Detection of a mollicute-like organism in kuruma shrimp, Penaeus japonicus (보리새우(Penaeus japonicus)에 감염된 Mollicute-like Organism)

  • A filamentous form of mollicute-like bacterium was detected during a routine health survey of wild adult kuruma shrimp Penaeus japonicus. The kuruma shrimp were native to Japan and were imported to Korea. The histology showed no pathological changes. Transmission electron microscopy revealed extensive infections of hepatopancreatic epithelial cell by a pleomorphic, filamentous intracellular bacterium. The filamentous bacterium was of about 60 nm in diameter and 300 nm to more than $1{\mu}m$ in length. Teh morphology of bacteria were filamentous and branched with terminal blebs, or knobs, on the branches. They lacked the cell wall, and were bounded by the plasma membrane. They contained typical prokaryotic ribosomes and fibrillar DNA-like strands. No additional internal structure has been observed. They are considered to be mollicutes, based upon the morphological appearance and upon the cellular location.

Oligonucleotide Array-based Detection and Genotyping of Mollicutes (Acholeplasma, Mycoplasma, and Ureaplasma)

  • An oligonucleotide array was developed to detect and genotype mollicutes based on the internal transcribed spacer (ITS) sequence. The results of the assay were compared with those of a PCR-RFLP assay. The proposed oligonucleotide array containing 5 genus- and 23 species-specific probes was able to detect Mycoplasma species, including M. penetrans and M. spermatophilum, that were not detected by the PCR-RFLP assay. Therefore, the results demonstrated that the proposed oligonucleotide array was effective for the detection and discrimination of 23 species, including an acholeplasma, 21 mycoplasmas, and a ureaplasma, and showed promise as a countermeasure to ensure that biological products are safe and of good quality.

Effects of Mixture Lactic Acid Bacteria and Sea Tangle on Anti-obesity and Gut Microflora in Rats (흰 쥐에서 복합 유산균과 다시마가 항비만 및 장내 미생물에 미치는 영향)

  • The present study was undertaken to investigate the effects of dietary provision of lactic acid bacteria (LB) and sea tangle (ST) on the obesity-associated intestinal microbiota in rats with obesity induced by a high-fat diet. Forty-eight 8-wk-old Sprague-Dawley rats were fed a basal diet (CON), a high fat diet (HFD; CON supplemented with 10% lard), HF supplemented with LB [HFL; $5{\times}10^8cfu$ of each of Lactobacillus rhamnosus, Lactobacillus johnsonii, Bifidobacterium longum and Bifidobacterium lactis], or HFL containing 10% ST (HFLS), with 4 replicates (cages) of 3 rats per dietary treatment, for 6 wk, and the intestinal microbiota were determined by pyrosequencing. The HFL and HFLS groups exhibited reduced rates of weight gain than the HF group, and the former groups had smaller ratios of Firmicutes and greater ratios of Bacteriodetes, with decreased Firmicutes/Bacteroidetes ratios, than the latter at the level of the phylum. Compared with the results for the HF group, HFL and HFLS had reduced ratios of the families of Roseburia, Mollicute, Erysipelotrichi, and Oscillibacter within Firmicutes associated with obesity and increased ratios of the families of Prevotella, Alistipes and Bacteroides within the Bacterioidetes phylum known to have an anti-obesity effect. The content of butyric acid in feces was greater in the HFLS group vs. HF and HFL. In conclusion, the present results suggest that dietary provision of LB plus ST has an anti-obesity effect and induced changes in intestinal microorganisms, and enhanced the content of butyric acid, which is an intestinal metabolite.

Investigation of COGs (Clusters of Orthologous Groups of proteins) in 1,309 Species of Prokaryotes (원핵생물 1,309종에 분포된 COGs (Clusters of Orthologous Groups of proteins) 연구)

  • Authors previously reported the results of analyses of COGs (Clusters of Orthologous Groups of proteins) in 711 prokaryotes. The data of COGs were significantly updated for 2020 using 1,309 prokaryotic genomes. Here, we report the results of analyses of 3,455,853 proteins comprising 4,877 updated COGs in terms of COGs and prokaryotes. The numbers of COGs in each prokaryote ranged from 97 to 2,281, with an average of 1,430.0 and a standard deviation of 414.2. Mean numbers of COGs at the phylum level were minimal 497.86 for Mollicutes and maximal 1,642.90 for Cyanobacteria. The top 10 species with the highest COG retention numbers were all Proteobacteria, and 9 out of the bottom 10 were those that could not be cultured in vitro. The numbers of proteins belonging to each COG ranged from 2 to 22,048, with over 12,000 proteins up to the top 11. Five of the top 11 were COGs that bind to DNA and were involved in the gene expression, indicating the importance of regulating gene expression in prokaryotes in a changing environment. COG data are expected to be widely utilized as they can be used for the identification of genes included in the genome and the selection of genes for the strain improvement.
