  • Effects of vitamins, metabolic intermediates and several inorganic mineral salts on the biomass and lipid accumulation of Mucor plumbeus were investigated after 15 days of incubation at $37^{\circ}C$ under static culture condition. The optimum concentrations of various vitamins were ${\gamma}/l$ for biotin, and 0.01 g/l for nicotinic acid, pyridoxine, thiamine and riboflavin. Among them pyridoxine was the most stimulatory. The maximum felt weight and lipid content per 50ml medium were $2.82{\pm}0.14\;g$ and 62.8%. Triglyceride content of neutral lipid produced under this condition was 64.9%. The major fatty acids were oleic acid (50.0%), linoleic (23.8%) and palmitic acid (13.9%). Malonic acid was considered not to be desirable even though it stimulated the biomass and lipid accumulation because triglyceride content was lowered considerably comparing with control. $MgSO_4{\cdot}7H_2O$ was the most stimulatory among the various magnesium salts and its optimum concentration was 5 g/l. Mucor plumbeus did not require $NaH_2PO_4$ for the stimulation of felt and lipid production. However, the addition of $MnCl_2$ at the concentration of 2 g/l was stimulatory to show $2.76{\pm}0.28\;g$ of felt/50 ml and 56.4% lipid content, and 73.9% triglyceride in the neutral lipid.

Thick Positive Electrode using Polytetrafluorethylene (PTFE) Binder for High-Energy-Density Lithium-ion Batteries (높은 에너지 밀도의 리튬이온 이차전지를 위한 PTFE 바인더를 적용한 고로딩 양극)

  • Many researchers have increased the loading level of electrodes to improve the energy density of secondary batteries. In this study, high-loading NCM523 (LiNi0.5Co0.2Mn0.3O2) positive electrode is manufactured using a polytetrafluoroethylene (PTFE) binder, not the conventional polyvinylidene fluoride (PVdF) binder, which has been commonly used in lithium-ion batteries. Through the kneading process using PTFE suspension, not the conventional slurry process using PVdF solution in N-methyl-2-pyrrolidinone (NMP), thick electrodes with high loading are easily manufactured. When the PTFE and PVdF-based electrodes are prepared at a loading level of 5.0 mAh/cm2, respectively, the PTFE-based electrode shows better cycle performance and rate capability than those of PVdF-based electrodes. The electrode manufactured by the kneading process using a PTFE binder has high electrode porosity due to insufficient roll-press, but the porosity can be lowered by high temperature roll-press over 120℃. However, there is no significant difference in cycle performance according to the roll press temperature. In addition, the cycle performance of the high loading electrode is slightly improved by increasing the content of the conductive material. Overall, the PTFE binder can improve the performance of the high loading electrode, but additional solutions will be needed.

Promoter Effect on Ni/YSZ Anode Catalyst of Solid Oxide Fuel Cell for Suppressing Coke Formation in the Methane Internal Reforming (고체산화물 연료전지용 Ni/YSZ 음극 촉매에서의 메탄 내부개질 반응 시 탄소 침적 억제를 위한 첨가제 영향)

  • Various additives were added in small amounts on Ni/YSZ anode of SOFC (solid oxide fuel cell) in order to improve reactivity and to inhibit deactivation due to coke deposition during methane reforming using a low mole ratio steam ($H_2O/CH_4=1.5$) at $800^{\circ}C$. Ni/YSZ catalysts added with various perovskites did not show any improvement but exhibited a gradual decrease in the methane conversion. K-doped Ni/YSZ showed a steady increase and maintenance of the conversion up to 42 hours, after which there was an abrupt deactivation of catalyst owing to potassium loss by volatilization. Addition of 5% of $K_2Ti_2O_5$ on Ni/YSZ showed a stable maintenance of the conversion without K loss, and was able to prevent coke formation during a long time operation. Deactivation of catalyst during the reaction was mainly caused by the accumulation of graphidic carbon on the catalyst surface.

  • To investigate the effects of paper sludge on seasonal variations of volatile lower fatty acids in paddy soil, paper sludge was applied to pots at the rate of either 300, 600, 900 or 1,200 kg/l0a which was either preadjusted at a C/N ratio of 30 : 1 or not adjusted. The decomposition rate of paper sludge, the evolution of $CO_2$, and the fractions of volatile lower fatty acids in the soil were determined. The results are summarized as follows: 1. Paper sludge was decomposed to $35{\sim}44%$, and its C/N ratio was $55{\sim}82$, respectively, at 120 days after treatment. 2. The evolution of $CO_2$, in the soil was proportional to the amount of paper sludge added. Significant positive correlations were observed the $CO_2$ evolution was compared with the decomposition rate of paper sludge, and volatile fatty acid contents in soil. 3. Acetic, propionic, butyric, i-butyric, valeric and i-valeric acids were identified in all the soils investigated. The content of the total volatile fatty acids in the soil increased with as the application of paper sludge increased. The formation of the acids was the highest at 25 days after treatment, and thereafter the contents of the acids decreased as time elapsed. 4. The volatile fatty acids in the soil inhibited the growth of paddy rice in early stages. The contents of acetic, propionic and i-valeric acids in the soil negatively, correlated with the uptake of N, $P_2O_5$, $K_2O$, CaO, MgO and $SiO_2$ in the paddy rice at 25 days after transplantation. In addition, the uptake of $P_2O_5$ and CaO in the paddy rice negatively correlated with the content of butyric acid in the soil. 5. The content of total volatile fatty acids positively correlated with the content of $Fe^{++}$ and $Mn^{++}$ in the soil at 25 days after transplantation. A significantly positive correlation was observed between $Fe^{++}$ and acetic acid contents in the soil.

Magnetic Tunnel Junctions with AlN and AlO Barriers

  • We studied the magnetotransport properties of tunnel junctions with AlO and AlN barriers fabricated using microwave-excited plasma. The plasma nitridation process provided wider controllability than the plasma oxidization for the formation of MTJs with ultra-thin insulating layer, because of the slow nitriding rate of metal Al layers, comparing with the oxidizing rate of them. High tunnel magnetoresistance (TMR) ratios of 49 and 44% with respective resistance-area product $(R{\times}A) of 3 {\times} 10^4 and 6 {\times} 10^3 {\Omega}{\mu}m^2$ were obtained in the Co-Fe/Al-N/Co-Fe MTJs. We conclude that AlN is a hopeful barrier material to realize MTJs with high TMR ratio and low $R{\times}A$ for high performance MRAM cells. In addition, in order to clarify the annealing temperature dependence of TMR, the local transport properties were measured for Ta $50{\AA} /Cu 200 {\AA}/Ta 50 {\AA}/Ni_{76}Fe_{24} 20 {\AA}/Cu 50 {\AA}/Mn_{75}Ir_{25} 100 {\AA}/Co_{71}Fe_{29} 40 {\AA}/Al-O$ junction with $d_{Al}= 8 {\AA} and P_{O2}{\times}t_{0X}/ = 8.4 {\times} 10^4$ at various temperatures. The current histogram statistically calculated from the electrical current image was well in accord with the fitting result considering the Gaussian distribution and Fowler-Nordheim equation. After annealing at $340^{\circ}C$, where the TMR ratio of the corresponding MTJ had the maximum value of 44%, the average barrier height increased to 1.12 eV and its standard deviation decreased to 0.1 eV. The increase of TMR ratio after annealing could be well explained by the enhancement of the average barrier height and the reduction of its fluctuation.

Mechanisms of Immobilization and Leaching Characteristics of Arsenic in the Waste Rocks and Tailings of the Abandoned Mine Areas (폐광산 지역 폐광석 및 광미에서 비소의 고정 메커니즘과 용출특성)

  • EPMA determined that Fe(Mn)-(oxy)hydroxides and well-crystallized Fe-(oxy)hydroxides and could contain a small amount of As $(0.3-11.0\;wt.\%\;and\;2.1-7.4\;wt.\%\;respectively)$. Amorphous crystalline Fe-(oxy) hydroxide assemblages were identified as the richest in As with $28-36\;wt.\%$. On the ternary $As_2O_5-SO_3-Fe_2O_3$ diagram, these materials were interpreted here as 'scorodite-like'. Dissolved As was attenuated by the adsorption on Fe-(oxy) hydroxides and Fe(Mn)-(oxy) hydroxides and/or the formation of an amorphous Fe-As phase (maybe scorodite: $FeAsO_4\cdot2H_2O$). Leaching tests were performed in order to find out leaching characteristics of As and Fe under acidic conditions. At the initial pHs 3 and 5, As contents dissolved from tailings of the cheongyang mine significantly increased after 7 days due to the oxidation of As-bearing secondary minerals (up to ca. $2.4\%$ of total), while As of Seobo mine-tailing samples was rarely released (ca. $0.0-0.1\%$ of total). Dissolution experiments at an initial pH 1 liberated a higher amount of As (ca. $1.1-4.2\%$ of total for Seobo tailings, $1.5-14.4\%$ of total for Cheongyang tailings). In addition, good correlation between As and Fe in leached solutions with tailings was observed. The kinetic problems could be the important factor which leads to increasing concentrations of As in the runoff water. Release of As from Cheongyang tailings can potentially pose adverse impact to surface and groundwater qualities in the surrounding environment, while precipitation of secondary minerals and the adsorption of As are efficient mechanisms for decreasing the mobilities of As in the surface environment of Seobo mine area.

A Study on the Evaluation of Fertilizer Loss in the Drainage(Waste) Water of Hydroponic Cultivation, Korea (수경재배 유출 배액(폐양액)의 비료 손실량 평가 연구)

  • Korean facility horticulture and hydroponic cultivation methods increase, requiring the management of waste water generated. In this study, the amount of fertilizer contained in the discharged waste liquid was determined. By evaluating this as a price, it was suggested to reduce water treatment costs and recycle fertilizer components. It was evaluated based on the results of major water quality analysis of waste liquid by crop, such as tomatoes, paprika, cucumbers, and strawberries, and in the case of P component, it was analyzed by converting it to the amount of phosphoric acid (P2O5). The amount of nitrogen (N) can be calculated by discharging 1,145.90kg·ha-1 of tomatoes, 920.43kg·ha-1 of paprika, 804.16kg·ha-1 of cucumbers, 405.83kg·ha-1 of strawberries, and the fertilizer content of P2O5 is 830.65kg·ha-1 of paprika, 622.32kg·ha-1 of tomatoes, 477.67kg·ha-1 of cucumbers. In addition, trace elements such as potassium (K), calcium (Ca), magnesium (Mg), iron (Fe), and manganese (Mn) were also analyzed to be emitted. The price per kg of each item calculated by averaging the price of fertilizer sold on the market can be evaluated as KRW, N 860.7, P 2,378.2, K 2,121.7, Ca 981.2, Mg 1,036.3, Fe 126,076.9, Mn 62,322.1, Zn 15,825.0, Cu 31,362.0, B 4,238.0, Mo 149,041.7. The annual fertilizer loss amount for each crop was calculated by comprehensively considering the price per kg calculated based on the market price of fertilizer, the concentration of waste by crop analyzed earlier, and the average annual emission of hydroponic cultivation. As a result of the analysis, the average of the four hydroponic crops was 5,475,361.1 won in fertilizer ingredients, with tomatoes valued at 6,995,622.3 won, paprika valued at 7,384,923.8 won, cucumbers valued at 5,091,607.9 won, and strawberries valued at 2,429,290.6 won. It was expected that if hydroponic drainage is managed through self-treatment or threshing before discharge rather than by leaking it into a river and treating it as a pollutant, it can be a valuable reusable fertilizer ingredient along with reducing water treatment costs.

Incubation Conditions Affecting Bacteriocin Production of Lactobacillus plantarum K11 Isolated from Dongchimi (동치미에서 분리된 Lactobacillus plantarum K11의 박테리오신 생산에 영향을 미치는 배양 조건)

  • The influence of incubation temperature, pH and media components on bacteriocin production by Lactobacillus plantarum K11 were investigated. The highest activity was observed in MRS broth, but no bacteriocin activity was obtained in TSB. The bacteriocin was produced from the exponential growth phase and its activity also reached a maximum in MRS broth, but then dropped after 16 hr because of degradation by extracellular proteolytic enzymes or exhaustion of medium nutrients. The optimal temperature and pH for production of bacteriocin were $37^{\circ}C$ and pH 7.0 in MRS broth, respectively. The addition of 0.5 or 1.0% glucose and $0.5{\sim}1.5%$ lactose to MRS resulted in the increase of the bacteriocin production. With 0.5% NaCl and $K_2HPO_4$, the activities were significantly higher than that of control, respectively. However, increasing nitrogen sources such as beef extract, casein, and tryptone and salts such as $NH_4PO_4$, $MgSO_47H_2O$, and $MnSO_4H_2O$, had detected a negative influence upon the bacteriocin production. Consequently, because the bacteriocin produced by L. plantarum K11 was affected by various incubation conditions, the bacteriocin activity of L. plantarum K11 applied in food as a novel starter will be dependent on environmental factors such as fermentation conditions and food ingredients.

Characteristics of Culture of Oyster Mushroom (Pleurotus ostreatus) on Addition Rate of Acer tegmentosum (산겨릅나무(Acer tegmentosum) 톱밥의 첨가량에 따른 느타리버섯의 재배적 특성)

  • This study was carried out to investigated optimum mixing ratio of Korean natural Acer tegmentosum for production of functional oyster mushroom. Total nitrogen and carbon sources of Acer tegmentosum were 0.2% and 44.4%, respectively and C/N ratio was 234. Total nitrogen source and pH of substrate mixed with Acer tegmentosum were 2.7~2.9 and 4.8~5.0, respectively. The contents of $P_2O_5$, $K_2O$ and MgO at Acer tegmentosum media were higher than that of the control. Mycerial growth was the fastest at Acer tegmentosum 10%, and slower by increase of Acer tegmentosum substrate. Yields of fruiting body was the highest to 159 g/850 mL at 10% of Acer tegmentosum and dimeter and thick of pileus were the highest, too. The L value of pileus and stipes were decreased by increase of Acer tegmentosum substrate, but there was no significant difference in the a-value and the b-value. The contents of $P_2O_5$ and $K_2O$ of fruiting body were increased at Acer tegmentosum substrate, but there was no significant difference in contents of CaO, MgO and $Na_2O$.

Effects of Simulated Acid Rain on the Growth, Nutrient Status of Korean Pine (Pinus koraiensis) Seedlings and Soil Acidification (인공산성우(人工酸性雨)가 잣나무 묘목(苗木)의 생장(生長), 영양상태(營養狀態) 및 토양산성화(土壤酸性化)에 미치는 영향(影響))

  • Three-year-old Pinus koraiensis seedlings, transplanted in brown forest soils originating from granite were treated with simulated acid rain of pH concentrations 5.6(control), 4.0, 3.0, 2.5 and 2.0 for 210 days from April 21 to November 17, 1999. Visible injury of the seedlings were observed at the pH 2.0 and pH 2.5 treatments. The total dry weight of the seedlings decreased at pH 2.0 treatment compared with that of the control, and T/R ratio increased at pH 2.0 treatment compared with others. The elements in each part of the seedlings, concentrations of Ca, P and content of chlorophyll in needles increased at the pH 2.0 treatment compared with the control. The concentration of N in the needles of the seedlings increased as the soil pH decreased. As the treated pH was lowered, soil pH has decreased, and concentrations of Ca, Mg, Al, and Mn increased, especially at pH 4.4. In addition, there was a strong correlation(r=0.90, p<0.05 ; r=-0.94, p<0.01) between the dry weight of the seedlings and the pH and Al concentration of the soils. Therefore, the pH and Al concentration in the soil may be useful indicator for assessing the effect of acid rain on the growth of woody plants.
