AMPKα, C/EBPβ, CPT1β, GPR43, PPARγ, and SCD Gene Expression in Single- and Co-cultured Bovine Satellite Cells and Intramuscular Preadipocytes Treated with Palmitic, Stearic, Oleic, and Linoleic Acid

  • We previously demonstrated that bovine subcutaneous preadipocytes promote adipogenic gene expression in muscle satellite cells in a co-culture system. Herein we hypothesize that saturated fatty acids would promote adipogenic/lipogenic gene expression, whereas mono- and polyunsaturated fatty acids would have the opposite effect. Bovine semimembranosus satellite cells (BSC) and intramuscular preadipocytes (IPA) were isolated from crossbred steers and cultured with 10% fetal bovine serum (FBS)/Dulbecco's Modified Eagle Medium (DMEM) and 1% antibiotics during the 3-d proliferation period. After proliferation, cells were treated for 3 d with 3% horse serum/DMEM (BSC) or 5% FBS/DMEM (IPA) with antibiotics. Media also contained $10{\mu}g/mL$ insulin and $10{\mu}g/mL$ pioglitazone. Subsequently, differentiating BSC and IPA were cultured in their respective media with $40{\mu}M$ palmitic, stearic, oleic, or linoleic acid for 4 d. Finally, BSC and IPA were single- or co-cultured for an additional 2 h. All fatty acid treatments increased (p = 0.001) carnitine palmitoyltransferase-1 beta ($CPT1{\beta}$) gene expression, but the increase in $CPT1{\beta}$ gene expression was especially pronounced in IPA incubated with palmitic and stearic acid (6- to 17-fold increases). Oleic and linoleic acid decreased (p = 0.001) stearoyl-CoA desaturase (SCD) gene expression over 80% in both BSC and IPA. Conversely, palmitic and stearic acid increased SCD gene expression three fold in co-cultured in IPA, and stearic acid increased $AMPK{\alpha}$ gene expression in single- and co-cultured BSC and IPA. Consistent with our hypothesis, saturated fatty acids, especially stearic acid, promoted adipogenic and lipogenic gene expression, whereas unsaturated fatty acids decreased expression of those genes associated with fatty acid metabolism.

Determination of Priority for Improvement Using the Theory of Two-dimensional Quality (품질의 이원론을 이용한 개선의 우선순위 결정)

  • The theory of two-dimensional quality, in particular, the Kano model that is developed by the analogy with the M-H theory, has been applied in various industry fields for more than three decades. Importance-Performance Analysis (IPA) assumes that the degree of physical fulfilment of quality attributes and the satisfaction of that attribute is linear, and therefore, it is applicable to the traditional one-dimensional attribute, not other quality types defined in the Kano's model such as attractive or must-be attribute. To solve this problem, the current study suggests a new importance-satisfaction analysis using a modified IPA in accordance with the three quality types and a diagonal method introduced by Slack (1999) to determine improvement priority. For this, I investigated 19 smartphone's quality attributes and conducted a survey of 334 university students for the results of Kano's model, which adopted from Song and Park (2012)'s study, and the importance/satisfaction of the quality attributes and the results of the priority for improvement of the 19 quality attributes. The results show that the proposed I-S priority model is better than the conventional IPA based on the comparison results of determination coefficient from the regression analysis of the two models.

Development of Selective Adsorption Process with Various Pore Size A-type Zeolite on Removal of Acetylenes for Isoprene Purification (제올라이트 A를 이용하여 이소프렌에서 아세틸렌 제거를 위한 선택적 흡착공정 개발)

  • This study focused on the development of effective adsorbent to remove acetylenes for the purification of isoprene. The adsorbents with various pore sizes from $4{\AA}$ to $5{\AA}$ were prepared to investigate the effect of pore size on selective adsorption of acetylene as an impurity. The pore size of zeolite A was adjusted by ion-exchange between Na and Ca ions. The pore size of adsorbents has affected the removal of acetylenes selectively because of the kinetic diameter of acetylenes, such as 2-methyl-1-butyne-3-yen (IPA) and 2-butyne. In a batch adsorption experiment, 5A zeolite with pore size of $5{\AA}$ showed the highest removal capacity of 2-butyne. However, IPA was hardly removed from isoprene by the A-type zeolites. For the adsorption isotherm, modified Langmuir model was well fitted with 2-butyne adsorption. Moreover, the regeneration of adsorbent was carried out to determine optimum method. The adsorbent heated for 12 h at $300^{\circ}C$ was regenerated significantly.

A Study on Physical Dispersion and Chemical Modification of Graphene (Graphene의 물리적 분산과 화학적 표면 개질 연구)

  • Graphene has a wide spectrum on its application field due to various and excellent physical properties. However, it is very difficult to apply that graphene exists as lump or fold condition in general organic solvents. Besides, graphene was difficult to maintain as uniform condition due to chemical inert and distributions with various size and shapes. Therefore, this study was focused to study dispersion and modifying methods of aggregated graphene. The dispersion methods contain as follow: i) physical milling using glass bead, ii) co-treatment of glass bead and ultrasonic waves, iii) dispersion in organic solvents, iv) modifying with dry-ice. Milling using glass bead with size 2.5 mm was effective to be size decrease of 36.4% in comparison with control group. Mixed treatment of glass bead (size 2.5 mm) and ultrasonic waves (225W, 10 min) showed relative size decrease of 76%, suggesting that the size decrease depends on the size of glass bead, intensity of ultrasonic waves and treatment time. Solvents of Ethyl acetate (EA) and Isoprophyl alcohol (IPA) were used in order to improve dispersion by modifying surface of graphene. IPA of them showed a favorable dispersion with more -CO functional groups in the FT-IR analysis. On the other hand, the oxygen content of graphene surface modified by dry-ice was highly increased from 0.8 to 4.9%. From the results, it was decided that the favorable dispersion state for a long time was obtained under the condition of -CO functional group increase in IPA solvent.

Determining Improvement Priorities of Barrier Free Components for Beaches through M-IPA (M-IPA를 이용한 해수욕장 Barrier Free 환경조성 요소의 개선 우선순위 분석)

  • A number of cities are committed to enhancing the barrier-free (BF) environments. However, no attempts have been made to examine the BF environment design concept by accounting for the universal BF components, which should be improved for providing both disabled people and non-disabled people with better BF environments. With this challenge in mind, this study determines the improvement priorities of BF components for beaches in the city of Busan by investigating the results of satisfaction survey, which was administered to disabled and non-disabled visitors. The modified importance-performance analysis (M-IPA) results show that "sanitation facilities" are the universal BF components having the highest improvement priority. The analysis results also indicate that "access facilities" have the second highest improvement priority for disabled people while "amenity facilities" have the second highest improvement priority for non-disabled people. It is highly expected that the suggested improvement priorities of BF components can be used for policy makers to develop more flexible plans when designing the BF beach.

An Analysis on Determining Quality of Service Criteria for Expressway Bus Passengers Using The Importance-Performance Analysis (IPA) - Focussing on Yong-in City : Suji - (IPA 분석을 이용한 간선급행버스 이용자 서비스 특성분석 - 용인 수지지구 중심으로 -)

  • The bus transfer system has been extended from Seoul to Gyonggi province to provide better Seoul metropolitan area transit service in September, 2008. Moreover, the curved bus routes, except existing rapid bus routes, which cause longer travel time have been straightened out. Also, the Skip Stop operation suggested by passengers has been introduced to main stops in the newly added express bus lines. This study surveyed passengers service satisfaction for the recent adopted bus policies such as the transfer discount system and the express bus system in Seoul Metropolitan area. The survey results may be important foundation for future strategies for improvement. The survey included questionnaires about the importance and the satisfaction level on both quantitative and qualitative factors. The results were statistically analyzed by the modified IPA (Importance-Performance Analysis). As the result of the survey, the newly adopted services such as fare system, fare discount on transferring, travel time savings and increased number of stops are economically feasible and satisfactory, whereas the accessibility to stops, ventilation and air quality in vehicles are the priorities to be improved. Also, the safety and the information system is in need of improvement.

A study on national cybersecurity policy agenda in Korea using national cyber capability assessment model (국가 사이버 역량평가 모델을 활용한 국내 사이버안보 정책 의제 도출 연구)

  • The National Cyber Capability Assessment(NCCA) could be used as meaningful information for improving national cyber security policy because it provides information on the elements necessary for strengthening national cyber capabilities and the level of each country. However, there were few studies on improving cyber capabilities using the NCCA result in Korea. Therefore, we analyzed the result of National Cyber Power Index(NCPI) conducted by Belfer Center of Harvard Univ. by applying modified-IPA method to derive cybersecurity policy agendas for Korea. As a result, the need to set agendas on surveillance and offensive cyber capability and improve the effectiveness of policy implementation for intelligence and defense was drawn. Moreover, we suggested need for in-depth study of each policy agenda deduced from preceding research data as a future tasks. And it is expected to increase practical use of NCCA for domestic policy analysis by developing and using our own NCCA model which considered analysis framework proposed in this study.

Quality Dynamics Using a Modified Satisfaction Index (수정된 고객만족지수를 이용한 품질속성의 동태성 분석)

  • It is well known that the Kano model measures customer satisfaction and classifies quality attributes into must-be, attractive as well as one-dimensional. The main purpose of this study is to investigate the dynamics of e-learning quality attributes by applying the proposed method using Kano's satisfaction index in the rapidly changing online learning environment. For this, the current study examined 27 e-learning quality attributes and conducted a comparative study using Kano's results obtained in 2013 and 2020. The result shows that the dynamics of quality attributes suggested by Kano(2001) is confirmed in the case of e-learning. The proposed approach shows better results in terms of Kano's direct classification method, and has potential application areas such as IPA(Importance-Performance Analysis) in the area of risk assemement. Some suggestions for better understanding of the proposed SI-DI diagram are also included in this study.

  • This Study analyzed heavy snow properties according to the area that was based by winter weather properties and the damage data by the heavy snow among each local government of Chungcheongbuk-do. The result of analysis, Jecheon-si and Boeun-gun are represented the highest dangerous regions by potential degree of risk by average amount of snowfall for 35 years. But, the potential degree of risk by maximum amount of snowfall for 35 years is different with it. Cheongju-si and Youngdong-gun, Goesan-gun, Boeun-gun are represented the highest dangerous regions. Examining the frequency of regions with potential danger factors according to the characteristics of heavy snowfall, Boeun-gun and Jecheon-si, Goesan-gun, Youngdong-gun, Cheongju-si is derived the highest dangerous regions in Chungcheongbuk-do.

Effect of H2O2 modification of H3PW12O40@carbon for m-xylene oxidation to isophthalic acid

  • The production of isophthalic acid (IPA) from the oxidation of m-xylene (MX) by air is catalyzed by $H_3PW_{12}O_{40}$ (HPW) loaded on carbon and cobalt. We used $H_2O_2$ solution to oxidize the carbon to improve the catalytic activity of HPW@C catalyst. Experiments reveal that the best carbon sample is obtained by calcining the carbon at $700^{\circ}C$ for 4 h after being impregnated in the 3.75% $H_2O_2$ solution at $40^{\circ}C$ for 7 h. The surface characterization displays that the $H_2O_2$ modification leads to an increase in the acidic groups and a reduction in the basic groups on the carbon surface. The catalytic capability of the HPW@C catalyst depends on its surface chemical characteristics and physical property. The acidic groups play a more important part than the physical property. The MX conversion after 180 min reaction acquired by the HPW@C catalysts prepared from the activated carbon modified in the best condition is 3.81% over that obtained by the HPW@C catalysts prepared from the original carbon. The IPA produced by the former is 46.2% over that produced by the latter.
