The present research program was conducted to characterize a strain of actinomycetes producing an anti methicillin-resistant Staphylococcus aureus (MRSA) antibiotic. Soil samples were collected from various sites in Korea and a number of actinomycetes were isolated from the soil samples by applying selective agar for actinomycetes. Among over 400 isolates, a strain (AMLK-135) producing anti-MRSA antibiotic against S. aureus TK 784 was selected. According to the morphological and physiological characteristics, the strain AMLK-135 was confirmed to belong to the genus Streptomyces. From the results of species identification with the TAXON program, the strain AMLK-135 was shown to belong to major cluster 5 (Streptomyces exfoliatus), but it had a low simple matching coefficient ($S_{SM}$ SM/) value to member organisms of major cluster 5. Percentage ($\%$) of strain further away of the strain AMLK-135 was low (1.9400) and it was placed further away than the outer-most members in major cluster 5. Therefore, the strain AMLK-135 was identified as a new species of the genus Streptomyces.
The stable structures of pure ethylene and mixed ethylene-ammonia cluster ions are studied using an electron impact ionization time-of-flight mass spectrometer. Investigations on the relative cluster ion distributions of $(C_2H_4)_n(NH_3)_m^+$ under various experimental conditions suggest that $(C_2H_4)_2(NH_3)_3^+$ and $(C_2H_4)_3(NH_3)_2^+$ ions have the enhanced structural stabilities, which give insight into the feasible structure of solvated ions. For the stable configurations of these ionic species, we report an experimental evidence that both $(C_2H4)_2^+(C_2H_4)_3^+$ clusters as the central cations provide three and two hydrogen-bonding sites, respectively, for the surrounding $NH_3$ molecules. This interpretation is based on the structural stability for ethylene clusters and the intracluster ion-molecular rearrangement of the complex ion under the presence of ammonia solvent molecules.
The molecular cluster model for the homogeneous bubble nucleation was extended to predict the bubble nucleation events in elastomers, polymers and polymer solutions. For the bubble nucleation in elastomers and polymers, the strain energy overcome by a critical bubble was also considered. The calculation results for the number of bubbles nucleated are in good agreement with observed ones.
IscA was proposed to be involved in the iron-sulfur cluster assembly encoded by the iscSUA operon, but the role of IscA in the iron-sulfur cluster assembly still remains controversial. In our previous study, the IscA from A. ferrooxidans was successfully expressed in Escherichia coli, and purified to be a [$Fe_4S_4$] -cluster-containing protein. Cys35, Cys99, and Cys101 were important residues in ligating with the [$Fe_4S_4$] cluster. In this study, Asp97 was found to be another ligand for the iron-sulfur cluster binding according to site-directed mutagenesis results. Molecular modeling for the IscA also showed that Asp97 was a strong ligand with the [$Fe_4S_4$] cluster, which was in good agreement with the experimental results. Thus, the [$Fe_4S_4$] cluster in IscA from A. ferrooxidans was ligated by three cysteine residues and one aspartic acid.
The Arches cluster is a young (2-4 Myr), compact (~1 pc), and massive (${\sim}2{\times}10^4M_{\odot}$) star cluster located ~30 pc away from the Galactic center (GC) in projection. Being exposed to the extreme environment of the GC such as elevated temperature and turbulent velocities in the molecular clouds, strong magnetic fields, and larger tidal forces, the Arches cluster is an excellent target for understanding the effects of star-forming environment on the initial mass function (IMF) of the star cluster. However, resolving stars fainter than ~1 $M_{\odot}$ in the Arches cluster partially will have to wait until an extremely large telescope with adaptive optics in the infrared is available. Here we devise a new method to estimate the shape of the low-end mass function where the individual stars are not resolved, and apply it to the Arches cluster. This method involves histograms of pixel intensities in the observed images. We find that the initial mass function of the Arches cluster should not be too different from that for the Galactic disk such as the Kroupa IMF.
Faba bean (Vicia faba L.) is one of the most important legume crops in Egypt. However, production of faba bean is affected by several diseases including fungal diseases. Fusarium wilt incited by Fusarium oxysporum Schlecht. was shown to be the most common wilt disease of faba bean in Assiut Governorate. Evaluation of 16 faba bean genotypes for the resistance to Fusarium wilt was carried out under greenhouse conditions. Three molecular marker systems (inter-simple sequence repeat [ISSR], sequence related amplified polymorphism [SRAP], and simple sequence repeat [SSR]) and a biochemical marker (protein profiles) were used to study the genetic diversity and detect molecular and biochemical markers associated with Fusarium wilt resistance in the tested genotypes. The results showed that certain genotypes of faba bean were resistant to Fusarium wilt, while most of the genotypes were highly susceptible. The percentage of disease severity ranged from 32.83% in Assiut-215 to 64.17% in Misr-3. The genotypes Assiut-215, Roomy-3, Marut-2, and Giza2 were the most resistant, and the genotypes Misr-3, Misr-1, Assiut-143, Giza-40, and Roomy-80 performed as highly susceptible. The genotypes Assiut-215 and Roomy-3 were considered as promising sources of the resistance to Fusarium wilt. SRAP markers showed higher polymorphism (82.53%) compared with SSR (76.85%), ISSR markers (62.24%), and protein profile (31.82%). Specific molecular and biochemical markers associated with Fusarium wilt resistance were identified. The dendrogram based on combined data of molecular and biochemical markers grouped the 16 faba bean genotypes into three clusters. Cluster I included resistant genotypes, cluster II comprised all moderate genotypes and cluster III contained highly susceptible genotypes.
Infrared multiphoton dissociation (IRMPD) spectra of various protonated 1,2-diaminoethane-water clusters DAE-$H^+-(H_2O)_n$ (n = 1-6) were measured in the wavelength range of 3000-3800 $cm^{-1}$. The IRMPD spectra of the well separated ionic clusters were simulated by the MP2 method employing various basis sets. Comparison of the IRMPD spectra with the theory indicates that each cluster may exist as several low-lying conformers, and the sum spectra of the various conformers reveal almost one to one correspondence between theory and experiment. Free N-H and O-H stretches are observed in the ranges of 3400-3500 and 3600-3800 $cm^{-1}$, respectively. The $O-H{\cdots}N$ and $N-H{\cdots}O$ stretches are, however, observed in the broad region of 3000-3600 $cm^{-1}$. The theoretical calculations on DAE-$H^+-(H_2O)_n$ (n = 1-4) show gradual decrease of the average binding energy between DAE-$H^+$ and $H_2O$ as the cluster size increases, attaining the lowest value of 55 kJ/mol when n = 4. We found a low energy barrier of 21 kJ/mol to the isomerization converting the lowest energy cluster of DAE-$H^+-(H_2O)_n$ to the second lowest one.
A semi-empirical fluctuation term is presented to improve a classical equation of state (EOS) for volumetric properties in the critical region. The term is based on the two assumptions: (1) The Helmholtz energy is individually divided into classical and long-range density fluctuation contribution (2) All molecules form cluster near the critical region due to long-range density fluctuation. To formulate such molecular cluster, we extended the Veytsman statistics originally developed for the cluster due to hydrogen bonding. The probability function in the statistics is modified to represent the characteristics of long-range density fluctuation vanishing far from critical region. The proposed fluctuation contribution was incorporated into the Sanchez-Lacombe EOS and the combined model with 6 adjustable parameters has been tested against experimental VLE data. The combined model is found to well represent flatten critical isotherm for methane and top of the coexistence curve for the tested components. The prediction results for caloric data are in good agreement with the experimental data.
We investigate the molecular and atomic gas properties of 20 Virgo cluster spiral galaxies by comparing with optical properties to assess the effect of the Virgo environment on the interstellar media of the Virgo disks. CO maps from FCRAO On-The-Fly (OTF) mapping survey and HI maps from VIVA (VLA Imaging of Virgo spirals in Atomic gas) are shown, and radial properties of molecular and atomic gas are compared. H2 deficiency along with HI is investigated, and gas evolution history of the Virgo cluster spirals is also examined.
By including the overlap integrals between atomic orbitals, the modified cluster orbitals for a metal cluster of face centered cubic lattice are found. The modified analytic solutions of the cluster are obtained from them with the assumption that the cluster orbitals with different state indices do not mix together. The physical properties-the HOMO levels and the unit electronic energies-of Ni, Pd, and Pt clusters of various size, calculated by the modified cluster orbital method, agree better with the results obtained by the Extended Huckel calculation than those of the previous(unmodified) cluster orbital method do. As a result, it is verified that the physical properties, at least those related to the energy levels, obtained by the Extended Huckel method may be reproduced by use of the modified cluster orbital method instead.
