Production and Characteristics of Lytic Enzyme against Streptococcus mutans Cell Wall from Alkalophilic Bacillus sp. 4830 (호알카리성 Bacillus sp. 4830이 생산하는 Streptococcus mutans 세포벽 분해효소의 분리와 특성)

  • To elucidate a method of preventing dental caries, strains producing lytic enzymes were isolated and their characteristics were investigated. Among 5,00 alkalophilic strains isolated from soil, 22 strains showed lytic activity against Streptococcus mutans. Strain No. 4830, with the highest lytic activity, was selected for further study. Strain 4830 showed 94% sequence homology with the 16S rDNA sequence of Bacillus alcalophilus, but it was concluded to be different from Bacillus alcalophilus because of its biochemical characteristics. The strain was named Bacillus sp. 4830. The lytic enzyme from Bacillus sp. 4830 was purified by ethanol precipitation and CM-agarose column chromatography. The molecular weight of the lytic enzyme was determined to be 28 kDa by SDS-PAGE. The lytic enzyme was stable between pH 5.0 and pH 11 and up to $40^{\circ}C$. The optimal pH and temperature for the lytic activity was 9.0 and $50^{\circ}C$, respectively.

A Study on Pyrolytic and Anatomical Characteristics of Korean Softwood and Hardwood (국산 침·활엽수재의 열분해 및 해부학적 특성에 관한 연구)

  • To investigate the pyrolytic and anatomical characteristics of Korean softwood, Pinus densi-flora, Pinus rigida and Larix leptolepis, and hardwood, Acer palntatum, Fraxinus rhynchophylla and Quercus variabilis, chemical components analysis, TG-DTA (Thermogravimetric Analysis & Differential Thermal Analysis), MBA (Methylene Blue Adsorption) test and SEM observation were carried out. For TG-DTA, samples were carbonized up to $800^{\circ}C$ at the heating rate of $10^{\circ}C$/min under $N_2$ flows 1 l/min using thermogravimetric analyzer. Chemical component analysis of all samples resulted in typical contents of major wood component. In TG-DTA results, softwood showed higher char yield than hardwood, and lignin displayed the highest char yield among the major wood components. All samples showed typical TGA, DTG and DTA curves for wood pyrolysis except a few differences between softwood and hardwood. Content of lignin influenced its pyrolysis characteristics, while molecular structure of lignin affected not only the weight loss but also the yield of char. In MBA test results, MBA of softwoods was higher than that of hardwoods. Char of Pinus densiflora showed the highest MBA, but its degree was lower than activated carbon or fine charcoal about 23 and 4 times, respectively. SEM observation showed carbonization process preserves wood structure and retain the micro-structure of wood fibers.

  • It is well established that dissolution is freruently the rate limiting step in the gastrointestinal absorpton of a drug from a solid dosage from. The relationship between the dissolution rate and absorption is particularly distinct when considering drugs of low solubility. Consequently, numerous attempts have been made to modify the dissolution characteristics of poorly water soluble drugs. Since dissolution rate is directly proportional to surface area, one may increase the rate by decreasing the particle size of the drug. Levy has considered a number of methods by which a drug may be presented to the GI fludids in finely divided from. The direct method is the utilization of microcrystalline or micronized particles. A second method involves the administration of solutions from which, upon dilution with gastric fluids, the dissolved drug will precipitate in the form of very fine particles. A more unique way of obtaining microcrystalline dispersions of a drug has been ercently suggested by Sekiguchi et al. They have first proposed the formation of a eutectic mixture of a poorly water soruble drug with a physiologically inert, easily soluble carrier. When such systems are exposed to water or GI fluids, the soluble carrier will dissolve rapidly and the finely dispersed drug particles will then be released. It has been suggested by Shefter and Higuchi that the formation of crystalline solvate could be a powerful tool in affecting rapid disslution of highly insoluble substances. Goldberg et al. have noted that the formation of solid solution could reduce the particle size to a minimum and increase the dissolution rate as well as the solubility of the durgs. It has also been shown that the rates of solution of drugs were appreciably increased by coprectipitating the drug with soluble polymers. The increase was found to be sensitive to the method of preparation, the molecular weight of polymer and the particular ratio of drugs to polymer. Although several investigations have demontrated that the solubility and/or dissolution rates of drugs can be increased in this manner, little information is available in the literature related to the in vivo absorption pattern of drugs orally administered as PVP coprecipitates. Recently, however, it was demonstrated that both the rate and extent of absorption of the insoluble drug could be markedly enhanced when orally administered to rats in the form of a coprecipitate with PVP. The purpose of the present investigation was to ascertain the general appility of soluble polymer coprectation technique as a method for enhancing the in vitro dissolution rate of hydrophobic indomethacin. To accomplish this aim, the dissolution characteristics of pure indomethacin, indomethcin-polymer physical mixtures and indomethacin-polymer coprecipitates were quantitatively studied by comparing their relative dissolution rates. The solubility and dissolution behavior of these systems were also examined.

Electrochemical Characteristics of Lithium Battery Anode Materials Using Petroleum Pitches (석유계 피치를 사용한 리튬전지 음극소재의 전기화학적 특성)

  • In this study, the molecular weight controlled pitches derived from pyrolyzed fuel oil (PFO) were prepared using solvent extraction and were carbonized. Electrochemical characteristics of lithium battery anode materials were investigated using these petroleum pitches. Three pitch samples prepared by the thermal reaction were 3903 (at $390^{\circ}C$ for 3 h), 4001 (at $400^{\circ}C$ for 1 h) and 4002 (at $400^{\circ}C$ for 2 h). The prepared hexane insoluble pitches were analysed by XRD, TGA, SEM and Gel permeation Chromatography (GPC). The electrochemical characteristics of the PFO-derived pitch as an anode material were investigated by constant current charge/discharge, cyclic voltammetry and electrochemical impedance tests. The coin cell using pitch (4001) and the electrolyte of $LiPF_6$ in organic solvents (EC : DMC = 1 : 1 vol%, VC 3 wt%) has better initial capacity (310 mAh/g) than that of other pitch coin cells. Also, this carbon anode showd a high initial efficiency of 82%, retention rate capability at 2 C/0.1 C of 90% and cycle retention of 85%. It was found that modified pitches improved the cycling and rate capacity performance.

An Ultrafiltration Study for the Recycling of Synthetic Water-Based Cutting Oil (수용성 합성 절삭유의 재사용을 위한 한외여과 연구)

  • In the present study the membrane filtration characteristics of a commercially available synthetic water-based cutting oil through two kinds of ultrafiltration membranes (HF1-45-CM50 and HF1-43-CM100) with molecular weight cut-offs of 50,000 and 100,000, respectively, have been investigated in detail. Among these membranes, the hydrophilic one (HF1-45-CM50) was found to show a satisfactory result for both the permeate flux and the permeability of oil components, whereas the permeate flux obtained with the hydrophobic membrane (HF1-43-CM100) appears to be significantly low, indicating that synthetic cutting oil was easily wetted on the hydrophobic membrane surface and induced more membrane fouling. The effect of material characteristics of the membrane on the filtration characteristics was found to be much more significant compared with the mean pore size of the membrane. Backflushing by nitrogen gas was applied to reduce the formation of a gel layer and membrane fouling. With the hydrophilic membrane, the backflushing was found to increase the permeate flux, whereas the backflushing resulted in a decrease in flux for the hydrophobic membrane. The flux recovery was observed to be highest when the membranes fouled with waste synthetic cutting oil were immersed into a cleaning solution for more than 72 hours and then backflushed by nitrogen gas.

Morphological Characteristics and Habitat Types of Rattus norvegicus and R. tanezumi Collected in Jeju Island (제주도에서 채집된 Rattus norvegicus와 R. tanezumi의 서식지 유형과 형태적 특성)

  • This study was investigated the morphological characters, secondary sexual dimorphism, and habitat patterns of Korean Rattus animals collected on Jeju Island from April 2005 to October 2012. Two wild rat species, namely R. norvegicus and R. tanezumi were identified on Jeju Island based on morphological characteristics and molecular data; however, R. rattus, which had recorded formerly, was not found in this study. Individuals of R. norvegicus were captured from urban, rural, and natural habitats, while those of R. tanezumi were specially found in animal farms and the surrounding areas. Comparing of morphological characters of two species, R. norvegicus had a shorter tail and ears than R. tanezumi (p<0.05), and the ratios of tail length and ear length to those of head-body length showed significantly differences between two species (p<0.05). The body weights (BW) of urban populations of R. norvegicus were significantly heavier than those of rural populations (p<0.05). No secondary sexual dimorphism was found in R. norvegicus, but females of R. tanezumi showed heavier BW than those of males (p<0.05). These findings suggest that it is necessary to revise the records for the existence of R. tanezumi and to confirm the animal fauna and elucidate the distribution and ecological characteristics from further studies using extensive sampling and detailed investigations on Jeju Island and also on the Korean Peninsula.

  • The characteristics of endogenous and exogenous ${\beta}-galactosidase\;({\beta}-Galase)$ produced from L. acidophilus were investigated as one of the serial studies on the fermentation of Chinese cabbage kimchi using L. acidophilus. Apparent molecular weight of endogenous and exogenous of the ${\beta}-Galase$ were investigated to be 550,000 and 740,000 daltons by the method of gel filtration and Km values of the both enzymes were 1.67mg/ml, 1.33mg/ml and $V_{max}$ were $8.5\;{\mu}\;mol/mg/30min.$, $2.65\;{\mu}\;mol/ml/30min.$, respectively. The optimum pH of the enzymes were 7 and 8, respectively. The optimum temperatures and salt concentrations of the both enzyme were the same and appeared to $30^{\circ}C$and$4{\sim}5%$, respectively. The activities of the endogenous and exogenous ${\beta}-Galase$ were decreased by increasing of temperature from $60^{\circ}C$to$90^{\circ}C$ and the decreasing rate of the enzyme activities on the processing of the heating times showed high at first 2 minutes of heating.

Predictive Thermodynamic Model for Gas Permeability of Gas Separation Membrane (기체 분리막의 투과 특성 예측 모델식 개발)

  • It is of special interest in our membrane separation technology due to its low energy consumption and cost, relatively simple equipment, low investment and operation cost, et al. Full scale utilization of such processes can be widely utilized to the various fields. Using the difference of permeability of gas molecules between the filter layers, it is able to separate effectually pure gases from the mixed gases. In this paper, the membranes of PDMS, ${\gamma}-radiated$ PDMS, PTFE, PTFE-X are chosen to develop the predictive model for the separation of pure gases such as oxygen, nitrogen, hydrogen, and other gases from mixed gases. By utilizing the thermodynamic gas properties($\sigma$, $\varepsilon/k$) and experimental data of gas transport characteristics for different polymer membranes, it is able to develop the predictive model equation under the influence of temperature, pressure and polymer characteristics. Predictive model developed in this research showed good agreement with experimental data of gas permeability characteristics for develop four different polymer membranes. The proposed model can also be extended to the general equation for predicting the separation of gases based on the properties of polymeric membranes.

Production and Fermentation Characteristics of Mukeunji with a Mixed Starter (혼합 스타터를 이용한 묵은지의 제조 및 발효 특성)

  • To develop a starter culture system for the fermentation of mukeunji, we introduced lactic acid bacteria and yeast isolated from mukeunji into kimchi fermentation as a single or a mixed culture. On evaluating mukeunji flavor, we found that the mixed starter kimchi prepared with two strains, ML17 and MY7, gave the best sensory score. These strains were identified as Lactobacillus (Lb.) curvatus ML17 and Saccharomyces (S.) servazzii MY7 by molecular identification method. The fermentative characteristics of starter kimchi were investigated by measuring changes in the physicochemical and microfloral characteristics during the fermentation. The decrease in pH and increase in acidity in the starter kimchi were faster compared to respective values of control kimchi. There was a gradual decrease in hardness of starter kimchi, which was still slow compared to hardness decrease in control kimchi. Microbial analysis of starter kimchi revealed that Lb. curvatus ML17 and S. servazzii MY7 were the dominant organisms during the entire fermentation period. The lactic acid and citric acid contents of starter kimchi were higher than those of the control kimchi after 90 days of fermentation. By sensory evaluation, the starter kimchi scored higher in appearance, mukeunji flavor, sourness, carbonated flavor, savory taste, texture, and overall acceptability, but lower in off-flavor than the control kimchi.

Studies on the Optical and the Electrical Characterization of Organic Electroluminescence Devices of Europium Complex Fabricated with PVD(Physical Vopor Deposition) Technique (진공 증착법에 의하여 제작한 Europium complex 유기 박막 전기발광소자의 광학적.전기적 특성에 관한 연구.)

  • Electroluminescent(EL) devices based on organic materials have been of great interest due to their possible applications for large-area flat-panel displays. They are attractive because of their capability of multi-color emission, and low operation voltage. An approach to realize such device characteristics is to use active layers of lanthanide complexes with their inherent extremely sharp emission bands in stead of commonly known organic dyes. In general, organic molecular compounds show emission due to their $\pi$-$\pi*$ transitions resulting in luminescence bandwidths of about 80 to 100nm. Spin statistic estimations lead to an internal quantum efficiency of dye-based EL devices limited to 25%. On the contrary, the fluorescence of lanthanide complexes is based on an intramolecular energy transfer from the triplet of the organic ligand to the 4f energy states of the ion. Therefore, theoretical internal quantum efficiency is principally not limited. In this study, Powders of TPD, $Eu(TTA)_3(phen) and AlQ_3$ in a boat were subsequently heated to their sublimation temperatures to obtain the growth rates of 0.2~0.3nm/s. Organic electrolumnescent devices(OELD) with a structure of $glass substrate/ITO/Eu(TTA)_3(phen)/AI, glass substrate/ITO/TPD/Eu(TTA)_3(phen)/AI and glass substrate/ITO/TPD/Eu(TTA)_3(phen)/AIQ_3AI$ structures were fabricated by vacuum evaporation method, where aromatic diamine(TPD) was used as a hole transporting material, $Eu(TTA)_3(phen)$ as an emitting material, and Tris(8-hydroxyquinoline)Aluminum$(AlQ_3)$ as an electron transporting layer. Electroluminescent(EL) and current density-voltage(J-V) characteristics of these OELDs with various thickness of $Eu(TTA)_3(phen)$ layer were investigated. The triple-layer structure devices show the red EL spectrum at the wavelength of 613nm, which is almost the same as the photoluminescent(PL) spectrum of $Eu(TTA)_3(phen)$.It was found from the J-V characteristics of these devices that the current density is not dependent on the applied field, but on the electric field.
