Morus genus including Morus alba L.. Also their varieties and hybrids are distributed so much. In sucession of previous report we collected control and marketing specimens of Mori Cortex Radicis, comparative experiments were pharmacognostically carried out to identify the control specimen by the differences of external and internal morphology. It was difficult to identify marketing specimens by external morphology, because they are similar in spite of conparating with control specimen which the origin is definite. In internal morphology, medullary ray is developed near the cambium to primary bark in control specimen A(Morus alba series) and C(M. Lhou series), but less developed in B(M. bombycis series). The difference of these three series was observed. The thickness of cork layer is almost the same($7{\sim}12$ layers) in A and C series, but B is thin layer and sample E(that on the market) is generally more thick and has a stick cork cell. The kinds of starch, Ca-oxalate and latex, cell centents were same, but it was easy to identify them by the differences of their distribution. The bast fibre of D(wild specimen) and E were light lignified, latex tube of A and C series was richer distributed than others. These results show that the origin of Mori Cortex Radicis on the market can be appreciated in four groups of Korean Morus genus which are M. alba, M. bambycis, M. Lhou series and the others.
