DETERMINATION OF PURINE AND PYRIMIDINE BASES IN RUMEN MICRO-ORGANISMS BY REVERSED PHASE HPLC AFTER HYDROLYTIC DIGESTION UNDER PRESSURE

  • A rapid and accurate method is described for the determination of nucleo-bases in rumen micro-organisms. A procedure to satisfactorily hydrolyse the micro-organisms involving reaction with a mixture of readily volatile organic acids (acetic and formic acids) under high pressure, is proposed, and optimal conditions for an analytical procedure with reversed phase HPLC is described. The following nucleobases contents (mmol/kg DM) of rumen micro-organisms were found: Adenine (Ade), 82.62; Guanine (Gua), 61.34; Cytosine (Cyt), 84.61; Thymine (Thy), 35.74; Uracil (Ura), 68.62; Hypoxanthine (Hxn), 13.06; Xanthine (Xn), 8.35. Total purine-N content (g/kg N) of rumen micro-organisms were 99.60. The nucleic acid N content (g/kg N) of microbial isolates were: RNA-N, 109.9; DNA-N, 50.9.

유용 미생물과 제올라이트를 이용한 시멘트 벽돌의 수질 정화 특성에 관한 실험적 연구 (An Experimental Study on Water-Purification Properties in Cement Bricks Using Effective Micro-Organisms and Zeolite)

  • 이 연구는 미생물 혹은 생물의 다양한 기능을 이용한 환경의 수복(bioremediation)을 활용한 자연 친화형 수질 정화 시멘트 벽돌로서의 가능성을 검토하고자 하였다. 현재 유용 미생물을 건축 소재에 활용하여 수질을 개선하는 연구들이 제안되어 왔다. 이 연구에서는 이러한 미생물의 수질 정화 능력을 이용하는 것으로서 선행 연구의 낫토균 이외에 된장균, 대구 S환경사업소에서 미생물을 탐색하여 16S rDNA 염기서열 분석법에 의해 동정된 유용 미생물 자원들을 이용하였으며, 이렇게 확보된 수질 정화 능력을 가진 유용 미생물 자원들에 대한 시멘트 벽돌에서의 수질 정화 특성을 검토하였다. 또한 시멘트 벽돌에 미생물을 흡착시키기 위하여 제올라이트를 사용 하였으며, SEM 분석을 통하여 제올라이트에 미생물이 흡착되어 있는 것을 확인 할 수 있었다. 실험 결과 유용 미생물을 이용 하였을 때 우수한 오염물질 제거율이 나타났기 때문에 자연 친화형 건설재료의 유용 미생물 활용이 가능하다고 판단되며, 이 연구에서 소개한 미생물 이외에도 다양한 미생물 자원들을 확보하여 건축 소재에 활용 가능한 수질 개선용 유용 미생물 자원들을 확보할 수 있을 것으로 기대된다.

콘택트렌즈 재질 및 침착 단백질에 따른 균 흡착 정도와 다목적용액의 살균력 비교 (Comparisons of Adherence Level of Micro-organisms According to Contact Lens Materials and Protein Deposition and Disinfection Efficacy of Multipurpose Solution)

  • 목적: 콘택트렌즈 재질 및 렌즈에 침착된 단백질 종류에 따른 균의 흡착력을 비교하고 관리용품의 살균력 차이를 측정하고자 하였다. 방법: FDA 평가기준 및 방법인 Disinfection Efficacy Testing의 Part 2. Regimen Procedure For Disinfecting Regimens에 따라 균 흡착 및 관리용품의 살균력 측정실험을 실시하였다. 결과: 포도상구균을 제외한 녹농균, 세라티아균, 칸디다균은 etafilcon A 렌즈에 더 많이 흡착되었으며, 4종의 다목적용액은 칸디다균을 제외한 모든 균에서 거의 완벽한 살균력을 나타냈다. 눈물 단백질을 침착시킨 렌즈에서 세라티아를 제외한 모든 균은 알부민 침착 렌즈에서의 흡착이 많았으며, 다목적용액의 살균력은 라이소자임 침착 렌즈에 흡착된 균보다 알부민 침착 렌즈에 흡착된 균에서 높은 것으로 나타났다. 또한, 균의 종류에 따라 다목적 용액의 살균력이 달랐다. 결론: 균의 종류, 콘택트렌즈 재질, 침착된 눈물 단백질의 종류가 콘택트렌즈에 흡착되는 균의 양에 영향을 주며, 눈물 단백질의 침착은 다목적용액에 의한 살균력에 변화를 초래할 수 있음을 밝혔다. 이러한 살균력에 영향을 주는 요인에 의해 콘택트렌즈 위생상태가 달라지며 부작용 발생 여부가 영향을 받을 수 있음을 제안한다.

  • A refined version of automatic micro-organism recognition and identification method, 'O.I.S.M.2' is proposed in this paper, using image processing based on an expert system. This proposed method is based on the segmentation of the organism image, characterizing segment features, which are independent of individual size and length. Complicated shapes of organisms are divided into basic shape segments defined in this paper such as lines, circles, ovals etc. Organisms can then be expressed simply in a set of segments, regardless their individual differences.

  • Normal human skin, constantly challenged by environmental micro-organisms, has an innate ability to fight invading microbes through antimicrobial peptides. These peptides, described in both plant and animal kingdoms are able to inactivate a broad spectrum of micro-organisms. Mammalian defensins constitute one of the most common antimicrobial peptide family. Among the three human beta-defensins hBD1, hBD2 and hBD3 produced in epithelia, only hBD2 and hBD3 are inducible and additionally have been described as expressed by differentiated keratinocytes at site of inflammation and infection. The aims of these studies were to define a cell culture model in which the basal production of hBD could be detected and up-regulated in order to enhance skin auto-protection against micro-organisms. A specific Polymerase Chain Reaction method have been developed for hBD2 and hBD3 mRNA detection in non-differentiated monolayer keratinocytes cell culture. We have been able to demonstrate that in vitro, hBD2 and hBD3 expression in normal human keratinocytes could be detected and enhanced by TNF-alpha and IFN-gamma, in hypercalcic culture conditions. This research opened the possibility of the development of cosmetic active compounds, able to induce the expression of skin natural antibiotic peptides responsible about microflora ecology of the skin.

충치에서 분리한 Streptococci에 관하여 (Characterization of Streptococci Isolated from Human Dental Plaque)

  • 충치 sample 50개에서 46 strains의 Streptococcus sp.를 분리하여 그 생리적 특징을 조사한 바 1. 대부분의 균주들이 non-haemolytic strain이었으며, 2. 기질로 maltose, inulin, mannitol, sorbisI, lactose, mannabinose를 분해하여 산을 생성하고 arabinose와 xylose는 균주에 따라 차이를 보였으며, 3. Sucrose broth에서는 거의 gel을 형성하였다.

  • 오늘날 산업의 발달과 함께 생활수준이 향상됨에 따라 삶의 질을 향상시키기 위한 안락하고 편안한 주거환경의 확보와 함께 주거환경주변에서 발생하는 각종오염문제의 해결에 관해서도 그 관심이 집중되고 있다. 그러나 최근 새집증후군 등 콘크리트제품에 대한 환경적인 안정성에 대한 논란이 심화되면서 콘크리트 경화시 발생하는 각종 물질들의 처리에 대하여 본 연구에서는 오염된물의 처리, 하수슬러지 및 오염된 토양의 중금속 및 악취제거에 사용되는 유용미생물을 콘크리트에 적용을 통하여 하천, 호수 등 각종 수변부에 적용되는 구조물에의 적용 가능성을 알아보기 위하여 유용미생물의 사용 유무에 따른 콘크리트의 수질정화성능(SS, BOD, COD, T-P, T-N)을 측정하였다. 유용미생물을 이용한 콘크리트의 수질정화 시험결과 각각의 제거율은 유용미생물이 함유된 콘크리트 블록의 경우가 일반콘크리트 블록에 비해 오염물질 제거율이 전체적으로 약 10% 정도 높게 나타나 유용미생물의 작용에 의하여 수질정화에 효과가 있는 것으로 사료된다.

EFFLUENT FROM RUSITEC INOCULATED WITH RUMEN LIQUOR OR COW FAECES AS SOURCES OF MICRO-ORGANISMS FOR IN VITRO DIGESTION OF FORAGES

  • The experiment investigated the possibility of using effluent from RUSITEC (rumen simulation technique) inoculated with rumen liquor or cow faeces as sources of micro-organisms for in vitro digestion of forages. Nine forages ${\times}3$ sources of inoculum were used in a factorial arrangement of treatments. Rumen liquor was collected from fistulated sheep and faeces was collected from cows. The RUSITEC apparatus consisted of 4 vessels, 2 vessels were charged with faecal liquor and 2 with rumen liquor. On the 8th day of the experiment RUSITEC effluent were collected to use in in vitro studies. In vitro OMD (g/kg) values using three sources of inoculum (fresh rumen liquor, RUSITEC effluent from rumen liquor or cow faeces) were statistically significant (p < 0.001). The regression relationships between OMD using fresh rumen liquor and RUSITEC effluent were highly significant ($R^2>0.90$). The results suggest that RUSITEC effluent either from rumen liquor or cow faeces can be used as a source of micro-organisms for in vitro digestion of forages.

活性슬러지의 嫌氣性消化에 미치는 影響因子에 관한연구 (A Study on Factors Affecting Anaerobic Digestion of Waste Activated Sludge)

  • This study was carried out to investigate factors affecting anaerobic digestion enhancement of waste activated sludge(WAS). In order to this investigation, the degradability and rupture of microorganisms cell present in WAS, and mesophilic anaerobic digestion(MAD) of these compounds, were also evaluated. The micro-organisms cell in WAS were ruptured by a mechanical jet stream and smashed under pressure of 30 bar. The rupture level of micro-organisms cell in WAS were determined using phosphate, soluble protein and soluble chemical oxygen demand (SCOD)concentrations. It was found that the rupture level of micro- organisms cell within WAS increased with increasing pretreatment times, and the pretreated WAS once under pressure of 30 bar resulted in an increase in VS removal and methane production of 5%, 9% over the intact WAS of 35%, 71%, respectively, in batchwise MAD of 6-day and 14-day retention time. With the pretreatment and MAD of 6-day retention time used, mesophlic bioconvertibility as the biogasification of WAS were found to be significantly higher biogas of 1, 850 ml than 300 ml under intact WAS. In conclusion it can be stated mechanical pretreatment enhances WAS bioconvertibility, while under identical treatment conditions, resulted in a considerable decrease in the bioconvertibility of intact WAS.

설태의 형광특성 - 설태 형광현상의 발현기전 소개 및 제안 - (Fluorescence of Tongue Coat - Mechanism of Tongue Coat Fluorescence and Suggestions on Development of Digital Tongue Diagnosis System -)

  • In traditional Korean medicine, inspection of the tongue is an important method of making medical diagnoses and determining prognosis. We surveyed the fluorescence characteristics of the tongue coat in the ultraviolet light. The tongue coat comprises micro-organisms, blood metabolites, leukocytes from periodontal pockets, large amounts of desquamated epithelial cells released from the oral mucosa and different nutrients. In the ultraviolet light tissues of the oral cavity generally emit weak red or green fluorescence, which is not easily seen by the human eye, but is readily detected. This fluorescence has been proved to be due to the production of porphyrins by oral micro-organisms. While the composition of motile micro-organisms on the dorsum of the tongue is not constant, variations also occur persistingly in the fluorescence characteristics of the tongue coat. But because live bacteria contain a variety of intracellular biomolecules that have specific excitation and emission wavelength spectra characterizing their intrinsic fluorescence, the tongue coat emits fluorescence. the tongue itself, on the other hand, emits very weak or not fluorescence. In conclusion, we suggests that the uncoated tongue area be eliminated from the coated tongue area with the difference between the fluorescence characteristics of the tongue and that of the tongue coat.
