Deoxynivalenol (DON), a Fusarium mycotoxin, causes health hazards for both humans and livestock. Therefore, the aim of this study was to investigate the metabolic profiles of the liver, serum, and urine of piglets fed DON using proton nuclear magnetic resonance ($^1H-NMR$) spectroscopy. The $^1H-NMR$ spectra of the liver, serum, and urine samples of the piglets provided with feed containing 8 mg DON/kg for 4 weeks were aligned and identified using the icoshift algorithm of MATLAB $R^2013b$. The data were analyzed by multivariate analysis and by MetaboAnalyst 4.0. The DON-treated groups exhibited discriminating metabolites in the three different sample types. Metabolic profiling by $^1H-NMR$ spectroscopy revealed potential metabolites including lactate, glucose, taurine, alanine, glycine, glutamate, creatine, and glutamine upon mycotoxin exposure (variable importance in the projection, VIP > 1). Forty-six metabolites selected from the principal component analysis (PCA) helped to predict sixty-five pathways in the DON-treated piglets using metabolite sets containing at least two compounds. The DON treatment catalyzed the citrate synthase reactions which led to an increase in the acetate and a decrease in the glucose concentrations. Therefore, our findings suggest that glyceraldehyde-3-phosphate dehydrogenase, citrate synthase, ATP synthase, and pyruvate carboxylase should be considered important in piglets fed DON contaminated feed. Metabolomics analysis could be a powerful method for the discovery of novel indicators underlying mycotoxin treatments.
Sanguinarine은 천연 항균성 물질로 의약품, 생활용품 그리고 화장품 등 그 이용은 다양하나 가격이 비싸고 수율이 따르지 못하는 점을 식물세포배양을 통하여 해결할 수 있을 것으로 기대된다. Sanguinarine의 생산을 극대화하기 위하여 전통적인 방법인 환경적 요인을 고려한 cell selection방법과 유전적 접근인 protein의 변화를 동시에 관찰하였다. Yeast extract를 elicitor로 이용하였을 경우에 control에서는 관찰 할 수 없었던 sanguinarine에 해당하는 오렌지색의 형광을 볼 수 있었으며, 실제 metabolite의 분석에서도 sanguinarine의 증가를 확인 할 수 있었다. 세포 내부에서는 sanguinarine이 약 8배의 증가를 보였으며 배지에서는 약 5배의 증가를 보였다. Protein 역시 2-D electrophoresis로 확인한 결과 intensity가 $5\%,\;88\%,\;6.4\%$의 증가, 일정, 감소를 보인 spot들이 elicitor 처리 후 세포에서는 $34\%,\;39.4\%\;26.5\%$로 intensity가 증가된 spot들이 더많이 검출되었다. 본 연구에서는 sanguinarine을 yeast extract를 이용해서 생산량을 증가시키고 sanguinarine의 생산과 관련된 protein의 변화에 대해서 알아보고자 하였다. 본 예와 같은 실험 연구방법이 식물이차대사산물 생산성에 관련된 protein군을 규명하는데 초석이 될 수 있을 것으로 기대된다.
반하(Pinellia ternata Breitenbach)는 천남성과(Araceae)의 약용 식물로 동아시아 일대가 원산지이며, 유럽과 북아메리카 일부 지역에도 널리 분포하고 있는 식물이다. 반하의 괴경(tuber)은 약재로 사용되어 구토, 염증, 외상을 치료하는 목적으로 사용되고 있다. 보고된 약리학적 작용으로는 항경련, 항종양, 살충, 세포독성 등이 있는 것으로 알려져 있다. 반하는 약리학적으로 유용한 약용식물로 잘 알려져 있음에도 불구하고, 원산지를 판별할 수 있는 신뢰성이 있고 표준화된 방법이 없는 실정이다. 이를 위한 한국산과 중국산 반하를 판별할 수 있는 유의적인 화합물을 탐색하기 위해 UPLC-PDA와 QTof-MS에 기반을 둔 대사체 표지법을 이용하였다. 한국산으로부터 원산지 판별 화합물로 예상되는 화합물을 반복적인 역상 칼럼크로마토그래피에 기반을 둔 활성유도 분획법을 통해 분리하였다. 그리고, NMR과 MS를 포함한 물리화학적, 분광학적 정보의 결과를 토대로 [6]-gingerol이라는 화합물의 구조를 동정하였다. [6]-gingerol은 원산지를 판별할 수 있는 능력과 melanin 생합성 저해 활성을 지닌 기능성 화장품 소재로서 가치가 있다고 판단되어짐에 따라 유효성분 원산지 판별 화합물로 Fingerprint법에 의해 선정되었다. 더욱이, 한국산과 중국산 반하 유래 [6]-gingerol의 함량 비교를 위해, 유효성이 검증된 분석법을 이용하여 이에 대한 검량곡선을 작성하여 그 함량을 비교하였다. 이것은 한국산 반하판별을 위한 화합물 선정과 성공적인 유효성 검증을 다룬 최초의 보고이다.
Red ginseng marc (RGM) has been used on primary industries using fertilizer or forage, and it mostly has been dumped. To improve utilization of RGM, the biological activities of RGM were examined. RGM was extracted and fractionated using various solvents and their biological activities were compared. The hexane fraction from the methanol extract of RGM (RGMMH) showed strong anti-cancer activity (58.56 ± 6.04% at 100 ㎍/mL) and anti-inflammatory effect (65.72 ± 1.33% at 100 ㎍/mL). But, oil extract of RGM extracted with hexane (RGMH) showed low activities (anti-cancer: 16.42 ± 3.33%, at 100 ㎍/mL, anti-inflammatory activity: 29.46 ± 2.10%, at 100 ㎍/mL). Their metabolites were analyzed using HPLC. Panaxydol known as anti-cancer compound of RGM was one of major compounds in RGMMH. Meanwhile, panaxydol was detected in trace amount in red ginseng marc oil (RGMH). In addition, RGMMH and RGMH showed big differences in HPLC profiling. This research suggests optimal extraction method of RGM oil.
This study investigated the effect of proline addition on the salt tolerance of Tetragenococcus halophilus. Salt stress led to the accumulation of intracellular proline in T. halophilus. When 0.5 g/l proline was added to hyperhaline medium, the biomass increased 34.6% (12% NaCl) and 27.7% (18% NaCl) compared with the control (without proline addition), respectively. A metabolomic approach was employed to reveal the cellular metabolic responses and protective mechanisms of proline upon salt stress. The results showed that both the cellular membrane fatty acid composition and metabolite profiling responded by increasing unsaturated and cyclopropane fatty acid proportions, as well as accumulating some specific intracellular metabolites (environmental stress protector). Higher contents of intermediates involved in glycolysis, the tricarboxylic acid cycle, and the pentose phosphate pathway were observed in the cells supplemented with proline. In addition, addition of proline resulted in increased concentrations of many organic osmolytes, including glutamate, alanine, citrulline, N-acetyl-tryptophan, and mannitol, which may be beneficial for osmotic homeostasis. Taken together, results in this study suggested that proline plays a protective role in improving the salt tolerance of T. halophilus by regulating the related metabolic pathways.

The levels of 12 isoflavones were measured in soybean (Glycine max (L.) Merrill) sprouts of 68 genetic varieties from three countries (China, Japan, and Korea). The isoflavone profile differences were analyzed using data mining methods. A principal component analysis (PCA) revealed that the CSRV021 variety was separated from the others by the first two principal components. This variety appears to be most suited for functional food production due to its high isoflavone levels. Partial least squares discriminant analysis (PLS-DA) and orthogonal projections to latent structures discriminant analysis (OPLS-DA) showed that there are meaningful isoflavone compositional differences in samples that have different countries of origin. Hierarchical clustering analysis (HCA) of these phytochemicals resulted in clusters derived from closely related biochemical pathways. These results indicate the usefulness of metabolite profiling combined with chemometrics as a tool for assessing the quality of foods and identifying metabolic links in biological systems.
Species are facing strong selection pressures to adapt to inhospitable high-altitude environments. Yaks are a valuable species and an iconic symbol of the Qinghai-Tibet Plateau. Extensive studies of high-altitude adaptation have been conducted, but few have focused on metabolism. In the present study, we determined the differences in the serum metabolomics between yaks and the closely related species of low-altitude yellow cattle and dairy cows. We generated high-quality metabolite profiling data for 36 samples derived from the three species, and a clear separation trend was obtained between yaks and the other animals from principal component analysis. In addition, we identified a total of 63 differentially expressed metabolites among the three species. Functional analysis revealed that differentially expressed metabolites were related to the innate immune activation, oxidative stress-related metabolism, and energy metabolism in yaks, which indicates the important roles of metabolites in high-altitude adaptation in yaks. The results provide new insights into the mechanism of adaptation or acclimatization to high-altitude environments in yaks and hypoxia-related diseases in humans.
국제 올림픽 위원회에서 금지하고 있는 합성 동화성 스테로이드로서 다양한 생물학적 효과를 나타내고 있는 danazol을 대상으로 androgenic 효과를 확인하기 위해 성인 남자 3명에게 100 mg씩 복용하게 한 후 그들의 소변을 채취하였다. 인체내 대사과정에서 생성되는 주요 대사체인 ethisterone의 배설에 따른 8가지의 내인성 스테로이드들의 배설정도를 GC/MS의 selected ion monitoring(SIM) 방법으로 동시분석 하였다. 그 결과 본실험방법에서 내인성 스테로이드들에 대한 회수율은 71.59%~93.56% 이었으며, within-a day 및 day-to-day 분석에서의 RSD 값은 각각 1.87%~10.48%와 1.32~11.25%이었다. 이때의 검출한계는 $0.01{\sim}0.05{\mu}g/mL$ 이었다. 각각의 내인성 스테로이드들과 ethisterone의 표준물질을 뇨시료에 0.1, 0.5, 1.0, 10, 20 그리고 $50{\mu}g/mL$를 첨가하여 작성한 검정곡선은 전체적으로 상관계수 0.963~0.991의 직선성을 보였다. 따라서 본 실험은 내인성 스테로이드들의 변화를 관찰함으로서 간접적으로 스테로이드 계열의 금지약물 복용여부를 판단할 수 있는 방법으로 매우 유용함을 알 수 있었다.
Background: The genus Panax in the Araliaceae family has been used as traditional medicinal plants worldwide and is known to biosynthesize ginsenosides and phytosterols. However, genetic variation between Panax species has influenced their biosynthetic pathways is not fully understood. Methods: Simultaneous analysis of transcriptomes and metabolomes obtained from adventitious roots of two tetraploid species (Panax ginseng and P. quinquefolius) and two diploid species (P. notoginseng and P. vietnamensis) revealed the diversity of their metabolites and related gene expression profiles. Results: The transcriptome analysis showed that 2,3-OXIDOSQUALENE CYCLASEs (OSCs) involved in phytosterol biosynthesis are upregulated in the diploid species, while the expression of OSCs contributing to ginsenoside biosynthesis is higher in the tetraploid species. In agreement with these results, the contents of dammarenediol-type ginsenosides were higher in the tetraploid species relative to the diploid species. Conclusion: These results suggest that a whole-genome duplication event has influenced the triterpene biosynthesis pathway in tetraploid Panax species during their evolution or ecological adaptation. This study provides a basis for further efforts to explore the genetic variation of the Panax genus.
식물 대사체 (plant metabolomics) 연구는 식물 세포 및 조직에 존재하는 모든 대사산물의 시간적, 공간적 변화를 추적 조사함으로써 식물의 복잡한 생리 현상을 총체적으로 이해하는 연구이다. 이와 같은 식물 대사체 연구는 최근 개발이 이루어지고 있는 여러 오믹스 연구 분야의 하나로 시스템생물학의 한 분야이다. 식물 대사체 연구는 시료로부터 순수 화합물 또는 복합물을 정제하거나 또는 정제가 이루어지지 않은 혼합액으로부터 대사체 스펙트럼 정보를 확보하여 분석이 이루어지므로 추출액 제조 및 얻어진 대사체 데이터의 분석과정의 표준화가 필수적으로 이루어져야 한다. 이는 대사체 분석 결과의 해상도 및 재현성의 확보의 핵심 요소 이다. 식물 대사체 연구는 기능유전체학의 연구 수단은 물론 식물의 종, 품종, 더 나아가 GM 식물의 식별, 대사조절 기작 규명, 유용물질 생산, 식물의 외부 환경 스트레스 요인에 대한 다양한 생리적 반응 이해 등 다양한 연구 분야에서 활용이 이루어지고 있다. 최근 식물 대사체 연구는 모델식물(벼, 애기장대)의 유전체 정보와 연계하여 돌연변이주의 분석을 통해 유전자의 기능 정의 수단으로 활용되고 있다. 따라서 향후 유전체 정보와 대사체 정보의 연계를 통해 복잡한 대사경로 규명이나 다양한 생리 현상 해석 연구가 더욱 활발하게 진행될 것으로 전망된다.
