고분자 담체에 부착된 미생물 형상

  • Optical microscope, SEM and fluorescent microscope were used for qualitative and morphological studies of the attachment bacteria on PE substratum under anaerobic condition. The observation of optical microscopic has demonstrated that the initial attachment of bacteria began in crevices of surface. In SEM photographs, shape and structure of biofilm could be observed, but bacteria species and methanogens was not identified. A large number of methanogenic bacteria were showed on the surface of PE substratum by fluorescence under 480nm of radiation. It was estimated that methanogenic bacteria was related to initial attachment of bacteria under anaerobic condition.

The Effects of T-2 Toxin on the Morphology of Mouse Organ (Mouse 장기의 형태에 미치는 T-2 Toxin의 영향)

  • To study the immunotoxicity of mouse injected with fungal mycotoxin, T-2 toxin (Fusarium mycotoxin) was treated to 6 week-old female C3H/He mouse and the body, organ weight and morphological change were investigated. The weights of body, liver and kidney of mouse injected the 2 mg/kg of toxin was decreased to 17, 20 and 3%, respectively, compared to control animal and the comsumption of feed was also decreased with lapsing the time. The fat dropleting phenomenon and destruction of Golgi apparatus in liver and histopathological changes of tissue and mitochondria in small intestine were found by scanning electron microscopic observation.

  • Scale is formed when hard water is heated or cooled in heat transfer equipments such as heat exchangers, condensers, evaporators, cooling towers, boilers, and pipe walls. When scale deposits in a heat exchanger surface, it is traditionally called fouling. The objective of the present study is to investigate the formation of fouling in a heat exchanging system. A lab-scale heat exchanging system is built-up to observe and measure the formation of fouling experimentally. Water analyses are conducted to obtain the properties of HAN river water. In the present study a microscopic observation is conducted to visualize the process of scale formation. Hardness of HAN-river water is higher than that of tap water in Seoul.

  • A fretting wear test rig for cross contacting tube specimens, which employs a piezoelectric actuator, has been developed. Along with the simple loading scheme using dead weights, the rig is very simple to be used also. The accuracy was found acceptable. Inconel 690 tubes were tested in room temperature and ambient condition. Normal load and sliding amplitude range up to 35N and $100{\mu}m$, respectively. The sizes of wear scar and the wear volumes were measured, and wear coefficients have been calculated based on those. A study on the fretting wear mechanism of the tubes has been attempted via microscopic observation. Rugged wear surfaces are induced by the separation and adhesion of particles and formation and subsequent fracture of surface layers. Lapped specimens were also tested and abrasive wear seems to be playing a dominant role.

  • Chemoautotrophic symbiotic bacteria of organisms inhabiting the hydrothermal vent and cold seep communities in the deep-sea were histologically examined using a transmission electron microscopy on symbionts of Calyptogena sp. A from the site east off Miyako (water depth at 1,700m), Calyptogena sp. B from the Calyptogena Site, vestimentiferan tube worm Lamellibrachia sp. A from Sagami Bay Lamellibrachia sp. B from Calyptogena Site of the Iheya Ridge, pogonophoran tube worms from Sagami Bay and Calyptogena Site of the Iheya Ridge, Bathymodiolus spp. from Sagami Bay, the Iheya Ridge and the North Fiji Basin. Based on the morphological microscopic observations, two species of Calyptogena from Miyako and the Iheya Ridge, two species of vestimentiferan tube worms from Sagami Bay and the Iheya Ridge, and pogonophoran tube worms from Sagami Bay and the Iheya Ridge observed to host sulfur-oxidizing symbiotic bacteria. The occurrence of chemosynthetic symbionts in these organisms was expected beforehand based on the ecological observations of their habitats. Other members of these groups from the world oceans, and the recent advances in the symbiosis at vents and seeps were reviewed.

  • Liver protective effects of ginsenosides as well as fractions of dammarane glycosides of Panax ginseng were studied using galactosamine (GalN)-induced cytotoxicity in primary cultured rat hepatocytes. Preventing effects on GalN-induced hepatotoxicity were found both microscopic observation and determination of GPT level with total dammarane glycosides fraction and $20(S)-ginsenoside-Rb_1$ as well as $20(S)-ginsenoside-Rg_1$ at the concentration of $50{\mu}g/ml$. The syntheses of both protein and RNA were significantly increased by the treatment of $50{\mu}g/ml$ of total dammarane glycoside fraction, $20(S)-ginsenoside-Rb_1$, -Rc, -Re and $-Rg_1$, respectively in both normal and GalN-induced cytotoxic hepatocytes.

Diagnosis of Potato Leafroll disease by Fluorescence Microscopic Detection of Callose Stained with Resorcin Blue (Resorcin Blue 염색 기법에 의한 감자 잎말림병의 형광 현미경적 진단)

  • Deep blue fluorescence of resorcin blue-stained callose was observed only in the potato leafroll virus (PLRV)-infected potato plants, but not in other potato viruses investigated. The plant sections stained with aniline blue showed non-specific fluorescence regardless of PLRV infection, which means that aniline blue is not suitable for the staining of callose induces by PLRV infection. The fluorescence of resorcin blue-stained callose was more easily detectable than autofluorescence by a direct fluorescence detection method because of its deep blue color. The lateral branch of lower leaves was turned out to be the best material for fluorescence observation of all plant parts tested. In comparison of diagnostic efficacy of this technique to enzyme-linked immunosorbent assay (ELISA), PLRV infected potato plants showed corresponding increment of the fluorescence of resorcin blue stained callose as absorption values in ELISA increased. In the future, the criteria measuring the fluorescence objectively are thought to be determined for the practical application to the diagnosis of potato leafroll disease.

Step-wise Anion-Exchange in Layered Double Hydroxide Using Solvothermal Treatment

  • Synthetic layered double hydroxides (LDHs), [$Mg_4Al_2(OH)_{12}]CO_3{\cdot}nH_2O$, were prepared in the submicron size of plate-like polycrystals. Anion-exchange reactions with various linear dicarboxylic acids were performed to produce LDH/organic hybrid materials by solvothermal treatment in toluene. X-ray powder diffraction spectra for the products indicated that the interlayer spacings of LDHs remarkably changed, up to 20 $\AA$ when 1,10-decanedicarboxylic acid anions were intercalated as an organic guest. Dicarboxylates-LDHs samples could be also re-exchanged consecutively with other dicarboxylic acids or carbonate without serious destruction of layer structure under the scanning electron microscopic observation.

  • T-2 toxin was given to rats(Spargue Dawley) with a 4mg/kg and 2mg/kg dose p.o. Data was based on the T-2 toxin 4mg/kg treated group except for counting of acidophil bodies. GPT activities increased significantly from 0.5 to 10 hours. GOT activities were also increased at 1 and 5 hours significantly and the relative weights of liver were increased at 0.5 and 5 hours significantly. There were slight necrotic foci in microscopic observation. There was dose-dependent trend for the frequency of the acidophil body between the 4mg/kg treated group and the 2mg/kg treated group.

Thermodynamic Calculations of High Temperature Bubble Formation at SiC/$SiO_2$ Interface (SiC/$SiO_2$ 계면의 고온 기공발생에 관한 열역학적 계산)

  • Numerous researchers have observed the bubble fromation at SiC/SiO2 interface from 130$0^{\circ}C$ to 1$700^{\circ}C$. According to thermodynamic calculation, the bubble could be formed from the microscopic impurities which result from the chemical vapor deposition of SiC. When C-impurity is present at the interface, it is calculated that the bubble is formed at 1511$^{\circ}C$ and when Si is present, the bubble is formed at 177$0^{\circ}C$. These results are very close to the prior observations, but the calculation can not explain the observation of bubble below 150$0^{\circ}C$.
