Storeability and Cooking Property of Dried Oak Mushroom Treated with Ethylene Oxide and Gamma Radiation (감마선 조사와 훈증 처리된 건조 표고버섯의 저장성 및 조리 적성)

  • ABSTRACT$.$ Ethylene oxide (E.O) fumigation and gamma irradiation were applied to compar$.$ ative researches on the microbiological, physical and cooking quality of dried oak mushroom stored at $25^{\circ}C$ and different relative humidities. The equivalent moisture contents of dried oak mushroom for the limiting growth of general molds and xerophilic mold at $25^{\circ}C$ were shown to be 17% and 27% respectively. Total aerobic bacteria, molds and coliforms were sterilized at 5 kGy irradiation but E.O. fumigation was proved insufficient to eliminate the molds. The hydration rate of dried oak mushroom increased according to the increase of irradiation dose and soaking temperatures. and an irradiation by 5 kGy could shorten the hydration time of the sample as compared to E.O. treatment and control group. Sensory evaluation for the irradiated cooked sample was not significantly different in flavour but the texture of the gamma irradiated sample was significantly better(p < 0.01) than that of E. O. fumigated sample.sample.

The Suppressive Effects of Calcium Compounds against Botrytis cinerea in Paprika (파프리카 양액재배에서 발생하는 잿빛곰팡이병 방제에 대한 칼슘제제의 효과)

  • Plant diseases including gray mold caused by Botrytis cinerea are often reduced when calcium compounds are used as alternative materials in paprika. However, much less information is available about the effects of calcium compounds on controlling of $B.$ $cinerea$. Seven calcium compounds such as calcium sulfate dihydrate, calcium chloride, calcium nitrate, calcium oxide, calcium hydroxide, calcium carbonate, and calcium hydride were evaluated for their effectiveness against $B.$ $cinerea$ on potato dextrose agar medium. The pH of selected calcium compounds was higher (pH 8.2-10) than that of the control (pH 6.6). Calcium carbonate, calcium oxide, calcium hydride, and calcium hydroxide among seven calcium compounds were more effectively inhibited the growth of $B.$ $cinerea$ than other calcium compounds. In the case of spraying the spore suspension on paprika applied with the selected four calcium compounds and supplied with the selected calcium supplements in a hydroponic culture system, the paprika treated with calcium compounds showed less severity of disease than those untreated plants. On the basis of our results, we propose that the suppressive effects of calcium compounds on $B.$ $cinerea$ in paprika resulted from the supply of calcium and a certain degree of salt stress.

Effect of Sorbitol on the Physicochemical and Microbial Characteristics of Chiffon Cake (소비톨 첨가 시폰 케이크의 품질 특성 연구)

  • Sucrose in chiffon cakes was replaced with different levels of sorbitol (0, 30, 50, 70, and 100%), and the effects on the physical properties and storage stability of the cakes were measured. The specific gravity and viscosity of the batter, as well as the volume and moisture content of the cakes, decreased with increasing sorbitol content. The hardness of the cakes slightly decreased as the amount of sorbitol increased, whereas the lowest cohesiveness was observed in the cakes with equal amounts of sucrose and sorbitol. Storage for one day slightly increased the hardness of the cakes and decreased their cohesiveness. Although the amount of mold and total microbial count increased with increasing storage days at room temperature, the replacement of sucrose with 70 or 100% sorbitol was effective in suppressing microbial growth. In sensory tests, the formulation with 70% sorbitol substitution in the chiffon cakes was the most acceptable.

Occurrence of Major Diseases in Vegetable Growing under the Furnihsed Condition in Southern Part of Korea (남부 시설원예지대의 주요병 발생생태에 관한 연구)

  • The study was conducted to obtain the basic information on the diseases epidemics of vegetable crops grown in Namji, Jinju, Gimhae and Suncheon under the extremely varied and specified conditions, plastic film house. The disease survey was conducted from the end of April, 1951 to April 1982. Leaf mold and late blight were serious foliar disease in tomato during the seedling stage, especially when the infected seeds were sown. The diseases increased rapidly 35 days after seeding. In both continuous cultivation of cucumber and rotation with upland crops, incidence of Fusarium wilt was severe while incidence of the disease was negligible in cultivations after paddy rice or grafting on pumpkin. Downy mildew of cucumber was severe in Jinju and Suncheon area, however, it was not so serious in Namji area where the growing season of cucumber was unfavorable for the maximum disease incidence. Cucumber mosaic virus disease was prevalent in the areas surveyed and the disease incidence was increased rapidly after June. Powdery mildew prevailed at the early stage of cucumber growth after transplanting in Namji area. Root rot and wilting caused by Phytophthora capsici was as the most destructive disease in pepper grown under the vinyl house, especially in Namji and Jinju area where the pepper has been cultivated intensively. The Phytophthora attacked most parts of young plants during the winter time and then induced crown rot on the adults plants. Cultivation of pepper in vinyl house was almost impossible because of the Phytophora disease by the end of June. Virus diseases to tomato plants were prevalent throughout the surveyed area and the damage was also severe. In Jinju and Gimhae area leaf mold and late blight showed high infection rate in tomato during the harvest time.

Preservative Safety Analysis of the Fresh Apple Snails and their Processing Products (우렁이 생체 및 가공제품의 저장안정성 조사)

  • This study investigated the microbial safety of apple snails and intermediate-moisture-content apple snail products subjected to various treatments, according counts of total bacteria, Escherichia coli, mold and yeast during storage at 4C Heating of raw apple snails at 95C for 5 min increased the storage period in relation to total bacteria, E. coli, mold and yeast-cell growth. Addition of 10% NaCl and 50% vinegar increased the microbial safety of apple snails. Microorganisms were not detected for 14 weeks in vacuum-packaged intermediate-moisture-content apple snail products stored at either 4C or 30C. These results suggest that heat treatment of raw apple snails, soaking in 10% NaCl and 50% vinegar solutions, and vacuum-packaging increases the shelf-life and microbial safety of apple snail products.

Isolation and characterization of antifungal violacein producing bacterium Collimonas sp. DEC-B5 (항진균활성 violacein 색소를 생산하는 Collimonas sp. DEC-B5 균주의 분리 및 특성)

  • Forty-nine pigments were extracted from the collections of 106 pigment producing bacteria from the plant rhizosphere soil. Antibacterial activity test was performed in the subjects of the extracted pigments with plant pathogenic bacteria including Xanthomonas axonopodis and Xanthomonas campestris, and with plant pathogenic fungi including Botrytis cinerea, Colletotrichum acutatum, and Fusarium oxysporum. The yellow pigment by Chryseobacterium sp. RBR9 and the red pigment by of Methylobacterium sp. RI13 showed the antibacterial activities against Xanthomonas axonopodis and Xanthomonas campestris. The violet pigment by Collimonas sp. DEC-B5 showed the antibacterial activity as well as the antifungal activities against Botrytis cinerea and Fusarium oxysporum. Especially, the violet pigment inhibited the growth of Botrytis cinerea more than 65% at MIC $20{\mu}M$. Upon the HPLC analysis result for the isolation of pigment with antifungal activity, violacein (91.6%) and deoxyviolacein (8.4%) were isolated for the pigment by Collimonas sp. DEC-B5. The production amount of the pigment was increased more than 10 times higher when D-mannitol 1.5% and yeast extract 0.2% were added as the nitrogen source to SCB medium. This study suggests that produced violacein by Collimonas sp. DEC-B5 will be effective to control strawberry gray-mold rot fungi by its preventive activity.

Temperature and microbial changes of corn silage during aerobic exposure

  • Objective: This study was conducted to estimate the temperature and microbial changes of corn silages during aerobic exposure. Methods: Kwangpyeongok (KW) and Pioneer 1543 (PI) corn hybrids were harvested at 29.7% of dry matter and chopped to 3 to 5 cm lengths. Homo (Lactobacillus plantarum; LP) or hetero (Lactobacillus buchneri; LB) fermentative inoculants at $1.2{\times}10^5$ colony forming unit/g of fresh forage was applied to the chopped corn forage which was then ensiled in quadruplicate with a $2{\times}2$ (hybrid${\times}$inoculant) treatment arrangement for 100 days. After the silo was opened, silage was sub-sampled for analysis of chemical compositions, in vitro digestibility, and fermentation indices. The fresh silage was continued to determine aerobic exposure qualities by recorded temperature and microbial changes. Results: The KW silages had higher (p<0.01) in vitro digestibilities of dry matter and neutral detergent fiber than those of PI silages. Silages applied with LB had higher (p<0.001) acetate concentration, but lower (p<0.01) lactate concentration and lactate to acetate ratio than those of LP silages. The interaction effect among hybrid and inoculant was detected in acetate production (p = 0.008), aerobic stability (p = 0.006), and lactic acid bacteria count (p = 0.048). The yeast was lower (p = 0.018) in LB silages than that in LP silages. During the aerobic exposure, PI silages showed higher (p<0.05) temperature and mold than KW silages, while LP silages had higher (p<0.05) lactic acid bacteria and yeast than LB silages. Conclusion: The results indicated that the changes of silage temperature during aerobic exposure seems mainly affected by mold growth, while applied LB only enhanced aerobic stability of PI silages.

Effect of Acidulant Treatment on the Quality and Storage Period of Topokkidduck (산미료 단독 처리가 떡볶이 떡의 저장기간 및 품질에 미치는 영향)

  • This study evaluated the effects of acidulant treatment on the quality and storage period of Topokkidduck. Two samples of Topokkidduck were prepared, one soaked in 10% acidulant (10SAT) and the other without soaking in the acidulant (NSAT). During the storage period, the two samples were tested for presence of microorganisms (aerobic bacteria, E.coli, and mold) and physicochemical properties (color value, texture profile analysis (TPA)). The 10SAT could be stored for 49 days without detection of E.coli and a mold level of 1.0 log CFU/g. NSAT could be stored for only 21 days. NSAT had an aerobic count of 2.27 log CFU/g as early as 7 days, and E.coli was detected at 21 days at a level of 4.15 log CFU/g. The presence of E.coli is not permitted according to the Ministry of Food and Drug Safety (MFDS). The hardness of the 10SAT increased during the storage period but to a much lesser extent compared to the NSAT. Thus the preparation of Topokkidduck by soaking in the acidulant controlled microbial growth for up to 49 days which is a much longer period compared to the control. Also, Topokkidduck soaked in the acidulant had a softer texture than the control during the storage period.

Monitoring for the Resistance of Botrytis cinerea Causing Gingseng Gray Mold to Procymidone and Its Multiple resistance with the Mixture of Carbendazim/Diethofencarb (인삼 잿빛곰팡이병균의 procymidone에 대한 감수성 변화와 carbendazim/diethofencarb 합제와의 다중 저항성)

  • Effects of fungicides on the mycelial growth of Botrytis cinerea isolated from ginseng leaves were investigated by an agar dilution method. By using a agar dilution method, it was investigated the effect of fungicides, procymidone, carbendazim and the mixture with both of carbendazim and diethofencarb, on the mycelial growth of Botrytis cinerea isolates, which were isolated from infected leaves of ginseng in 2005 and 2006. With MIC (minimum inhibiton concentration) of procymidone against B. cinerea, pathogens were divided into two groups. While one showed the low MIC between 0.8 and $4.0{\mu}g/ml$, the other showed higher MIC above $20{\mu}g/ml$. In terms of the inhibition ratio of mycelial growth at the indicated concentration of procymidone, isolates of B. cinerea were divided into three groups; the sensitive, the intermediate resistant, and the resistant group. Each group was differentiated by $EC_{50}$; the sensitive group showed below $2.0{\mu}g/ml$, the intermediate resistant group between 2.0 to $5.0{\mu}g/ml$, and resistant group above $5.0{\mu}g/ml$. Compared with the ratio of resistant isolates of B. cinerea in 2005, the ratio in 2006 increased from 19.3% to 27.5%. Furthermore, the average $EC_{50}$ value of them increased from $10.0{\mu}g/ml$ in 2005 to $237.3{\mu}g/ml$ in 2006. The ratio of isolates showing the multiple resistance between procymidone and carbendazim was 40.2%, whereas the ratio was 4.0% showing the multiple resistance in the mixture.

Effect of agrochemicals on mycelial growth and spore germination of a hyperparasite, Ampelomyces quisqualis 94013 for controlling cucumber powdery mildew (농약이 오이흰가루병 방제용 중복기생균 Ampelomyces quisqualis 94013의 균사생장 및 포자발아에 미치는 영향)

  • A promising hyperparasite, Ampelomyces quisqualis 94013(AQ94013) was selected as a biological control agent to cucumber powdery mildew caused by Sphaerotheca fusca. Effect of agrochemicals on mycelium growth and spore germination of AQ94013 and effect of spread stickers on hyperparasitical activity of AQ94013 to powdery mildew pathogen were evaluated. Finally it was confirmed that mycelial growth and spore germination of AQ94013 on potato dextrose agar amended with two fungicides for controlling powdery mildew, triadimefon and pyrazophos; five fungicides for controlling downy mildew, dimethomorph, kasugamycin+copper oxychloride, dichlofluanid+copper oxychloride and tribasic copper sulfate; three fungicides for controlling gray mold, iprodione, vinclozolin and procymidone; and six insecticides immidacloprid, teflubenzuron, bifenthrin, ethofenprox, deltamethrin and phenthoate were slightly reduced. Addition of mineral oil in the spore suspension of AQ94013 enhanced 7.9% control value to cucumber powdery mildew.
