닭 크립토스포리디움의 형태계측을 위한 전자현미경적 연구 (Morphometrical analysis of chicken Cryptosporidium on electron microscopy)

  • Morphometrical analysis of chicken Cryptosporidium baileyi in various stages of life cycle in the bursa of Fabricius were carried out by electron microscope to establish a differential point for identification of C baileyi. By avidin-biotin complex method, protozoans of the bursa of Fabricius were identified Cryptosporidium spp. The size and area on each developmental stages of C baileyi, as measured by Morphomat 10 attached to electron microscope were as follows. Trophozoites' size with range of $3.21{\pm}0.70{\times}2.49{\pm}0.59{\mu}m$, area with range of $118.82{\pm}41.92{\mu}m^2$; meronts' size $3.99{\pm}1.07{\times}2.96{\pm}0.52{\mu}m$, area $210.11{\pm}57.11{\mu}m^2$; merozoites' size $1.98{\pm}0.43{\times}0.60{\pm}0.18{\mu}m$, area $24.10{\pm}5.97{\mu}m^2$; microgametes' size $1.36{\pm}0.83{\times}0.50{\pm}0.23{\mu}m$, area $20.23{\pm}6.73{\mu}m^2$; macrogametes' size $4.57{\pm}0.65{\times}4.02{\pm}0.55{\mu}m$, area $258.37{\pm}51.83{\mu}m^2$; oocytes' size $4.39{\pm}0.56{\times}3.44{\pm}0.50{\mu}m$, area $187.21{\pm}62.68{\mu}m^2$. In conclusion, the size and area on each developmental stages of Cryptosporidium baileyi is different from that of other Gryptosporidia spp. It suggests, with considering tissue tropism and life cycle, morphometrical analysis can be quite a good way to identify C baileyi.

Morphological types and morphometrical measurements of the suprascapular notch in both dry bones and human cadavers: anatomical study to improve the outcomes of the diagnostic and interventional procedures in the shoulder region

  • Understanding the anatomy of suprascapular area helps the clinicians and surgeons in management of any disability at the shoulder region. This work aimed to clear the different morphological and morphometrical types of suprascapular notch (SSN). Unknown 120 dry human scapulae of both sides and 60 formalin-embalmed cadaveric upper limbs (40 males and 20 females) were used in the present study. Three main morphological forms of SSN were reported: J, U, and V-shaped. J-shaped notch showed the highest incidence followed by U-shaped then V-shaped one. Morphometrically, type (III) notch was the most prevalent in both dry bones and cadavers, while the incidence of type (II) was the lowest form. Also, the measurements of superior transverse diameter, middle transverse diameter and vertical dimension of the different types of the notch showed no side or sex significant difference. The suprascapular foramen with ossified superior transverse scapular ligament (STSL) was seen in 5.8% of dry bones and 10% of cadaveric specimens. Fan and band-shaped ossified transverse scapular ligaments were reported. Absence of SSN was seen in 10.8% of dry bones, 7.5% of male and 10% of female specimens with left side predominance. V-shaped, absence, and ossified STSL were considered as predisposing factors of suprascapular nerve entrapment syndrome. Knowledge of the morphology and morphometric parameters of SSN is of great clinical significance for anatomists, radiologists, physiotherapists, orthopedics and neurosurgeons to perform good diagnosis and best planning for surgical or arthroscopic interventions within the shoulder region.

Morphometric Characteristic between Diploid and Spontaneous Triploid Carp in Korea

  • We used flowcytometry to ploidy verification after that investigate difference between diploid and spontaneous triploid through the truss dimension and classical dimension at crucian carp, Carassius auratus, crucian carp C. cuvieri and common carp, Cyprinus carpio collected from Hangang river, Hantangang river, Imjingang river, Geumgang river, Yeongsangang river and Nakdonggang river, Korea. There were significant differences among the three species for the truss dimensions anterior origin of dorsal fin $(2){\times}$ anterior origin of anal fin (5), $2{\times}$ anterior origin of pelvic fin (6), $2{\times}$ origin of pectoral fin (7), posterior origin of dorsal fin $(3){\times}5$, $3{\times}6$, and $3{\times}7$ (P<0.05). There were no significant differences among the three species in the truss dimensions dorsal fin length ($2{\times}9$) and eye diameter (ED)(P>0.05). On the other hand, there were no significant differences in the several classical dimensions of each species (P>0.05). Three classical dimensions, most anterior extension of the head $(1){\times}2$, $1{\times}6$ and $2{\times}$ most posterior scale in lateral line (4) did not differ between the C. auratus diploid and spontaneous triploid (P>0.05). Two classical dimensions, $1{\times}6$ and longest length between most anterior extension of the head and gill cover ($1{\times}8$) did not differ between the C. cuvieri diploid and spontaneous triploid (P>0.05). One classical dimensions, $1{\times}2$ did not differ between the common carp diploid and spontaneous triploid (P>0.05). There were significant differences in the each diploid and triploid species (P<0.05). These results suggest that the classification of each species and classification between diploid and spontaneous triploid morphometrical parameters used in this study are useful indices of morphometrical status in the each species from major river of Korea.

Effect of Sodium Chloride on Biology of Catenaria anguillulae

  • Growth studies of Catenaria anguillulae isolates in response to sodium chloride indicated that all the isolates grew in linseed oil-cake agar medium containing sodium chloride up to 1.0%. Medium with 1.5% sodium chloride, however, completely checked the growth of all the isolates. The size of zoosporangia greatly increased with abundant zoospore production in medium containing sodium chloride at 0.5%.

  • Complexities of testis structure and function are emphasized in morphometrical and genotoxic evaluation by statistical analysis. F-344 rats were treated with azinphos methyl, cyclophosphomide, and dichlorvos. And Brdu was injected with intrapertionially before sacrifice. The existence and degree of DNA damage were measured by Brdu labeling index which represented relative amount of Brdu incorporated in DNA, morphometric change was evaluated by the relative length of tubular diameter in circular seminiferous tubules and the number of spermatogonia per Sertoli cell in stage IX seminiferous tubules.

Effect of Temperature on Biology of Different Isolates of Catenaria anguillulae

  • Growth of ten isolates of Catenaria anguillulae on linseed oil-cake agar medium was studied at 10, 13, 15, 20, 25, 30, 35, 40, 44 and $46^{\circ}C$. The cardinal temperatures of these isolates were also determined. Observations clearly revealed that the isolates differed in their temperature requirements. Isolate GA was found to grow best at $40^{\circ}C$, whereas VF isolate showed best growth at 35 and $40^{\circ}C$. Isolates PA, KA, CHP, KO, MA and SWP grew best at $35^{\circ}C$. The other isolates(KP and MMT) showed their best growth at $30^{\circ}C$. Based on radial growth, isolates were categorized as fast growing, medium growing and slow growing. Studies on effect of different temperatures on pathogenicity was made using only three isolates: VF, KP and MA against Xiphinema basin. VF isolate caused maximum infection at $40^{\circ}C$, whereas KP and MA isolates caused maximum infection at $30^{\circ}C$. The morphology of sporangia varied with temperature in isolates producing elongate-elliptical or filamentous sporangia. In VF isolate the sporangia were largely iilamentous to elongate elliptical at $30^{\circ}C$. The width of the sporangia increased with increasing and decreasing temperatures. At $44^{\circ}C$ the sporangia of this isolate were mostly broadly elliptical or spherical. This clearly indicates that sporangia vary in morphology with temperature. From the morphometrical studies it was understood that sporangial morphology was more consistent and reliable for grouping of isolates. Based on the morphology of sporangia the isolates of C. anguillulae were characterized in three groups viz., Mamentouslelongate elliptical(VF, KA, GA and SWP), spherical(PA, KP, MA and MMT) and vertically elongate sporangia(CHP and KO).

Morphometrical Changes on Korean Rose Bitterling, Rhodeus uyekii, in Early Growth Period

  • We investigated the process of yolk absorption in Korean rose bitterling, Rhodeus uyekii, and determined the changes in its morphometric characteristics. The R. uyekii from 1 days post hatching (DPH) to 21 DPH, the eye diameter (ED) was 5.4 at 5 DPH. Thereafter, the ED/total length (TL) ratio increased to 10.7 at 21 DPH (p<0.05). The yolk length (YL) decreased from 95.4 to 1.1 by 21 DPH, and this rate of decrease was greater than that for any other dimension (p<0.05). 12 morphometric dimensions/TL for the R. uyekii were measured at each sampling day from 21 DPH to 170 DPH. At just hatching, the average TL and BW were $6.1{\pm}0.09mm$ and $4.9{\pm}0.07mg$, respectively. At 53 DPH, the average TL was $12.9{\pm}0.28mm$ and the average BW was $14.7{\pm}0.72mg$; the total length growth equation was $TL=5.507e^{0.038t}$ ($R^2=0.916$). Further, the body weight growth equation was $BW=3.3647e^{0.0296t}$ ($R^2=0.9354$). The dimensions with regard to body depth showed the greatest growth rates in the external characteristics of the fish (p<0.05). The patterns of the morphometric characteristics measured in this study can be classified in three ways. The patterns were shown to be increased (y=0.0992x+12.07, $R^2=0.8333$), decreased (y=-0.0569x+42.029, $R^2=0.8395$) or maintained (y=0.005x+18.263, $R^2=0.3678$) from 21 DPH to 170. These results will provide useful indices for the successful rearing of the R. uyekii.

산천어, Oncorhynchus masou 2배체와 유도 3배체의 형태학적 차이 (Morphometrical Differences between Diploid and Induced Triploid Cherry Salmon, Oncorhynchus masou)

  • 산란기 직전의 산천어, Oncorhynchus masou 2배체와 유도 3배체를 대상으로 체장과 체중 및 형태측정을 실시하였다. 체중, 내장괴 제거 체중, 체장, dressing 비, 비만도, 생식소 무게와 생식소 지수, 복강두께 및 내장괴 무게와 내장괴 지수가 조사되었으며 체둘레 측정이 등지느러미, 배지느러미 및 등지느러미의 각 종단면에서 이루어졌다. 종단면 면적과 2가지 복강두께 형질이 등지느러미, 배지느러미 및 뒷지느러미의 종단면에서 조사되었으며 아울러 각 종단면에서의 높이, 전체높이, 폭, 3가지 체형질과 9가지 종단절단형질이 조사되었다. 2배체의 체장과 체중은 유도 3배체와 비교시 크게 나타났으며 dressing 비, 생식소 무게, 생식소지수, 내장괴 지수 및 간 지수 역시 2배체가 유도 3배체 보다 크게 나타났다. 반면 유도 3배체는 2 배체에 비해 가슴지느러미 및 등지느러미 종단면에서의 1가지 복강두께 형질과 일부 종단형질에서 높은 치를 나타내었다. 2배체에 비해 유도 3배체의 이러한 형질특성은 유도 3배체 자체의 불임에 기인된 것으로 산란기 직전 유도 3배체의 경제적 가치성을 시사한다고 할 수 있다.

Spatial Heterogeneity and Long-term Changes in Bivalve Anadara broughtoni Population: Influence of River Run-off and Fishery

  • A comparison was made of population of the economically important cockle Anadara (=Scapharca) broughtoni (Bivalvia, Arcidae) inhabiting different areas of the Razdolnaya River estuary at the head of Amurskii Bay (Peter the Great Gulf, East Sea). Also, changes in cockle population density and structure, as well as in cockle growth rates during the last 20 years were studied. In all years of investigation, the morphometrical parameters and growth rates of cockles were smaller at the sites located close to the River mouth than farther down-estuary. The differences can be attributed to higher concentration of suspended particulate matter, decreased salinity and water temperature, as well as a longer exposure to these unfavorable environmental factors at sites located close to the River mouth, compared to farther sites. For two decades, cockle population density had decreased by almost 30 times at some sites in the River estuary. The main reason for this population decline is commercial over-fishing of the cockle. Besides, for the last 20 years, linear parameters of the cockles in the population decreased approximately by 30% and weight parameters, almost two times. Cockle growth rates also decreased for this period. Evidently, these facts are due to the damaging effect of dredging.
