Assessment of the ozonation against pathogenic bacteria in the effluent of the quarantine station

  • This study investigated how ozone treatment can successfully inactivate pathogenic bacteria in both artificial seawater and effluents discharged from the fishery quarantine station in Pyeongtaek Port, Korea. Vibrio sp. and Streptococcus sp. were initially inoculated into the artificial seawater. All microbes were almost completely inactivated within 10 min and 30 min by injecting 6.4 mg/min and 2.0 mg/min of ozone, respectively. It was discovered that the water storing Pleuronichthys, Pelteobagrus, and Cyprinus imported from China contained the indicator bacteria, Vibrio sp., Enterococcus sp., total coliforms, and heterotrophic microorganisms. Compared to the control, three indicator bacteria were detected at two to six times higher concentrations. The water samples displayed a diverse microbial community, comprising the following four phyla: Bacteroidetes, Proteobacteria, Firmicutes, and Actinobacteria. Almost all indicator bacteria were inactivated in 5 min at 2.0 mg/min of ozonation; comparatively, 92.9%-98.2% of the less heterotrophic microorganisms were deactivated within the same time period. By increasing the dosage to 6.4 mg/min, 100% deactivation was achieved after 10 min. Despite the almost complete inactivation of most indicator bacteria at high doses after 10 min, several bacterial strains belonging to the Proteobacteria have still been found to be resistant under the given operational conditions.

Sensitization of Periodontitis Disease Causing Bacteria by Low Power He-Ne Laser Radiation

  • The present investigation is an attempt to create an optimized protocol for a bactericidal modality of different powers of He-Ne laser radiation to eliminate periodontitis disease causing bacteria from dental plaques. Periodontitis is most prevalent infectious disease of men and caused by a limited number of Gram negative oral bacteria. Porphyromonas gingivalis and Streptococcus sanguis are the important bacteria responsible for periodontitis diseases. Effect on periodontitis disease causing bacteria were produced by the exposure of different powers of He-Ne laser light i.e. 9 mW, 17 mW and 26 mW of red colour of wavelength 632.8 nm in two different periods of time i.e. 10 min. and 20 min. in the presence of dye Methylene blue (MB) used as a photosensitizer. The results have been shown in terms of percentage inhibition of colony forming units (cfu.) of bacteria. This study has shown that maximum inhibition of cfu. were observed in Laser+MB-20 min. exposure time. This inhibition was followed by Laser+MB-10 min., but minimum inhibition was seen in Laser only at 10 min. exposure. In case of effect of methylene alone on the cfu. of bacteria, it was seen that MB have not shown more inhibition of cfu. and it had shown that the no. of cfu. are very similar to that of control. The above observation of the present study was seen in case of every 3 different type of used powers of laser for both the bacteria. Maximum percentage inhibition of cfu. were seen in case of 26mW powers of He-Ne laser, which was 67. 28% to 61.42% for Porphyromonas gingivalis and Streptococcus sanguis respectively. So, increasing the power of laser (safe range for dentistry is 3-30 mW) under conditions shows an increased percentage inhibition of cfu. Thus the present investigation may be a useful adjunct with mechanical debridement in the prevention of recolonization of subgingival lesions by pathogenic microorganisms which are harmful and drug resistant.

A Study on Inactivation of Pathogenic Bacteria for Nutrient Solution Recycling Using Advanced Oxidation Processes

  • This study was conducted to test the applicability of hydroxy radical reactor system, which applied advanced oxidation processes, to sterilize pathogenic bacteria for nutrient solution recycling in closed hydroponics. Removal efficiency was tested on 25 L of nutrient solution maxed with 10 mL culture solution of bacteria, E. coli, and R. solanacearum in a pilot tank. The testing conditions included various levels of hydroxy radicals resulting from air flow rates of 40, 80, and $120L\;min^{-1}$, and 12 hours processing time. The removal of bacteria, E. coli, and R. solanacearum by hydroxy radical in nutrient solution was significantly increased with an increase in the flow rate of the air from $40L\;min^{-1}$ to $120L\;min^{-1}$. The optimum removal efficiency was achieved at an air flow rate of $120L\;min^{-1}$ for 2 hours treatment. There were no significant differences in removal efficiency among bacteria, E. coli, and R. solanacearum for tested level and time of hydroxy radical. These results verified the efficiency of hydroxy radical in removing the pathogenic bacteria and the applicability of hydroxy radical reactor system in the field.

유전체장벽방전 플라즈마에 의한 주요식중독세균의 살균 효과 (Antibacterial Activity of Dielectric Barrier Discharge Plasma against Main Food-borne Bacteria in Suspensions)

  • Dielectric barrier discharge (DBD) plasma is one of the promising next generation non-thermal technologies for food sterilization. The present study investigated the effects of DBD plasma on the reduction of most common food-borne pathogenic bacteria (Staphylococcus aureus, Bacillus cereus, Vibrio parahaemolyticus, Salmonella enterica) and sanitary indicative bacteria (Escherichia coli) in the suspension (initial inoculum of approx. 9 log CFU/mL). The bacterial counts were significantly (P<0.05) reduced with the increase in the treatment time (1-30 min) of DBD plasma in the suspension. The D-values (time for 90% reduction) of DBD plasma by first-order kinetics for S. aureus, B. cereus, V. parahaemolyticus, S. enterica, and E. coli were 17.76, 19.96, 32.89, 21.55, and 15.24 min, respectively (R2>0.90). These results specifically showed that 30 min of DBD plasma treatment in > 90% reduction of seafood-borne pathogenic and sanitary indicative bacteria. This suspension study may provide the basic data for use in seafood processing and distribution.

Bactericidal Effects of CaO (Scallop-Shell Powder) on Foodborne Pathogenic Bacteria

  • This study was investigated the bactericidal effects of calcium oxide (CaO) on three common foodborne pathogenic bacteria: Escherichia coli, Listeria monocytogenes, and Salmonella typhimurium. Each bacteria level was determined in a CaO solution (0.01, 0.03, 0.05, 0.10, 0.15, and $0.20\%$ [w/v]) exposed for either 15 sec, 30 sec, 1 min, 2 min, 3 min, 5 min, 10 min, or 30 min. All three bacteria were not greatly affected by CaO solutions at concentrations of 0.01 and $0.03\%$, however, the decline of E. coli $(99\%;\;2.78\;log_{10}CFU/mL)$, L. monocytogens $(45\%;\;1.44\;log_{10}CFU/mL)$, and S. typhimurium $(70\%;\;2.08\;log_{10}CFU/mL)$ was greatest when they were exposed to $0.05\%$ CaO solution for 10 min. Moreover, the bactericidal action of CaO was maintained for at least 24 h of storage. The results of this study provide evidence that CaO, as a substitute for synthetic chemical substances has potential for use in the disinfection and sanitization of foods and food processing equipment.

무균 돈사 환경 모니터링을 위한 대기 중 미생물 탐지기법 확립 (Optimization of monitoring methods for air-borne bacteria in the environmental conditions of pig facilities)

  • Experimental animals have been used to biological and medical purposes and the animals must be, for these purposes, healthy and clean to microbial infection. However, the animals can be easily exposed to pathogenic microorganism via several routes. Of the routes, environmental conditions are the most important factors to keep the animals healthy and clean, especially air condition. Monitoring of air-condition has been required to keep the animal healthy and clean. However, any guideline is not available for experimental conditions with pigs. Therefore, the sampling times and points were compared in different conditions to establish an optimal protocol for monitoring of air borne bacteria. Tryptic soy agar(TSA), blood agar containing 5% defibrinated sheep blood and Sabraud dextrose agar(SDA) were used as media to capture total bacteria, pathogenic bacteria and fungi, respectively. Two methods, compulsive capture using an air-sampler and capturing fall-down bacteria were used to capture the microorganisms in the air. The points and time of capturing were different at each experiment. Air borne microorganisms were captured at three and five points in the open and closed equipments, respectively. Air was collected using an air-sampler for 1 min and 5 min and the agar plates as open status were left from 30 min to 2hr. At first, we monitored an experimental laboratory which dealt with several pathogenic bacteria and then, a protocol obtained from the investigation was applied to open or close experimental conditions with pigs. Number of bacteria was high from 10:00 to 15:00, especially on 13:30-15:30 but sharply decreased after 17:00. The tendency of the number of bacteria was similar between two methods even though the absolute number was higher with air sampler. Critical difference in the number of cells was observed at 5 min with air sampler and 2 hr with fall-down capturing method. However, 1 min with air sampler and 1 hr with fall-down capturing were the best condition to identify bacterial species collected from the air. Number of bacteria were different depending on the sampling points in closed condition but not in opened condition. Based on our results, a guide-line was suggested for screening air-borne microorganism in the experimental conditions with pigs.

양배추 Coleslaw 제조 중 HACCP 설정을 위한 식초를 이용한 E. coli 제어 (Control of E. coli in Cabbage during Producing Coleslaw by Vinegar for HACCP)

  • Cabbage, the main material of coleslaw was not safely washed because it is uneven and tightly layered. In this study, vinegar was used to control the number of E. coli in cabbage for coleslaw production. When cabbage was soaked in 10% acetic acid, The number of bacteria was reduced by $10^{-3}$ was survived after 5min and $10^{-6}$ after 30min. After soaking, 15-20% (w/w of cabbage) of 10% acetic acid was remained in chopped cabbage. Spraying 10% acetic acid up to 3% of cabbage weight decreased the number of bacteria by 1/1000 after 60min. Spraying 3% (v/w of cabbage) of vinegar containing 10% acidity decreased the bacteria by 2/3 after 20min but no further decrease was observed. This result indicated that vinegar can be used only in the case that contamination is low.

  • The purpose of this paper is to study on the morphological and physiological nature generally observed for the identifications, and the four strains of thermal resistanting aerobic bacteria isolated from swelled cans and the different soils collected from the wide area in Korea. The results obtained in the light of the Bergey's Manual for the identification of the bacteria, have been shown that the four strains of bacteria are pertained to Bacillus subtilis. The optimum temperature, pH and the thermal resistance (2.5 min. at $121^{\circ}C$) of the bacteria have been measured.

초고압 처리에 의한 좁쌀탁주의 미생물 살균 및 효소 불활성화 (Inactivation of Microorganisms and Enzymes in Foxtail Millet Takju by High Hydrostatic Pressure Treatment)

  • 좁쌀탁주를 초고압으로 처리하여 미생물 살균 및 효소불활성화의 효과를 측정하였다. 무처리 좁쌀탁주의 세균은 $6.8{\times}10^7\;CFU/mL$, 젖산균은 $1.3{\times}10^8\;CFU/mL$, 효모는 $8.4{\times}10^7\;CFU/mL$이었다. 좁쌀탁주를 상압에서 열처리($65^{\circ}C$/30분)하였을 때 젖산균과 효모는 완전히 사멸되었으나 세균은 $2.2{\times}10^5\;CFU/mL$ 잔존하였다. 좁쌀탁주를 초고압으로 처리한 결과 젖산균과 효모는 압력의 증가에 따라 급격히 감소하였으며, 400 MPa에서는 완전히 사멸되었다. 세균은 상온에서 압력을 600 MPa로 높여도 멸균되지 않았으며, 400 MPa/10분에서 처리온도의 증가에 따라 급격히 감소하는 경향을 보였으나, $66^{\circ}C$에서도 완전히 사멸되지 않았으며, $66^{\circ}C/400$ MPa에서 60분, 600 MPa에서 10분 처리로 완전히 사멸되었다. ${\alpha}-amylase$의 활성은 처리압력의 증가에 따라 감소하였으며, 상온/600 MPa/10분에서 73.2% 잔존한 반면, glucoamylase의 활성은 압력의 증가에 따라 상승하는 경향을 보였다. 400 MPa에서 처리온도의 증가에 따라 ${\alpha}-amylase$와 glucoamylase 모두 활성이 감소하였는데, $66^{\circ}C$에서 ${\alpha}-amylase$는 59.7%, glucoamylase는 20.5% 불활성화되었다. 가열처리($65^{\circ}C$, 30 분)에 의한 ${\alpha}-amylase$의 잔존활성은 14.1%이었는데 반하여, $66^{\circ}C/600\;MPa$에서 30분 처리로 잔존활성이 1.2% 이하가 되었다.
