  • Trichosporon cutaneum lipase의 작용기작(作用機作) 및 위치특이성(位置特異性)을 thin-layer chromatography 및 densitometer를 사용(使用)하여 검토(檢討)하였던 바 다음과 같은 결과(結果)를 얻었다. 1. 본(本) lipase 지질분해속도(脂質分解速度)는 triolein>diolein>monooleie의 순(順)이었다. 2. 본(本) lipase의 diolein분해속도(分解速度)는 triolein의 그것과 비슷한 정도(程度)로 빨랐다. 3. 본(本) lipase의 monoolein분해속도(分解速度)는 triolein 및 diolein의 그것에 비(比)하여 현저(顯著)히 늦었다. 4. 본(本) lipase는 극(極)히 미약(微弱)한 ${\alpha}$-위치특이성(位置特異性)을 나타내었다.

Monoolein액정상이 Retinylpalmitate의 안정성과 경피전달에 미치는 효과에 관한 연구 (Study on the Influence of Cubic Liquid Crystalline Phases of Monoolein on the Stability and Transdermal Delivery of Retinylpalmitate)

  • Retinoids have many important and diverse functions and particularly, have been widely used as anti-aging agent and for the treatment of acne and psoriasis in cosmetics. However, retinoids have low stability against the air, light, water, oxygen and heat, thus, to stabilize the retinoids in formulations is very critical procedure. In this study, cubic liquid crystalline phase of monoolein was applied to stabilize the retinylpalmitate (RP) and to enhance the transdermal permeation. Cubic liquid crystalline phase significantly enhanced the stability of RP. After 15 days, the content of RP in the cubic formulation was 94.7% while the content of RP in ethanol solution was below 0.5% at room temperature. Although BHT containing crystalline phase showed the slightly increased stability of RP, there were no significant differences in RP stability between with or without antioxidants (ascorbic acid, ${\alpha}$-tocopherol, BHT, BHA) at $40^{\circ}C$. The skin retention of RP in crystalline formulations was approximately $5.3{\sim}6.4$ times greater than that of o/w cream formulation. Incorporation of RP into cubic liquid crystalline phase of monoolein effectively stabilized the RP and worked as excellent topical vehicle for RP. Liquid crystalline phase is considered to be suitable formulation for RP for topical delivery system as a stabilizer and permeation enhancing agent.

Monoglyceride의 지방산(脂肪酸) 조성(組成)이 Ice Cream 물성(物性)에 미치는 영향 (Effect of Fatty Acid Compositions by Monoglyceride on Rheological Properties of Ice Cream)

  • Ice cream용 유화제로 사용되는 monoglyceride의 지방산 조성에 따른 ice cream의 물성에 미치는 영향에 대해 검토하였다. 일반적으로 monolaurin부터 monostearin의 포화지방산 monoglyceride에는 각기 성질에 큰 차이가 없었으나 monocaprin및 불포화지방산인 monoolein에서는 ice cream의 점도가 현저히 상승되어 적합한 overrun과 보형성이 양호한 ice cream이 만들어지며 이의 원인은 지방구의 응집 때문으로 확인되었다. 한편 포화지방산 monoglyceride는 overrun이 freezing time의 초기에 발생되기 쉽지만 시간이 경과함에 따라 차츰 저하되는 반면 불포화지방산 monoglyceride는 액상 또는 저융점으로 인해 기름에 대한 융해성이 커짐으로써 overrun이 초기에는 어려우나 freezing time이 경과함에 따라 지방구가 입체적인 망상구조를 이룰뿐 아니라 지방구의 응집을 나타냄으로써 overrun이 개선되며 ice cream에 보형성을 증진시키는 결과를 얻었다. 또한 monostearin과 monoolein의 비율에 있어서는 30:70의 부근에서 지방구의 응집이 극대점을 나타내고 있음을 확인하였다.

Simultaneous Quantification of Oleins (triolein, diolein and monoolein) in Mouse Feces using Liquid Chromatography-Electrospray Ionization/Mass Spectrometry

  • Fat malabsorption is an important cause of poor growth in infancy and childhood. Steatorrhea tests have been developed using various methods. Traditional measurements of stool fat, however, require large samples and it often takes as a week to complete the analysis. In this paper, a liquid chromatography-electrospray ionization/mass spectrometry (LC-ESI/MS) method was developed for simultaneous quantitative analysis of triacylglycerols, triolein, diolein and monoolein, in mouse feces. Moreover, the procedure was rapid, simple as well as compatible with LC-ESI/MS. Chloroform-isopropyl alcohol solution was used for fat-soluble sample extraction. After centrifugation and filtration, an analytical solution was prepared. Triolein, diolein and monoolein were separated using non-aqueous reversed-phase column with the mobile phase consisting of A (methanol) and B (acetone-isopropyl alcohol). The precision (% CV) and accuracy (% bias) of the assay were 3.8-14.7% and 85.2-114.9%, respectively. This method has been successfully applied to simultaneous determination of triolein, diolein and monoolein in feces from 30 mice. This method can therefore be applied to measure triacylglycerols in mouse feces accurately and precisely by LC-ESI/MS, thereby helping to predictive biomarker in fat malabsorption and diagnostic research.

  • Monoglycerides, diglycerides, triglycerides와 free fatty acids를 대상으로 brassinosteroid검정용 생물검정법인 rice inclination test에 의해 생물활성을 검정하였다. Monoglycerides 뿐만 아니라 diglycerides, triglycerides 그리고 free fatty acids의 일부도 이 생물검정법에 양성을 나타냈다. 활성을 나타낸 물질중 ${\alpha}$-monoolein, diolein은 동일 농도의 indole-3-acetic acid 보다 높은 생물활성을 나타내어 olein 관련물질이 특히 높은 생물활성을 갖는 흥미로운 사실을 확인할 수 있었다.

Effect of Cubic Liquid Crystalline Systems on Skin Localization of Oregonin and Hirsutanonol

  • Monoolein-based cubic liquid crystalline systems were formulated for the local delivery of oregonin and hirsutanonol for the treatment of atopic dermatitis. The liquid crystalline phase and its nanodispersion containing drugs were prepared. The skin permeation and deposition properties of the drugs were examined in normal and delipidized rat skin. The proportion of oregonin (%) deposited in normal skin after topical administration of the drugs in the form of aqueous solution, cubic phase or cubic nanodispersions were $1.53\;{\pm}\;0.46$, $3.62\;{\pm}\;0.17$ and $5.13\;{\pm}\;0.73$, and those of hirsutanonol were $2.46\;{\pm}\;0.02$, $5.44\;{\pm}\;0.27$ and $17.28\;{\pm}\;2.19$, respectively. The greater lipophilicity and thus greater skin affinity of hirsutanonol than oregonin contributed the greater amount of skin deposition. The monoolein-based liquid crystalline phases significantly increased the amount of both drugs permeated and deposited. Approximately 3.2, 2.1 and 3.0 times greater amount of oregonin, and 3.4, 2.1 and 2.2 times greater amount of hirsutanonol were deposited in delipidized skin after administration of each drug in the form of aqueous solution, cubic phase and cubic nanodispersions system, respectively, because of lowered barrier function of the delipidized skin. In this study, the effects of drug property, vehicles type and skin condition on skin deposition and permeation properties of drug were examined and concluded that monoolein-based liquid crystalline systems would be a promising formulation for the local delivery of drugs.

Bioproduction and Anticancer Activity of Biosurfactant Produced by the Dematiaceous Fungus Exophiala dermatitidis SK80

  • A new biosurfactant producer was isolated from palm-oil-contaminated soil and later identified through morphology and DNA sequencing as the yeast-like fungus Exophiala dermatitidis. Biosurfactant production was catalyzed by vegetable oil, supplemented with a basal medium. The culture conditions that provided the biosurfactant with the highest surface activity were found to be 5% palm oil with 0.08% $NH_4NO_3$, at a pH of 5.3, with shaking at 200 rpm, and a temperature of $30^{\circ}C$ for a 14-day period of incubation. The biosurfactant was purified, in accordance with surfactant properties, by solvent fractionation using silica gel column chromatography. The chemical structure of the strongest surface-active compound was elucidated through the use of NMR and mass spectroscopy, and noted to be monoolein, which then went on to demonstrate antiproliferative activity against cervical cancer (HeLa) and leukemia (U937) cell lines in a dose-dependent manner. Interestingly, no cytotoxicity was observed with normal cells even when high concentrations were used. Cell and DNA morphological changes, in both cancer cell lines, were observed to be cell shrinkage, membrane blebbling, and DNA fragmentation.

Effects of Stirring and Addition of Chemical Compounds on Glycerolysis of Triglyceride in Reversed Micelles

  • Glycerolysis of triolein by lipase from Chromobacterium viscosum lipase was studied batchwise in AOT-isooctane reversed micelles. The reaction mixture was extracted with chloroform and the content of triolein, 1, 2-diolein, 1, 3-diolein, 1-monoolein, and free fatty acid in the condensed chloroform solution was determined using high performance liquid chromatography (HPLC). The effect of agitation speed on the initial rate of conversion was examined. As the speed of agitation increased up to 700 rpm, the reaction rate increased. However, above 700 rpm, the rate approached maximum and did not increase that much. The glycerolysis activity and the stability of the enzyme were affected by stirring and addition of histidine or copper. Addition of histidine and copper increased the rates of glycerolysis but they are detrimental to the operational stability in reversed micelles.

좁은잎천선과 잎 추출물 유래 항산화 활성 성분의 동정 및 효능 확인 (Isolation and Evaluation of Anti-oxidative Constituents from the Extracts of Ficus erecta var. sieboldii King Leaves)

  • 본 연구에서는 좁은잎천선과 잎 추출물 및 극성별 용매 분획물의 항산화 활성을 검색하고 유효성분을 분리하여 화학구조를 규명하였다. 좁은잎천선과 잎 추출물 및 용매 분획물의 DPPH 및 $ABTS^+$ 라디칼 소거 활성을 측정한 결과 에틸아세테이트 분획물에서 우수한 라디칼 소거 활성을 확인하였다. 활성이 좋은 에틸아세테이트 분획물에서 유효성분을 찾고자 vacuum liquid chromatography (VLC), silica gel column chromatography를 실시하였으며, 분리된 화합물은 1H 및 13C NMR 데이터 분석 및 문헌치 비교를 통하여 총 5개의 화합물을 동정하였다. 분리된 화합물은 monoolein (1), oleic acid (2), lutein (3), afzelechin (4), catechin (5)으로 확인되었으며 이들은 모두 좁은잎천선과에서 처음으로 분리된 화합물이다. 분리된 화합물에 대한 DPPH 및 $ABTS^+$ 라디칼 소거 활성 실험 결과 afzelechin (4) 및 catechin (5)에서 활성이 우수하게 나타났으며 특히 catechin (5)의 경우 대조군인 비타민 C보다 더 좋은 라디칼 소거 활성이 있음을 확인하였다. 또한 HPLC 분석을 통해 좁은잎천선과 잎에서 분리된 catechin의 함량을 확인한 결과 추출물에서 3.8 mg/g, 에틸아세테이트 분획물에서 20.8 mg/g이 함유되어 있는 것으로 확인되었다. 이상의 연구 결과로부터 좁은잎천선과 잎을 이용한 천연 항산화 소재로의 개발이 가능할 것이라 사료된다.
