  • Since the cultivation of resistance cultivar is essential for organic agriculture, Phytophthora late blight resistance of 16 advanced potato clones obtained from the potato breeding laboratory at the National Institute of Highland Agriculture was evaluated. Resistance of the clone was examined through artificial inoculation in the laboratory and in the field. Dominant isolates of the pathogen in Gangwon province produced abundant sporangia on leaves of most clones showing susceptibility. The number of sporangia ranged from $10^{7{\sim}8}sporangia/ml$ in the susceptible clones at 7 days after inoculation. However, one clone is resistant in organic farming fields. Disease incidence was 2.3% using the resistant clone. The area under disease progress curve(AUDPC) was 75.5. Contrarily, disease incidence of the susceptible variety was 100% and the AUDPC was 1773.5 during the same cultivation period. The resistance clone named as 'Haryeong' is considered suitable for organic potato cultivation in Gangwon province in Korea.

  • Despite the high potency of the retrovirus vector system in gene transfer, one of the main drawbacks of has been difficulty in preparing highly concentrated virus stock. Numerous efforts to boost the virus titer have ended in unsatisfactory results mainly due to fragile property of retrovirus envelope protein. In this study, to overcome this problem, we constructed our own retrovirus vector system producing vector viruses encapsulated with VSV-G (vesicular stomatitis virus G glycoprotein). Concentration process of the virus stock by ultracentrifuge did not sacrifice the virus infectivity, resulting in more than 108 to 109 CFU (colony forming unit) per ml on most of the target cell lines tested. Application of this high-titer retrovirus vector system was tested on chicken embryos. Injection of virus stock beneath the blastoderms of pre-incubated fertilized eggs resulted in chick embryos expressing E. coli LacZ gene with 100% efficiency. Therefore, our results suggest that it is possible to transfer the foreign gene into chicken embryo using our high-titer retrovirus vector.

  • Abstract Griseofulvin has been used as an antifungal antibiotic for the treatment of mycotic diseases of humans and veterinary animals. The purpose of this work was to identify a griseofulvin-producing endophytic fungus from Abies holophylla and evaluate its in vivo antifungal activity against plant pathogenic fungi. Based on nuclear ribosomal ITS1-5.8SITS2 sequence analysis, the fungus was identified and labeled as Xylaria sp. F0010. Two antifungal substances were purified from liquid cultures of Xylaria sp. F0010, and their chemical identities were determined to be griseofulvin and dechlorogriseofulvin through mass and NMR spectral analyses. Compared to dechlorogriseofulvin, griseofulvin showed high in vivo and in vitro antifungal activity, and effectively controlled the development of rice blast (Magnaporthe grisea), rice sheath blight (Corticium sasaki), wheat leaf rust (Puccinia recondita), and barley powdery mildew (Blumeria graminis f. sp. hordei), at doses of 50 to 150 ${\mu}$g/ml, depending on the disease. This is the first report on the production of griseofulvin and dechlorogriseofulvin by Xylaria species.

  • This paper has considered the location estimation problem in two dimension space by using a non-linear filter under non-Gaussian noise in underwater acoustic sensor networks(UASNs). Recently, the extended Kalman filter (EKF) is widely used in location estimation. However, the EKF has a lot of problems in the non-linear system under the non-gaussian noise environment like underwater environment. In this paper, we propose the improved Two-Dimension Particle Filter (TDPF) using the re-interpretation distribution techniques based on the maximum likelihood (ML). Through the simulation, we compared and analyzed the proposed TDPF with the EKF under the non-Gaussian underwater sensor networks. Finally, we determined that the TDPF's result shows more accurate localization than EKF's result.

  • In this paper, the capacity of the clustered response model for correlated MIMO-OFDM fading channel is investigated. We compare the capacities achieved by correlation at receiver and achieved by correlation at both the transmitter and the receiver for the case where the channel is known and unknown at the transmitter are considered. It is found that the capacity achieved by correlation at receiver is better than the other. It is also shown that the capacity using the water-filling methed is larger than that using the uniform power allocation due to the water-filling gain. But it is negligible when the number of clusters is over the maximum rank of the sum correlation matrix and SNR is high.

  • family (CMTM) is a novel family of proteins linking classical chemokines and the transmembrane 4 superfamily (TM4SF). Our earlier studies indicated several CMTM members (such as CKLF1 and CMTM2) have a secreted form. This is the first report of the secreted form of CMTM5-v1, the major RNA splicing form of CMTM5, which is produced as small vesicles (<100 nm diameter) and floats at a peak density of 1.19 g/ml on continuous sucrose gradients. CMTM5-v1 has no obvious co-localization with CD63 or Golgi complex. In addition, brefeldin A but not wortmannin can inhibit the secretion of CMTM5-v1. Our results suggest that CMTM5-v1 might be secreted via a different vesicle-mediated secretory pathway, which will be helpful for the studies of vesicle-mediated secretion and MARVEL domain-containing proteins.

  • The purpose of this study was to observe the effects of the polysaccharide(G009) obtained from liquid cultured Ganoderma lucidum IY009 on the lipidperoxidation in rat liver microsome. It is well known that the polysaccharide of G. lucidum have the hepatoprotective activity, antitumor activity etc., which was thought to have the relationship to anti-lipidperoxidation. In order to the estimate the effects of anti-lipidperoxidation of the polysaccharide obtained from G. lucidum IY009, enzymatic and nonenzymatic reaction were performed, in vitro, in rat liver microsome. In enzymatic lipid peroxidation reaction by ADP/FeCl$_3$/NADPH and $CCl_4$/NADPH, G009(1mg/ml) inhibited 77.4%, 39.4%, respectively, and the nonenzymatic reaction strongly exhibited 97.4% inhibition. And also, in enzymatic and nonenzymatic inducers treated with G009, the formation of MDA was progressively greater decreased by raising G009 concentration. These results suggest that anti-lipidperoxidation by G009 treatment may be play an important part in liver protection action.

  • To understand effect of inoculum size, cell density, sucrose concentration and concentrations of MS basal on suspension culture and protoplast isolation of wild viola(Viola patrinii DC.) callus from petiole segments this experiment was conducted. In the lot of 30 mesh inoculum size, two observations were; One was that a considerable increase in the fresh and dry weight of callus was determined. Another was that the callus mass was relatively compact compared with others. A recommendable cell density was 0.4g for 20ml culture medium and the higher sucrose concentration, the higher fresh and dry weight were obtained. The dilution of MS basal salt was differently affected on fresh and dry weight; the highest fresh weight was found in 1x MS salt, while the higest dry weight was in 1/3x dilution. The addition of casein hydrolysate(3g/L) was more effective to increase of both fresh and dry weight. THe contents of protein was great in the inoculum lots with larger inoculum sizeand higher concentration of MS basal salts contenting 3g/L of casein hydrolysate and higher sucrose compared with others. The greatest protoplasts were isolated from the lot of 10 mesh size treated with 1%pectinase SE-150 and 2% cellulase YC. In general, for optimal protoplast isolation the followingconditions were recommended; 1) Use of smaller cell size cultured for 2-5 weeks, and 2) more than 5 hours incubation using the combined mixture of the enzymes with proper concentrations.

  • Objectives : This study investigated whether the water extract of Spatholobi Caulis (SCE) has the ability to protect hepatocyte against oxidative stress induced by tert-butylhydroperoxide (tBHP) in vitro and $CCl_4$ in vivo. Methods : In vitro, HepG2 cells pre-treated with Spatholobi Caulis water extract (1, 3, 10, $30{\mu}g$/ml) for 12h and further incubated with tBHP ($100{\mu}M$) for the next 12h. Cell viability was assessed by MTT assay. In vivo, rats were orally administrated with the aqueous extract of Spatholobi Caulis (SCE; 50, 100 mg/kg) for 4 days and then, injected with $CCl_4$ 1 mg/kg body weight to induce acute liver damage. Results : Treatment with SCE inhibited cell death induced by tBHP, as evidenced by alterations in the levels of the proteins associated with apoptosis:SCE prevented a decrease in $Bcl_2$, and cleavage of poly(ADP-ribose)polymerase and pro-caspase-3. Moreover, SCE inhibited the ability of tBHP to generate $H_2O_2$ production, thereby restoring GSH content. Moreover, SCE treatments in rats effectively decreased liver injuries induced by a single dose of $CCl_4$, as evidenced by decreases in hepatic degeneration and inflammation as well as plasma alanine aminotransferase and lactate dehydrogenase activities. Consistently, treatments of SCE also protected liver in rats stimulated by $CCl_4$, as indicated by restoration GSH and prevention of MDA in the liver. Conclusions : SCE has the ability 1) to protect hepatocyte against oxidative stress induced by tBHP and 2) to prevent $CCl_4$-inducible acute liver toxicity. Present findings may be informative not only in elucidating the pharmacological mechanism of Spatholobi Caulis, but in determining its potential application for oxidative cellular damage in the liver.
