Isolation of Cholesterol-Lowering Lactic Acid Bacteria from Pig and Human Feces

  • Elevated level of serum cholesterol in humans is a risk factor correlated with the development of coronary heart disease. We have screened lactic acid bacteria from pig and human feces for the development of probiotics which have an anti-cholesterol effect. We have used special media to isolate only lactic acid bacteria and they were subjected to the experiments such as oxgal test, carbohydrate fermentation test. Results from the acid tolerance test and growth test in the presence of oxgal demonstrated that some strains would likely survive in thuman stomach, where acidity is high, and in small intestine, where bile fluid is present. In conclusion, we were able to screen lactic acid bacteria which were tolerant against bile acid and supposed to be prominent to lower the cholesterol level in human serum.

결장암 예방에 대한 유산균의 기능 (The Functions of Lactic Acid Bacteria in Colon Cancer Prevention)

  • Certain lactic acid bacteria have anti-tumor activity, especially colon cancer. The fermented milk products containing that kind of lactic acid bacteria have to be recommended for human health as excellent health functional foods. This paper have been classified by 5 regions on the functions of lactic acid bacteria related to prevention of colon cancer. 1) Enhancing of host's immune response; Production of cytokines. 2) Binding and degradation of potential carcinogens; Binding and degradation of mutagenicity. 3) The changes of intestinal microflora and production of antitumorigenic or antimutagenic compounds; Production of azoxymethane. 4) Alteration of the metabolic activity of intestinal microflora; Decrease of harmful enzymes in intestinal tract. 5) Alteration of physicochemical conditions in the colon; Decrease of pH and bile acids contents.

Biotransformation of Ginseng Extract to Cytotoxic Compound K and Ginsenoside $Rh_2$ by Human Intestinal Bacteria

  • When saponin extracts of dried ginseng and red ginseng were anaerobically incubated with human intestinal microflora, these extracts were metabolized to compound K and ginsenoside $Rh_2$, respectively. However, when these extracts were incubated with commercial lactic acid bacteria, these did not metabolize these ginsenosides to compound K or ginsenoside $Rh_2$. Among some intestinal bacteria isolated from human feces, Bacteroides C-35 and C-36 transformed these saponin extracts to compound K and ginsenoside $Rh_2$, respectively. These bacteria also transformed water extracts of dried ginseng and red ginseng to compound K and ginsenoside $Rh_2$, respectively, similarly with that of the saponin extracts. Among transformed ginsenosides, compound K and 20(S)-ginsenoside $Rh_2$ exhibited the most potent cyotoxicity against tumor cells.

Detailed Analysis on the Toxic Effect of Bisphenol A to the Liver and Testis in the Rat

  • Environmental endocrine disruptors are very toxic to the animals including humans. They are hormone-like acting chemicals which can be found in our normal daily life. We have examined the toxic effect of bisphenol A and if lactic acid bacteria could suppress this toxic effect. Thirty rats were divided into three groups (control, bisphenol A treated, bisphenol A and lactic acid bacteria treated). Treatments were carried out at an interval of 12 hours for each group. Control group showed normal and clear morphology of tissues. Cells were fine in their shape and color, and density was high enough for the normal function. However, bisphenol A treated group was abnormally destructed in cell morphology. In the testis, sperms were totally destructed. When treated with lactic acid bacteria together, the toxic effect of bisphenol A was clearly decreased. This study indicated that bisphenol A was toxic in any concentration especially for the liver and testis, however, lactic acid bacteria could suppress the toxic effects of bisphenol A.

  • Recently there have been extensive research activities on the development of on-the-spot detection technologies for bacteria from foods due to growing high demand for food safety. In particular, on-the-spot detection devices using biosensors with rapid, highly sensitive and multiplexed sensing capability are promising for portable or mobile applications. Firstly, issues related to on-the-spot bacteria detection are discussed. Then, detection methods for bacteria, types of biosensors depending on transducing principle and receptors, and platforms for integration of biosensors and signal readers are reviewed. Finally, prospects for development of on-the-spot detection devices are summarized.

  • Geniposide, an iridoid glucoside, has been isolated from the butanol fraction of Gardenia jasminoides Ellis (Rubiaceae). The component was found to be transformed into the blue pigments by some aerobic bacteria, suggesting that geniposide is the precursor for the formation of pigments after converting into genipin, an aglycon of geniposide, by ${\beta}-glucosidase$. Some bacteria having a ${\beta}-glucosidase$ activity did not form the pigments, which may mean that the formation of pigments can only be occurred by the reaction of any enzyme or compound in the pigment-producing bacteria.

Characterization of Bacterial Community Dynamics during the Decomposition of Pig Carcasses in Simulated Soil Burial and Composting Systems

  • Soil burial is the most widely used disposal method for infected pig carcasses, but composting has gained attention as an alternative disposal method because pig carcasses can be decomposed rapidly and safely by composting. To understand the pig carcass decomposition process in soil burial and by composting, pilot-scale test systems that simulated soil burial and composting were designed and constructed in the field. The envelope material samples were collected using special sampling devices without disturbance, and bacterial community dynamics were analyzed by high-throughput pyrosequencing for 340 days. Based on the odor gas intensity profiles, it was estimated that the active and advanced decay stages were reached earlier by composting than by soil burial. The dominant bacterial communities in the soil were aerobic and/or facultatively anaerobic gram-negative bacteria such as Pseudomonas, Gelidibacter, Mucilaginibacter, and Brevundimonas. However, the dominant bacteria in the composting system were anaerobic, thermophilic, endospore-forming, and/or halophilic gram-positive bacteria such as Pelotomaculum, Lentibacillus, Clostridium, and Caldicoprobacter. Different dominant bacteria played important roles in the decomposition of pig carcasses in the soil and compost. This study provides useful comparative date for the degradation of pig carcasses in the soil burial and composting systems.

Evaluation of Lactic Acid Bacteria for the Resistance to Endocrine Disruptors

  • Endocrine disruptors are chemicals which can be found in our normal daily life. They can be easily ingested through plastic food containers, pesticides, etc. They include DDT, bisphenol A, benzophenone and phenylphenol, etc. Endocrine disruptor can be very harmful and toxic because it disrupts the normal function of the endogenous endocrine system. It has been reported that endocrine disruptor can cause the fatal strike in reproductive system central nervous system and the other part of the body. We have examined if the growth of lactic acid bacteria could be resistant to the endocrine disruptor. We have used Lactobacillus delbruekii as an experimental strain and benzophenone and phenylphenol for the comparison purpose. Experiments included the evaluation of turbidity, absorbance and actual cell counts. Although Lactobacillus delbruekii showed the higher resistance to benzophenone than phenylphenol it was still resistant to both benzophenone and phenylphenol. Because the experimental concentrations of benzophenone and phenylphenol were so high to compare with the actual concentration we meet in daily life, Lactobacillus delbruekii was considered to be sufficient to survive in the environmental concentration of these endocrine disruptors. This study should contribute to the development of fermented beverage with beneficial effect by lactic acid bacteria.

군산인근 해역에서 해양세균의 균체수와 Biovolume (Determination of Marine Bacterial Number and Biovolume in the Intertidal Zone of the Yellow Sea near Kunsan, Korea)

  • As a baseline survey to clarify the structure and function of bacterial populations in the intertidal waters of the Yellow Sea near Kunsan in 1987/1988, annual variation of bacterial number and biovolume was estimated by AODC method. Rod-shaped bacteria were more than 74% of all cells during the investigation period. Average biovolume of sampled bacteria ranged from ${3.19}\pm0.59*10^{-2}$ to $6.19\pm0.76*10^{-2}$ .$\mu$$m^{3}$ for coccoid bacteria, and from 4.57.+-.0. $17*10^{-2}$ to $12.94\pm0.21*10^{-2}$ .$\mu$$m^{3}$ for rod-shaped ones. It showed that larger biovolume occurred at surface in case of coccoid bacteria, while at bottom in case of rod-shaped ones. There was little correlation between population density and biovolume. Diameters of edlls were below 0.5$\mu$m among 77-89% of all measured coccoid bacteria, and lengthes of cells more than 70% of all measured rod-shaped ones were below $1\mu$m. It explains the high percentage of "mini-bacteria" was distributed in the investigation area.tion area.
