PCE, TCE로 오염된 지하수내 미생물 특성 및 분포

  • Chlorinated aliphatic hydrocarbons (CAHs) especially perchlorethylene (PCE) and trichlooethylene (TCE) are common groundwater contaminants in Korea. PCE and TCE were often reductively dechiorinated in an aquifer. Several isolates dechlorinate PCE to TCE or cis-1,2 dichloroethylene (c-DCE) were obtained from contaminated and pristine sites in USA and Europe. However in Korea, no information on indigenous microorganism being involved in reductive dechlorination of PCE and TCE is available and different dechlorinating microorganisms might be reside in Korea, since geochemical, and hydrogeological conditions are different, compared to those in the other sites. So we evaluate that: 1) if reductive dechlorinating microorganisms are present in PCE-contaminated site in Korea, 2) if so, what kinds of microorganisms are present; 3) to what extent PCE is reductively dechlorinated. As a results in some PCE-contaminated aquifers in Korea other dechlorinating microorganisms but Dehalococcoides ethenogenes may be responsible for PCE dechlorination. More detailed molecular works are required to evaluate that different dechlorinating microorganisms would reside in Korea.

Distribution of Rhizosphere and Endosphere Fungi on the First-Class Endangered Plant Cypripedium japonicum

  • Endangered native plant habitats and populations are rapidly disappearing because of climate and environmental changes. As a representative, the abundance of the first-class endangered wild plant, Cypripedium japonicum, has been rapidly decreasing in Korea. The purpose of this study was to evaluate the distribution of rhizosphere and endophytic fungi on C. japonicum in its native habitat. A total of 440 rhizosphere and 79 endosphere fungi isolates were isolated and identified on the basis of their molecular characteristics. Sixty-five genera and 119 fungi species were identified in this study. The genus Trichoderma showed the highest abundance among both rhizosphere and endosphere fungi. Mortierella, Hypocrea, and Penicillium spp. were also relatively dominant species on C. japonicum. The community structures of rhizosphere and endosphere fungi were similar, but endosphere fungi showed greater diversity.

Synthesis of Poly(1,6-heptadiyne) Derivatives Containing Bulky Substitutents by Metathesis Polymerization

  • Poly(l,6-heptadiyne) derivatives with phenoxy and 3,7-dimethyloctyloxyphenoxy substituents were synthesized using metathesis polymerization. Polymerizations using MoCl$\sub$5/ as the sole catalyst gave high yields and the resulting polymers were completely soluble in organic solvents. The polymers' structures and thermal properties were characterized using NMR and UV-Vis spectroscopy, as well as with TGA and DSC thermograms. From the analysis of the high-resolution $\^$13/C-NMR spectra, we was found that these polymers contain six-membered rings. The number-average molecular weights and polydispersities of the polymers were ca. 7.0∼20 ${\times}$ 10$^3$ and 3.1∼5.8, respectively. The glass transition temperatures of the polymers were found to be in the range 91∼159$^{\circ}C$.

  • Near infrared (NIR) spectroscopy is a powerful technique for non-destructive analysis that can be obtained in a wide range of environments. Recently, NIR measurements have been utilized as probe for quantitative analysis in agricultural, industrial, and medical sciences. In addition, it is also possible to make practical application on NIR for molecular structural analysis. In this work, Fourier transform near infrared (FT-NIR) measurements were carried out to utilize extensively in the relative amounts of different secondary structures were employed, such as Iysozyme, concanavalin A, silk fibroin and so on. Several broad NIR bands due to the protein absorption were observed between 4000 and $5000\;^{-1}$. In order to obtain more structural information from these featureless bands, second derivative and Fourier-self-deconvolution procedures were performed. Significant band separation was observed near the feature at $4610\;^{-1}$ ,. Particularly the peak intensity at $4525\;^{-1}$ shows a characteristic change with thermal denaturation of fibroin. The structural information can be also obtained by mid-IR and CD spectral. Correlation of NIR spectra with protein structure is discussed.

  • Intramolecular charge transfer (ICT) system has received great attentions due to their promising optoelectronic properties For the efficient ICT of the chromophore, their organic compound mainly consists of strong electron donors (e.g. $NR_2$ or OR groups) and acceptors (e.g. CN or $NO_2$ groups). According to the molecular design and synthesis, the ICT compounds can be extended in many application fields. In this study, we have synthesized ICT compounds having a strong electron acceptor and donor. These novel ICT compounds were easily synthesized by a previously described method with some modifications. Their single crystals were grown and their structures were solved and described in this presentation.

Structural and Optical Properties of Self-assembled InAs Quantum Dots as a Function of Rapid Thermal Annealing Temperature (급속 열처리 온도에 따른 자발 형성된 InAs 양자점의 구조 및 광학 특성)

  • We present the effects of rapid thermal annealing (RTA) temperature on the structural and optical properties of self-assembled InAs quantum dot (QD) structures grown on GaAs substrates by molecular beam epitaxy (MBE). The photoluminescence (PL) measurements are performed in a closed-cycle refrigerator as a function of temperature for the unannealed and annealed samples. RTA at higher temperature results in the increase in island size, the corresponding decrease in the density of islands, and the redshift in the PL emission from the islands. The temperature dependence of the PL peak energy for the InAs QDs is well expressed by the Varshni equation. The thermal quenching activation energies for the samples unannealed and annealed at $600^{\circ}C$ are found to be $25{\pm}5meV$ and $47{\pm}5$ meV, respectively.

  • We have performed Liquid Chromatography-Solid Phase Extraction-NMR (LC-SPE-NMR) analysis for liquid crystalline mixture and elucidated the structures of selected components by NMR spectra. Combining the results of one-dimensional 1H experiments as well as homonuclear and heteronuclear two-dimensional experiments, we could analyze the molecular structure of the liquid crystal singles whose structure had not been interpretable by mass spectrometry alone.

Effect of chain structure of polypropylenes on the melt flow behavior

  • Rheological Properties of polypropylenes having different molecular structures (linear polypropylene (PPL) and branched one (PPB)) were studied. Both the extensional flow and oscillatory shear flow properties were checked. Especially, the melt strength of polypropylenes having various shear history were investigated by using in-house-made Rheometer (called SMER). Compared to linear polypropylene, the branched polypropylene shows enhanced melt strength during extensional flow due to the retarded relaxation of molecules. When the slope of melt tension was plotted against take up speed of melt strand, the characteristic peak was observed in case of branched polypropylene, while the linear polypropylene shows only monotonously decreasing pattern. This entanglement was partially disrupted by physical forces such as shear during melt extrusion. However, the melt strength of PPB after multiple extrusion is still higher than PPL, implying the loss of elasticity during multiple extrusion is not so comprehensive. On dynamic experiments, PPB shows typical shear thinning behavior and the tangent delta of PPB is lower than PPL, reflecting high elasticity of PPB.

Proteomic Studies in Plants

  • Proteomics is a leading technology for the high-throughput analysis of proteins on a genome-wide scale. With the completion of genome sequencing projects and the development of analytical methods for protein characterization, proteomics has become a major field of functional genomics. The initial objective of proteomics was the large-scale identification of all protein species in a cell or tissue. The applications are currently being extended to analyze various functional aspects of proteins such as post-translational modifications, protein-protein interactions, activities and structures. Whereas the proteomics research is quite advanced in animals and yeast as well as Escherichia coli, plant proteomics is only at the initial phase. Major studies of plant proteomics have been reported on subcellular proteomes and protein complexes (e.g. proteins in the plasma membranes, chloroplasts, mitochondria and nuclei). Here several plant proteomics studies will be presented, followed by a recent work using multidimensional protein identification technology (MudPIT).
