Ensiling Characteristics and the In situ Nutrient Degradability of a By-product Feed-based Silage

  • This study was conducted to evaluate the ensiling characteristics and the in situ degradability of a by-product feed (BF)-based silage. Before ensilation, the BF-based mixture was composed of 50% spent mushroom substrate, 21% recycled poultry bedding, 15% ryegrass straw, 10.8% rice bran, 2% molasses, 0.6% bentonite, and 0.6% microbial inoculant on a wet basis and ensiled for up to 4 weeks. The BF-based silage contained on average 39.3% moisture, 13.4% crude protein (CP), and 52.2% neutral detergent fiber (NDF), 49% total digestible nutrient, and 37.8% physically effective $NDF_{1.18}$ on a dry matter (DM) basis. Ensiling the BF-based silage for up to 4 weeks affected (p<0.01) the chemical composition to a small extent, increased (p<0.05) the lactic acid and $NH_3$-N content, and decreased (p<0.05) both the total bacterial and lactic acid bacterial counts from $10^9$ to $10^8$ cfu/g when compared to that before ensiling. These parameters indicated that the silage was fermented and stored well during the 4-week ensiling period. Compared with rice or ryegrass straws, the BF-based silage had a higher (p<0.05) water-soluble and filterable fraction, a lower insoluble degradable DM and CP fraction (p<0.05), a lower digestible NDF (p<0.05) fraction, a higher (p<0.05) DM and CP disappearance and degradability rate, and a lower (p<0.05) NDF disappearance and degradability rate. These results indicated that cheap, good-quality BF-based roughage could be produced by ensiling SMS, RPB, rice bran, and a minimal amount of straw.

Co-ensiling garlic stalk with citrus pulp improves the fermentation quality and feed-nutritional value

  • Objective: Ensiling is a simple and effective method for long-term preservation; however, less information exists about the ensilability characteristics of garlic stalk (GS). Therefore, the objectives were to examine the ensiling feasibility of GS. Methods: The GS was ensiled alone or inoculated with Lactobacillus plantarum KU5 in the presence or absence of 5% molasses and ensiled for 7, 14, and 28 d. As an alternative storage method, GS was co-ensiled with wet citrus pulp (CP) at different proportions (GS:CP: 70:30, 60:40, 50:50, and 40:60). Analysis was made on physicochemical, fermentative, and nutritional parameters. Results: The GS was found to be a biomass which is difficult to ensile. A combination of microbial inoculant and molasses was successful in the improvement of the silage fermentation quality of GS. Co-ensiling of GS with wet CP at the mixing ratio of 50:50 provided the most desirable silage fermentation parameters, including the substantial lactic acid formation, low final pH, minor effluent loss, and the more favorable organoleptic properties. Conclusion: Co-ensiling GS with CP appears to be a simple and viable method of conservation, enabling the more efficient utilization of these by-product resources over a prolonged period.

수박 만할병균에 길항하는 Bacillus sp. YJ-3에 의한 대목용 참박 생육촉진효과 (Plant Growth-Promoting Effects of Antagonistic Bacillus sp. YJ-3 against Fusarium Wilt of Watermelon-Rootstock Gourd)

  • 여러 작물 재배지 토양에서 각종 근권미생물을 분리하여 이들 분리미생물 중 수박 만할병균인 Fusarium oxysporum f. sp. niveum(KCCM34741)에 길항하는 미생물인 YJ-19, YJ-45, YJ-3 등 12 균주를 분리하였다. 길항미생물들은 MS agar 배지 실험 및 미생물제제 첨가 상토 실험에서 모두 참박의 발아율을 향상시켰으며, 이들 중 분리주 YJ-3 균주는 참박의 발아율과 초기생육에 가장 큰 영향을 주는 미생물로 선별되었다. YJ-3 균주를 제제화하여 참박 재배 기간별로 식물체 생육실험을 행한 결과 대조구인 일반 상토 및 시판 미생물혼합 상토 보다 각각 46%와 13%정도 생육을 촉진하는 것으로 나타났다. 분리주 YJ-3 균주는 간균의 gram 양성균으로서 Bacillus sp.로 동정되었다. Bacillus sp. YJ-3은 수박 만할병균인 F. oxysporum f. sp. niveum에 대해 강한 억제효과를 나타내었고, 그외 수박 병원균인 Alternaria cucumerina, Botrytis cinerea, Colletotrichum orbiculare, Didymella bryoniae, Rhazoctonia solani 등에서도 우수한 길항력을 나타내어 본 Bacillus sp. YJ-3 균주는 수박 만할병균에 길항하는 동시에 참박의 생육을 촉진하는 미생물로 확인되었다.

A Two-Strain Mixture of Rhizobacteria Elicits Induction of Systemic Resistance Against Pseudomonas syringae and Cucumber Mosaic Virus Coupled to Promotion of Plant Growth on Arabidopsis thaliana

  • We evaluated a commercial biopreparation of plant growth-promoting rhizobacteria (PGPR) strains Bacillus subtilis GB03 and B. amyloliquefaciens IN937a formulated with the carrier chitosan (Bio Yield) for its capacity to elicit growth promotion and induced systemic resistance against infection by Cucumber Mosaic Virus (CMV) and Pseudomonas syringae pv. tomato DC3000 in Arabidopsis thaliana. The biopreparation promoted plant growth of Arabidopsis hormonal mutants, which included auxin, gibberellic acid, ethylene, jasmonate, salicylic acid, and brassinosteroid insensitive lines as well as each wild-type. The biopreparation protected plants against CMV based on disease severity in wild-type plants. However, virus titre was not lower in control plants and those treated with biopreparation, suggesting that the biopreparation induced tolerance rather than resistance against CMV. Interestingly, the biopreparation induced resistance against CMV in NahG plants, as evidenced by both reduced disease severity and virus titer. The biopreparation also elicited induced resistance against P. syringae pv. tomato in the wild-type but not in NahG transgenic plants, which degrade endogenous salicylic acid, indicating the involvement of salicylic acid signaling. Our results indicate that some PGPR strains can elicit plant growth promotion by mechanisms that are different from known hormonal signaling pathways. In addition, the mechanism for elicitation of induced resistance by PGPR may be pathogen-dependent. Collectively, the two-Bacilli strain mixture can be utilized as a biological inoculant for both protection of plant against bacterial and viral pathogens and enhancement of plant growth.

Evaluation of in vitro ruminal fermentation of ensiled fruit byproducts and their potential for feed use

  • Objective: Ensiling of tannin-rich fruit byproducts (FB) involves quantitative and qualitative changes in the tannins, which would consequently change the rumen fermentation characteristics. This study aimed to evaluate whether ensiled FBs are effective in mitigating methane emission from ruminants by conducting in vitro assessments. Methods: Fruit byproducts (grape pomace, wild grape pomace, and persimmon skin) were collected and subjected to four-week ensiling by Lactobacillus buchneri inoculant. A defined feed component with or without FB samples (both fresh and ensiled material) were subjected to in vitro anaerobic culturing using rumen fluid sampled from beef cattle, and the fermentation parameters and microbial populations were monitored. Results: Reduced methane production and a proportional change in total volatile fatty acids (especially enhanced propionate proportion) was noted in bottles containing the FBs compared with that in the control (without FB). In addition, we found lower gene copy number of archaeal 16S rRNA and considerably higher levels of one of the major fibrolytic bacteria (Fibrobacter succinogenes) in the bottles containing FBs than in the control, particularly, when it was included in a forage-based feed. However, in the following cultivation experiment, we observed that FBs failed to exhibit a significant difference in methane production with or without polyethylene glycol, implying that tannins in the FBs may not be responsible for the mitigation of methane generation. Conclusion: The results of the in vitro cultivation experiments indicated that not only the composition but also ensiling of FBs affected rumen fermentation patterns and the degree of methane generation. This is primarily because of the compositional changes in the fibrous fraction during ensiling as well as the presence of readily fermented substrates, whereas tannins in these FBs seemed to have little effect on the ruminal fermentation kinetics.

Effect of feeding fermented soybean meal on broiler chickens' performance: a meta-analysis

  • Objective: The present study aimed to quantify the effects of fermented soybean meal (FSBM) on broiler chickens' performance by employing a meta-analysis approach. Methods: A total of 16 studies were included in the database after being systematically selected using a PRISMA protocol. Hedges' g effect size was used to quantify pooled standardized mean difference (SMD) using random-effects models at 95% confidence intervals (95% CI). Publication bias among studies was computed with Egger's test and visualized using funnel plots. Results: Results indicated that dietary FSBM inclusion increased final body weight (BW) (SMD = 0.586, 95% CI: 0.221 to 0.951, p = 0.002) of broiler chickens, particularly in starter period (SMD = 0.691, 95% CL: 0.149 to 1.233, p = 0.013) while in the finisher period, the effect was weaker (SMD = 0.509, 95% CI: 0.015 to 1.004, p = 0.043). Average daily gain (ADG), feed intake (FI), and feed conversion ratio (FCR) were not affected with FSBM inclusion when compared to control. Subgroup analysis revealed that FI increased in starter period (SMD = 0.582, 95% CI: 0.037 to 1.128, p = 0.036). When considering types of microorganism as moderating variables in the subgroup analysis, we found that Aspergillus oryzae, mixed probiotics+bromelain protease, Bacillus subtilis, and Lactobacillus bacteria significantly increased ADG and FI (p<0.01). Additionally, either Bacillus subtilis+protease or Bacillus subtilis alone decreased FCR (p<0.001). However, meta-regression analysis showed that levels of FSBM inclusion had no effects on final BW (p = 0.502), ADG (p = 0.588), FI (p = 0.861), and FCR (p = 0.462). Conclusion: Substituting SBM in broiler chickens' diet with FSBM improved BW of broiler chickens, especially in the starter period whereas the effects on ADG, FI, and FCR were mostly dependent on microbial strains used for fermentation.

Effects of Microbial Inoculants on the Fermentation, Nutrient Retention, and Aerobic Stability of Barley Silage

  • Fermentation characteristics, nutrient retention and aerobic stability of barley silages prepared using 6 commercial inoculants were evaluated using 126 mini-silos (3-L) in a completely randomized design. Whole barley forage was chopped, wilted to 39% DM and treated with water (control, S) or one of six inoculants: A (containing Lactobacillus plantarum); B (L. plantarum and Enterococcus faecium); C (L. plantarum and Pediococcus cerevisiae); D (L. plantarum, Pediococcus pentosaceus and Propionibacterium freudenreichii, plus hydrolytic enzymes); E (Lactobacillus buchneri plus hydrolytic enzymes); F (L. buchneri and P. pentosaceus plus hydrolytic enzymes). Samples of treated forage were collected for analysis at the time of ensiling, and then 18 silos of each treatment were filled, capped and weighed. Triplicate silos were weighed and opened after 1, 3, 5, 7, 33, and 61 d. On d 61, $400{\pm}5g$ of material from each silo was placed in 1-L styrofoam containers, covered with cheesecloth and held at room temperature. Silage temperature was recorded hourly for 14 d via implanted thermocouple probes. Chemical composition of the forage at ensiling was consistent with previously reported values. At d 61, pH was lowest (p<0.01) in silage S. Ammonia-N was lower (p<0.05) in silage A than in silages S, B, E, or F. Compared to pre-ensiling values, water soluble carbohydrate concentrations were elevated in silages S, A, B, C and D, and decreased in E and F. Lactic acid concentrations were similar (p>0.10) across treatments. Acetic acid levels were highest (p<0.01) in silage E and lowest (p<0.01) in silage D. Recovery of DM was lower (p<0.01) in silage F than in silages S, A, B, C, or D. On d 61, yeasts were most numerous (p<0.01) in silage D, which was the only silage in which temperature rose more than $2^{\circ}C$ above ambient during aerobic exposure. Silage D also had the highest (p<0.01) pH and ADIN content after aerobic exposure. Lactic acid and WSC content of silage D decreased dramatically during the 14-d aerobic exposure period. Yeast counts (at d 14 of exposure) were lowest (p<0.01) in silages E and F. In general, the commercial inoculants did not appear to enhance the fermentation of barley silage to any appreciable extent in laboratory silos.

  • 고수분 유기성 물질인 맥주박, 베지밀박 등의 농산가공부산물들은 상온에서 변패성이 강하여 가축의 사료로 이용하기위해서는 보관성증진을 위한 첨가제의 처리가 중요하다. 본 연구에서는 맥주박과 베지밀박을 소금, 유기산, 발효미생물 및 메치오닌수산화물(MHA)을 처리하여 고온다습한 7월중순경에 상온에서 20일간 방치한 후 보관일수에 따른 시료의 표면연화현상과 곰팡이 발생양상, 시료내부 pH, 암모니아태질소 함량변화를 조사하여 각 첨가제의 시료에 대한 변패방지효과를 비교한 결과, MHA 5%이상 처리구에서 특별한 효과가 있는 것으로 나타내었다.

항진균능을 가진 불용성 인산 가용화 세균의 분리 및 특성 (Isolation and Characterization of Insoluble Phosphate-Solubilizing Bacteria with Antifungal Activity)

  • 다기능성 농업용 미생물 제제를 개발하기 위하여 항진균능을 가진 불용성 인산 가용화 세균을 식물의 근권 토양으로부터 분리하였다. 분리균주의 분류학적 위치를 검토한 결과, Pseudomonas fluorescens RAF15로 동정되었다. P. fluorescens RAF15는 농작물의 잿빛곰팡이병균 Botrytis cinerea와 줄기썩음병균 Rhizoctonia solani의 생육을 억제할 수 있었다. 불용성 인산인 $Ca_3(PO_4){_2}$로부터 가용성 인산을 생성하기 위한 최적 배지 및 배양조건은 glucose 1.5%, urea 0.005%, $MgCl_2{\cdot}6H_2\;0.3%,\;MgSO_4{\cdot}7H_2O\;0.01%,\;CaCl_2{\cdot}2H_2O$ 0.01%, NaCl 0.05%, 배양온도 $30^{\circ}C$ 및 초기 pH 7.0이었으며, 최적조건에서 배양 5일 후 863 mg/L의 가용성 인산이 생성되었다. 불용성 인산 가용화는 유기산 생성에 의한 배양액의 pH 감소와 밀접한 관계가 있었다. P. fluorescens RAF15는 $10-35^{\circ}C$, 1-4% 염 농도 및 pH 2.0-11.0의 범위에서도 가용성 인산을 생성할 수 있었다. 본 균주는 $CaHPO_4,\;Ca_3(PO_4){_2}$ 및 hydroxyapatite에서 각각 971-1121 mg/L, 791-908 mg/L 및 844 mg/L의 가용성 인산을 생성하였다. 그러나 $FePO_2$$AlPO_4$의 경우, 생성된 가용성 인산의 농도는 각각 18 mg/L, 5 mg/L이었다.

Mobilization of Heavy Metals Induced by Button Mushroom Compost in Sunflower

  • Button mushroom compost (BMC) was prepared by fermenting the mixture of waste button mushroom bed collected from Boryeong area in South Korea (4): sawdust (8) : pig and fowl manure (1) for 40 days at $30^{\circ}C$. The BMC compromised diverse microorganisms including aerobic bacteria $8.1{\times}10^6cfu\;g^{-1}$, Gram negative bacteria $1.7{\times}10^7cfu\;g^{-1}$, genus Bacillus $6.4{\times}10^6cfu\;g^{-1}$, genus Pseudomonas $1.5{\times}10^4cfu\;g^{-1}$, actinomycetes $1.0{\times}10^4cfu\;g^{-1}$, and fungi $3.5{\times}10^3cfu\;g^{-1}$. BMC was used as a microbial inoculant for estimating the mobilization of heavy metals in soil or plant. When metal solubilization potential of BMC was assessed in a batch experiment, the inoculation of BMC was shown to increase the concentrations of water soluble Co, Pb, Cd, and Zn by 29, 26, 27, and 43% respectively, than those of non-inoculated soils. BMC-assisted growth promotion and metal uptake in sunflower (Helianthus annuus) was also evaluated in a pot experiment. In comparison with non-inoculated seedlings, the inoculation led to increase the growth of H. annuus by 17, 15, 18, and 21% respectively in Co, Pb, Cd, and Zn contaminated soils. Moreover, enhanced accumulation of Co, Pb, Cd, and Zn in the shoot and root systems was observed in inoculated plants, where metal translocation from root to the above-ground tissues was also found to be enhanced by the BMC. The apparent results suggested that the BMC could effectively be employed in enhancing phytoextraction from the soils contaminated with heavy metals such as Co, Pb, Cd, and Zn.
