팽이버섯 수확후배지의 사료화에 가장 적합한 저장법을 제시하기 위하여 미생물과 개미산을 처리한 팽이버섯 수확후배지를 퇴적, 뒤집기, 비닐피복 등의 방법으로 저장한 다음 미생물과 개미산 처리가 팽이버섯수확후배지의 저장성에 미치는 영향을 조사하였다. 저장기간에 따른 처리구별 팽이버섯 수확후배지의 외관적 성상변화를 종합해보면 저장기간 동안 모든 처리구에서 버섯 균사체가 버섯수확후배지의 표면에 우점하고 있었으며 호기적 조건보다는 혐기적 조건에서 저장성이 향상되었고 개미산 처리와 같은 화학적 처리법보다는 미생물을 접종한 처리구의 저장성이 향상됨을 확인할 수 있었다. 미생물과 개미산을 처리한 버섯수확후배지를 퇴적, 비닐피복, 뒤집기 등의 방법으로 8일 동안 저장하면서 버섯수확후배지의 온도, pH, 일반세균 수 등을 조사한 결과, 온도, pH, 일반세균 수는 미생물과 개미산 무처리구를 뒤집기한 경우에 가장 높게 나타났다.
Haloperoxidase를 생산하는 미생물을 분리하기 위하여 국내 연근해와 남북극 등의 해양시료에서 분리된 방선균 균주를 대상으로 탐색을 수행하여 남해 백도 해조류 추출물로부터 분리된 한 종류의 방선균(#1460)에서 높은 haloperoxidase 활성이 확인되었다. 본 균주의 생리.생화학적 특성은 Streptomyces 속과 유사하며 생산되는 haloperoxidase는 세포 조 추출물로부터 ammonium sulfate precipitation, High-Q column chromatography, gel permeation chromatography, Hydroxyapetite chromatography 그리고 hydrophobic interaction chromatography를 통하여 42%의 수율과 purification fold 70으로 정제하였다. 본 효소의 최적 반응 pH는 7이고 pH 8에서 더 높은 안정성을 보여 $60^{\circ}C$에서 1시간 반응에 효소활성의 50%가 생존한다. 또 cyanide와 azide 이온에 의해 강한 저해현상을 보인다.
Bacillus brevis CD162가 생산하는 cyclodextrin glycosyltransferase (CGTase)를 ammonium sulfate 침전, DEAE-Sephadex CL-6B 및 Sephadex G-150 컬럼 크로마토그래피를 수행하여 분리정제하였다. 정제된 CGTase는 분자량이 약 74,000, 등전점은 약 6.3인 단백질이었고, 정제된 단백질을 SDS-PAGE한 후 변성된 단백질을 재활성시켜 zymogram을 수행한 결과 cyclodextrin glycosyltransferase임을 확인할 수 있었다. 이 효소의 최적활성 pH와 온도는 각각 8.0과 $55^{\circ}C$이었으며, pH $5.5{\sim}9.0$과 $50^{\circ}C$까지 안정한 활성을 보였다. 또한, CGTase의 $NH_2$-말단 부위의 아미노산서열은 Ser-Val-Thr-Asn-Lys-Val-Asn-Tyr-Ser-Lys-Asp-Val-Ile-Tyr-Gln 이었으며, 전분으로부터 cyclodextrin으로의 전환률을 분석한 결과, ${\alpha}$-cyclodextrin은 1.3%, ${\beta}$-cyclodextrin은 33.9% ${\gamma}$-cyclodextrin은 9.7% 이었다.

The present study aimed to analyze the metaproteome of the microbial community comprising harmful algal bloom (HAB) in the Daechung reservoir, Korea. HAB samples located at GPS coordinates of 36°29'N latitude and 127°28'E longitude were harvested in October 2013. Microscopic observation of the HAB samples revealed red signals that were presumably caused by the autofluorescence of chlorophyll and phycocyanin in viable cyanobacteria. Metaproteomic analysis was performed by a gelbased shotgun proteomic method. Protein identification was conducted through a two-step analysis including a forward search strategy (FSS) (random search with the National Center for Biotechnology Information (NCBI), Cyanobase, and Phytozome), and a subsequent reverse search strategy (RSS) (additional Cyanobase search with a decoy database). The total number of proteins identified by the two-step analysis (FSS and RSS) was 1.8-fold higher than that by one-step analysis (FSS only). A total of 194 proteins were assigned to 12 cyanobacterial species (99 mol%) and one green algae species (1 mol%). Among the species identified, the toxic microcystin-producing Microcystis aeruginosa NIES-843 (62.3%) species was the most dominant. The largest functional category was proteins belonging to the energy category (39%), followed by metabolism (15%), and translation (12%). This study will be a good reference for monitoring ecological variations at the meta-protein level of aquatic microalgae for understanding HAB.
Recently the roof gardening has become very popular and even on the apartment-veranda and on the roofs in high buildings many kinds of vegetables and berries are cultivated. And the demand of the nutritional supplement for the effective plant growth is also increasing. The general urban conditions are to be adapted for plant growth. A different hygroscopic and temperature-conditions after regions, sunshine and wind have strong influences on the plant growth and usually it is not optimal enough. It is because why a nutritional condition in soil for plant growth so important and essential. The usual compound-soil or -fertilizer cannot offer enough quantities of nutrients for plant growth and additional soil conditioner becomes more necessary. There are many kinds of soil conditioners like hydrogel in the market and we studied on Geohumus, Montigel and Geko, which are widely used in Europe and other countries. Water absorption and microbial immobilization with effective microorganisms were tested and compared. The EM solution was identified as bacteria, fungi and azotobacter etc. and they were immobilized at the soil conditioners at first. And the cultivated and immobilized at the soil conditioners EM-solution was added to the plant soil. 1 g of the soil conditioners absorbed ca. 20 g of water. The plant grew 10 cm more, got 3 times more branches and 2 times more fruits in the soil with soil conditioners immobilized with microorganisms. With water addition the plant with both conditions in the soil could stay fresh in comparison to without soil conditioners.
The effect of biofertilizer (compound of microbial inoculants or groups of micro-organisms) on growth and yield of rice was investigated. The experiment was carried out in a randomized complete block design with 3 replications and 7 treatments namely: $RF=N-P_2O_{5-}K_2O$ (11-5.5-4.8 kg $10a^{-1}$); half of the recommended fertilizer rate, $HRF=N-P_2O_5-K_2O$ (5.5-2.75-2.4 kg $10a^{-1}$); HRF+Bio 250=HRF combined with 250 kg biofertilizer $10a^{-1}$; HRF+Bio 500=HRF combined with 500 kg biofertilizer $10a^{-1}$; Bio 250=250 kg biofertilizer $10a^{-1}$; Bio 500=500 kg biofertilizer $10a^{-1}$; and NF = No Fertilizer. Results showed that the recorded values of plant height, tiller number and chlorophyll content at 40 to 60 days after transplanting (DAT) in HRF+Bio 500 were significantly higher than those recorded in the RF treatment. Similar observations between these two treatments were only recorded from 60 DAT onwards. Yield components were also superior in HRF+Bio 500 treatment and comparable to that of RF. The highest grain yield obtained in HRF+Bio 500 treatment (785.8 kg $10a^{-1}$) was statistically similar to that of RF (739.8 kg $10a^{-1}$) but significantly higher than that of NF (506.7 kg $10a^{-1}$). Finally, head grain recovery (90.9) was low while chalkiness (0.03) was high at HRF+Bio 500 treatment as compared with RF, which were (96.1) and (0.3), respectively. Results showed that combined treatment of HRF and 500 kg biofertilizer $10a^{-1}$ has similar effects on the growth and yield of rice with that of RF.

우유는 유화나 colloid상태 때문에 우유의 다양한 정량을 위해서는 투명화 이용이 대단히 유용하다. 지방구 butanone에 의하여 분산될 수 있고 casein 복합체는 우유내에서 동시에 SDS 와 NaOH의 작용의 도움으로 Triton 작용에 의하여 단량체로 분해될수 있는데 왜냐하면 butanone은 양친매성이고 물에 섞일수 있는 용매이기 때문이다 용매 배합비율 T:B/8:2 와 T:B/9:1을 제외하고 모든 비율들이 각종 유제품을 투명화시킬수 있다. 용매가 세포와 접촉생장이 억제됨에도 불구하고 이배합은 균수 측정에 사용될수있는데 왜냐하면 세포벽을 공격하지 않기 때문이다. 이러한 이유 때문에 이기술은 원유품질관리, 유대지불, 제품관리, 국가적 낙농 계획 조절에 유용하다.
BACKGROUND: Application of urea may increase $CO_2$ emission from soils due both to $CO_2$ generation from urea hydrolysis and fertilizer-induced decomposition of soil organic carbon (SOC). The objective of this study was to investigate the effects of increasing urea application on $CO_2$ emission from soil and mineralization kinetics of indigenous SOC. METHODS AND RESULTS: Emission of $CO_2$ from a soil amended with four different rates (0, 175, 350, and 700 mg N/kg soil) of urea was investigated in a laboratory incubation experiment for 110 days. Cumulative $CO_2$ emission ($C_{cum}$) was linearly increased with urea application rate due primarily to the contribution of urea-C through hydrolysis to total $CO_2$ emission. First-order kinetics parameters ($C_0$, mineralizable SOC pool size; k, mineralization rate) became greater with increasing urea application rate; $C_0$ increased from 665.1 to 780.3 mg C/kg and k from 0.024 to 0.069 $day^{-1}$, determinately showing fertilizer-induced SOC mineralization. The relationship of $C_0$ (non-linear) and k (linear) with urea-N application rate revealed different responses of $C_0$ and k to increasing rate of fertilizer N. CONCLUSION(s): The relationship of mineralizable SOC pool size and mineralization rate with urea-N application rate suggested that increasing N fertilization may accelerate decomposition of readily decomposable SOC; however, it may not always stimulate decomposition of non-readily decomposable SOC that is protected from microbial decomposition.
아라키돈산을 고농도로 함유한 유지를 생산하는 Mortierella sp. M-12로부터 얻은 균체 지방질을 식용에 적합하도록 정제하기 위하여 탈색 공정을 최적화하였다. 동결건조한 균체로부터 Folch's solvent를 사용하여 균체 지방질 조유를 얻은 후 탈검과 탈산을 거쳐 얻은 탈산유를 50-100 mmHg 진공 하에서 탈색을 진행하였다. 활성백토를 1% 수준으로 첨가하여 $90^{\circ}C$에서 20분간 수행하였을 때 식용유지 규격에 적합한 색상을 갖는 탈색유를 얻을 수 있었다. 탈색 공정 중에 부수적으로 일어난 탈검과 탈산 작용으로 인하여 탈색유의 인 함량은 31.7% 감소하였으며, 산값은 0.5로 감소하여 식용유지의 규격에 적합한 수준이 되었다. 모르티에렐라 단세포유지는 일반적인 유지의 탈색공정을 통하여 식용급 동식물 유지와 같은 수준의 탈색유를 얻을 수 있었다.
과산화수소의 이스라엘잉어에 대한 급성독성 시험을 실시하여 반수치사농도 ($LC_{50}$)를 구하고 기생충 제거 효과와 살균력을 조사하였다. 이스라엘잉어 (평균체중 0.44 g)는 80 $\mu\ell/\ell$에서 24시간 동안 폐사가 발생하지 않았으며 24시간 $LC_{50}$은 148.9 $\mu\ell/\ell$ 이었다. 이스라엘잉어 (평균 체중 0.1 g)에 기생된 트리코디나는 40 $\mu\ell/\ell$의 과산화수소로 1회 약욕 하였을 때 모두 제거되었다. 30종의 어병 임상균주를 사용하여 시험한 결과, 과산화수소에 대한 어병 세균의 최소억제 농도는 거의 40 $\mu\ell/\ell$보다 낮았으며 과산화수소에 의한 세균 증식 억제 효과를 확인 할 수 있었다.
