Spectroscopic Analysis of Partially Folded State of Ubiquitin (유비퀴틴 단백질의 부분적으로 폴딩된 구조에 대한 분광학적 분석)

  • Hydrophobic core variant of ubiquitin appeared to have partially folded structure at pH around 2. The intrinsic tryptophan fluorescence emission maximum of this ubiquitin variant at pH 2 showed slight blue shift compare to that of unfolded state, suggesting that some residual tertiary structures remain in this solvent condition. At the same solvent condition, this ubiquitin variant binds with hydrophobic dye, 8-anilinonaphthalene-1-sulfonic acid(AMS), which is known to bind to exposed hydrophobic surface. Furthermore, far-UV circular dichroic spectrum of this ubiquitin variant in the diminished pH was remarkably different from the far-UV CD spectrum of the native state or unfolded state. Based on the molar ellipticity at 220 nm, this ubiquitin variant at pH 2 appeared to have significant amount of secondary structures. All these observations suggest that this ubiquitin variant in the diminished solvent pH has loosely folded hydrophobic core with some secondary structures, which are key features of molten globule conformation. Since molten globule has long been considered as a protein folding intermediate, it is considered that this hydrophobic core variant ubiquitin will serve as a valuable model to study protein folding process.

Cyclic AMP Receptor Protein Adopts the Highly Stable Conformation at Millimolar cAMP Concentration (높은 cAMP 농도에서 cAMP 수용성 단백질의 열 안정화)

  • Cyclic AMP receptor proteins(CRP) activate many genes in Escherichia coli by binding of cAMP with not fully known mechanism. CRP existed as apo-CRP in the absence of cAMP, $CRP;(cAMP)_2$$_2$ at low(micromolar) cAMP concentration, or $CRP;(cAMP)_4$ at high(millimolar) concentration of cAMP. This study is designed to measure the thermal stability of S83G CRP, which substituted glycine for serine at amino acid 83 position, with CD spectrapolarimeter at 222nm by the constant elevation of temperature from $20^{\circ]C\; to\; 90^{\circ}C\; at\; 1^{\circ}C/min$. The non-linear regression analysis showed that melting temperatures were 68.4, 72.0, and $82.3^{\circ}C$ for no cAMP, 0.1mM cAMP, and 5mM cAMP, respectively. Result showed the strong thermal stability of CRP by binding of additional cAMP molecules to region between the hinge region and helix-turn-helix(HTH) motif at 5mM cAMP concentration.

The Effect of Acidic pH on the Spectral Properties of Bacteriorhodopsin (산성 pH가 박테리오로돕신의 분광학적 성질에 미치는 효과)

  • Purple membrane from Halobacterium halobium was incorporated into 7.5% polyacrylamide gels. Absorption and circular dichroic spectra of purple membrane incorporated with gels were obtained at various pH. The spectra of these gels measured at pH 7.0 were essentially identical with those obtained in the aqueous suspension of purple membrane. Acid titration of the gels showed the transition to a form absorbing at 605nm $(bR_{605}^{acid}$) at pH 2.6, and to a second form at 565nm $(bR_{565}^{acid})$ at pH 0.8. Dark-adapted gels showed an isosbestic point for each transition whereas light-adapted gels did not. Visible CD spectra of $bR_{570}^{LA},\;bR_{305}^{acid}\;and\;bR_{565}^{acid}$ all showed the typical bilobed pattern. CD spectra measured at UV wavelength region were also independent of the variation of pH in terms of molar ellipticity and spectral shape. The protonated species $bR_{605}^{acid}$ may be one of the intermediates formed during the normal photochemical cycle of purple membrane. Most probably, the species $bR_{605}^{acid}$ is considered to be $O^{640}$ in the cycle.
