To observe the intervening effect of resveratrol on coxsackie virus B3m-induced myocarditis in Balb/c mice and explore the mechanism of intervening effect. Using an animal model of viral myocarditis induced by coxsackie virus B3m (CVB3m), with Ribavirin and Astragalan as comparison, to examine the changes of general condition, mortality, the weights of heart, liver and spleen, serum MDA and NO levels, and cardiac histology in Balb/c mice. By comparison with Ribavirin and Astragalan, it was found that in the mice model of viral myocarditis induced by coxsackie virus B3m resveratrol significantly improved the changes of general condition, mortality, the weights of heart, liver and spleen, serum MDA and NO levels, and cardiac histology. It suggested that resveratrol may have some chemopreventive and chemotherapeutic effects in the treatment of viral myocarditis.
The importance of MICE information sharing has recently emerged in MICE industry. However it is very difficult to provide a solution that would enable MICE industry to come to an agreement to share MICE information. In this context, the purpose of this study is to analyze the major problems in information sharing process of MICE industry and to develop the paradigm model by ground theory approach and to propose a guideline for MICE information sharing which have not been treated as a research topic in the domestic MICE areas sofar.
Neonatal infection caused by Cronobacter species can result in serious illnesses such as bacteremia, septicemia, meningitis, and death in at-risk infants who are orally fed contaminated reconstituted powdered infant formulas. The objective of this study was to compare the virulence among three Cronobacter species strains by using an animal model for human neonatal Cronobacter species infections. We acquired timed-pregnant ICR mice and all owed them to give birth naturally. On postnatal day 3, each pup was administered orally a total dose of $1{\times}10^7$ CFU Cronobacter species strain 3439, CDC 1123-79, and 3231. Mice were observed twice daily for morbidity and mortality. At postnatal day 10, the remaining pups were euthanized, and brain, liver, and cecum were excised and analyzed for the presence of Cronobacter species. Cronobacter species were isolated from cecum and other tissues in inoculated mice. In the tissues of Cronobacter species infected mice, meningitis and gliosis were detected in the brain. In this study, we identified the virulence among Cronobacter species strains by using a neonatal mice model which was a very effective animal model for human neonatal Cronobacter species infections.
Ulcerative colitis (UC) is a serious gastrointestinal tract disease characterized by recurrent chronic inflammation and mucosal damage of the gastrointestinal tract. The conventional therapies of choice are anti-inflammatory agents, steroids and anti-TNF-${\alpha}$ therapy. However, inherent limitations in these therapies have steered many UC patients to supplement existing therapies with alternative medicinal products. In the current study, we tested the efficacy of Gingko bilola extract (EGb 761) in abating colonic inflammation in a DSS-induced murine model of colitis. C57BL/6 mice were administered 2% DSS in the drinking water for 7 days, then regular water for 7 days, and then 2% DSS for an additional 7 days. EGb 761 (1 mg/dose) was oral gavaged daily for the duration of the experiment. At the termination of the experiment, mice treated with EGb+DSS showed higher body weight, lower spleen weight and longer colon length compared to mice treated with DSS alone. HE-stained colon tissues also exhibited less histologic inflammation in mice treated with EGb+DSS mice compared to mice treated with DSS alone. The serum levels inflammatory cytokines, KC and TNF-${\alpha}$, were also decreased in mice treated with EGb+DSS compared to mice treated with DSS alone. Finally, addition of EGb 761 to TNF-${\alpha}$ treated colonic cell line (HT29/c1) decreased secretion of IL-8 in vitro. These results collectively suggest that EGb 761 abates induction of colitis in DSS-induced model of colitis in mice.
This study was undertaken to develop a subacute murine model for predicting occurrence of systemic anaphylaxis and to evaluate efficacy of various immunological parameters as the monitoring indices for the occurrence of anaphyalxis. The murine anaphyalxis model was developed through intraperitoneally sensitizing 100 $\mu\textrm{g}$ ovalbumin (OVA) in the presence of 1 mg alum and 300 ng cholera toxin twice a week interval followed by challenging 500 $\mu\textrm{g}$. OVA intravenously. Typical anaphylaxis symptoms were demonstrated at the both BALB/c mice, a strain prone to type-2 response, and C57BL/6 mice. a strain prone to type-1 response. Level of plasma histamine was approximately 50-fold or 30-fold higher in the mice sensitized with OVA than the mice sensitized with alum plus cholera toxin or the saline-treated mice after OVA challenge, respectively. Sensitization and challenge with OVA significantly enhanced plasma leukotriene $B_4$ level but not IgE levels in comparison with the control mice, which indicated the role of leukotriene $B_4$ for progression of anaphyalxis. Furthermore, among mice suffered from anaphylaxis, levels of OVA-specific IgGl were significantly higher in the BALB/c mice than in the C57BL/6 mice, which implied the genetic susceptibility for the induction of systemic anaphylaxis. Conclusively, simultaneous evaluation of histamine, leukotriene $B_4$, and allergen-specific IgG isotype may serve as more efficient monitoring tool for vaccine-related anaphyalxis.

Objective : A mouse model of spinal cord injury (SCI) could further increase our basic understanding of the mechanisms involved in injury and repair of the nervous system. The purpose of this study was to investigate whether methods used to produce and evaluate photochemical graded ischemic SCI in rats, could be successfully adapted to mice, in a reliable and reproducible manner. Methods : Thirty female imprinting control region mice (weighting 25-30 g, 8 weeks of age) were used in this study. Following intraperitoneal injection of Rose bengal, the translucent dorsal surface of the T8-T9 vertebral laminae of the mice were illuminated with a fiber optic bundle of a cold light source. The mice were divided into three groups; Group 1 (20 mg/kg Rose bengal, 5 minutes illumination), Group 2 (20 mg/kg Rose bengal, 10 minutes illumination), and Group 3 (40 mg/kg Rose bengal, 10 minutes illumination). The locomotor function, according to the Basso-Beattie-Bresnahan scale, was assessed at three days after the injury and then once per week for four weeks. The animals were sacrificed at 28 days after the injury, and the histopathology of the lesions was assessed. Results : The mice in group 1 had no hindlimb movement until seven days after the injury. Most mice had later recovery with movement in more than two joints at 28 days after injury. There was limited recovery of one joint, with only slight movement, for the mice in groups 2 and 3. The histopathology showed that the mice in group 1 had a cystic cavity involving the dorsal and partial involvement of the dorsolateral funiculi. A larger cavity, involving the dorsal, dorsolateral funiculi and the gray matter of the dorsal and ventral horns was found in group 2. In group 3, most of the spinal cord was destroyed and only a thin rim of tissue remained. Conclusion : The results of this study show that the photochemical graded ischemic SCI model. described in rats, can be successfully adapted to mice, in a reliable and reproducible manner. The functional deficits are correlated an increase in the irradiation time and, therefore, to the severity of the injury. The photothrombotic model of SCI, in mice with 20 mg/kg Rose bengal for 5 minutes illumination, provides an effective model that could be used in future research. This photochemical model can be used for investigating secondary responses associated with traumatic SCI.

Ionizing radiation causes the massive generation of reactive oxygen species, resulting in cellular and tissue damage. The present study was performed to evaluate ${\gamma}$-irradiation induced cellular damage in ICR mice. The mice were divided into four groups with ten mice in each group. Group 1 served as an unexposed control group. Groups 2, 3, and 4 were exposed to 3, 5, and 7 Gy of ${\gamma}$-radiation, respectively. Five mice per group were sacrificed 1 and 7 days after ${\gamma}$-radiation. Exposure to ${\gamma}$-irradiation resulted in hematopoietic damage in a dose-dependent manner when compared with the unexposed control group, which featured a significantly decreased spleen index. However, the exposed mice showed no significant differences in their serum AST, ALT and in the histopathological change of their liver. These results suggest that ${\gamma}$-irradiation is a good tool to prepare a hematopoietic damage model. This animal model can be employed to study the hematopoietic efficacy of biologically active compounds.
The altered amino acid (AA) levels as neurotransmitter closely correlate to neurodegenerative conditions including Alzheimer's disease (AD). Target profiling analysis of nineteen AAs in brain cortex samples from three Tg2576 mice as AD model and three littermate mice as control model was achieved as their N(O,S)-ethoxycarbonyl/tert-butyldimethylsilyl derivatives by gas chromatography. Subsequently, star pattern recognition analysis was performed on the brain AA levels of AD mice after normalization to the corresponding control median values. As compared to control mice, $\gamma$-aminobutyric acid among ten AAs found in brain samples was significantly reduced (P < 0.01) while leucine was significantly elevated (P < 0.02) in AD mice. The normalized AA levels of the three AD mice were transformed into distorted star patterns which was different from the decagonal shape of control median. The present method allowed visual discrimination of the three AD mice from the controls based on the ten normalized AA levels.

Purpose: Adenoid cystic carcinoma (ACC) is a relatively rare tumor that arises in glandular tissues of the head and neck region and sometimes has a protracted clinical course with perineural invasion and delayed onset of distant lung metastasis. Treatment failure of salivary ACC is most often associated with perineural and hematogenous tumor spread. However, very little has been known about the cellular and molecular mechanisms of perineural invasion and hematogenous distant metastasis of parotid ACC. This study was designed to develop an orthotopic tumor model of parotid adenoid cystic carcinoma in athymic nude mice. Experimental Design: A melanoma cell line was injected into the parotid gland of athymic mice to determine whether such implantation was technically feasible. A parotid ACC cell line was then injected into the parotid gland or the subcutaneous tissue of athymic mice at various concentrations of tumor cells, and the mice were thereafter followed for development of tumor nodule. The tumors were examined histopathologically for perineural invasion or regional or distant lung metastasis. We used an oral squmous cell carcinoma cell line as control. Results: Implantation of tumor(melanoma) cell suspension into the parotid gland of nude mice was technically feasible and resulted in the formation of parotid tumors. A parotid ACC cell line, ACC3 showed no significantly higher tumorigenicity, but showed significantly higher lung metastatic potential in the parotid gland than in the subcutis. In contrast, mucosal squmous cell carcinoma cell line doesn’t show significantly higher lung metastatic potential in the parotid gland than in the subcutis. The ACC tumor established in the parotid gland seemed to demonstrate perineural invasion of facial nerve, needs further study. Conclusion: An orthotopic tumor model of salivary ACC in athymic nude mice was successfully developed that closely recapitulates the clinical situations of human salivary ACC. This model should facilitate the understanding of the cellular and molecular mechanisms of tumorigenisis and metastasis of salivary ACC and aid in the development of targeted molecular therapies of salivary ACC.
The discovery of the immunodeficient mice has provided a tool for establishing animal models as hosts for in vivo analysis of AML. Various model systems have been established in the last few decades, and it is essential that murine AML models are developed to exploit more specific, targeted therapeutics. In this review, we concentrate on the models of AML and discuss the development of immunodeficiency models for understanding of leukemogenesis, describe those now available and their values and document the methods used for establishing and identifying AML mice models, as well as factors influencing engraftment of human AML in immunodeficient mice. Thus, the function of this article is to provide clinicians and experimentalists with a chronological, comprehensive appraisal of all AML model systems.
