Analysis of Nitrosation Inhibition and Antioxidant Effect by Phyto-Extract Mixture (식물추출 혼합제재인 phyto-extract mixture의 니트로세이션 억제능과 항산화능 분석)

  • The most representative nitrosamine derived from nicotine, nitrosamine-4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone(NNK), has been reported to cause lung cancer in A/J mice. It has been also demonstrated that NNK-induced lung tumorigenesis involves $O^6-methylguanine(O^6MeG)$ formation, leading to $GC\;{\rightarrow}\;AT$ transitional mispairing during DNA replication. Our in vitro experiment, modified from the method of DBA assay, examined the ability of phyto-extract mixture to inhibit the metabolism of nicotine to nitrosamines. The production of nitromorpholine from morpholine was inhibited about 75% at the concentration of 20 mg/mL of phyto-extract mixture, which was lower than vitamine C and green tea powder. NNK, which is a pro-carcinogen in laboratory animals, is hydroxylated primarily in liver and lung by CYP 1A2, 2A6 and 3A4. A critical phase. of NNK activation is its change to an unstable metabolite methyl-diazohydroxide via CYP-mediated ${\alpha}-hydroxylation$; and then it provides a methyl group to the DNA to form DNA adducts which can easily induce mutations. $Aroclor^R$ 1254 was used to induce CYPs in the liver of a Sprague-Dawley rat. The ability of various test samples to inhibit CYPs that participate in NNK activation was evaluated, following the removal of the liver from the rat. Microsomal CYPlA2 catalyzing the conversion of NNK into strong carcinogenic chemicals was inhibited more efficiently by phyto-extract mixture than green tea powder. These results indicate that phyto-extract mixture can be used to reduce $O^6MeG$ DNA adducts for chemoprevention.

  • The effect of garlic extract and vitamin C mixture on the various cancer cell lines in vitro and in vivo have been examined. Proliferation of human colon cancer (HT-29), human rectal cancer (HRT-18) and human hepatoma (HepG2) cells was inhibited by garlic extract and vitamin C, respectively. Based on the cytotoxic activity, mixture of garlic extract and vitamin C was demonstrated to possess a synergistic growth inhibition on HT-29, HRT-18 and HepG2 cancer cells. Mixture of garlic extract and vitamin C significantly arrested G2/M phase cells in the HepG2 cell cycle. Oral administration of mixture of garlic extract and vitamin C to sarcoma-180 tumor-bearing mice prolonged survival time compared to that of control group. These results suggested that addition of vitamin C enhances anticancer activity of garlic extract in vitro, and mixture of garlic extract and vitamin C has antitumor effect in vivo.

  • The effects of extract mixture of red ginseng and steamed Rehmaniae radix prepared for antidiabetic activity was examined in streptozotocin-Induced diabetic rats. The increased blood glucosec level in the streptozotocin-induced diabetic rats was significantly decreased by the treatment with the mixture (800, 1600 mg/kg, p.o.). However, neither red ginseng extract nor steamed Rehmaniae radix extract alone showed significant hypoglycemic effects. The mixture prevented a weight loss in streptozotocin-induced diabetic rats. These results suggest that the mixture has the relieving action against streptozotocin-induced hyperglycemia.

  • The purpose of this study was performed to evaluate antioxidant activity of the Artemisia asiatica Nakai and Moringa oleifera Lam mixture extract. Mixture extracts were manufactured by concentration and compared with a single extract (only the Artemisia asiatica Nakai mixture and only the Moringa oleifera Lam mixture). The experiments conducted Total polyphenol measurements, Total flavonoid measurements, DPPH radical scavenging activty, ABTS radical scavenging activty and LDH assay. The LDH assay assessment shows that all extracts are cells compared to controls. The toxicity was weak. Finally, The antioxidant capacity was rated higher than mixture extract of a single extract. Also, the optimized mixture was determined AM5 (Artemisia asiatica Nakai mixture: Moringa oleifera Lam mixture = 3:1). For extracts of AM5, Total phenol and flavonoid contents were 271.769±18.087 mg/g and 45.384±5.026 mg/g. and DPPH and ABTS scavenging activity were 70.8±6.496% and 77.1±9.634%. Therefore, it is expected that the value of the extract will increase as it increases its antioxidant activity if it is manufactured according to the appropriate ratio.

A Study on the Anti-microbacterial Activity, Anti-inflammatory and Anti-allergic Effects of mixture of Rheum coreanum Scutellaria baikalensis Phellodenron amurense (대황(大黃), 황금(黃芩), 황백(黃柏) 복합 추출물의 항균, 항염 및 항알레르기 효과에 대한 실험적 연구)

  • Herbal mixture water extract of (Rheum coreanum Scutellaria baikalensis, Phellodendron amurese), which exhibit several beneficial effects including acne and skin diseases, was tested for anti-microbial activity and anti-inflammation effects. The herbal mixture extract showed antimicrobial activity against Stapylococcus epidermis and Propionbacterium acne. The growth of Stapylococcus epidermis and Propionibacterium acne was inhibited completely by addition of 1.0% of the extract. Also in the present study we examined the mixture extract on compound 48/80 induced allergy and LPS induced cyclooxygenase-2(COX-2) gene expression in RAW264.7 macrophage. The results indicated the ear swelling and histamine release induced by compound 48/80 were dose-dependently reduced, ranging 18-36% and 10-61%, respectively. Furthermore the extract inhibited the expression of LPS-induced COX-2 proteins and mRNAs without an appreciable cytotoxic effects on RAW264.1 cells. The cytotoxicity of the extract using MTT assay showed the cytotoxicity of 6% and 13% against L929 cell line. Based on these results, it is concluded that the herbal mixture water extract can be applied to this acne and skin diseases therapy.

Anti-oxidant and Anti-proliferative Effects of Water Extract Mixture of Cordyceps Militaris and Allium Tuberosum (동충하초 및 부추 혼합 물추출물의 항산화 및 암세포 증식억제 효과)

  • The present study was performed to evaluate the anti-oxidant and anti-proliferating activity of the water extract mixture of Cordyceps militaris (CM) and Allium tuberosum (AT). The water extract mixture rate of CM and AT was optimized by means of a sensory evaluation test. The optimized mixture rate were decided at 70% of CM, 30% of AT, and 10% of apple concentrate as an additive. The values of total acidity, pH, sugar contents, and turbidity of the water extract mixture were 0.1%, 4.28, 9.10 °Brix, and 1.48 respectively. The water extract mixture had effective DPPH radical scavenging activity, reducing power effect, and ABTS radical activity. DPPH radical activities of the water extract and the water extract mixture were 43.2% and 51.7% respectively; their reducing power (OD700) was 1.14 and 1.43 respectively; and ABTS.+ radical activities were 47.1% and 62.2% respectively. Also, the water extract mixture showed a higher anti-proliferating effect than the AT extract on human prostate cancer cells. These results provided experimental evidence that the water extract mixture of CM and AT is a better source of anti-oxidant and anti-cancer ingredients than a single extract of CM. In conclusion, the water extract mixture of CM and AT will be beneficial in development of a functional drink.

Cytotoxicity of Garlic and Onion Methanol Extract on Human Lung Cancer Cell Lines (인체 폐암세포주에 대한 마늘과 양파 메틴올추출물의 세포독성)

  • This study was designed to investigate the cytotoxic effect of methanol extract of garlic, onion and those mixture on two kinds of human lung cancer cell lines (NCI-H522, NCI-H596) using MTT assay. MeOH extract of garlic, onion and those mixture showed cytotoxic effect on both NCI_H522 and NCI-H596. The growth of the cancer cells exposed to medium containing garlic, onion extracts and those mixture was inhibited dose-dependently. The growth of NCI-H522 was inhibited more in the garlic extract than in the onion extract, but that of HCI-H596 was inhibited highese in the onion extract. IC50 values of garlic extract on NCI-H522 and NCI-H596 were 0.84 mg/mL, 0.88 mg/mL and those of onion extract were 1.04 and 0.79, respectively. and the mixture of garlic and onion extracts also inhibited the growth of both NCI-H522 and NCI-H596 cells.

  • In this study, antioxidant and anti-inflammatory of fermented Houttuynia cordata Thunb and scoria mixture extract were investigated in vitro. Radical-scavenging activities of the ethanol extracts were examined by using ${\alpha},{\alpha}$-diphenyl-${\beta}$-picrylhydrazyl (DPPH) and 2,2-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid (ABTS)) radicals assay. Consequently, we confirmed that fermented Houttuynia cordata Thunb and scoria mixture extract dependent removed DPPH and ABTS radical. Also, to confirm anti-inflammatory activity of ethanol extract, we treated fermented Houttuynia cordata Thunb and scoria mixture extract on BV-2 cell with LPS. The result showed that fermented Houttuynia cordata Thunb and scoria mixture extract concentration-dependent inhibited NO production. Therefore, fermented Houttuynia cordata Thunb and scoria mixture extract showed inhibition radical oxygen activities and inflammatory and have available for a pharmacological composition on neuritis-protection and treatment.

Tissue regenerative activity of Magnolia and Zizyphi fructus extract mixtures (후박 및 대조추출혼합물이 골조직 재생에 미치는 영향)

  • The purpose of this study was to perform on the biological activity of Magnolia and Zizyphi fructus extract mixtures on the wound healing of defected rat calvaria. For the determination of the mixture ratio of two extracts for oral administration, preliminary experiments were performed with the mixture combination of 2000 and $3000{\mu}g/ml$ of Magnolia extract, and also 20, 30, 200, 300, 2000 and $3000{\mu}g/ml$ of Zizyphi fructus extract, respectively and divided into 6 groups. The combination of extracts mixture were tested on the enhancing effect of cellular activity. The effect of the extracts mixture on the cellular activity was evaluated using MTT method and measured on the results with optical density by ELISA reader. The ability to tissue regeneration of the extracts mixture was performed by measuring new bone and new connective tissue regeneration on the 5mm defected rat calvaria for 1, 2 and 3 weeks after oral administration of 2 different dosages groups : 10:1(0.1g/kg) and 10:1(0.5g/kg). It was employed the same dosages of unsaponifiable fraction of Zea Mays L as positive controls. Each group of rat was sacrificed and en bloc section for histological examination. The effect on the cellular activity of each mixture ratio showed significantly higher in $2000{\mu}g/ml$ of Magnolia extract and $200{\mu}g/ml$ of Zizyphi fructus extract group to compare with other groups. These preliminary results showed that appropriate mixture ratio of two extracts was 10:1 of Magnolia and Zizyphi fructus extract. Histological examination on the activity of tissue regeneration of each group showed that 2weeks and 3weeks specimens of 0.5g/kg of 10:1 extract mixture of Magnolia and Ziziphi fructus administrated rat calvaria revealed significantly more osteoid and new bone formation of defected calvaria with unification of defected area than the specimens of any other negative and positive controls. Even though the specimen administrated the same dosages of unsaponifiable fraction of Zea Mays L, positive controls, showed the trend that they promote significantly the repair of calvarial defect, their bone reparative activities were less inductive than the same dosages of Magnolia and Ziziphi fructus extract mixture. These results implicated that the mixture of Magnolia and Zizyphi fructus extracts should be highly effective on the wound healing of bony defected site and might have potential possibilities as an useful drug to promote periodontal tissue regeneration.

  • The purpose of this study is to investigate vasodilatation effect of complex saponin separated from Scrophulariae radix, Asparagus cochinchinensis and Liriope platyphylla mixture extract on rabbit carotid artery. In this study, to determine vasodilatation effect of complex saponin separated from Scrophulariae radix, Asparagus cochinchinensis and Liriope platyphylla mixture extract on rabbit carotid artery, arterial rings with intact or damaged endothelium were used for experiment using organ bath, and were contracted by endothelin. complex saponin, major active constituents of Scrophulariae radix, Asparagus cochinchinensis and Liriope platyphylla mixture extract, showed a moderate vasodilatation effect on the basilar arteries of rabbits. Therefore, treatment with complex saponin separated from Scrophulariae radix, Asparagus cochinchinensis and Liriope platyphylla mixture extract may selectively accelerate cerebral blood flow through dilatation of the basilar artery. Theseis result suggest a potential role of complex saponin separated from Scrophulariae radix, Asparagus cochinchinensis and Liriope platyphylla mixture extract as source of vasodilatation agent.
