Optimal Condition for the Production of Exopolysaccharide by Marine Microorganism 96CJ10356

  • Marine microorganism strain 96CJ10356 produced exopolysaccharides, designated as EPS-R. To optimize culture conditions for the production of EPS-R, carbon, nitrogen, mineral salt, temperature, and pH were examined. STN medium was suggested as follow; sucrose 20, tryptone 10, NaCl 10, $MgSO_4$ 5, $CaCl_2$ 1, $KH_2PO_4$ 0.0076, $K_2HPO_4$ 0.0083, $FeCl_2$ 0.005, $MnCl_2$ 0.001, $NaMoO_4$ 0.001, $ZnCl_2$ 0.001 (g/1) and pH 7.0 About 9.23 g/l of EPS-R was obtained from the STN medium after cultivation for 120 h at $25^{\circ}C$ in 5-liter jar fermentor with an aeration rate of 0.17 vvm. Apparent viscosity and flocculation activity of the culture broth were increased with the production of the EPS-R and the maximal values were reached to 415 cp and 1400 units/ml against 0.5 % activated carbon, respectively.

Effect of Cold Plasma on Total Polyphenol Content and Anti-Inflammatory Activities of Peanut (Arachis hypogaea L.) Hull

  • In recent studies, cold plasma has been used to induce exudation of polyphenols and flavonoids from food materials, leading to enhancement of functional properties. And it is known that polyphenols interact with inflammation related metabolism. The objectives of this study were to investigate the effects of cold plasma treatments on the increase of total phenolic content (TPC), total flavonoid content (TFC), and anti-inflammatory activities of 'Sinpalkwang' peanut (Arachis hypogaea L.) hull. Plasma treatments were carried out using a dielectric barrier discharge gas exchange system at different radicals and temperatures (O3-25℃, O3-150℃, NOx-150℃). Significant differences in TPC, TFC, and inflammatory mediator such as nitric oxide (NO) and tumor necrosis factor a (TNF-α) in lipopolysaccharide stimulated Raw 264.7 macrophages were observed between treated and non-treated peanut hull samples (p < 0.001). Cold plasma treated samples showed higher content (TPC: 2.87-2.93 mg/g sample, TFC: 0.96-0.98 mg/g sample) than non-treated sample (TPC: 2.47 mg/g sample, TFC: 0.78 mg/g sample). Cold plasma treated samples showed lower content of NO (3.3-5.0 uM) and TNF-α (141.4-162.2 ng/mL) than non-treated sample (NO: 11.1 uM, TNF-α: 210.2 ng/mL). This study suggests that cold plasma has potential to improve functionalities of food materials and that cold plasma treated peanut hull can be used as immune enhancing materials.

Effects of Paeoniae Radix Rubra on CRF, c-Fos and TH in the Forced Swimming Test (적작약(赤芍藥)이 강제수영부하시험에서 CRF, c-Fos 와 TH에 미치는 영향)

  • Objectives : For the purpose of verifying the anti-depressant effect of Paeoniae Radix Rubra(PR), the expression of corticotropin-releasing factor(CRF), c-Fos and tyrosine hydroxylase(TH) was evaluated after performing the Forced Swimming Test(FST). Methods : Spraque-Dawley rats were ingested PR extract(100mg/kg, 400mg/kg, p.o.) for 3 times prior to FST. And the expression of corticotropin-releasing factor(CRF), c-Fos in the paraventricular nucleus(PVN) and tyrosine hydroxylase(TH) in the locus coelureus(LC) and ventral tegmental area(VTA) was measured immunohistochemically after FST. Results : The duration of immobility was significantly decreased in PR 100mg/kg Group and PR 400mg/kg Group, in comparison with the control group (p<0.001). The expression of CRF in the PVN was significantly decreased in PR 400mg/kg Group in comparison of the control group (p<0.05). The expression of c-Fos in the PVN was rather significantly increased in PR 100mg/kg Group in comparison with the control group, while almost no change was demonstrated in PR 400mg/kg Group. The expression of TH was significantly decreased in VTA in comparison with the control group (p<0.05), but the number of expression cells in LC was slightly decreased in case of PR 100mg/kg group while it was increased in case of PR 400mg/kg Group. Conclusion : Judging from the result of the aforementioned tests, Paeoniae Radix Rubra has decreased immobility. In addition, it has also decreased the expression of CRF and the expression of TH in VTA, while the expression of c-Fos and of TH in LC has no significance. Therefore, it is believed that Paeoniae Radix Rubra has an anti-depressant effect by decreased immobility through the reduced expression of CRF and TH in VTA.

  • The analysis of Jerusalem artichoke showed that it contains 12.09% of Inulin. The results obtained from the examination of the conditions for fructose production by cultivating Pencillum sp 1 in the Jerusalem articoke medium were as follows: 1. The optimum amount of water added to Jerusalem artichoke was 2.5 $\ell$ of distilled water per ㎏ of fresh Jerusalem artichoke. It this case, the concentration of Inulin was 4% (w/v). 2. The optimum temperature was $30^{\circ}C$, the initial optimum pH was 5.0 and the optimum cultural period was 72 hours. 3. Shaking culture with 50 ml of the medium and 120 oscills/min in 500 ml shaking flask was most effective as the culture method. 4. 0.1% of $NH_4H_2PO_4$ as a nitrogen source, 0.001 of $FeSO_47H_20$ and 0.001% of $MgSO_47H_2$ as metal salts were most effective. 5. Fructose production continued to increase for 72 hours under the optimum conditions for cultivation and the highest production rate to the Inulin was 95.25%.

  • A strain producing extracellular endo-type inulase was selected from Actinomycetes isolated from soil, and identified as Streptomyces sp. The maximum inulase production was obtained with medium containing inulin 1.0%, yeast extract 1.0%, (NH$_4$)$_2$HPO$_4$ 0.4%, NH$_4$H$_2$PO$_4$0.8%, KCl 0.05%, MgSO$_4$ㆍ7$H_2O$ 0.05%, FeSO$_4$ㆍ7$H_2O$ 0.001% at 96 hours culture in jar fermentor. The endo-type inulase was considered to be an inducible enzyme produced by inulin only.

  • We describe methods for depositing high temperature superconducting films on textured metal tapes by reactive co-evaporation (RCE). We discuss how RCE can be used to deposit on moving tape in a continuous fashion in a Garching-style process. Results are presented on films deposited by RCE at Los Alamos on IBAD-MgO textured tapes. The performance achieved, attaining over 500A/cm-width in self-field at 75.5 K, is competitive with the best results obtained by other processes for coated conductors. Tape production throughput is critical for the economics of the process and high deposition rates achieved in RCE are attractive for this. We present a detailed cost analysis model for HTS deposition using an RCE Garching process. The results indicate that HTS deposition can cost $<$5/kA{\cdot}m$ in a scaled up manufacturing environment.

Warm Tensile Deformation Behavior and Constitutive Equation of Supersaturated Solid-Solutionized Al-9Mg Extruded Alloy

  • In this paper, as a purpose to apply the supersaturated solid-solutionized Al-9Mg alloy to the structural sheet parts of automotive, tensile tests were conducted under the various conditions and a constitutive equation was derived from the tensile test results. Al-9Mg alloy was produced using a special Mg master alloy containing Al2Ca during the casting process and extruded into the sheet. In order to study the deformation behavior of Al-9Mg alloy in warm temperature forming environments, tensile tests were conducted under the temperature of 373 K-573 K and the strain rate of 0.001/s~0.1/s. In addition, by using the raw data obtained from tensile tests, a constitutive equation of the Al-9Mg alloy was derived for predicting the optimized condition of the hot stamping process. Al-9Mg alloy showed uncommon deformation behavior at the 373 K and 473 K temperature conditions. The calculated curves from the constitutive equation well-matched with the measured curves from the experiments particularly under the low temperature and high strain rate conditions.

Heavy Metals and Pesticides with Maturing Process of Rehmanniae Radix Preparata (숙지황 제조과정에서 중금속과 잔류농약)

  • To obtain the risk assessment of hazardous materials with maturing process of Rehmanniae Radix Preparata, the residual heavy metals and pesticides in samples are surveyed. Concentration of As, Cd and Pb during the maturing process of Rehmanniae Radix Preparata of Korea are $0.147{\pm}0.007\;mg/L,\;0.025{\pm}0.001\;mg/L\;and\;0.193{\pm}0.021\;mg/L$, respectively. Concentration of As, Cd and Pb during the maturing process of Rehmanniae Radix Preparat of China are $0.0145{\pm}0.014\;mg/L,\;0.023{\pm}0.002\;mg/L\;and\;0.299{\pm}0.013\;mg/L$, respectively. Concentration of As, Cd and Hg remain unchanged during the maturing process of Rehmanniae Radix Preparata. Concentration of As, Cd and Pb in Rehmanniae Radix Preparat on the Korea are 3.53%, 6.66% and 5.16% for Korea Food & Drug Administration (KFDA) advisory level, respectively. The residual pesticides, such as aldrin, $BHC-{\alpha},\;BHC-{\beta},\;BHC-{\gamma},\;BHC-{\delta}$, captan, chlorfenapyr, chlorpyrifos, DDD, DDE, DDT-o,p, DDT-p.p, dieldrin, endosulfan-sulfate, $endosulfan-{\alpha}$, $endosulfan-{\beta}$, fenitrothion, penthoate, permethrin, quintozene, tolyfluanid and vinclozolin, are not detected in Rehmanniae Radix Preparat on the Korea and China. The hazardous materials in Rehmanniae Radix Preparata on the Korea and China are verified the safety of the residual heavy metals and pesticides compare with Korea Food & Drug Administration (KFDA) advistory level.

Anti-fibrotic effects of Orostachys japonicus A. Berger (Crassulaceae) on hepatic stellate cells and thioacetamide-induced fibrosis in rats

  • BACKGROUND/OBJECTIVE: Orostachys japonicus A. Berger (Crassulaceae) has been used in traditional herbal medicines in Korea and other Asian countries to treat various diseases, including liver disorders. In the present study, the anti-fibrotic effects of O. japonicus extract (OJE) in cellular and experimental hepatofibrotic rat models were investigated. MATERIALS/METHODS: An in vitro hepatic stellate cells (HSCs) system was used to estimate cell viability, cell cycle and apoptosis by MTT assay, flow cytometry, and Annexin V-FITC/PI staining techniques, respectively. In addition, thioacetamide (TAA)-induced liver fibrosis was established in Sprague Dawley rats. Briefly, animals were divided into five groups (n = 8): Control, TAA, OJE 10 (TAA with OJE 10 mg/kg), OJE 100 (TAA with OJE 100 mg/kg) and silymarin (TAA with Silymarin 50 mg/kg). Fibrosis was induced by treatment with TAA (200 mg/kg, i.p.) twice per week for 13 weeks, while OJE and silymarin were administered orally two times per week from week 7 to 13. The fibrotic related gene expression serum biomarkers glutathione and hydroxyproline were estimated by RT-PCR and spectrophotometry, respectively, using commercial kits. RESULTS: OJE (0.5 and 0.1 mg/ mL) and silymarin (0.05 mg/mL) treatment significantly (P < 0.01 and P < 0.001) induced apoptosis (16.95% and 27.48% for OJE and 25.87% for silymarin, respectively) in HSC-T6 cells when compared with the control group (9.09%). Further, rat primary HSCs showed changes in morphology in response to OJE 0.1 mg/mL treatment. In in vivo studies, OJE (10 and 100 mg/kg) treatment significantly ameliorated TAA-induced alterations in levels of serum biomarkers, fibrotic related gene expression, glutathione, and hydroxyproline (P < 0.05-P < 0.001) and rescued the histopathological changes. CONCLUSIONS: OJE can be developed as a potential agent for the treatment of hepatofibrosis.
