Purple-colored shell individuals were discovered among normal green-colored shell individuals in artificial seed of Pacific abalone, Haliotis discus hannai, reared on an ordinary type of diatom and artificial diet. In the present study, factorial mating experiments were designed to clarify the genetic control of the variant (purple type) and normal (green type) of shell color. The parental population of purple type and green type individuals were derived from a single family between a female and male of each type of coloration. The all mating families were reared in same tank for the same breed environment. The individual of 4 type families were distinguished by paternity test using microsatellite DNA. In factorial mating experiments, all individuals offspring of GG (green type female and green type male), GP (green type female and purple type male) and PG (purple type female and green type male) mating types appeared to green type. In only PP (purple type female and purple type male) mating type, all individuals offspring appeared to purple type. The results suggested that the purple shell color is controlled by recessive purple type allele and a dominant green type allele at a single locus.

We studied the genetic impact of natural regeneration practices, such as Single seed tree, Group seed tree, Patch clear cutting and Alternate strip clear cutting systems, by comparing the nuclear microsatellite(nSSR) variation of post-practice natural regeneration one-year old seedlings of Pinus densiflora to that of pre-practice mature trees. The levels of genetic diversity of seedlings (A=13.6, $A_e$=4.3, $H_o$=0.571, $H_e$=0.597) were similar to those of mature trees (A=13.4, $A_e$=4.3, $H_o$=0.596, $H_e$=0.598) and the differences in the level of genetic diversity between seedlings and mature trees for each of the practices were not statistically significant. The degree of genetic differentiation between seedlings and mature trees was very low ($F_{ST}$=0.002) and the pairwise $F_{ST}$ values between seedlings and mature trees for all practices were less than 0.01. Overall, the natural regeneration practices appeared to have only minor impacts on the genetic diversity and the genetic composition in the studied P. densiflora stands. For a better understanding of the genetic effects of natural regeneration practices, subsequent studies such as temporal genetic variation of seedlings formed by crossing among post-practice mature trees should be considered.
The leptin receptor gene(LEPR) produces a high affinity receptor that mediates the regulation of the leptin gene. Leptin secreted from adipose tissue plays an important role in regulating feed intake and energy balance. In this study, a microsatellite marker within LEPR was selected and genotyped for the F2 population composed of 354 individuals from an intercross between Korean Native boars and Landrace sows. Totally, six alleles (255, 259, 261, 263, 265 and 267bp) and nineteen genotypes were detected in the population, of which the CE (261/265), CC (261/261) and EE (265/265) types were observed by 20.0%, 10.1% and 9.6%, respectively. Relationships between their genotypes and economic traits were analyzed. We found specific genotypes associated with economic traits such as body weight at 12 weeks of age/body fat including abdominal and trimmed fat/shear force (P〈0.001), body weight of 30 weeks of age (P〈0.01) and body weight of 3 weeks of age/back fat thickness (P〈0.05). The DD (263/263) and DF (263/267) types were associated with body weight at 3, 5, 12 and 30 weeks of age. The DF (263/267) type showed a highly significant effect on back fat thickness and body fat including abdominal and trimmed fat. The DF (263/267) type showed positive effect on shear force, whereas the BB (259/259) and DD (263/263) types negatively affected on tenderness.
The level of genetic variation and relationships in three native Korean goat populations (Dangjin, Jangsu, and Tongyeong) as well as the populations of a farm were analyzed, based on 30 microsatellite markers. A total of 277 distinct alleles were observed across the four goat populations, and 102 (36.8%) of these alleles were unique to only one population. The mean observed heterozygosity and polymorphism information content were calculated as 0.461~0.651 and 0.462~0.679, respectively. In the NJ tree constructed based on Nei's $D_A$ genetic distance, the four populations represented four distinct groups. However, the genetic distances between each Korean native goat population and the farm population were two times those among the three native Korean breeds. The genetic structure within the three Korean native goat populations was also investigated. Cluster analysis, using the STRUCTURE software, suggested three clusters. The molecular information of genetic diversity and relationships in this study will be useful for the evaluation, conservation, and utilization of native Korean goat breeds as genetic resources.
(Table Omitted)

Simple sequence repeats (SSRs) were isolated from major Pleurotus pulmonarius cultivars in Korea, namely 'HS47' (monokaryon, gamete of 'Santari'), 'GB19' (monokaryon, gamete of 'Santari'), 'Hosan,' 'Yeoleumneutali1,' 'Sambok,' 'Gangsan,' 'Yaksan,' 'Jasan,' 'Hyangsan,' and 'Yeoleumneutali2,' and characterized via HiSeq genome sequencing and bioinformatic analysis. The genome sizes of the monokaryons 'HS47' and 'GB19' were estimated to be 37.3 and 37.2 Mb, respectively, and those of the other dikaryotic cultivars ranged from 47.1 to 61.1 Mb. A total of 711 (smallest) and 1,106 (1.5 times the smallest) SSRs were found in the 'HS47' and 'Gangsan' genomes, respectively. Hexanucleotide and octanucleotide motifs accounted for the top two fractions of all SSRs. CGA/TCG, A/T, and CTC/GAG were the most frequently detected nucleotides in the SSRs. Most of the SSRs were 21~30 nucleotides long (hypervariable for application), accounting for 70% of all lengths of SSRs.
A number of Korean native chicken(KNC) populations were registered in FAO (Food and Agriculture Organization) DAD-IS (Domestic Animal Diversity Information Systems, http://www.fao.org/dad-is). But there is a lack of scientific basis to prove that they are unique population of Korea. For this reason, this study was conducted to prove KNC's uniqueness using 25 Microsatellite markers. A total of 548 chickens from 11 KNC populations (KNG, KNB, KNR, KNW, KNY, KNO, HIC, HYD, HBC, JJC, LTC) and 7 introduced populations (ARA: Araucana, RRC and RRD: Rhode Island Red C and D, LGF and LGK: White Leghorn F and K, COS and COH: Cornish brown and Cornish black) were used. Allele size per locus was decided using GeneMapper Software (v 5.0). A total of 195 alleles were observed and the range was 3 to 14 per locus. The MNA, $H_{\exp}$, $H_{obs}$, PIC value within population were the highest in KNY (4.60, 0.627, 0.648, 0.563 respectively) and the lowest in HYD (1.84, 0.297, 0.286, 0.236 respectively). The results of genetic uniformity analysis suggested 15 cluster (${\Delta}K=66.22$). Excluding JJC, the others were grouped in certain cluster with high genetic uniformity. JJC was not grouped in certain cluster but grouped in cluster 2 (44.3%), cluster 3 (17.7%) and cluster8 (19.1%). As a results of this study, we can secure a scientific basis about KNC's uniqueness and these results can be use to basic data for the genetic evaluation and management of KNC breeds.

To provide a ecological restoration strategy considering genetic diversity of Abies koreana in Mt. Jiri, the genetic diversity and the genetic differentiation among sub-populations such as Banyabong, Byeoksoryeong, and Cheonwangbong were investigated. The average number of alleles (A) was 7.8, the average number of effective alleles ($A_e$) was 4.9, observed heterozygosity ($H_o$) was 0.578, and expected heterozygosity ($H_e$) was 0.672, respectively. The level of genetic diversity within sub-populations ($H_e=0.672$) was lower than those of both population ($H_e=0.778$) and species ($H_e=0.759$) level. However, the level of genetic diversity was high compared those of Genus Abies. Genetic differentiation was 0.014 from F-statistics ($F_{ST}$) and was 0.004 from AMOVA analysis (${\Phi}_{ST}$). There was no almost genetic differentiation among sub-populations in Mt. Jiri from bayesian clustering. Therefore, If the seeds are sampled sufficiently by selecting the parameters from three sub-populations, it is possible that we could obtain genetically appropriate materials for ecological restoration.
Novel cpSSR primers were developed based on the sequence information of the Pinus koraiensis chloroplast genome. A total of 30 cpSSR loci were detected in the chloroplast genome, and a total of 30 primer sets flanking those loci were designed. All primer sets were successfully amplified for chloroplast DNA in P. koraiensis. The cross-species transferability of the 30 primer sets was considerably high in P. pumila (100%) and P. paviflora (97%) belonging to the same Subgenus (Strobus) of P. koraiensis. Meanwhile, the transferability was relatively low (73%) in P. densiflora and P. sylvestris belonging to Subgenus Pinus. A total of 13 cpSSR loci out of the 30 loci were polymorphic in the Mt. Jumbong population of P. koraiensis. The mean of haploid diversity(H) was 0.512. The number of haplotypes(N) and the haplotype diversity($H_e$) were 25 and 0.992, respectively. Of the 25 haplotypes, 22 were unique in the analyzed population. The unique haplotypes differentiated 22 individuals (79%) from the total of 28 individuals. In conclusion, the novel cpSSR primers developed in this study would be applicable to other Pinus species, especially the subgenus Strobus, and provide a high level of polymorphism for the study of genetic variation of P. koraiensis.

The invasive red swamp crayfish, Procambarus clarkii, is native to south-central United States and northeastern Mexico. Recently, it has been being spreading in the wild in South Korea. However, its primary sources, introduction routes, establishment, and expansion in South Korea remain unclear. Here, we analyzed genetic diversity and population genetic structures of its domestic natural populations during early invasion, commercial stock from local aquaria (a suspected introduction source), and original United States population using mitochondrial COI gene sequences for 267 individuals and eight microsatellite markers for 158 individuals. Natural and commercial populations of P. clarkii showed reduced genetic diversity (e.g., haplotype diversity and allelic richness). The highest genetic diversity was observed in one original source population based on both genetic markers. Despite a large number of individuals in commercial aquaria, we detected remarkably low genetic diversity and only three haplotypes among 226 individuals, suggesting an inbred population likely originating from a small founder group. Additionally, the low genetic diversity in the natural population indicates a small effective population size during early establishment of P. clarkii in South Korea. Interestingly, genetic differentiation between natural populations and the United States population was lower than that between natural populations and aquarium populations. This suggests that various genetic types from the United States likely have entered different domestic aquariums, leading to distinct natural populations through separate pathways. Results of our study will provide an insight on the level of genetic divergence and population differentiation during the initial stage of invasion of non-indigenous species into new environments.
