  • "Chung Wi Dan" is a Chinese patent medicine, which is used for various purposes in Korea. According to traditional publications, they are mixtures of several powders made of herb medicines. Chung Wi Dan of 19 kinds of powder is used for catarrh of the gastrointestines, indigestion, a pain in the chest. nausea. For the identification of individual ingredients in such powdery mixtures, microscopic method may advantageously be used as it requires only a small amount of specimens. (omitted)

  • Objectives : A morphological classification among Adenophora triphylla var. japonica, Codonopsis lanceolata, Adenophora remotiflora and Codonopsis pilosula was made through microscopic observation. Method : The slice of the tested material made by paraffin section technique was colored with Safranine Malachite Green contrast methods, and then observed and photographed by olymphus-BHT. Result : 1. Codonopsis pilosula is like a long cone, with amole-like projected parts at the beginning of the roots. 2. Adenophora triphylla var. japonica, Codonopsis lanceolata, Adenophora remotiflora, and Codonopsis pilosula are all similar as following: 1) Cork layers are composed of a few to over 10 subsidiary cells, with individual or a group of stone cells among them. 2) Phloem is wide and has a milk tube. Conclusion : Adenophora triphylla var. japonica, Codonopsis lanceolata, Adenophora remotiflora, and Codonopsis pilosula are all similar and difficult to be distinguished through microscopic observation, so that chemical experiments should be conducted for their identification.

Method : The slice of the tested material made by paraffin section technique was colored with Safranine Malachite Green contrast methods, and then observed and photographed by olymphus-BHT. Result : 1. The cross-sections of Sophora tonkinensis and Indigofera kirilowii have piths, but Menispermum dauricum does not have any. 2. The lateral parts of Sophora tonkinensis cortex include the square crystal of calcium oxalate, but Indigofera kirilowii does not. Conclusion : The number of piths and the presence of the square crystal of calcium oxalate can be used to distinguished Sophora tonkinensis from Indigofera kirilowii.

  • Objectives : A morphological classification among Rheum palmatum Linne, Rheum undulatum Linne and Rumex crispus was made through microscopic observation. Methods : The slice of the tested material made by paraffin section technique was colored with Safranine Malachite Green contrast methods, and then observed and photographed by olymphus-BHT. Results: 1. Stellate spots were found on the intersection of Rheum palmatum, composed of heteromorphy vascular bundles. 2. Stellate sponts were not found in Rheum undulatum and Rumex crispus, because they do not have heteromorphy vascular bundles. Conclusion : Rheum palmatum can be distinguished with Rheum undulatum and Rumex crispus, based on the presence of Stellate spots on its intersection.

  • Objectives : Araliae Continentalis Radix(AC) is a medicinal herb belonging to the drug efficacy group treating musculoskeletal disorders(MSD) with anti-inflammatory, analgesic and antipyretic action. However, due to morphologic and onomastic similarity, adulterants(Angelicae Pubescentis Radix: AP, Gypsophilae Oldhamianae Radix: GO, Levistici Officinalis Radix: LO) have been included or replaced the standard. Methods : Multilateral methods were carried out on the identification of AC and its adulterants. Macroscopic and microscopic characteristics were observed by using stereoscope and microscope. For the comparison of chromatogram pattern, standard compounds were analyzed simultaneously using high performance liquid chromatography. Results : 1. The macroscopic identification of original plants was determined by the phyllotaxis type, the inflorescence type, the leaf margin and the color of flowers. The macroscopic identification of herbal materials was examined by oil spots, the cambium, heteromorphic vascular bundles, and the pholem. 2. For the microscopic identification, the fact whether its xylem ray is proliferated or not was first determined. Then medicinal herbs were secondly divided by cellular inclusions, fiber bundles, the distribution of secretary canals and the shape of cambium. 3. AC and its adulterants showed different chromatographic fingerprints. AC was containing continentalic acid and kaurenoic acid. AP was containing osthole and columbianadin. LO was containing osthole and falcarindiol. None of the compounds were found in GO. Conclusions : This recent identification keys of might be helpful to discriminate the pharmacopoeia standard and its adulterants for the right usage in clinics.

  • Damage evolution in the form of void nucleation, propagation and coalescence is the primary cause that is responsible for the ductile failure of microalloyed steels. The Gurson-Tvergaard-Needleman (GTN) damage model has proven to be extremely robust for characterizing the microscopic damage behavior of ductile metals. Nonetheless, successful applications of the model on a given metal type are limited by the correct identification of damage parameters as well as the validation of the calculated void growth rate. The purpose of this study is two-fold. First, we aim to identify the damage parameters of the GTN model for Q345 steel (Chinese code), due to its extensive application in mechanical and civil industries in China. The identification of damage parameters is facilitated by the well-suited response surface methodology, followed by a complete analysis of variance for evaluating the statistical significance of the identified model. Second, taking notched Q345 cylinders as an example, finite element simulations implemented with the identified GTN model are performed in order to analyze their microscopic damage behavior. In particular, the void growth rate predicted from the simulations is successfully correlated with experimentally measured acoustic emissions. The quantitative correlation suggests that during the yielding stage the void growth rate increases linearly with the acoustic emissions, while in the strain-hardening and softening period the dependence becomes an exponential function. The combined experimental and finite element approach provides a means for validating simulated void growth rate against experimental measurements of acoustic emissions in microalloyed steels.

  • Single cell PCR analysis and light and scanning electron microscopic techniques were utilized to identify free living bivalve larvae in the coastal waters of Tae-an, on the west coast of Korea. Through DNA sequencing, venerid clam larvae were isolated and identified as Ruditapes philippinarum (99% similarity) and Meretrix lusoria (99%). Under microscopic observation, the D-veliger stage of R. philippinarum exhibited symmetrical shoulder angles and an elliptical ventral form. In contrast, M. lusoria displayed asymmetrical shoulder angles and a round ventral form in the umbonal stage. Size of the R. philippinarum larvae was $156{\pm}22{\mu}m$ in length, $126{\pm}12{\mu}m$ in height, $92{\pm}14{\mu}m$ in width with a length: height ratio of 1.23. Meretrix lusoria was $202{\pm}44{\mu}m$ in length, $161{\pm}35{\mu}m$ in height, $96{\pm}38{\mu}m$ in width with a length: height ratio of 1.25. Experimental results indicate that morphological and molecular characteristics provide evidence for the larval identification of these two venerid clam larvae species in nature.

  • Microscopic method was compared with X-ray diflraction analysis for the identification of rockforming minerals using 11 main rock samples in Korea. 1. There was no difference between X-ray diffraction analysis and microscopic one in major minerals, but some accessary minerals. 2. The rock-forming minerals of main rocks presented in this study occured almost in crystalline state so that they could be easily identified by X-ray analysis alone.

  • Data obtained from X-ray diffractometry, thermal analysis, IR spectroscopy and microscopic observation were used for the identification and characterization of four crystalline modifications of piroxicam. form a was crystallized from sodium hydroxide-hydrochloric acid and from c was obtained by crystallization from toluene. Form b and d was crystallized from methanol under the different temperature conditions. Relative rates of dissolution and solubility of four crystal forms of piroxicam in distilled water were measured.
