Characteristics of an imperfect fungus, Microsporum nigricans Yeeh sp. nov. (불완전 진균 Microsporum nigricans Yeeh sp.nov의 특성)

  • An imperfect fungus recognized as a dermatophyte was isolated from soil samples and the macroscopic, microscopic and physiological characteristics were examined. From the comparisons of this isolate with the known species, the strain belonged to the genus Microsporum. She showed mant different characteristics from the related species, including colony color and shape, or number in a cell of macrocondium. This isolate was considered to be a novel species in the genus Microsporum. Therefore the name was given to her as Microsporum nigricans Yeeh sp. nov. because of dark blackish color from the macroscopic observation.

Detection of Adenovirus from Respiratory and Alimentary Tract in Pusan, 1999

  • Adenovirus which is an important infectious viral agent in respiratory and alimentary tract was investigated in Pusan, 1999. Fifteen cases of adenovirus were detected from stools and throat swabs of suspected patients. Two cases of enteric adenovirus were detected from a 5 years old boy and a 6-month-old boy. Thirteen cases of respiratory adenoviruses were detected from children aged under 10 years old and one adult. From respiratory specimens, 1 case of adenovirus type 2, 1 case of type 5, and 11 cases of type 3 were found. Enterotype 41 was detected from fecal preparations. Adenoviruses appeared mostly during winter months, January, February and December. Adenovirus showed a slowly progressive cytopathic effect on HEp-2 cells, Vero cells and BGM cells at 37$^{\circ}C$, in a 5-7% $CO_{2}$ incubation. An electron microscopic observation exhibited non-enveloped icosahedron with a diameter of 70nm. No significant differences on cytopathic effect and morphological features have been found from specimens of either alimentary tract or respiratory secretions.

  • Fracture behaviors of SCM435 high strength bolt have been studied including macroscopic and microscopic fracture observation, Energy Dispersive X-ray Spectroscopy, Vickers hardness test and applied stress evaluation. cracks (ratchet marks) were generated by the repetitive loads acting on the bolts, initial stress of bolt and the stress concentration. The applied stress was found to be slightly higher than the fatigue limit of the material. The initial stress of bolt must be removed, and the mold temperature during the process must be maintained by room temperature. Bolts are recommended to be peened to improve fatigue limit.

  • The needs to machine hardened steels with high productivity and good sufrace integrity have been increased in the dies & molds industry. This paper presents some experimental results on the CBN ball endmilling for hardened tool steel. This investigation concerns on the effects of cutting fluids, cutting speed, and feed on the cutting performance such as cutting forces, tool wear, and surface finish. The wear of CBN ball endmill for each cutting conditions were also examined through the microscopic observation. It has been found that the higher cutting speeds with cutting fluids result in better cutting performance.

First Discovery of Stereostratum corticioides Causing Rust on the Culm of the Bamboo Pseudosasa japonica

  • A fungus strain Stereostratum corticioides PKVL1, belonging to the family Pucciniaceae that causes rust in plants, was discovered on the sheath of the bamboo Pseudosasa japonica leading to the death of the infected bamboo in the following year. Microscopic observation of the yellow fungal mass revealed teliospores with an oval, one-septate (two-celled) structure. The average length and width of teliospores were 31.83±3.57 ㎛ and 20.74±1.72 ㎛, respectively. The large-subunit ribosomal RNA gene was amplified using the LR0R and LR7 primers, showing that the strain PKVL1 had a similarity of 99.34% to previously reported S. corticioides. In particular, the two Stereostratum strains form a separate cluster among the fungi in the family Pucciniaceae. This is the first report in the Republic of Korea of fungal rust occurring on the culm of bamboo rather than on the leaves.

XRF Analysis and Polarizing Microscopic Study of the Lava Cave Formation, Korea, Japan and Russia (한국, 일본, 러시아 용암동굴 형성층의 형광X선 분석과 편광현미경적 연구)

  • (1) Kaeusetgul Cave in Kimnyong-Ri, Jeju-Do, Korea. Kaeuset-gul Cave (KC) is situated in NNE area of the Manjang-gul cave (125m a.s.l.). Kaeuset-gul Cave lies at $126^{\circ}45'22"$ E in longitude and $33^{\circ}33'09"$ N in latitude. The coast belong Kimnyeong-Ri, Kujwa-eup, Jeju-Do. Altitude of the cave-entrance is 10m and length of the cave is 90m. Lava hand-specimens of KC are studied by X-ray fluorescence analysis (XRF). Average major chemical components of specimens from KC is as follows (wt.%); $SiO_2=47.03$, $TiO_2=3.16$, $Al_2O_3=18.41$, FeO*=13.53, MnO=0.14, MgO=5.05, CaO=8.66, $Na_2O=2.81$, $K_2O=0.67$, $P_2O_5=0.55$ in KC. Polarizing microscopic studyindicates that these specimens are described of alkali-basalt. (2) Tachibori Fuketsu (Cave) in Shizuoka Prefecture, Fuji Volcano, Japan Tachibori Fuketsu lies attoward the south in skirt of the Fuji volcano, $138^{\circ}42'04"$ east longitude and $35^{\circ}18'00"$ north latitude. The location of cave entrance is 2745, Awakura, Fujinomiya-shi, Shizuoka Prefecture. The above sea level and length of Tachibori Fuketsu are 1,170m and 82m. Average major chemical components of specimens from cave areas follows (Total 100 wt.%) ; ($SiO_2$=50.52, $TiO_2$=1.69, $Al_2O_3$=15.47, FeO*=13.13, MnO=0.20, MgO=5.97, CaO=9.17, $Na_2O$=2.52, $K_2O$=0.94 and $P_2O_5=0.40).$ Polarizing microscopic study indicates that these specimens may belong to tholeiite-basalt series. According to polarizing microscopic study, Au (Augite), P1 (Plagioclase), and O1 (Olivine) are contained as phenocryst minerals. (3) Gorely Cave in Kamchatka Peninsula, Russia Gorely caldera is located at the southeastern part of Kamchatka Peninsula, about 75km southwest of Petropavlovsk-Kamchatskiy.. Gorely lava caves are situated in NHE area of Mt. Gorely volcano (1829m a.s.1.). One of lava cave (Go-9612=K-1) lies at $158^{\circ}00'22"$ east longitude and $52^{\circ}36'18"$ north latitude. The elevation of cave entrance is about 990m a.s.1. and the main cave extends in the NNW direction for about 50m by 15m wide and 5m in depth. The cave of K-3is near the K-1 cave. "@Lava hand-specimens K-1 and K-3 caves are studied by X-ray fluorescence analysis and polarizing microscopic observation. Average major chemical components of specimens from these caves are as follows (wt.%) ;($SiO_2$=55.12, $TiO_2$=1.25, $Al_2O_3$=16.07, T-FeO* =9.41, MnO=0.16, MgO=5.01, CaO=7.21, $Na_2O$=3.39, $K_2O$=1.92, $P_2O_5$=0.45) and these values indicate that the Gorely basaltic andesite belong to high alumina basalt. Polarizing microscopic study indicates that these specimens are described of Augite andesite.

Characteristics of Microbial Decomposition of Bast Fibers by Wood Rot Fungi (목질분해균에 의한 인피섬유의 미생물분해 특성)

  • In order to use bast fibers of mulberry tree at a pulp source of Hanji, the bast fibers were microbiologically treated with several wood rot fungi, and the microscopic characteristics of bast fibers depending on treatment days were evaluated. By wood rot fungi, Phanerochaete chrysosporium and Trametes versicolor, the weight reduction ratio was approximately 50 percent within incubation for 20 days. occurring together with decomposition of useful fibers. However, Hwterobasidion insularis and Stereum hirsutum have completely decomposed the utmost layer of black blue colored bast fibers, and not caused the damage if fibers. Until incubation for 10 days, the cellulose content of vast fibers by Stereum hirsutum was 78.9 percent with lignin content of 7.2 percent, showing an appropriate decomposition for useful fibers. By microscopic observation, the bundled fibers were separated to single fiber within treatement days 30 by Pleurotus ostreatus, and there were no damage on the surface of fiber by treatment days 50.

A Study on a Morphological Identification of Pinelliae Tuber, Typhonii Flagelliformis Rhizoma and Arisaematis Rhizoma (반하(半夏), 수반하(水半夏) 및 천남성(天南星)의 형태(形態) 감별에 관한 연구)

  • Objectives : A morphological classification among Pinelliae Tuber, Typhonii Flagelliformis Rhizoma and Arisaematis Rhizoma was made through microscopic observation. Method : The slice of the tested material made by paraffin section technique was colored with Safranine Malachite Green contrast methods, and then observed and photographed by olymphus-BHT. Result : 1. Pinelliae Tuber is like a ball with a $1{\sim}1.5cm$ diameter, with a concave apex and pockmarked root trace around. 2. Typhonii Flagelliformis Rhizoma is somewhat oval or like a cone, with a $4{\sim}8mm$ diameter, a $7{\sim}15mm$ height, a projected round apex, and a pin-pointed back. 3. Arisaematis Rhizoma is somewhat flat round with a $2{\sim}2.5cm$ diameter, a little than Pinelliae Tuber, and its apex has incomplete lobopodium and root trace. 4. Fundamental tissues of Pinelliae Tuber, Typhonii Flagelliformis Rhizoma, and Arisaematis Rhizoma have starch grains and mucilage cells, and their mucilage cells include the raphides of calcium oxalate. Conclusion : Internal forms of Pinelliae Tuber, Typhonii Flagelliformis Rhizoma, and Arisaematis Rhizoma are difficult to distinguish from one another, but Pinelliae Tuber and Arisaematis Rhizoma can be distinguished by their external forms and sizes, and Pinelliae Tuber and Typhonii Flagelliformis Rhizoma were distinguished by the forms of apex.

  • Since the widespread application of hyperbaric oxygenation in clinical medicine, the problems of oxygen toxicity have been attracting a deep interest from the researchers on hyperbaric medicine as a practical issue. Among extensive research trials, the study on the protective agents oxygen toxicity occupied one of the most challenging field. As the mechanisms of oxygen toxicity, the role of the oxygen free radicals produced by peroxidation process are strongly accepted by the leading researchers on oxygen toxicity, the probable protective effects of antioxidant against oxygen toxicity are sustaining a sufficient rational. Maltol ($2-methyl-3-hydroxy-{\gamma}-pyrone$) which is known to be a component of Korean red ginseng has been reporting to have an antioxidant action. But, further study is needed to provide definite evidence for this compound to be an antioxidant, since the action was based on the results which were obtained under in vitro experiment. In this study, the author attempted to evaluate the effect of maltol as protective agent against oxygen toxicity through the observation of death rate, convulsion rate, time to convulsion and microscopic pathological changes in some organs of experimental rats exposed to various conditions. The findings observed are as follows : 1) The death rate, convulsion rate, time to convulsion, lung/weight ratio and microscopic pathological finding of lung were identified as reliable objective and quantitative indices for oxygen toxicity. 2) Maltol showed excellent protective effect against pulmonary oxygen toxicity as an antioxidant.

A STUDY OF BONE APPOSITION AND MARGINAL ALVEOLAR BONE LOSS AROUND IMMEDIATE IMPLANSTS (발치 직후 매식 임프란트의 골침착과 변연골 상실에 대한 연구)

  • The purpose of this study was to observe bone apposition and marginal bone loss and to check the possibility of success on titanium implant, HA coated implant and the implant with natural coral that were place immediately after teeth extraction in dogs. Experimental subjects were divided into 4 groups ; the 1st group is the titanium implant, second the HA coated implant, third the implant with natural coral, and the last the control group that was prepared in the extraction sockets. After 12 weeks, the dogs were sacrificed for visual observation and microscopic examination approaching histologic and histomorphometric analysis. The results were as follows : 1. Neither the infection nor the exposure of implant was found at the sites of all implant. 2. In a histomorphometric analysis, mean percentage of direct bone contact with the titanium implant was 80.7% and the HA coated implant showed 81.5% apposition, but the implant with natural coral showed 64.9% apposition(P<0.05). 3. In a microscopic examination, mature lamellated bone was found around the immediate implants and control group, while unabsorbed natural coral around the immediate implants with natural coral was found. 4. All immediate implant groups showed the loss of marginal bone in order from implant with natural coral, titanium implant, and HA coated implant. 5. Implant with natural coral that was placed by the type I interface of the Barzilay's classification immediately after teeth extraction showed low percentage of direct bone contact area, low success rate and a lot of marginal bone loss. Above results suggested that the immediate implants are osseointegrated successfully, although slightly marginal bone was loss.
