The adsorption equilibria of nitrogen on a region of nanoporous carbonaceous adsorbent with local molecular orientation (LMO) were calculated by grand canonical Monte Carlo simulation at 77.16 K. Regions of LMO of identical size were arranged on a regular lattice with uniform spacing. Microporosity was predominately introduced to the model by removing successive out-of-plane domains from the regions of LMO and tilting pores were generated by tilting the basic structure units. This pore structure is a more realistic model than slit-shaped pores for studying adsorption in nanoporous carbon adsorbents. Their porosities, surface areas, and pore size distributions according to constrained nonlinear optimization were also reported. The adsorption in slit shaped pores was also reported for reference. In the slit shaped pores, a clear hysteresis loop was observed in pores of greater than 5 times the nitrogen molecule size, and in capillary condensation and reverse condensation, evaporation occurred immediately at one pressure. In the LMO pore model, three series of local condensations at the basal slip plane, armchair slip plane and interconnected channel were observed during adsorption at pore sizes greater than about 6 times the nitrogen molecular size. In the hysteresis loop, on the other hand, evaporation occurred at one or two pressures during desorption.

This study examined the transition process of clinical laboratory technologists' national examination and job definition in Korea and compared the differences in the national examination between Korea and neighboring countries, such as Japan and Taiwan. In Korea, the number of questions made for it was 200 (1965), 200 (1977), 300 (1982), 250 (1992), 330 (2006), and 280 (2015). The practice of clinical physiology is important for real-time monitoring, given the characteristics of physiological testing. On the other hand, there are conflicts between other occupations in the working area. Clinical molecular biology needs to be established as a new major subject considering the diagnostic importance of molecular biological tests and the speed of science and technology development. Clinical laboratory operations provide policy and guidance recommendations to technologist staff. The proposed clinical laboratory technologists' national examination comprises major subjects: clinical biochemistry, clinical hematology, clinical transfusionology, clinical immunology, clinical microbiology, clinical molecular biology, clinical histology, clinical cytology, clinical physiology, and clinical laboratory operations. In addition, this study proposes the job definition of clinical laboratory technologists, revising various chemical or physiological testing to biomedical or physiological testing required for medical practice.

The sperm quality is determined by the kinetic characteristics and acrosome integrity of the sperm. In the previous studies, analysis of semen quality had large errors because those experiments by using microscope had been conducted by people. In recent years, the molecular biological methods have been newly developed to complement the previous techniques. The ZAR1 gene is known to be a gene that affects early embryonic development in vertebrates, but there is no study of the association with semen. In this study, we analyzed the association between the kinetic characteristics and ZAR1 single nucleotide polymorphism (SNP) genotype. To detect the SNPs, we performed sequencing using genomic DNA from the whole bloods of Duroc pigs. We identified an SNP in the ZAR1 gene g.2540T>C. ZAR1 SNP genotypeing in 105 pigs revealed that the major and minor alleles were T and C, respectively. After we analyzed the association between the kinetic characteristics of sperm and the ZAR1 SNP genotype, we found a significant association in MOT (p<0.01), VSL (p<0.05) of the kinetic characteristics in the Duroc boars. It was confirmed that the boars with T allele were lower in MOT and VSL than C allele. Therefore, pigs with C allele are judged to be better at the MOT and VSL of semen. Based on these results, ZAR1 SNP genotyping may be a useful molecular biomarker to improve semen quality by applying molecular breeding technology.
A humic acid(HA, Aldrich Co) sample was subjected to ultrafiltration for molecular size fractionation and three fractions of different nominal size($F_1$: 1,000-10,000 daltons; $F_2$: 10,000-50,000 daltons; $F_3$: 100,000-300,000 daltons) were obtained. The structural characteristics of the size-fractionated HA were analyzed using their IR and solid state C-13 NMR spectral data, and the carboxylate group contents of the humic acids were determined using their pH titration data. The $^7F_0-{^5}D_0$ excitation spectra of Eu(III) complexes of the size-fractionated mgHA in aqueous solution were acquired($[Eu(III)]=1.0{\times}10^{-4}mol\;L^{-1}$, $(HA)=470-970mg\;L^{-1}$) at pH 5.0 using a pulsed tunable laser system, in which metal binding properties of the size-fractionated HA were elucidated and compared on another. Characterization of the IR and C-13 NMR spectral data indicated that the fraction($F_3$) with molecules of larger size were primarily aliphatic, while the fractions($F_1$, $F_2$) with smaller molecules of less than 50,000 daltons were predominantly aromatic. Titration data were consistent with an increase in the number of carboxylate groups per unit mass as molecular size became smaller. The $^7F_0-{^5}D_0$ excitation spectral data of Eu(III)-humate complexes showed that the peak maxima on these spectra were shifted toward lower energies with increasing molecular size of HA, indicating the higher degree of bindings of the Eu in the molecules of larger size. We also discussed the relationship of the lower energy shifts of the maximum peaks with increasing the molecular size of HA with the structural differences of the size-fractionated HA.
The study was conducted to evaluate the effect of chromium nanoparticle (CrNano) on growth, carcass characteristics, pork quality, and tissue chromium in finishing pigs. A total of 100 Duroc$\times$Landrace$\times$Yorkshire barrows (average initial BW of 64.78$\pm$0.83 kg) were divided into 10 pens, 10 pigs per pen. 5 pens of pigs were fed with control diet (corn-soybean meal, 1.05% Lys) and others were fed with control diet addition with 200 $\mu$g/kg Chrome (Cr) from CrNano. During the trial, all pigs were given free access to feed and water. After 35 days trial, 10 pigs from each each treatment (weight similar to average group weight, 91.34$\pm$1.31 kg for control group, and 92.26$\times$0.56 kg for CrNano treated group) were sacrificed to measure carcass characteristics, pork quality, and to collect the samples of longissimus muscle, liver, kidney and heart. The results indicated that the addition of 200 $\mu$g/kg CrNano decreased feed: gain ratio by 3.56% (p<0.05) compared to the control group. Pigs fed CrNano had 14.06% higher (p<0.05) carcass lean percentage, 19.96% (p<0.05) larger longissimus muscle area and 25.53% lower (p<0.05) carcass fat percentage, 18.22% lower (p<0.05) backfat thickness. Drip loss in chops from pigs fed CrNano was decreased by 21.48% (p<0.05) and weights of longissimus muscle and semimembranosus were increased by 16.33% (p<0.05) and 14.87% (p<0.05) respectively. In addition, supplemental CrNano resulted in 184.11% (p<0.05), 144.99% (p<0.05), 88.13% (p<0.05) and 52.60% (p<0.05) increment of Cr concentration in longissimus muscle, liver, kidney and heart, respectively. These results suggest that supplemental CrNano has beneficial effects on carcass characteristics, pork quality and individual skeletal muscle weight, increase tissue chromium concentration in selected muscle and organs.
Background : Plant breeding requires the collection of genetically diverse genetic resources. Studies on the characteristics of Platycodon grandiflorum resources have not been carried out so far. The present study was carried out to discriminate P. grandiflorum based on morphological characteristics and genetic diversity using simple sequence repeat (SSR) markers. Methods and Results :We collected 11 P. grandiflorum cultivars: Maries II, Hakone double white, Hakone double blue, Fuji white, Fuji pink, Fuji blue, Astra white, Astra pink, Astra blue, Astra semi-double blue and Jangbaek. Analyses of the morphological characteristics of the collection were conducted for aerial parts (flower, stem and leaf) and underground parts (root). Next, the genetic diversity of all P. grandiflorum resources was analyzed using SSR markers employing the DNA fragment analysis method. We determined that the 11 P. grandiflorum cultivars analyzed could be classified by plant length, leaf number and root characteristic. Based on the genetic diversity analysis, these cultivars were classified into four distinct groups. Conclusions : These findings could be used for further research on cultivar development using molecular breeding techniques and for conservation of the genetic diversity of P. grandiflorum. Moreover, the markers could be used for genetic mapping of the plant and marker-assisted selection for crop breeding.
This study was carried out to investigate the structural characteristics of kraft lignin and the wood degrading characteristics, the productivity of ligninolytic enzymes and the enzymatic degradation of kraft lignin by white-rot fungi. To purify kraft lignin, precipitation of kraft pulping black liquors of pitch pine meal was done by titration with lN $H_{2}SO_{4}$ reaching to pH 2, and isolation of the precipitates done by centrifugation. The isolated precipitates from pitch pine were redissloved in lN NaOH, reprecipitated by titration with lN $H_{2}SO_{4}$, washed with deionized water, and kept ofr analysis after freeze drying. Fractionation of the precipitates in solution by successive extraction with $CH_{2}Cl_{2}$ and MeOH, and the fractionates were named SwKL, SwKL I, SwKL II, and SwKL III for pitch pine kraft lignin. The more molecular weights of kraft lignin increased, the less phenolic hydroxyl groups and the more aliphatic hydroxyl groups. Because as the molecular weights increased, the ratio of etherified guaiayl/syringyl(G/S ratio) and the percentage were increased. The spectra obtained by 13C NMR and FTIR assigned by comparing the chemical shifts of various signals with shifts of signals from autherized ones reported. The optimal growth temperature and pH of white-rot fungi in medium were $28^{\circ}C$ and 4.5-5.0, respectively. Especially, in temperature and pH range, and mycelial growth, the best white-rot fungus selected was Phanerochaete chrysosporium for biodegradation. For the degradation pathways, the ligninolytic fungus jcultivated with stationary culture using medium of 1% kraft lignin as a substrate for 3 weeks at $28^{\circ}C$. The weight loss of pitch pine kraft lignin was 15.8%. The degraded products extracted successively methoanol, 90% dioxane and diethyl ether. The ether solubles were analyzed by HPLC. Kraft lignin degradation was initiated in $\beta$-O-4 bonds of lignin by the laccase from Phanerochaete chrysosporium and the degraded compounds were produced from the cleavage of $C\alpha$-$C\beta$ linkages at the side chains by oxidation process. After $C\alpha$-$C\beta$ cleavage, $C\alpha$-Carbon was oxidized and changed into aldehyde and acidic compounds such as syringic acid, syringic aldehyde and vanilline. And the other compound as quinonemethide, coumarin, was analyzed. The structural characteristics of kraft lignin were composed of guaiacyl group substituted functional OHs, methoxyl, and carbonyl at C-3, -4, and -5 and these groups were combinated with $\alpha$ aryl ether, $\beta$ aryl ether and biphenyl. Kraft lignin degradation pathways by Phanerochaete chrysosporium were initially accomplished cleavage of $C\alpha$-$C\beta$ linkages and $C\alpha$ oxidation at the propyl side chains and finally cleavage of aromatic ring and oxidation of OHs.
Characteristics and pathogenicity of Yersinia sp. strain isolated from diseased cultured olive flounder, Paralichthys olivaceous were studied. The causative organisms were identified as Yersinia sp. by biochemical and biophysical characteristics. The strain was named KF-1, and it showed the optimal growth rate at pH 9.0 and 1% NaCl. Total cell protein peptide bands of the Yersinia sp. KF-1 were between 14.4-100.6 Kd in molecular weight by the electrophoretic analysis, and a total of 28 bands appeared. The band found at 32.8 Kd in molecular weight was the major one of electrophoretic phase. In the pathogenicity test of the isolate to the flounder injecting with $1.0{\times}10^7$ cfu/fish 9 died out of 10 within 60 hrs, and in the group with $1.0{\times}10^7$ cfu/fish 5 died within 60 hrs. Thus the $LD_{50}$ was presumed to be $1.0{\times}10^7$ cfu/fish. In the drug sensitivity test KF-1 strain was sensitive to chloramphenicol, gentamycin, kanamycin, streptomycin and pefloxacin, and intermediate to erythromycin, and resistant to ampicillin, cephalothin, sulfamethoxazole/trimethoprim, tetracycline and vancomycin.
Sixteen soil samples around six areas (residental area, traffic area, power plant area, incineration area and factory area) where the stationary and mobile sources of polycyclic aromatic hydrocarbons (PAHs) are estimated to be emitted in Jeju City, were collected during Feburuary to March, 2004, and analyzed for 16 PAHs recommended by US EPA as primary pollutants to investigate their distribution characteristics. The concentrations of total PAHs (t-PAHs) and total carcinogenic PAHs $(t-PAH_{CARC})$ in soils of Jeju City were in the range of $21.7\sim264.2ng/g$ on a dry weight basis with a mean value of 87.2 ng/g and $6.3\sim118.0ng/g$ with a mean value of 33.4 ng/g, respectively. The concentrations of t-PAHs were low in comparison with those in soils of other domestic and foreign countries. The mean concentrations of t-PAHs and $(t-PAH_{CARC})$ with area decreased in the following sequences: traffic area> incineration area > factory area > power generation area > harbor area enli residental area. The correlation between t-PAHs and $(t-PAH_{CARC})$ were very high $(\gamma^2=0.9701)$, indicating that $(t-PAH_{CARC})$ concentration increases in proportion with t-PAHs. Comparing the distribution ratio of ring PAHs with area among 16 PAHs, it decreased in the order of 4-ring > 5-ring > 6-ring > 3-ring > 2-ring in all the areas except for harbor area. whitens for harbor area it was similar among 3-, 4- and 5-ring with high value. Low and no correlations between t-PAHs and soil compositions (organic matter content and particle size distribution) were observed, which is considered to be caused by the complex factors, such as the loading and characteristics of PAHs and diverse soil environment change, etc. From the examination of the three PAH origin indices, such as LMW/HMW (low molecular weight $2\sim3$ ring PAHs over high molecular weight $4\sim6$ ring PAHs), phenanthrene/anthracene ratio and fluoranthene/pyrene ratio, it can be concluded that the soil PAH contaminations were ascribed to strong pyrogenic origin in ail areas except for harbor area and to both pyrogenic and petrogenic origins.

The 18 isolates of soybean red crown rot pathogen(Calonectria ilicicola) were isolated from infected soybean plants showing chlorosis symptom and red perithecia sign in 2002 and 2003 season in Korea. The morphological characteristics of the typical isolate SC03-15, such as macroconidia and macrovesicles in anamorph as well as perithecia, asci and ascospores in teleomorph, were observed from red crown rot pathogens. The perithecia were red-brown globose to ovoid, and $357.9(312.5{\sim}391.5.7){\mu}m$ high and $474.3(419{\sim}535){\mu}m$ diameter. Asci were hyaline and clavate and included 8 ascospores, which were straight to slightly curved, 1(-3)-septate, and $36.9(31.1{\sim}49.4){\times}5.2(4.5{\sim}6.1){\mu}m$. Macroconidia in anamorph were cylindrical, hyaline and in size $73.2(68.1{\sim}77.6){\times}5.8(4.9{\sim}6.8){\mu}m$. Further identification process was performed through molecular method, such as a phylogenetic analysis by rDNA sequencing of ITS and $\beta$-tubuline genes of all isolates tested in this study. All isolates were identified as Calonectria ilicicola by DNA sequencing of ITS and $\beta$-tubuline genes, and morphological characteristics.
