한의학계(韓醫學界)의 암연구동향(癌硏究動向)과 연구(硏究) 전략(戰略)에 대한 연구(硏究) (Study on trends of cancer study in TKM and its research strategy in future)

  • For the purpose of designing more successful cancer research, the strategy for cancer study in the field of Traditional Korean Medicine(TKM) during the 21th century was examined from the analysis of trends on cancer study in traditional korean medicine. The results were summarized as follows: 1. So far cancer research in TKM was chiefly done on cytotoxicity, side-effects by chemotherapy, tumor immunology, apoptosis, survival time with S-180 and pulmonary colonization assay and also clonogenic assay, cell adhesion assay, angiogenesis, cell-differentiation and side-effect by radiotherapy were partly performed. 2. It may be ideal that we should study synergistic effect between constituent drugs of prescriptions, tumor immunology, combined therapy between western and oriental medicines by reducing side-effect by radiotherapy and chemotherapy and antimetastasis according to the characteristics of oriental medicine chiefly and also supplement the studies on molecular biology, gene therapy, angiogensis and signal transduction. 3. We had better do specific-field research in cooperation between oriental medical colleges and Korea Institute of Oriental Medicine(KIOM) as well as study a target cancers such as hepatic cancer, pulmonary cancer and gastric cancer more intensively than all cancers domestically. 4. Our country must keep communication with China having many clinical data, Taiwan chiefly doing the combined tharapy between oriental and western medicines, Japan having done basic study actively on cancer.

Characterization of Toxicological Properties of L-Lysine Polymers in CD-1 Mice

  • We recently showed that polylysine, the polymer of lysines, retains anti-prion activity. Although the effectiveness of prion inhibition by polylysine was demonstrated with the regimen tolerated in mice, a determination of quantitative polylysine toxicity is necessary to precisely address the in vivo toxicity level of polylysine. In this communication, we report the results of body weight monitoring and hematologic tests performed in CD-1 mice that received two different tolerable dosages of polylysine for an either 5-day or 4-week period. We found that there was no significant alteration in overall serum chemistry, blood cell count, and body weight gain compared with controls. The only notable quantitative change with statistical significance was the decrease of platelet numbers in mice subchronically administered with polylysine. Our results suggest that polylysine is harmless in mice if administered for a short period, but administrations of polylysine in mice may require considerate attention for safety in future investigations as mice chronically receive tolerable doses of polylysine.

Analysis of Vasopressin-Induced $Ca^{2+}$ Increase in Rat Hepatocytes

  • To analyze vasopressin-induced $Ca^{2+}$ increase in liver cells, rat hepatocytes were isolated and attached to collagen-coated cover slips. Using fura-2, a $Ca^{2+}$-sensing dye, changes in intracellular $Ca^{2+}$ concentration by vasopressin were monitored. Results in this communication suggested that vasopressin-induced $Ca^{2+}$ increase were composed of both $Ca^{2+}$ release from internal $Ca^{2+}$ stores and influx from the plasma membrane. The $Ca^{2+}$ influx consisted of two distinguishable components. One was dependent on the presence of vasopressin and the other was not. SK&F96365 blocked vasopressin-induced $Ca^{2+}$ influx in a dose-dependent manner. Vasopressin-induced $Ca^{2+}$ release from internal stores diminished in a primary culture of hepatocytes according to the culture time. However, changes in vasopressin-induced $Ca^{2+}$ influx across the plasma membrane differed from changes in the $Ca^{2+}$ release from internal stores, suggesting two separate signalings from receptor activation to internal stores and to the plasma membrane.

프로모터 염기서열 분석을 위한 데이터 마이닝 기법 (Data Mining Techniques for Analyzing Promoter Sequences)

  • 최근 지놈(Genome) 프로젝트를 통해 DNA 염기서열에 대한 정보가 밝혀짐에 따라 분자 수준의 유전자 정보를 다루는 기법이 활발히 연구되고 있다. 그리고 밝혀진 서열정보들의 방대함으로 미루어볼 때 이들 정보를 데이터베이스화하고 효과적인 분석을 행하기 위한 새로운 컴퓨터의 알고리즘의 개발 또한 시급한 일이다. 이러한 측면에서 본 논문에서는 분자생물학에서 매우 중요한 연구 대상으로 삼고있는 프로모터 서열과 유전자간의 연관성으로 발현되는 특징을 알아내기 위한 연관 규칙 탐사 알고리즘을 연구한다. 기존의 탐사 알고리즘은 트랜잭션 데이터를 대상으로 하지만 본 논문에서는 생물학적 데이터를 대상으로 하였기때문에 데이터의형태와 생물학적인 특성을 수용하는 변형된 연관규칙 알고리즘을 설계한다. 본 연구를 통하여 얻어진 결과는 실제 생물학적 실험대상의 후보조합을 최소화 하므로써 많은 시간과 노력 비용을 절감할 수 있다.

둔상성 외상 후 양측 간내 담관에서 담즙 누출의 치료 사례 1례 (Management of Bile Leaks from Bilateral Intrahepatic Ducts after Blunt Trauma)

  • Bile leaks are complications that are much more frequent after a high-grade liver injury than after a low-grade liver injury. In this report, we describe the management of bile leaks that were encountered after angiographic embolization in a 27-year-old man with a high-grade blunt liver injury. He had undergone an abdominal irrigation and drainage with a laparotomy on post-injury day (PID) 16 due to bile peritonitis and continuous bile leaks from percutaneous abdominal drainage. He required three percutaneous drainage procedures for a biloma and liver abscesses in hepatic segments 4, 5 and 8, as well as endoscopic retrograde cholangiopancreatography with biliary stent placement into the intrahepatic biloma via the common bile duct. We detected communication between the biloma and the bilateral intrahepatic duct by using a tubogram. Follow-up abdominal computed tomography on PID 47 showed partial thrombosis of the inferior vena cava at the suprahepatic level, and the patient received anticoagulation therapy with low molecular weight heparin and rivaroxaban. As symptomatic improvement was achieved by using conservative management, the percutaneous drains were removed and the patient was discharged on PID 82.

SHORT-ROOT Controls Cell Elongation in the Etiolated Arabidopsis Hypocotyl

  • Transcriptional regulation, a core component of gene regulatory networks, plays a key role in controlling individual organism's growth and development. To understand how plants modulate cellular processes for growth and development, the identification and characterization of gene regulatory networks are of importance. The SHORT-ROOT (SHR) transcription factor is known for its role in cell divisions in Arabidopsis (Arabidopsis thaliana). However, whether SHR is involved in hypocotyl cell elongation remains unknown. Here, we reveal that SHR controls hypocotyl cell elongation via the transcriptional regulation of XTH18, XTH22, and XTH24, which encode cell wall remodeling enzymes called xyloglucan endotransglucosylase/hydrolases (XTHs). Interestingly, SHR activates transcription of the XTH genes, independently of its partner SCARECROW (SCR), which is different from the known mode of action. In addition, overexpression of the XTH genes can promote cell elongation in the etiolated hypocotyl. Moreover, confinement of SHR protein in the stele still induces cell elongation, despite the aberrant organization in the hypocotyl ground tissue. Therefore, it is likely that SHR-mediated growth is uncoupled from SHR-mediated radial patterning in the etiolated hypocotyl. Our findings also suggest that intertissue communication between stele and endodermis plays a role in coordinating hypocotyl cell elongation of the Arabidopsis seedling. Taken together, our study identifies SHR as a new crucial regulator that is necessary for cell elongation in the etiolated hypocotyl.

Recent advances in spatially resolved transcriptomics: challenges and opportunities

  • Single-cell RNA sequencing (scRNA-seq) has greatly advanced our understanding of cellular heterogeneity by profiling individual cell transcriptomes. However, cell dissociation from the tissue structure causes a loss of spatial information, which hinders the identification of intercellular communication networks and global transcriptional patterns present in the tissue architecture. To overcome this limitation, novel transcriptomic platforms that preserve spatial information have been actively developed. Significant achievements in imaging technologies have enabled in situ targeted transcriptomic profiling in single cells at single-molecule resolution. In addition, technologies based on mRNA capture followed by sequencing have made possible profiling of the genome-wide transcriptome at the 55-100 ㎛ resolution. Unfortunately, neither imaging-based technology nor capture-based method elucidates a complete picture of the spatial transcriptome in a tissue. Therefore, addressing specific biological questions requires balancing experimental throughput and spatial resolution, mandating the efforts to develop computational algorithms that are pivotal to circumvent technology-specific limitations. In this review, we focus on the current state-of-the-art spatially resolved transcriptomic technologies, describe their applications in a variety of biological domains, and explore recent discoveries demonstrating their enormous potential in biomedical research. We further highlight novel integrative computational methodologies with other data modalities that provide a framework to derive biological insight into heterogeneous and complex tissue organization.

Potential application of biomimetic exosomes in cardiovascular disease: focused on ischemic heart disease

  • Cardiovascular disease, especially ischemic heart disease, is a major cause of mortality worldwide. Cardiac repair is one of the most promising strategies to address advanced cardiovascular diseases. Despite moderate improvement in heart function via stem cell therapy, there is no evidence of significant improvement in mortality and morbidity beyond standard therapy. The most salutary effect of stem cell therapy are attributed to the paracrine effects and the stem cell-derived exosomes are known as a major contributor. Hence, exosomes are emerging as a promising therapeutic agent and potent biomarkers of cardiovascular disease. Furthermore, they play a role as cellular cargo and facilitate intercellular communication. However, the clinical use of exosomes is hindered by the absence of a standard operating procedures for exosome isolation and characterization, problems related to yield, and heterogeneity. In addition, the successful clinical application of exosomes requires strategies to optimize cargo, improve targeted delivery, and reduce the elimination of exosomes. In this review, we discuss the basic concept of exosomes and stem cell-derived exosomes in cardiovascular disease, and introduce current efforts to overcome the limitations and maximize the benefit of exosomes including engineered biomimetic exosomes.

The Role of Extracellular Vesicles in Senescence

  • Cells can communicate in a variety of ways, such as by contacting each other or by secreting certain factors. Recently, extracellular vesicles (EVs) have been proposed to be mediators of cell communication. EVs are small vesicles with a lipid bilayer membrane that are secreted by cells and contain DNA, RNAs, lipids, and proteins. These EVs are secreted from various cell types and can migrate and be internalized by recipient cells that are the same or different than those that secrete them. EVs harboring various components are involved in regulating gene expression in recipient cells. These EVs may also play important roles in the senescence of cells and the accumulation of senescent cells in the body. Studies on the function of EVs in senescent cells and the mechanisms through which nonsenescent and senescent cells communicate through EVs are being actively conducted. Here, we summarize studies suggesting that EVs secreted from senescent cells can promote the senescence of other cells and that EVs secreted from nonsenescent cells can rejuvenate senescent cells. In addition, we discuss the functional components (proteins, RNAs, and other molecules) enclosed in EVs that enter recipient cells.

전리방사선이 세포질 소기관의 미세구조변화와 기전에 미치는 영향 (The Effect of Ionizing Radiation on the Ultrastructural Changes and Mechanism on the Cytoplasmic Organelles)

  • 전리방사선은 물질과 상호작용하여 원자내의 중성자나 양성자, 궤도전자를 충분히 제거시킬 수 있는 에너지이다. 이와 같은 전리방사선은 DNA이나 세포질 소기관과 세포질의 방사선 분해 생성물을 통해 직접적, 간접적으로 상호작용하여 분자구조를 변화시키는 산화적 대사를 일으킨다. 이러한 전리 현상은 분자수준에서 조직을 손상시키고, 세포 기능을 파괴할 수 있다. 따라서, 전리방사선에 의해 유도된 이온채널과 수송 변형이 보고되고 있다. 이러한 현상이 항상성을 유지하기 위한 생화학적 과정을 무너뜨리면, 유도된 생물학적 변화가 지속되고 후대로 영향을 미친다. 또한, 전리방사선의 영향으로 형성된 활성산소는 세포연접을 통해 인접세포로 퍼져 나갈 수 있다. 방사선량, 선량률, 선질에 따라 이러한 메커니즘이 충분히 방어할 수도 있고 그렇지 않을 수도 있다. 본 총설에서는 방사선생물학의 개념을 뒷받침하는 전리방사선의 세포수준에서 생물학적 효과에 대한 보고서를 간략히 알아보았다. 전리방사선의 생물학적 효과를 잘 이해하면 방사선을 잘 이용할 수 있고, 방사선피폭으로부터 더 나은 방호를 할 수 있을 것이다.
