크실렌 노출로 인한 요중 메틸마뇨산 배설에 미치는 유전자 다형성 연구 (A Study on Polymorphism Affecting Excretion of Urinary Methylhippuric Acid due to Xylene Exposure)

  • Objectives : The purpose of this study was to investigate the effect of genetic polymorphism of cytochrome P450 2E1 (CYP2E1) and aldehyde dehydrogenase 2 (ALDH2) on the xylene metabolism. Methods : Among 247 workers, 116 were occupationally exposed to xylene and 131 were not. Workers exposed to xylene had different work such as spray, touch-up, mix & assist, and pre-treat. Questionnaire variables were age, sex, use of personal protective equipment, smoking, previous night's drinking and work duration. The urinary methylhippuric acid was measured in the urine collected in the afternoon and corrected by urinary creatinine concentration. The genotypes of CYP2E1 and ALDH2 were investigated by using PCR-RFLP (polymerase chain reaction-restriction fragment length polymorphism) methods with DNA extracted from venous blood. Results : 1. The urinary concentrations of o-, m-, and p-methylhippuric acid and total methylhippuric acid in the exposed group were significantly higher than those in the non-exposed group (p<0.001). 2. In multiple regression analysis, the urinary methylhippuric acid concentration was significantly influenced by exposure grade (Job-exposure matrixes), smoking, drug use and kind of protective equipment (p<0.1). 3. Genetic polymorphism of CYP2E1 and ALDH2 did not affect urinary methylhippuric acid level in the exposed group (p>0.05). Conclusions : Exposure grade, smoking, drug use and kind of protective equipment affected urinary methylhippuric acid level, whereas genetic polymorphism of CYP2E1 and ALDH2 did not. However, further investigation for the effect of genetic polymorphism on the metabolism of xylene with a larger sample size is needed.

특수 건강진단기관의 유기용제 분석 외부정도관리 실태 (The Status of Analytical External Quality Assurance Program of Organic Solvents for the Special Health Examination Laboratories)

  • It has been well known that organic industrial solvents such as toluene, xylene and styrene are environmental health hazards causing occupational diseases to workers. The detection of biological metabolites of toluene, xylene and styrene in blood, urine, breast milk, saliva, and hair of workers exposed to these organic industrial solvents is a useful tool for assessing the amount of organic industrial solvents. Therefor, the external analytical quality assurance program is extremely important work for increasing reliability of the data analyzed in the special health examination laboratory. We evaluated the results of analysis by Korean analytical quality assurance program from the second half 2007 to the first half 2009. The number of participants for analytical quality assurance program for both mandatory item and one of optional items, hippuric acid and methylhippuric acid were 116-122, 116-122 and 105-115 laboratories, respectively. The proficient rate of the special health examination laboratories analyzed both mandatory item and optional item, hippuric acid and methylhippuric acid were 92-98%, 92-98% and 96-98%, respectively. We can conclude from our data that the analytical quality assurance program is a essential tool for improvement of reliability on the data analyzed in the special health examination laboratories.

흰쥐에 Xylene반복 투여가 Xylene의 대사에 미치는 영향 (Effect of the Repeated Treatment of Xylene to the Rats on the Xylene Metabolism)

  • 실험동물에 xylene의 반복 투여가 이물질의 대사에 어떠한 영향을 미치는지를 알아보기 위하여 흰쥐에 m-xylene과 olive oil의 동량혼합액을 체중 100 g당 0.25 ml씩 2일 간격으로 1, 4, 8, 12 및 16회 복강으로 투여한 다음 마지막 투여 24시간 후에 처치하여 다음과 같은 결과를 얻었다. 요 중 m-methylhippuric acid함량은 m-xylene 4회 투여군의 경우 1회 투여군에 비하여 약 56%의 유의한 증가를 보였으며 이후 12회 투여까지 유사한 결과를 나타내었으나 m-xylene 16회 투여시에는 xylene 1회 투여군 치와 유사한 치로 감소되었다. 그리고 간조직의 microsomal aniline hydroxylase와 alcohol dehydrogenase 활성은 m-xylene 12회 투여시 까지는 대체적으로 점진적인 증가를 보였으나 이후 16회 투여시에는 12회 투여군에 비하여 유의한 감소를 보였다. 또한 aldehyde dehydrogenase 활성은 m-xylene투여 회수에 비례해서 전 실험기간 동안 감소되었으며 특히 16회 투여군에서 본 효소활성의 현저한 감소를 보였다. 한편 본 실험조건에서 투여기간에 따른 전자현미경적 미세구조의 변화는 초기에 활면소포체의 증식이 보이다가 16회 투여군에서는 활면소포체가 감소되고 조면소포체가 증가되었다. 이상 실험결과는 흰쥐에 있어서 xylene투여 회수에 따라서 요 중 m-methylhippuric acid의 농도 변동이 초래되며 이는 효소단백 유도에 따른 xylene 대사효소 활성 변동에 기인된 결과로 생각된다.

분석 방법에 따른 요중 마뇨산의 분석값 비교 (Comparison of analytical methods of hippuric acid in urine)

  • 이 연구의 목적은, 톨루엔에 노출된 근로자의 생물학적모니터링에 있어 혼재하는 유기산의 종류와 분석방법에 따른 요중 마뇨산 분석값을 비교하여, 다른 유기산이 혼재할 경우 UV법으로 분석했을 때 일어날수 있는 방해효과를 측정하는데 있다. 마뇨산만 포함된 소변 표준시료와 마뇨산, 메틸마뇨산, 페닐글리옥실산, 만델산 등 각각 다른 유기용제의 대사물이 포함된 소변 표준시료를 UV법, HPLC법으로 분석하고 유기산의 방해효과(IE; Inteference Efect)를 계산하였다. 또한 이들 소변 표준시료에 대해 전국 100개 검진기관을 대상으로 한 외부정도관리 결과, 참가기관들이 사용한 분석방법에 따른 요중 마뇨산 분석값을 비교하였다. 톨루엔과 크실렌에 노출된 근로자의 소변을 UV법으로 분석할 경우 크실렌 대사물인 메틸마뇨산은 마뇨산 분석값에 영향을 주어, 마뇨산 농도 0.33 g/L에서 0.5-2.0 g/L의 농도로 공존시 60-82%의 방해 영향을 가진 것으로 나타났다. 그러나 스티렌 대사물인 만델산, 페닐글리옥실산은 마뇨산 분석값에 거의 영향이 없었다. 따라서 톨루엔과 크실렌이 함께 노출되는 경우 소변 모니터링시 UV법을 사용하는 것보다 GC법이나 HPLC법을 사용하는 것이 바람직하다. 특히 특수검진기관의 의무 보유장비인 GC법은 경제적이고 신속하며 정확도가 높은 방법으로 UV법에 대체하여 사용할 수 있는 것으로 나타났다.

도료 제조업 근로자들의 복합유기용제 폭로농도에 관한 연구 (Evaluation of Mixed Organic Solvent Exposures in Painting Plants)

  • The exposure levels of mixed organic solvents for 66 exposed workers in six paint manufacturing plants were evaluated. In 66 exposed workers and 30 control subjects, we also determined the concentrations of toluene and xylene metabolites, hippuric acid, ($o^-$, $m^-$, and $p^-$)methylhippuric acid. The results were as follow ; 1. Seven organic compounds, which on averge accounted for approximately 90% of the identified mass in each painting plants air samples, were selected for quantification : methyl ethyl ketone, ethyl acetate, methyl isobutyl ketone, toluene, butyl acetate, ethyl benzene, ($o^-$, $m^-$, $p^-$)xylene. 2. The average mixed organic solvent exposure levels in 66 points with workplce were 3.8ppm of MEK, 12.2ppm of ethyl acetate, 4.0ppm of MIBK, 28.7ppm of toluene, 3.8ppm of butyl acetate, 10.2ppm of ethyl benzene, 14.6ppm of xylene, respectively. 3. For the total 66 points with workplace, the rate of them of which mixed solvents in air was exceeded th TLV of 1.0 were obtained for 23%(15/66 point). 4. The concentrations of hippuric acid in urine of exposed group and control were $0.94{\pm}0.65g/g$ of creatinine, $0.16{\pm}0.11g/g$ of creatinine, respectively. 5. There was a linear correlation between the end shift hippuric acid acid levels in urine and exposed toluene in air : y=0.02079X+494.2, r=0.6488, n=55 y:hippuric acid in urine(mg/g of creatinine), x:toluene levels in air(ppb) Toluene levels of 100ppm in air have been caculated to hippuric acid of 2.57g/g of creatinine in urine. 6. There was a linear correlation between the end shift methylhippuric acid acid levels in urine and exposed xylene in air : y=0.01664X+31.6, r=0.7264, n=55 y:methylhippuric acid in urine(mg/g of crea.), x:xylene levels in air(ppb) Xylene levels of 100ppm in air have been caculated to methylhippuric acid of 1.69g/g of creatinine in urine.

Determination of Derivatives of Phenol with a Modified Electrode Containing β-Cyclodextrin

  • Voltammetric determination of phenol derivatives, such as phenol, o-, m-, and p-cresols was studied with a β-Cyclodextrin (β-CD) modified-carbon paste electrode composing of the graphite powder and Nujol oil. Phenol derivatives were chemically deposited via the complex formation with β-CD by immersing the CME into a sample solution. The resulting surfaces were characterized with cyclic and differential pulse voltammetry. Treating the CME with 1 M nitric acid for five sec after a measurement could regenerate the electrode surface. Linear sweep and differential pulse voltammograms were recorded for the above system to optimize the experimental parameters for analysing the phenol derivatives. In this case, the detection limit for phenols was 5.0×10-7M for 25 min of the deposition time with differential pulse voltammetry. The relative standard deviation was ±5.2% of 3.0×10-6M (four repetitions). The interference effect of the following organic compounds was also investigated; Bezoic acid, hippuric acid, o-, m-, and p-methylhippuric acid. Adding the organic compounds into the sample solution reduces the peak current of the phenols to about 25%.

A Correlation between Urinary Methylhippuric Acid and Hematologic Finding and Urinalysis Index due to Xylene Exposure

  • This study was performed to investigate the urinary methyl hippuric acid (UMHA) excretion among aircraft maintenance workers exposed to xylene by examination of blood test, urine test and biochemical tests. Study subjects (301) were divided into 2 groups; non-exposed group (n=123, who were not exposed to xylene), and exposed group (n=178, who were exposed to xylene). The average of blood level and MHA level was normal. The ${\gamma}$-GTP level was 35.5 IU/L (45.7%) in the exposed group and 27.9 IU/L (44.7%) in the non-exposed (control) group. The proportion of total cholesterol (TG) level higher than 150 mg/dl were 34.3% in the exposed group and 21.1% in the non-exposed group. These results were showed statistically significant difference (${\chi}^2$ = 6.10, P<0.05). Other items showed no statistically significant differences. The results of urine tests were no statistically significant differences in protein, occult blood and glucose level.

External Quality Assessment Scheme for Biological Monitoring of Occupational Exposure to Toxic Chemicals

  • Objectives: In this study, we summarized the External Quality Assessment Scheme (EQAS) for the biological monitoring of occupational exposure to toxic chemicals which started in 1995 and continued until a $31^{st}$ round robin in the spring of 2010. The program was performed twice per year until 2009, and this was changed to once a year since 2010. The objective of the program is to ensure the reliability of the data related to biological monitoring from analytical laboratories. Methods: One hundred and eighteen laboratories participated in the $31^{st}$ round robin. The program offers 5 items for inorganic analysis: lead in blood, cadmium in blood, manganese in blood, cadmium in urine, and mercury in urine. It also offers 10 items for organic analysis, including hippuric acid, methylhippuric acid, mandelic acid, phenylglyoxylic acid, N-methylformamide, N-methylacetamide, trichloroacetic acid, total trichloro-compounds, trans,trans-muconic acid, and 2,5-hexanedione in urine. Target values were determined by statistical analysis using consensus values. All the data, such as chromatograms and calibration curves, were reviewed by the committee. Results: The proficiency rate was below 70% prior to the first round robin and improved to over 90% for common items, such as PbB and HA, while those for other items still remained in the range of 60-90% and need to be improved up to 90%. Conclusion: The EQAS has taken a primary role in improving the reliability of analytical data. A total quality assurance scheme is suggested, including the validation of technical documentation for the whole analytical procedure.

흰쥐 성장기간에 따른 Xylene의 독성에 관한 연구 (A Study on the Effect of Aging on the Xylene Toxicity in Rats)

  • 실험동물에 있어서 연령 차이에 따라서 xylene 독성이 어떠한 차이가 있는지를 검토하는 일환으로 5주령 및 12주령 흰쥐에 50% m-xylene을 체중 100 g 당 0.25 ml씩 1회 투여한 다음 24시간 후에 처치하여 다음과 같은 결과를 얻었다. Xylene투여로 인한 요 중 methylhippuric acid 함량은 5주령군이 12주령군에 비해 현저하게 높게 나타났다. 그리고 간조직 중 cytochrome P-450 함량은 대조군에 있어서 5주령군이 12주령군 보다 약 50% 정도 낮게 나타났으나 xylene 투여로 인한 cytochrome P-450 함량 증가율은 12주령군 보다 5주령군에서 높게 나타났다. 간 alcohol dehydrogenase 활성치도 대조군에 있어서는 5주령군이 12주령 보다 약 35% 정도 낮게 나타났으나 xylene 투여로 인한 본 효소의 활성 증가율은 5주령군에서 오히려 높게 나타났다. 그러나 간 aldehyde dehydrogenase 활성치는 대조군 및 xylene 투여군 모두 5주령과 12주령간에 유의한 차이를 나타내지 않았다. 한편 xylene 투여시 체중 당 간무게, 간조직 malondialdehyde 함량 및 혈청 ALT 활성 변동을 통하여 간손상 정도를 상호 비교 관찰하였을 때, 12주령군이 5주령 실험동물 보다 간손상이 다소 심하게 나타남을 알 수가 있었다. 이상 실험결과는 연령에 따라 xylene에 의한 간손상의 차이는 이물질의 생체내 대사율이 달리 나타나기 때문일 것으로 생각된다.
