Nano-Micelle of Moringa Oleifera Seed Oil Triggers Mitochondrial Cancer Cell Apoptosis

  • Cancer, a worldwide epidemic disease with diverse origins, involves abnormal cell growth with the potential to invade other parts of the body. Globally, it is the main cause of mortality and morbidity. To overcome the drawbacks of the commercially available chemotherapies, natural products-loaded nano-composites are recommended to improve cancer targetability and decrease the harmful impact on normal cells. This study aimed at exploring the anti-cancer impacts of Moringa oleifera seed oil in its free- (MO) and nano-formulations (MOn) through studying whether it mechanistically promotes mitochondrial apoptosis-mediating cell death. Mitochondrial-based cytotoxicity and flow cytometric-based apoptosis analyses were performed on cancer HepG2, MCF7, HCT 116, and Caco-2 cell lines against normal kidney BHK-21 cell line. The present study resulted that MOn triggered colorectal cancer Caco-2 and HCT 116 cytotoxicity via mitochondrial dysfunction more powerful than its free counterpart (MO). On the other side, MOn and MO remarkably induces HCT 116 mitochondrial apoptosis, while sparing normal BHK-21 cells with minimal cytotoxic effect. The present results concluded that nano-micelle of Moringa oleifera seed oil (MOn) can provide a novel therapeutic approach for colorectal and breast cancers via mitochondrial-mediated apoptosis, while sparing normal and even liver cancer cells a bit healthy or with minimal harmful effect. Intriguingly, MOn induced breast cancer not hepatocellular carcinoma cell death.

Aqueous phase removal of ofloxacin using adsorbents from Moringa oleifera pod husks

  • Chemically activated and carbonized adsorbents were prepared from Moringa oleifera pod husks (MOP), characterized and evaluated for their ability to remove a common antibiotic - ofloxacin (OFX) from aqueous solution. The pulverized precursor was steeped in a saturated ammonium chloride solution for a day to give the chemically activated adsorbent (AMOP). A portion of AMOP was pyrolyzed in a muffle furnace at 623 K for 30 min to furnish its carbonized analogue (CMOP). The adsorbents showed favorable physicochemical attributes. The effects of operational parameters such as initial OFX solution pH and concentration, adsorbent dosage, temperature and contact time on OFX removal were investigated. At equilibrium, optimal removal efficiencies of 90.98% and 99.84% were achieved at solution pH 5 for AMOP and CMOP, respectively. The equilibrium adsorption data fitted into both the Langmuir and Freundlich isotherms. Gibbs free energy change (${\Delta}G^o$), enthalpy change (${\Delta}H^o$) and entropy change (${\Delta}S^o$) indicated that the adsorption of OFX was feasible, spontaneous, exothermic and occurred via the physisorption mode. Adsorption kinetics obeyed the Blanchard pseudo-second-order model. The results may find applications in the adsorptive removal of micro-contaminants of pharmaceutical origin from wastewater.

Measurement of Engineering Properties Necessary to the Design of Drumstick (Moringa oleifera L.) Pod Sheller

  • Purpose: Designing equipment for processing, sorting, and other post-harvest operations of agricultural products requires information about their physical properties. This study was conducted to investigate some of the mechanical and physical properties of Moringa oleifera L. pods and seeds. Methods: Properties such as the length, width, thickness, bulk density, porosity, mass, static coefficient of friction, and angle of repose were determined as a function of moisture content. Statistical data and force-deformation curves obtained at each loading orientation and moisture level were analyzed for bioyield point, bioyield strength, yield force, rupture point, and rupture strength using a testrometric machine. Result: The basic dimensions (length, width, and thickness) of moringa pods and seeds were found to increase linearly from 311.15 to 371.45 mm, 22.79 to 31.22 mm, and 22.24 to 29.88 mm, respectively, in the moisture range of 12 to 49.5% d.b. The coefficient of friction for both pods and seeds increased linearly with an increase in moisture content on all the surfaces used. The highest value was recorded on mild steel, with 0.581 for pods and 0.3533 for seeds, and the lowest on glass for pods, with a value of 0.501, and of 0.2933 for seeds on galvanized steel. The bioyield and rupture forces, bioyield and rupture energies, and deformation of the pods decreased with an increase in moisture content to a minimum value, then increased with further decrease within the moisture content range, while the yield force increased to a maximum value and then decreased as the moisture content increased. Conclusion: These results will help to determine the most suitable conditions for processing, transporting, and storing moringa pods, and to provide relevant data useful in designing handling and processing equipment for the crop.

Screening Moringa species focused on development of locally available sustainable nutritional supplements

  • BACKGROUND/OBJECTIVES: The leaves of Moringa oleifera (MO) and Moringa stenopetala (MS) commonly grown in Ethiopia possess potential nutritional and medicinal value. The aim of this study was to evaluate the nutritional and functional characteristics of the dried leaf powder from two Moringa species to develop sustainable nutritional supplements for Ethiopians from locally grown plant sources. MATERIALS/METHODS: Freshly harvested and air-dried MO and MS leaves were authenticated and the nutritional contents, such as protein, ash, lipids, and selected vitamins and minerals, were analyzed using standard analytical procedures. Amino acid compositions were also determined by an amino acid analyzer. Nine-week-old mice were randomly divided into four groups to investigate the anti-obesity effects of Moringa. The first group was fed a basal diet, the second group a high-fat diet, and the others were fed a high-fat diet containing 0.1% Moringa leaf powder from each species. After seven weeks, serum indices related to lipid profiles from each mouse were analyzed. RESULTS: The present study revealed high protein (28-29%) and ash (7-11%) contents. Glutamic acid, aspartic acid, proline, and leucine were the most abundantly found amino acids in both species. The predominant minerals in the leaf powder were calcium (826-1,530 mg/100 g), potassium (794-904 mg/100 g), and magnesium (286-431 mg/100 g). Pyridoxine (475.06 mg/100 g) and vitamin E (34.2 mg/100 g) were found only in MS. Niacin was found only in MO at 32.21 mg/100 g, whereas ascorbic acid was found in both species (3.89 and 6.19 mg/100 g dry weight for MO and MS, respectively). The results of the animal study showed that mice on a high-fat diet containing 0.1% MO leaf powder alleviated the elevation of cholesterol, triglycerides, and low-density lipoprotein cholesterol induced by the high fat diet. MO was more effective than MS in preventing hypercholesterolemia and fat deposition. CONCLUSION: The findings in this work confirmed that Moringa leaves of both MO and MS possessed high nutritional value but MO was better at preventing the harmful effects of the high-fat diet than MS.

모링가 추출물에 대한 화장품약리활성 검증 (Studies on Cosmeceutical Activity of Extracts of Moringa oleifera Extract)

  • 본 연구에서는 화장품 천연소재로서 모링가 에탄올 추출물의 이용 가능성을 확인하였다. Tyrosinase와 elastase 저해활성을 측정한 결과 각각 $1,000{\mu}g/mL$에서 47%, 39%의 활성을 나타내었다. 모링가 에탄올 추출물에 대한 collagenase 저해활성을 측정한 결과 $1,000{\mu}g/mL$에서 31%의 활성을 확인하였다. 세포 생존율을 MTT 분석법으로 확인한 결과 대식 세포(Raw264.7)와 멜라노마 세포(B16F10)의 농도 구간이 $100{\mu}g/mL$ 일 때 각각 94.2%, 94.8%의 생존율을 보였다. 항염증 활성을 확인하기 위해 griess 분석에 의하여 대식 세포에 lipopolysaccharides (LPS)를 처리하였다. 그 결과 모링가 에탄올 추출물의 농도가 증가함에 따라 NO 발현 억제효과를 확인하였다. Western blot을 통한 단백질 발현 억제 효과를 측정하기 위해 25, 50, $100{\mu}g/mL$ 농도의 모링가 에탄올 추출물과 ${\beta}-actin$을 사용하였다. 그 결과, iNOS, COX-2, MITF, TRP-1, TRP-2, tyrosinase의 단백질 발현양이 $1 00{\mu}g/mL$에서 85.8%, 57.5%, 80.7%, 30%, 29.9%, 23.6%로 억제됨을 확인하였다. 따라서 미백 및 항염증 효과가 우수함을 확인하였고, 모링가 에탄올 추출물의 화장품 소재로서의 가능성을 확인하였다.

모링가 부위별 추출물의 항산화 활성 및 파이토케미컬 함량 (Antioxidant activity and phytochemical contents of the extracts from different parts of Moringa oleifera)

  • 열대 및 아열대 기후에서 자생하는 모링가가 기후변화로 인하여 국내에서도 재배가 되고 있으면서 관심이 증가하고 있다. 다양한 나라에서 식품 및 기능성제품으로 사용이 되면서 국내로 수입되는 모링가에 대한 분석이 필요하여 본 연구를 실시하게 되었다. 모링가의 부위별(잎, 가지 및 종자) 항산화 능력과 페놀 및 플라보노이드 함량은 잎과 가지에서 높았으며, 정의 상관관계를 나타내었다. 무기물 함량 역시 잎과 가지에서 풍부하였으며 특히 칼륨 이온이 가장 많았다. 또한 7종의 페놀산과 4종의 플라보노이드 성분이 검출이 되었으며 페놀산에서는 페룰릭산이, 플라보노이드에서는 쿼세틴 함량이 높게 나타났다. 이 연구결과는 향후 국내 모링가 재배농가의 비교자료로 유용하게 이용될 것으로 기대된다.

모링가 잎 분말을 첨가한 머핀의 품질 특성 (Quality Characteristics of Muffins Added with Moringa (Moringa oleifera Lam.) Leaf Powder)

  • 본 연구에서는 머핀의 품질향상을 위한 기능성 천연소재의 개발을 위해 당뇨병과 고혈압 예방, 항산화 작용 및 해독작용이 있는 것으로 알려진 모링가 잎 분말을 0%, 1%, 3%, 5% 및 7%를 첨가한 머핀의 이화학적 및 관능적 특성과 생리활성을 탐색하였다. 머핀의 중량은 64.69~65.81 g으로 모링가 잎 분말의 첨가량이 증가할수록 증가하는 경향이었고, 높이는 5.58~6.08 cm로 첨가량이 증가할수록 유의적으로 감소하였으며(P<0.05), 굽기 손실률은 첨가량이 증가할수록 감소하는 경향이었다. 머핀의 pH는 첨가량이 증가함에 따라 유의적으로 감소하였으며(P<0.05), 수분함량은 3% 첨가군에서 30.48%로 가장 높았으나 첨가군들 사이에서는 유의적인 차이가 없었다. 머핀의 기계적 특성에서 경도는 무첨가군에서 가장 높게 나타났으나 1% 첨가군과는 유의적인 차이가 없었고, 응집성은 첨가량이 증가함에 따라 감소하였다. 탄력성은 무첨가군이 가장 높게 나타났으나 모든 시료간에 유의적인 차이는 없었고, 씹힘성과 파쇄성은 무첨가군에서 가장 높게 나타나 1~5% 첨가군들과는 유의적인 차이가 있었다(P<0.05). 모링가 잎 분말을 첨가한 머핀의 총페놀 함량은 첨가량이 증가함에 따라 유의적으로 증가하였고(P<0.05), DPPH 라디칼 소거능 또한 첨가량이 증가할수록 유의적으로 증가하였다(P<0.05). 모링가 잎 분말을 첨가한 머핀의 알코올 분해 활성을 측정한 결과 ADH 및 ALDH 활성 모두 첨가량이 증가할수록 유의적으로 증가하였다(P<0.05). 머핀의 관능적 특성을 측정한 결과 외관에 대한 기호도는 7% 첨가군에 가장 높게 나타났고, 내부 색과 맛에 대한 기호도는 무첨가군, 향은 5% 첨가군, 조직감은 1% 첨가군에서 가장 높게 나타났다. 전반적인 기호도는 3% 첨가군에서 5.43으로 가장 높게 나타났고 7% 첨가군에서 4.70으로 가장 낮게 나타났지만, 시료 간에 유의적인 차이는 없었다. 이상의 결과를 종합해보면 모링가 잎 분말을 3% 수준까지 첨가하여 머핀을 제조한다면 관능적 특성에 긍정적인 영향을 줄 뿐만 아니라 생리활성이 향상된 머핀을 제조할 수 있을 것으로 판단되며, 이는 모링가 잎을 이용한 식품개발의 기초자료로 제공될 수 있을 것으로 생각한다.

Estimating Apparent Nutrient Digestibility of Diets Containing Leucaena leucocephala or Moringa oleifera Leaf Meals for Growing Rabbits by Two Methods

  • This study aimed to evaluate the nutrient digestibility of growing rabbits fed diets with different levels of either Leucaena leucocephala (LLM) or Moringa oleifera (MOLM) leaf meals and also to compare total collection and $TiO_2$ marker methods for estimating digestibility. A total of 30 California growing rabbits ($1.81{\pm}0.19kg$ live weight on average) were randomly distributed into five experimental groups of six rabbits each and were housed in individual cages. The groups were control, 30% LLM, 40% LLM, 30% MOLM, and 40% MOLM. All groups received pelleted diets for two weeks; diets also contained 4 g/kg titanium dioxide as dietary marker. Daily feed intake was recorded during the whole experimental period and total feces were collected daily and weighed individually during four days. The results showed that there were no difference (p>0.05) in feed, dry matter (DM), organic matter (OM), crude protein (CP), digestible energy, and crude fiber (CF) intake between the control group and the other experimental groups. The apparent digestibility values of DM, OM, CP, CF, acid detergent fiber, and gross energy were the highest for control group (p = 0.001), meanwhile MOLM diets had generally higher nutrient digestibility coefficients than LLM diets. Increasing the inclusion level of leaf meal in the diet from 30% to 40% improved the digestibility of CF from 45.02% to 51.69% for LLM and from 48.11% to 55.89% for MOLM. Similar results for apparent nutrient digestibility coefficients were obtained when either total collection or indigestible marker method was used. In conclusion, the digestibility of MOLM containing diets were better than LLM diets, furthermore $TiO_2$ as an external marker could be used as a simple, practical and reliable method to estimate nutrients digestibility in rabbit diets.

Processing Effects on the Antioxidant Activities of Beverage Blends Developed from Cyperus esculentus, Hibiscus sabdariffa, and Moringa oleifera Extracts

  • The discovery of bioactive compounds in foods has changed the dietary lifestyle of many people. Cyperus esculentus (tigernut) is highly underutilized in Africa, yet tigernut extract is highly profitable in Europe. This study aims to add value to tigernut extract by revealing its health benefits and food value. In this study, tigernut tubers were germinated or roasted and the extracts were combined with Moringa oleifera extract (MOE) or Hibiscus sabdariffa extract (HSE) and spiced with ginger to produce functional drinks. The drinks were evaluated for physicochemical characteristics, sensory parameters, and antioxidant potentials. The total phenolic content of each beverage was measured by the Folin-Ciocalteu method, and the antioxidant activity of each beverage was determined by the 2,2-diphenyl-1-picrylhydrazyl and 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid assays. The beverages from the germinated tigernut extracts had the highest titratable acidity and the lowest pH, while beverages containing the roasted tigernut extract had the highest $^{\circ}Brix$. Germination and roasting significantly enhanced the total phenolic content of the drinks. The beverage containing HSE and germinated tigernut extract had a total phenolic content of 45.67 mg/100 mL gallic acid equivalents, which was significantly higher than the total phenolic content of all other samples. The DPPH inhibition activity of the beverages prepared with germinated tigernut extracts was significantly higher than the DPPH inhibition activity of the beverages prepared with fresh tigernut extract. The taste and overall acceptability of drinks containing the roasted tigernut extract were preferred, while the color and appearance of drinks with the germinated samples were preferred. Roasting or germinating tigernuts before extraction and addition of MOE or HSE extracts is another way to add value and enhance the utilization of tigernuts.

모링가 지하부의 항혈전 활성 (Anti-thrombosis Activities of the Root Extract of Moringa oleifera Lam)

  • 열매, 잎, 꽃, 봉우리, 꼬투리 등의 지상부를 약용 및 식용으로 사용하고 있는 모링가는 miracle tree라고 불리고 있다. 그러나, 현재까지 주로 잎과 씨에 대한 연구가 진행된 바, 지하부에 대한 성분 및 효능 연구는 초보적인 상태이다. 본 연구에서는 모링가 지하부의 유용 생리활성을 평가하기 위해 지하부의 ethanol 추출물 및 이의 순차적 유기 용매 분획물을 조제하고 이의 혈액응고 저해 및 혈소판 응집저해 활성을 평가하였다. 그 결과, 모링가 지하부 ethanol 추출물은 항응고 활성이 미약하고, 혈소판 응집 촉진효과를 나타내어 지혈작용을 나타내었으나, 추출물의 hexane 및 ethylacetate 분획에서는 우수한 트롬빈, 프로트롬빈, 혈액응고인자 저해와 함께 혈소판 응집저해 활성을 나타내었다. 또한 모링가 지하부 추출물 및 분획물들은 1.0 mg/ml 농도까지 인간 적혈구 용혈활성이 없음을 확인하여, 상기 분획물들이 신규의 항혈전제로 사용 가능함을 제시하였다.
