Enzymatic Characteristics of Two Forms of the Purified Glucoamylase from Rhizopus oryzae (Rhizopus oryzae로부터 정제한 두가지형의 Glucoamylase의 효소적(酵素的) 특성(特性))

  • These experiments were conducted to investigate general enzymatic characteristics of two forms(glucoamylase I and glucoamylase II) of the purified glucoamylase produced by Rhizopus oryzae. Molecular weights of glucoamylase I and glucoamylase II estimated by Sephadex G-100gel filtration, were approximately 101,000 and 115,000, respectively, and those estimated by SDS-polyacrylamide gel electrophoresis being 120,000 and 127,000, respectively. Isoelectric points of the above enzyme were pH 7.25 and pH 7.75. The optimum temperature was $50^{\circ}C$ and the enzyme was stable below $45^{\circ}C$. Optimum pH of both glucoamylase I and glucoamylase II was about pH 5.0. The stable pH range of them were pH 3.5-8.0 and 4.5-8.0, respectively. Michaelis constants of glucoamylase I and glucoamylase II toward souluble starch were 4.545 mg/ml and 5.560 mg/ml, respectively. $Hg^{++}$, $Pb^{++}$, p-CMB and IAA were inhibitors of glucoamylase I and $Hg^{++}$, $Mn^{++}$, p-CMB and IAA were inhibitors of glucoamylase II.

Analysis of Hospital Admissions Related to Adverse Drug Events Using ADE Signals

  • Adverse drug events (ADEs) are the most common type of adverse events in medical practice. Hospital admissions related to ADEs cost high and should be monitored to prevent them. While concerns about the ADEs are increasing, the frequency and characteristics of admissions related to ADEs have not been reported in Korea. The objective of the study was to assess the rate of hospital admission related to ADEs and their characteristics through ADE signal-based retrospective reviews of medical records. As results, a total of 1,420 patients had ADE signals suggesting potential ADEs from 3,494 patients who discharged from an academic medical center over one month period. Six pharmacists independently assessed the presence of ADEs after the review of patients' medical records. Among the 3,494 discharges, 62 admissions (1.8%) were found to be realted to ADEs. Of admissions with ADEs, 83.9% were moderate (category F by the NCC MERP classification), 37.2% were preventable, and 85.5% were type A reaction. The most frequent suspected drugs causing ADEs were antineoplastics (48.9%), and the most frequent ADE signal detecting hospital admissions related to ADEs was white blood cell count (24.2%). Hospital admissions related to ADEs were found through screening the ADE signals. The ADE signal-based retrospective review could be a practical approach for identifying hospital admissions related to ADEs.

  • The adsorption characteristics of gaseous molecules on the carbonaceous adsorbent have been investigated at various temperature and pressure with different pore sizes using Grand Canonical Monte Carlo (GCMC) simulation method. The geometrical parameters and spectroscopic properties of adsorbates have been computed using density functional theory (DFT). At higher temperatures is the adsorption amount of adsorbates is decreased due to their vaporization. Considering the pore size effect, the adsorption characteristic depends on the adsorbate size, polarity and interaction between adsorbates, etc. At all cases employed in this study, the adsorption amount of adsorbates on the carbonaceous adsorbent is increased in the order $NH_3$ < $H_2S$ < $CH_3SH$, and this result is qualitatively in good agreement with the experimental observation.

  • Development of various medical systems was accomplished through the progress of biotechnological method for therapy of human diseases. Furthermore, drug delivery systems have been investigated to carry the bioactive materials such as drug or gene in the body effectively. The most important thing in this system is to develop biomedical polymers having biocompatibility, biodegradability, and non-toxicity. Chitosan, a natural polymer, has been importantly considered as biomedical materials due to its good biocompatibility and various bio-active characteristics. Since the property of chitosan is differently explained according to the crystalline structures of chitin, the study for structural analysis of chitin has to proceed to apply as a biomaterial. From this point of view, this article introduced the analysis of crystalline structural of chitin, general property of chitosan and potential characteristics of low molecular weight water-soluble chitosan (LMWSC) as a biomaterials. Furthermore, chemical modification of LMWSC using various functional groups was also performed to enhance its bioavailability and emphasize their potential as drug delivery carriers (DDS).

Selection of Starter Cultures and Optimum Conditions for Lactic Acid Fermentation of Onion

  • Lactic acid bacteria (LAB) isolated from various fruits and vegetables were screened in order to determine appropriate fermentation starters for manufacturing functional fermented onion juice. From the initial screening test comprising more than 700 isolated LAB, 16 isolates were selected based on their acid production rate. Among the selected isolates, the fermentation broth of KC-007 exhibited the highest electron donating and nitrite scavenging activities, with values at pH 1.2 of 95.6 and 68.7%, respectively. From the overall results obtained in this study, we finally selected the bacterium KC-007 as a fermentation starter. This bacterium was identified and named as Pediococcus pentosaceus based on its morphological and physiological characteristics, carbon-utilization pattern (as assessed using an API 50CHL kit), and molecular genetic characteristics (as assessed using the nucleotide sequence of the 16S rRNA gene). The optimal temperature, pH, and starter inoculation concentration (v/v) required for growth of the isolated strain were $40^{\circ}C$, pH 4.0-6.0, and 2%(v/v), respectively.

Flocculation Characteristics of the Polycondensate of 1-Butylamine and Epichlorohydrin(PBE) (1-부틸아민과 에피클로로히드린의 축합체인 PBE의 응집작용 특성)

  • A water soluble polymer, polycondensate of 1-butylamine and epichlorohydrin (PBE), was synthesized by condensation of 1-butylamine and epichlorohydrin. The characteristics of PBE were determined by IR spectroscopy, low angle light scattering measurement, and $\zeta$ potential measurement. Its interactions with colloidal bentonite particles in aqueous medium were also studied. The results of the studies are as follows : PBE is a cationic polyelectrolyte carrying tertiary ammonium ions on its backbone. The average molecular weight of PBE is found to be about 1,600. The adsorption of PBE on the colloidal bentonite particles are well described with Langmuir adsorption isotherm. As the amounts of PBE adsorbed on the bentonite particles increase, the $\zeta$ potential of the particles changes its sign from negative to positive. This inversion of charge confirms that PBE is cationic in nature. The adsorption of PBE onto the bentonite particles was found to occur through cation exchange reaction. It has been shown that PBE has flocculation effects on the colloidal suspension of bentonite. It has also enhanced effects of filtrability on the digested sludge.

  • Electrochemical characteristics and physical properties of polymer electrolyte which immobilized lithium salts such as $LiClO_4$ and $LiCF_3SO_3$ and plasticizers such as ethylene carbonate(EC) and propylene carbonate(PC) in high molecular weight poly(ethylene oxide)[PEO] polymer was investigated. PEO-Li based polymer electrolyte with plasticizers showed ionic conductivity of $10^{-4}S/cm$ at room temperature and high electrochemical stability up to 4.5 V(vs. $Li^+/Li$), so it can be applied to lithium secondary battery. The crystallinity of PEO decreased with the addition of lithium salts and plasticizers, especially $LiClO_4$ and PC showed more effective than and $LiCF_3SO_3$ and EC. Glass transition temperature($T_g$) of polymer electrolyte increased with increasing lithium salt concentration whereas melting temperature ($T_m$) decreased. Polymer electrolyte with plasticizers crystallized at $6^{\circ}C$.

The Content and Chemical and Physical Properties of the Pectin in Tangerine Peel (한국산 감귤류 폐과피 내의 펙틴함량과 펙틴의 특성에 관한 연구)

  • The pectin content in tangerine peel and the characteristics of extracted pectin and its gel were determined. The pectin contents of citrus peel and citrus albedo were 11.52% and 9.3% on a dry weight basis, respectively The equivalent weight, methoxyl content and acetyl content of the extracted citrus peel pectin were 596, 13.41% and 0.125% respectively. The intrinsic viscosity and molecular weight of extracted citrus peel pectin were higher than those of commercial citrus pectin samples. The anhydrouronic acid content and jelly grade of the extracted pectins were 98.4% and 138.1, respectively. These were higher than those of commercial citrus pectin. In the textural characteristics of gels, the gel made from the extracted pectin was shown to have lower values in hardness, cohesiveness, adhesiveness and gumminess than the gels made from commercial citrus pectins.

A Study on the Electrical and Optical Characteristics of CLN-PZT Ceramics (CLN-PZT 세라믹스의 전기, 광학 특성에 관한 연구)

  • This paper was studied on the effects of Ca-La-Nb substitution and Zr/Ti ratio variation to Pb(Zr, Ti)$O_3$ system on structural, electrical, optical and sound level characteristics in order to develope the piezoelectric and electrooptic ceramic devices. Also the specimens were prepared by the two stage sintering technique. The molecular formular was X($CaO{\cdot}1/4La_{2}O_{3}{\cdot}1/4Nb_{2}O_{5}){\cdot}(1-X)Pb(Zr_{Y}Ti_{1-Y})O_{3}$(x=100X, y=100Y), and the variation of x was $6{\sim}12$, y was 60${\sim}$49 and second stage sintering time was 20${\sim}$40 hours. The experimental results obtained from this study are as follows : 1. The density was decreased, the grain size was increased according to increase of Ca-La-Nb substitution. 2. The crystal structure was rhombohedral in composition 6/60/40, and the crystal structure was tetragonal and cubic according to increase of Ca-La-Nb substitution. 3. The Ca substitution of PZT system enhanced the sintering property. The Pb site vacancy resulting from the substitution of La-Nb increased the dielectrical constant, the piezoelectric charge constant, the dielectric loss and decreased the coercive field. 4. The resistivity of PZT system which has the P type conduction mechanism increased according to substitution of La-Nb because of the substituent acting as donor. 5. The PZT ceramics varied from ferroelectric substance according to increase of Ca-La-Nb substituent. The coercive field and saturation remanent polarization decreased, and at last straight line according to increase of La-Nb substitution. 6. The amount of Ca-La-Nb substitution to improve the light transmittance of speciment was 10 mol%, the Zr/Ti ratio was 49/51, and the second stage sintering time was 40 hours. 7. According to Ca-La-Nb substitution, the specimens was to be transparent. The 7.5/51/49 specimen was suitable for transparent sound vibrator because it had 58% light transmittance (thick 0.2[mm], wave length 700[mm]) and 48% electromechanical coupling factor.

Preparation and Characteristics of Polyurethane Hybrid Sealant Modified with Polydimethylsiloxane (Polydimethylsiloxnae 변성 Polyurethane Hybrid Sealant의 제조와 그 특성)

  • Three isocyanatopropyldimethoxysilylpolydimethylsiloxanes(IDMSi-PDMS) were synthesised from the reaction of isocyanatopropyltrimethoxysilane with monohydroxyl group termainated PDMS having different molecular weight($M_n$=5000, 10000, and 20000). Then PDMS modified polyurethane hybrid elastomer(PSMPH) were prepared from the reaction of IDMSi-PDMS with ${\alpha}$, ${\omega}$-hydroxyl group terminated polyurethane. PSMPH sealant was prepared by compounding PSMPH elastomer with additives such as plasticizer, adhesion promoter, crosslinking agent, vicosity increasing agent, inorganic filler, and catalyst at room temperature under nitrogen atmosphere. The methoxy group in the PSMPH sealant should be crosslinked with the hydroxyl group in the building stone or moisture by typical sol-gel reaction. The adhesive strength of the sealant having PDMS of $M_n$=5000 showed 40.28 kg of maxium load and 20.14 kg of break load. The shrinkage rate of the sealant having PDMS of $M_n$=20000 was 5.7% as the best result. Also, their skin over time, slump, oil content after 8 days under oil soaked paper and alkaline resistance characteristics show good results.
