Industrial Application for Aquaculture of Fermented Soybean with Bacillus polyfermenticus SCD (양식업에서 바실러스 폴리퍼멘티쿠스 콩발효물의 상업적 응용)

  • Bacillus polyfermenticus SCD (B. polyfermenticus SCD) has been appropriately used for the therapy of long-term intestinal disorders, because live strains in the form of active endospores can successfully reach the target intestine in humans. B. polyfermenticus SCD produces the most antibiotic-related materials. In the present study, B. polyfermenticus SCD was fermented with soybeans (BPFS) and its probiotic properties were investigated. B. polyfermenticus SCD and BPFS showed a broad spectrum of antimicrobial activity against pathogenic Gram-positive (Streptococcus parauberis, Streptococcus iniae, Lactococcus garviae) and Gram-negative (Flexibacter tractuosus, Vibrio harveyi, Vibrio vulnificus, Vibrio ordalii) bacteria and moulds (Aspergillus niger, Aspergillus oryzae). Sebastes schlegeli were used to examine survival rate and cleanup action by BPFS. Bacterial infection resulted in a mortality of up to 99% in the commercial fodder fed groups. BPFS both enhanced the growth rate of fry by improving their appetite and had cleaned by decreased eutrophication. Therefore, it seems appropriate that BPFS should be developed as an antibiotic replacement, favorable fodder additive, and antifungal material in fish farming systems.

Effects of Dietary Supplementation with Garlic Extract on Immune Responses and Diseases Resistance of Olive Flounder, Paralichthys olivaceus (사료 내 마늘 추출액 첨가가 넙치(Paralichthys olivaceus)의 비특이적 면역반응 및 질병저항성에 미치는 영향)

  • In this study, we investigated the effects of dietary 1% garlic extract on nonspecific immune responses and fish diseases (Viral Hemorrhagic Septicemia Virus, Vibrio anguillarum, Streptococcus iniae and Edwardsiella tarda) resistance in olive flounder Paralichthys olivaceus. Fish were fed a commercial diets supplemented with 1% garlic extract for 4 weeks. After the 4 weeks feeding experiment, the artificial infection was made by V. anguillarum, S. iniae, E. tarda and VHSV. And the cumulative mortality was monitored for 2 weeks after artificial infection. The cumulative mortalities decreased in all experiments except for group of E. tarda compared to control group. We observed significantly higher levels of the hematocrit, glucose, total protein, lysozyme activity and the macrophage activity in the experimental group compared to the control group. In the experiments of drug sensitivity and MIC using the three bacteria (V. anguillarum, S. iniae and E. tarda), 1% garlic extract was more effective than the previously reported fermented garlic powder. These results suggested that garlic extract can increase the disease resistance of olive flounder against V. anguillarum, S. iniae and VHSV and the ability of nonspecific immune responses.

Influence of Perilla frutescens var. acuta Water Extract on the Shelf Life and Physicochemical Qualities of Cooked Beef Patties

  • This study investigated the effects of Perilla frutescens var. acuta water extract (WEP) on the shelf life and physicochemical qualities of cooked beef patties. The WEP contained phenolic compounds (80.65 mg gallic acid equivalents/g) and had half-maximal effective concentrations of 0.437 and 4.509 mg/mL for scavenging of DPPH and ABTS+ radicals, respectively. Treatment with 0.6% WEP inhibited the growth of Escherichia coli O157:H7 (p<0.05). Based on the result of the antioxidative potential and antimicrobial potential of WEP, beef patties were prepared with three treatment groups: (1) beef patties without added antioxidant (control); (2) beef patties with 0.02% ʟ-ascorbic acid (BAA); and (3) beef patties with 0.6% WEP (BWEP). The pH and cooking loss of BWEP were lower and higher than those in the control, respectively (p<0.05). When cooked beef patties were stored for 21 d at 4℃, the total number of aerobic bacteria in BWEP was lower than those in the control on all days except day 14 (p<0.05). The TBARS values in BWEP were lower than those of controls on days 7, 14, and 21 (p<0.05). Compared to control and BAA, BWEP had lower L* and b* values and higher a* values throughout the storage period (p<0.05). Except on day 0, acceptability was higher in BWEP than in control and BAA (p<0.05). According to results, WEP can be used as a natural ingredient that improves the shelf life and sensorial qualities of meat products.

  • Addition of starch syrup, table sugar, potato powder, skim milk powder an parched soybean powder to melon flesh was suitable as fermented melon base. The manufacturing process of fermented melon was as follows: Pasteurization for 10 min at $95^{\circ}C$, use of 1% starter culture, fermentation for 12 hours at $35^{\circ}C$ and ripening for 3 days at $8^{\circ}C$. The growth and acid production of Pediococcus acidilactici among several starter cultures were most active for the first 12 hours, but such activities were disappeared during ripening. In the case of Lactobacillus plantarum, the activities were not high during fermentation, which, however, increased during ripening. Throughout the whole manufacturing process, the fermented melon with a mixed culture of P. acidilactici and L. plantarum showed more cell number of each bacterium and higher titratable acidity than that with single cultures. Also P. acidilactici surpressed the growth of L. plantarum during ripening.

Effects of Pyroligneous Acid on Control of Large Patch in Zoysiagrass (목초액을 이용한 한국잔디 Large Patch병 방제효과)

  • This study was carried out to investigate control efficacy of fungicides mixed with pyroligneous acid on Large patch caused by Rhizoctonia solani AG2-2. R. solani AG2-2 was not inhibited by concentration of 500 times and 1,000 times diluents of pyroligneous acid. It is thought that R. solani AG2-2 was not inhibited by Pyroligneous acid alone. Pencycuron and tebuconazol mixed with pyroligneous acid had a good effect in prevention of R. solani AG2-2 in zoysiagrass. Concurrently, it was observed that those were reduced $50\%$ of usage of fungicides. 500 times and 1,000 times diluents of pyroligneous acid treated group had a significantly effect compared to control turf in the leaf length, leaf width and dry weight. In conclusion, usage of fungicides could be reduced about $50\%$ by a fungicides mixed with pyroligneous acid for protection of R. solani AG2-2. Therefore, It is thought that fungicides mixed with pyroligneous acids can be reduced an economic load and also an environmental pollution such as a tolerance to pathogenic bacteria and a destruction of an ecological system of soil microbes according to use fungicides.

  • Components of extracellular matrix and the matrix-degrading enzymes are some of the key regulators of tumor metastasis and angiogenesis. Hyaluronic acid (HA), a matrix glycosaminoglycan, is known to promote tumor adhesion and migration, and its small fragments are angiogenic. Until now, we have compared levels of hyaluronidase, an enzyme that degrade HA, in normal adult prostate, benign prostate hyperplasia and prostate cancer tissues and in conditioned media from epithelial explant cultures, using a substrate (HA)-gel assay and ELISA-like assay (Kim et al., unpublished results). The present review described an overall characterization of hyaluronidases and its application to human diseases. The hyaluronidases are a family of enzymes that have, until recently, deed thorough explication. The substrate for these enzymes, hyaluronan, is becoming increasingly important, recognized now as a major participant in basic processes such as cell motility, wound healing, embryogenesis, and implicated in cancer progression. And in those lower life forms that torment human beings, hyaluronidase is associated with mechanisms of entry and spread, e.g. as a virulence factor for bacteria, for tissue dissection in gas gangrene, as a means of treponema spread in syphilis, and for penetration of skin and gut by nematode parasites. Hyaluronidase also comprises a component of the venom of a wide variety of organisms, including bees, wasps, hornets, spiders, scorpions, sh, snakes and lizards. Of particular interest is the homology between some of these venom hyaluronidases and the enzyme found in the plasma membrane of mammalian spermatozoa, attesting to the ancient nature of the conserved sequence, a 36% identity in a 300 amino acid stretch of the enzyme protein. Clearly, hyaluronidase is of biological interest, being involved in the pathophysiology of so many important' human disorders. Greater effort should be made in studying this family of enzymes that have, until recently, been overlooked. Also, oriental medical application of the hyaluronidase will be discussed with respect to inhibition and suppression of inflammation and malignacy.

  • The composting is a biological process that converts organic matter into useful resources such as fertilizers. It is a continuous transition of microbial communities to adapt changes in organic matter and environmental conditions (carbonation rate, temperature, humidity, oxygen supply, pH, etc.). Most of the composting plants are located in the proximity of the residential areas. It is a general scenario where government authorities receive complaints from the local residents due to release of odor from the composting, and has become a social problem in Korea. Identification of dominant microorganisms, understanding change in microbial communities and augmentation of specific microorganism for composting is vital to enhance the efficiency of composting, quality of the compost produced, and reduction of odor. In this paper, we suggest the optimum operation conditions and methods for compost depot to reduce odor generation. The selection of the appropriate microorganisms and their rapid increase in population are effective to promote composting. The optimal growth conditions of bacteria such as aeration (oxygen), temperature, and humidity were standardized to maximize composting through microbial degradation. The use of porous minerals and moisture control has significantly improved odor removal. Recent technologies to reduce odor from the composting environment and improved composting processes are also presented.

Molecular Analysis of Microbial Community in Soils Cultivating Bt Chinese Cabbage (분자생물학적 분석을 통한 Bt 배추의 토양미생물상 영향 비교평가)

  • The aim of this study was to investigate the possible impact of Bt Chinese cabbage on the soil microbial community. Microbial communities were isolated from the rhizosphere of one Bt Chinese cabbage variety and four varieties of conventional ones and were subjected to be analyzed using both culture-dependent and molecular methods. The total counts of bacteria, fungi, and actinomycetes in the rhizosphere of transgenic and conventional Chinese cabbages were observed to have an insignificant difference. Denaturing gradient gel electrophoresis (DGGE) analysis of PCR-amplified 16S rRNA genes revealed that the bacterial community structures were very similar to each other and this genetic stability of microbial communities was maintained throughout the culture periods. Analysis of dominant isolates in the rhizosphere of transgenic and conventional Chinese cabbages showed that the dominant isolates from the soil of transgenic Chinese cabbage belonged to the Bacilli and Alphaproteobacteria, while the dominant isolates from the soil of conventional cabbage belonged to the Holophagae and Planctomycetacia, respectively. These results indicate that the Bt transgenic cabbage has no significant impact on the soil microbial communities.

  • Methicillin-Resistant Staphylococcus aureus(MRSA) is a multidrug-resistant(MDR) strain. (+)-Usnic acid(UA) is uniquely found in lichens, and is especially abundant in genera such as Usnea and Cladonia. UA has antimicrobial activity against human and plant pathogens. Therefore, UA may be a good antibacterial drug candidate for clinical development. In search of a natural products capable of inhibiting this multidrug-resistant bacteria, we have investigated the antimicrobial activity of UA against 17 different strains of the bacterium. In this study, the effects of a combination of UA and permeable agents against MRSA were investigated. For the measurement of cell wall permeability, UA with concentration of Ethylenediaminetetraacetic acid(EDTA) was used. In the other hand, Sodium azide($NaN_3$) was used as inhibitors of ATPase. Against the 17 strains, the minimum inhibitory concentrations(MICs) of UA were in the range of $7.81-31.25{\mu}g/ml$. EDTA or $NaN_3$ cooperation against MRSA showed synergistic activity on cell wall. UA and in combination with EDTA and $NaN_3$ could lead to the development of new combination antibiotics against MRSA infection.

A Study of the correlation between breeding density and the azimuth of cattle sheds affected calf diarrhea disease (송아지 설사에 영향을 미치는 사육밀도 및 축사 방위와의 상관관계에 관한 연구)

  • This study was conducted to determine the effective environmental control measures in preventing calf diarrheal disease in farms. It was found that the high numbers of calves in a cow house was directly related with the high incidence rate of diarrheal diseases. When the number of calves was increased in a cow house, it was difficult to reduce the diarrheal diseases of calves. It was revealed that the incidence rates of calf diarrheal disease were dramatically reduced when the delivery house which faced south. The periodical changes of bedding materials seemed beneficial good to control the possible resident infectious agents, including pathogenic viruses and bacteria. The control of the delivery time was important for reduction of calf diarrhea. It was revealed that September and October were the best time as the delivery period. In order to use old cow house as a delivery house, it should be sterilized by the disinfectant and the flame. When the diarrheal feces were removed as soon as possible, the transmission of diarrheal diseases among calves In the same house was reduced. When the diarrheal feces were not disposed, the incidence rate of diarrhea reached up to 50%. When the morbid calves were not quarantined, the incidence rate of diarrhea was 100%. In contrast, when the diseased calves were quarantined from other calves, the incidence rate of diarrhea was reduced, remarkably. The mean recovery time from the diarrhea of the treated calves in the southern cow house was much faster than that in the north cow house. The treatment of calves in a clean and well-ventilated cow house with dry bedding was more effective than that in a cold, wet, and dark house. It could be postulated that the most important environmental factor in the reduction of diarrheal diseases of calves is the blocking of the entry of the infectious agents. The complete sanitation and control of the delivery house in the using farm is more important than building a new cow house, for prevention of calf diarrheal diseases.
