  • Euonymus alatus (EAE) and Euonymus alatus for. ciliatodentatus (Franch. & Sav.) Hiyama (EACHE) belong to the Celastraceae family and are one of the medicinal herbs used in traditional medicine as a therapeutic agent for blood circulation diseases. In this study, we prepared leaves methanolic extract of EAE (L-EAE), twigs methanolic extract of EAE (T-EAE), leaves methanolic extract of EACHE (L-EACHE) and twigs methanolic extract of EACHE (T-EACHE), and compared their anti-inflammatory activities. To analyze the anti-inflammatory activities, Raw 264.7 cells were used, since they are immature-macrophages and easily matured by lipopolysaccharide (LPS) stimulation. All extracts showed anti-inflammatory activities in the activated Raw 264.7 cells. That is, we showed that L-EACHE and T-EACHE are potent inhibitors of the production of nitric oxide and pro-inflammatory cytokines. These results are expected to serve as a guide for future studies on the ability of Celastraceae family to inhibit acute and chronic inflammatory diseases.

  • From the methanolic extract of defatted tender leaf of Camellia sinensis a new 4-hydroxy angular furocoumarin $C_{12}H_8O_5$, m.p. $212^{\circ}C$, was isolated using high-speed counter-current chromatographic technique. The structure of the compound was established as 4-hydroxy-2'-methoxy angular furocoumarin on the basis of physical methods viz. $^1H$ NMR, $^{13}C$ NMR and MS.

Inhibitory Effect of Trans-N-p-Coumaroyl Tryamine from the Twigs of Celtis chinensis on the Acetylcholinesterase

  • The methanolic extract of the twigs of Celtis chinensis was found to show inhibitory activity on acetylcholinesterase (AChE), an enzyme that plays a role in the metabolic hydrolysis of ACh. Bioassay-guided fractionation of the methanolic extract resulted in the isolation of N-p-coumaroyl tyramine. as an inhibitor on AChE. This compound inhibited AChE activity in a dose-dependent manner, and the $IC_50$ value of trans-N-p-coumaroyl tyramine was 34.5 $\mu$g/mL (122 $\mu$M).

Protective effect of methanolic extract of Ganoderma lucidum P. Karst. Reishi from South India against doxorubicin-induced cardiotoxicity in rats

  • Doxorubicin is a powerful anticancer antibiotic extensively used in the treatment of several types of cancers. Long-term administration of this drug results in cumulative dose related cardiotoxicity due to enhanced production of free radicals leading to oxidative stress. Our earlier investigations have demonstrated significant antioxidant, anti-inflammatory and antitumour properties of Ganoderma lucidum extracts. We extended our investigations to evaluate the protective effect of Ganoderma lucidum extract against doxorubicin-induced cardiotoxicity. Administration of 3 doses of doxorubicin, 6 mg/kg body weights, i.p. per each dose, alternative days, showed dear signs of cardiotoxicity in rats. The drug enhanced serum creatine kinase (CK) activity and lipid peroxidation in tissue drastically. The drug also induced significant decrease in GSH level and activities of CAT, SOD and GPx. Administration of methanolic extract of G.lucidum (500 and 1,000 mg/kg body weight) significantly increased the level of GSH and activities of CAT, SOD and GPx. Activity of CK was significantly lowered in a dose dependent manner. The treatment also caused significant decrease in lipid peroxidation (MDA). The results thus indicated that methanolic extract of G.lucidum prevented oxidative stress caused by doxorubicin administration and the increase in serum CK activity and lipid peroxidation in the tissue. The experimental findings suggest the therapeutic potential of G.lucidum as adjuvant in cancer chemotherapy.

포도씨 추출물과 분획물의 Tyrosinase 저해활성 (Inhibitory Effect of Methanol Extract and Its Fractions from Grape Seeds on Mushroom Tyrosinase)

  • 본 연구에서는 포도 가공 부산물인 포도씨를 이용하여 피부노화 및 색소침착에 관련된 tyrosinase 저해활성을 알아보고자 하였으며 폴리페놀 화합물과 총항산화력과의 상관관계를 비교·분석하고자 하였다. 포도씨 80% 메탄올 추출물은 hexane, chloroform, ethyl acetate, water 층의 순서로 분획 되었다. Tyrosinase 저해활성은 각 분획물보다 crude한 형태인 methanol 추출물의 효과가 더 높았으며 분획물 중 ethyl acetate 층이 가장 높은 tyrosinase 저해활성을 나타내었고 추출물과 분획물 모두 농도 의존적인 경향을 나타내었다. 포도씨 분획물 중 ethyl acetate층이 polyphenol과 flavonoid 함량이 가장 높았고 ABTS 라디칼 소거능 또한 가장 높은 결과를 나타내었으며 water 분획층도 비교적 높은 활성을 나타내었다. 본 연구 결과 tyrosinase 활성과 항산화 성분,항산화 활성 경향이 일치하는 것을 볼 수 있었으며 포도씨는 미백, 항노화 효과로 화장품 산업에서 천연 물질로 잠재적인 기능성을 가지고 있을 것으로 사료된다.

대두(Glycine maxim) 추출물이 티로시나아제 프로모터 활성에 미치는 효과 (Effects of Glycine maxim Extract on the Activity of Tyrosinase Promoter)

  • The methanolic extract of Glycine maxim increased the expression of the promoter in B16 mouse melanoma cells harboring a tyrosinase promoter. Extract concentrations of 10 ${\mu}g/m{\ell}$ and 100 ${\mu}g/m{\ell}$ resulted in tyrosinase promoter expression rates of approximately 113% and 184%, respectively, as compared to the control. The fraction layers consisting of butyl alcohol and methylene chloride improved expression effects on the tyrosinase promoter. In particular, the butyl alcohol fraction evidenced a high expression rate at 100 ${\mu}g/m{\ell}$. In the MTT assay, the methanolic extract did not evidence cytotoxicity at concentrations under 500 ${\mu}g/m{\ell}$. Therefore, the results observed with the extract of Glycine maxim showed that the substance exerted a positive effect on the tyrosinase promoter.

Preliminary antimicrobial and cytotoxic activities of Amoora cucullata extractives

  • Amoora cucullata (Meliaceae), a mangrove plant, has folkloric reputation as a medicinal agent in Bangladesh. In this study, the n-hexane, ethyl acetate and methanolic extracts of the stem bark of this plant were subjected to microbiological investigation and brine shrimp lethality bioassay. In case of antimicrobial screening, the ethyl acetate and methanolic extracts appeared to be potent in terms of both zone of inhibition and spectrum of activity showing the average zones of inhibition 8 - 14 mm and 9 - 16 mm, respectively. In the brine shrimp lethality bioassay, the methanolic extract demonstrated highest cytotoxicity having $LC_{50}$ of $0.549{\mu}g/ml$, whereas the ethyl acetate and n-hexane extract showed $LC_{50}$ of 7.943 and $17.180{\mu}g/ml$, respectively.

Characterization of Antioxidant Potential of a Methanolic Extract and Its Fractions of Highbush Blueberry (Vaccinium corymbosum L.)

  • The antioxidant potential of a $75\%$ methanolic extract of highbush blueberry (Vaccinium corymbosum L.) and its different fractions was investigated using different reactive oxygen species (ROS), nitric oxide (NO.), metal chelating and lipid peroxidation assays. Methylene chloride and $75\%$ methanol fractions showed equally high activities $(IC_{50} 0.010 mg/mL)$ for hydroxyl radical (HO) scavenging. Higher hydrogen peroxide $(H_2O_2)$ scavenging values were reported for the ethyl acetate and methylene chloride fractions and their $IC_{50}$ values were 0.20 and 0.15 mg/mL, respectively. Nitric oxide (NO.) and 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) scavenging activities were higher in ethyl acetate and methylene chloride fractions. Chloroform and water fractions showed higher activities in superoxide $(O_2.)$ scavenging. All fractions showed strong metal chelating capacities compared with the commercial antioxidants tested. The $0.1\%$ ethyl acetate fraction showed notable capacity to suppress lipid peroxidation in both fish oil and linoleic acid. Phenolic content was measured in all the fractions and methanolic extract. Among the fractions, ethyl acetate fraction showed the highest phenolic content.

  • Methanolic extract of the leaves of Cassia tom was evaluated for its hepatoprotective potentials in rats by inducing hepatotoxicity with carbon tetrachloride. The extract at a dose of 400 mg/kg orally showed significant protective effect by lowering the serum levels of transaminase (SGOT and SGPT), bilirubin and alkaline phosphatase (ALP). The effects produced were comparable to that of a standard hepatoprotective agent.

Antihyperglycemic activity of Biophytum sensitivum (L.) DC in alloxan diabetic rats

  • The study was to evaluate the antihyperglycemic activity of Biophytum sensitivum (L.) DC in different extracts. Albino Wistar rats with alloxan hydrate induced diabetes were divided into 7 groups of 6 each. Both aqueous and methanolic extract of Biophytum sensitivum were prepared and given individually at different doses to different batches of rats (both normal and diabetic rats) after an overnight fast. Methanolic extract at the dose of 200 mg/kg body weight showed maximum blood glucose lowering effect in diabetic rats. The same dosages did not produce any hypoglycemic activity in normal rats. The antihyperglycemic activity of Biophytum sensitivum was compared with a standard drug Glibenclamide, an oral hypoglycemic agent. The above results suggest that maximum hypoglycemic effect was found only with a dose of up to 200 mg/kg b.w. of methanolic extract which is therefore the optimum dose for hypoglycemia and was used in all the experiments of the present study.
