The inverted retina of the Onchidium dorsal eye (DE) is composed only of ciliary-type photoreceptor cells (CC's). The outer segment (OS) of the CC is a concentric lamellar structure consisting of many modified ciliary membranes and stains positively with anti-$\beta$-tubulin antibody. Near the base of the OS there are about 30 basal bodies each connecting individually to a cilium. The cilia are rod-shaped at the base, progressing upwards to a flattened sheet-like shape with increasing surface area. Three-dimensional analysis on serial sections demonstrates the ladle-shape of a modified cilium. Many modified cilia wrap around each other like the leaves of a cabbage. Nine pairs of microtubules (MT's) are located regularly in a ring at the base of the cilium, gradually losing their regular arrangement towards the periphery, where they separate into two subgroups that are contained within two swollen portions of a modified cilium. Within the CC of the Onchidium DE, MT's in the modified cilium exist as two poles extending longitudinally in a thin expanded ciliary membrane. This arrangement may support the photoreceptive OS and serve to maintain its structural integrity.
The internal ultrastructural changes of germ cells and external morphology of spermatozoon during the spermatogenesis in the rockfish, Sebastes inermis were studied using transmission and scanning electron microscope. The testis is seminiferous tubule type in internal structure. Seminiferous tubule consist of many cyst which contain numerous germ cells in same developmental stage. Spermatogonium contained a large nucleus with single nucleolus in interphase. Primary spermatocyte identified by the presence of synaptonemal complex in nucleus and the contained a number of mitochondria, endoplasmic reticula and Golgi bodies in cytoplasm. The nucleoplasm of secondary spermatocyte was more concentrated than that of the previous phase. Spermatids were more condensed in nucleus and cytoplasm, and show the long-spherical shape. In the cytoplasm of spermatid mitochondria located to lower portion of the nucleus and Golgi bodies located to upper portion, but proacrosomal granule is not appeared. The spermatozoon consist of the head and tail. No acrosome could be found in the head. The cytoplasmic collar of posterior part in sperm head contained mitochondria which surrounded axial filament. The well developed axonemal lateral fins were identified in sperm flagellum, and the axial filament of the flagellum consist of nine pairs of peripheral microtubules and one pair of central microtubules.
In this study, we found that sperm ball of Urechis unicinctus consisted of a somatic cell and spermatogenic cells. After separation from the sperm ball, individual spermatid floated freely in the coelomic fluid and differentiated into a mature sperm. Because of many nuclear vacuoles, spermatid nucleus was observed to be heterogeneous. Later, the spermatid nucleus condensed into the homogeneous round nucleus of the mature sperm. Perinuclear microtubules could be seen but did not seem to be organized into manchette microtubules. To understand the nature of nuclear condensation during spermiogenesis, the sperm and spermatids (spermiogenic cells) were treated with FITC-phalloidin, or anti-actin-FITC, or labeled with antiactin immunogold particles (AAIP; 10 nm) followed by transmission electron microscopy or confocal laser scanning microscopy. The anti-actin-FITC and FITC-phalloidin reactions occurred distinctly in the nuclei of both spermiogenic cells. FITC-phalloidin reacted more intensely with acrosomes. The AAIP were incorporated mainly into nuclei of both cells sometimes showing local distribution in the nucleus. Nuclear vacuoles of spermatids disappeared progressively with condensation of the nucleus, as the number of incorporated $AAIP/{\mu}m^2$ increased. These results suggest that nuclear actin microfilaments might be closely related to nuclear condensation.
Fine structural changes of germ cell during the gametogenesis of Korean rockfish, Sebastes schlegeli sampled in west coast of Korea were investigated from September 1993 to August 1994. In a layer of microvilli of oocyte with active yolk duplication, many pinocytotic vesicles containing protein granules regarded as yolk precursors were observed. The multivesicular bodies were formed by gathered mitochondria. They are participated in formation of the primary yolk globules homogeneously filled with high dense particles and enclosed within a limiting membrane. The precursors of yolk globule appeared to be formed by modification of mitochondria and they developed into the primary yolk globules with participation of large and dense pinocytotic vesicles. Yolk globules in mature oocyte were consisted of three components: the crystalline type main body, the superficial layer with dense and fine granules, and the limiting membrane. Steroid hormone secreting cells were recognized in the interstitial cells of growing testis. Numerous endoplasmic reticula and large mitochondria with well developed tubular cristae appeared in their cytoplasms. The axoneme in the tail flagellum of spermatozoon consisted of nine pairs of microtubules at the periphery and one pair at the center, and they were covered with doublet microtubules.

Cytoskeleton components play key role in maintaining cell structure and in giving shape to the cell. These components include microtubules, microfilaments and intermediate filaments. Among these filaments intermediate filaments are the most rigid and bear large compressive force. Actually, these filaments are surrounded by other filaments like microtubules and microfilaments. This network of filaments makes a layer as a surface on intermediate filaments that have great impact on buckling behavior of intermediate filaments. In the present article, buckling behavior of intermediate filaments is studied by taking into account the effects of surface by using Euler Bernoulli and Timoshenko beam theories. It is found that effects of surface greatly affect the critical buckling force of intermediate filaments. Further, it is observed that the critical buckling force is inversely proportional to the length of filament. Such types of observations are helpful for further analysis of nanofibrous in their actual environments within the cell.
In Arabidopsis thaliana, the microtubule-associated protein AtMAP65-1 shows various functions on microtubule dynamics and organizations. However, it is still an open question about whether AtMAP65-1 binds to tubulin dimers and how it regulates microtubule dynamics. In present study, the tubulin-binding activity of AtMAP65-1 was investigated. Pull-down and co-sedimentation exp eriments demonstrated that AtMAP65-1 bound to tubulin dimers,at a molar ratio of 1 : 1. Cross-linking experiments showed that AtMAP65-1 bound to tubulin dimers by interacting with $\alpha$-tubulin of the tubulin heterodimer. Interfering the bundling effect of AtMAP65-1 by addition of salt and monitoring the tubulin assembly, the experiment results indicated that AtMAP65-1 promoted tubulin assembly by interacting with tubulin dimers. In addition, five truncated versions of AtMAP65-1, namely AtMAP65-1 $\Delta$N339 (amino acids 340-587); AtMAP65-1 $\Delta$N494 (amino acids 495-587); AtMAP65-1 340-494 (amino acids 340-494); AtMAP65-1 $\Delta$C495 (amino acids 1-494) and AtMAP65-1 $\Delta$C340 (amino acids 1-339), were tested for their binding activities and roles in tubulin polymerization in vitro. Four (AtMAP65-1 $\Delta$N339, $\Delta$N494, AtMAP65-1 340-494 and $\Delta$C495) from the five truncated proteins were able to co-sediment with microtubules, and three (AtMAP65-1 $\Delta$N339, $\Delta$N494 and AtMAP65-1 340-494) of them could bind to tubulin dimers in vitro. Among the three truncated proteins, AtMAP65-1 $\Delta$N339 showed the greatest activity to promote tubulin polymerization, AtMAP65-1 $\Delta$N494 exhibited almost the same activity as the full length protein in promoting tubulin assembly, and AtMAP65-1 340-494 had minor activity to promote tubulin assembly. On the contrast, AtMAP65-1 $\Delta$C495, which bound to microtubules but not to tubulin dimers, did not affect tubulin assembly. Our study suggested that AtMAP65-1 might promote tubulin assembly by binding to tubulin dimers in vivo.
Oryzalin은 미세소관을 분열시키는 dinitroaniline계의 제초제이다. 미세소관과 미세섬유는 평형석 침강과 세포벽을 구성하는 세포골격들이다. 평형석은 뿌리 끝에 있는 columella 세포에서 중력 인지 조절을 한다. 본 연구는 oryzalin이 옥수수 일차 뿌리에서 ethylene 생성을 통하여 굴중성 반응에 미치는 영향을 연구하였다. 뿌리 끝 부분에 10-4 M oryzalin의 처리는 뿌리 성장과 굴중성 반응을 저해하였으나, 신장대에 처리하게 되면 저해현상은 관찰되지 않았다. 10-4 M oryzalin을 뿌리 끝에 15시간 처리하면 뿌리 끝의 생장이 억제되고 둥근 형태로 부풀었다. 에틸렌의 전구물질인 ACC를 뿌리 끝에 처리하여도 굴중성 반응이 억제되었다. Oryzalin의 작용과 에틸렌 생성에 대한 관련성을 연구하기 위하여 oryzalin 처리 후 에틸렌 생성을 측정하였다. Oryzalin 처리에 의해 ACC oxidase와 ACC synthase의 활성이 증가되어 에틸렌 생성이 촉진되었다. Oryzalin은 ACO와 ACS의 유전자의 발현도 증가 시켰다. Indole-3-acetic acid (IAA)는 굴중성 반응 동안 관찰되는 비 대칭적 신장에 중요한 역할을 한다. 이러한 연구 결과는 oryzalin이 뿌리 끝에서 IAA transport를 억제하여 뿌리 신장대의 윗면과 아랫면의 IAA 양의 차이를 감소시키고, 또한 에틸렌 생성을 촉진하며 미세소관의 배열을 방해하여 뿌리 글중성과 생장을 억제할 가능성을 제시하고 있다.
본 연구에서는 체외수정, 난자내 정자 직접주입, 난자내 정자 두부 미두 주입 후의 핵과 미세소관의 변화를 관찰하였다. 핵과 미세소관의 움직임은 형광염색을 실시한 후 공초점주사현미경을 이용하여 관찰하였다. 체외수정에서 관찰된 바와 동일하게 정자를 난자에 직접주입 한 직후 정자 중편부에서 성상체가 형성되었고, 이 성상체에 의해 자성 웅성 전핵이 융합되는 것으로 관찰되었다. 그러나 난자내 정자를 직접주입하였을 경우 웅성전핵으로 발달하는 비율이 낮았다. 이는 주입된 정자가 원형질막과 perinuclear theca에 싸인 체 난자내로 들어가 난자내의 sperm nucleus decondensing factor와 정자 핵과의 반응이 억제되기 때문으로 생각된다. 정자 두부 만을 주입하였을 경우 성상체가 형성되지 않았지만 자성 웅성 전핵 사이 또는 그 주위에서 두터운 미세소관층이 관찰이 되었다. 따라서 소에 있어서는 정자의 중편부에 위치하여 microtubule organizing center (MTOC)의 역할을 하는 중심립 또는 중심체 없이도 모계에서 유래된 미세소관이 형성되어 이것이 전핵의 융합과 세포분열에 관여하는 것으로 생각된다. 정자의 미부 만을 주입하였을 경우 성상체가 형성이 되지 않았으며, 자성핵 사이에 형성된 미세소관과 떨어져서 관찰되었다. 따라서 주입된 정자의 꼬리는 미세소관형성과 관련이 없는 것으로 생각된다. 이러한 결과는 소에 있어서, 수정 시 정자로부터 유래되는 중심립 또는 중심체가 없이도 미세소관을 형성하여 미세소관에 의해 이후의 배발달이 정상적으로 일어남을 보여주고 있다.
The ultrastructure of freeze-substituted Malpighian tubule cells of German cockroach after rapid freezing was compared to that fixed with conventional chemical fixation. As results, freeze-substitution proved to be the superior technique revealing much more structural details of the tissue cells. Especially, all kinds of membrane structures including infoldings, microfilament, microtubules and ribosomes were best preserved in this material.
Microtubules and microfilaments are major cytoskeletal components in mammalian ova that provide the framework for chromosomal movement and cellular division. Extensive changes of cytoskeletal organization occur during maturation and fertilization. The changes in cytoskeletons are essential for the normal meiotic maturation and for the formation of the biparental diploid genome of the embryo, and thus are repeated at each cell cycle during embryonic development. Disturbance of the cytoskeletal organization could result in abnormal gamete development and early embryonic death.
