Establishing the Genotoxicological Safety of Gamma-irradiated Egg White and Yolk (감마선 조사 계란의 유전독성학적 안전성 평가)

  • The genotoxicological safety of gamma-irradiated egg white and yolk was examined to ensure that required safety parameters were met, and in an effort to further apply gamma-irradiation for improvement of the hygienic qualities of eggs. Egg white and yolk were irradiated at 20 kGy, much higher than the legally approved dose (less than 5 kGy), and possible genotoxicity was evaluated using in vitro and in vivo tests. The SOS chromotest employing Escherichia coli PQ37, and a chromosomal aberration test in cultured Chinese hamster lung (CHL) cells, were performed in vitro with or without metabolic activation (S9). An in vivo micronucleus development test was conducted using mouse bone marrow cells. Negative results were obtained in the SOS chromotest. The incidence of chromosomal aberration in CHL cells and the frequency of micronuclear developmentin mouse bone marrow cells treated with irradiated samples were not significantly different from those of non-irradiated controls. Thus, it may be concluded that up to 20 kGy of gamma irradiation applied to egg white and yolk did not show any genotoxic effects under our experimental conditions.

Relationships of Obesity in Childhood to Plasma Lipids, Blood Pressure and Blood Glucose (아동기 비만이 혈장 지질, 혈압 및 혈당에 미치는 영향)

  • In order to investigate the relationships of obesity in childhood to plasma lipid, blood pressure and blood glucose concentration, we selected 21 subjects for the moderate obese group(MO), 9 for mild obese group(MI), and 19 for the control group(C) among children aged 10~12. While the level of plasma triglyceride and VLDL-cholesterol of the MO group was much higher than that of group C, a significantly lower percentage of HDL-cholesterol was found in the MO compared to the percentage found in group C. Also the level of the total cholesterol and LDL-cholesterol of the MI group as well as the MO was much higher than that of C. The elevated total-cholesterol level of the Mi group was due to increased LDL-cholesterol and that of the MO was due to increases in both VLDL-cholesterol and LDL-cholesterol. As the result of these differences, the atherogenic index of the MO was significantly higher than that of C. The incidence of hypercholesterolemia( 200mg/dl) of the MI and MO was 60.0% and 77.8% respectively, All of the physical parameters and indexes except height were positively correlated with plasma lipid levels, systolic blood pressure, and blood glucose concentration. The analyses of the correlated with plasma lipid levels, systolic blood pressure, and blood glucose concentration. The analyses of the correlation indicated that central fat to peripheral fat ratio and waist to thigh girth ratio seemed to be closely associated to plasma lipid levels and atherogenic index. The MO had significantly higher systolic blood pressure than C and significantly higher blood glucose concentration was found in both MI and MO than in C. These results confirmed that obesity in childhood may be relevant to chronic metabolic diseases such as abnormal lipid metabolism, atherosclerosis, high blood pressure and diabetes mellitus.

Effect of Fructose-1,6-diphosphate[FDP] on Red Blood Cells after Extracorporeal Circulation (체외순환후 fructose-1,6-diphosphatate[FDP]가 적혈구에 미치는 영향)

  • Extracorporeal cardiopulmonary bypass[CPB] has been associated with a wide variety of hematologic derangements, including a transient deformation and hemolysis of red blood cells[RBCs], which is supposed to be due to mechanical trauma and/or metabolic alterations. Since membrane integrity is, in part, maintained by energy requiring process, inadequate function of erythrocyte glycolytic pathway, which is inevitalble during CPB, may cause depletion of high energy phosphate pool and result in hemolysis. The authors performed an investigation to assess whether administration of Fructose-l, 6-diphsphate [FDP], which has been known to enhance intracellular glycolytic activities, could counteract erythrocyte hemolytic events caused by CPB. Sixty pateints with cyanotic congenital heart diseases, who underwent open heart surgery under CPB longer than 60 minutes, were randomly divided into two groups depending on whether use of FDP[Group FDP] or not[Group Control]. The age, sex, CPB time, preoperative hemoglobin level, disease entities were all similar[Table 1], and membrane type oxygenators were used in all patients. In Group, FDP, a dose of 250mg/kg body weight of FDP was administered by intravenous dripping every 12 hours from the morning of the operation to postoperative 48 hours, To demonstrate the degree and pattern of hemolysis of erythrocyte, reticulocyte count, indirect /direct bilirubin, haptoglobin, plasma hemoglobin, lactate dehydrogenase were measured every 12 hours from the time of cessation of CPB to 48 hours and RBC morphologic study, osmotic fragility test were done every 24 hours. All parameters revealed less hemolytic in group FDP [Fig. 1~5], though the differences between two groups were not significant, except plasma hemoglobin, lactate dehydrogenase changes. A pattern of sequential changes of plasma hemoglobin, lactate deh-ydrogenase showed the highest level at the time of CPB stop and abrupt decrease in following 24 hours in both groups, and statistically significant differences were demonstrated in group FDP at least for the first 12 hours postoperatively[p<0.05]. The authors conclude that they can expect the benificial effect of FDP on the maintenance of membrane stability of RBC probably by energy enhancement during the shock status of CPB, but FDP could not completely prevent the damaging effect on RBC by cardiopulmonary bypass

  • This study was conducted to investigate the application possibility of leaf and stem extract(LSE) extracted from mixture of leaf and stem of ginseng radix (Panax Ginseng C.A. Meyer). We conducted analysis of the ginsenoside content by HPLC. Also we investigate the effects of the LSE on the reduction of serum lipid and improvement of blood parameters in rats fed high fat diet 5 weeks. We examined by analyzing the serum total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride and atherogenic index and hematological datas and serum metabolic variables. Sprague-Dawley rat weigh $150\;g\;{\pm}\;15\;g$, were ramdomly assigned to 4 groups, basal diet only(BDG), high fat diet weithout LSE(FDCG), high fat diet and 10% LSE(FD10G), high fat diet and 20% LSE(FD20G). The result of this study were as follow. Hematological datas of 4 groups were same level, which were not significant. The activities of ALP, GOT and LDH level were significantly different. Total cholesterol, LDL-cholesterol, triglyceride contentrations in serum and atherogenic index were remarkably reduced in LSE supplemented groups as compared high fat control groups. These result imply that LSE could be used as possible for decrease of serum lipid concentration.

Simulation of the Effect of Protein Quality at the Different Protein Intake Level on Protein Metabolism (각기 다른 단백질섭취 수준에서 본 식이단백질의 질이 단백질대사에 미치는 영향 -Simulation Model을 이용하여-)

  • This study was designed to describe the effect of the protein quality at different intake level of protein on the protein metabolism in the whole body of growing pigs with a simulation model. Varying to the protein level in feeds, four simulations were conducted. The feed protein level, represented as proportions of digestible protein to the metabolic energy (DP/ME, g/MJ), were 6-8, 11-13, 17-19, and 23-25 DP/ME, respectively. Two protein quality and six weeks of growth time were used at each simulation. The objective function for the simulations was protein deposition in the whole body, which was calculated from the experimental results. The parameters in the simulation were determined by the parameter estimation technique. The results obtained from the simulation were as follows: The protein synthesis and breakdown rates(g/day) in the whole body was increased with the increase of protein quality only at lower or required level of protein intake. They showed a parallel behavior in the course of growth, irrespective of quality and level of feed protein intake. The simulated protein deposition and protein synthesis showed a linear relationship between them at different protein quality and level. The affinity parameter showed a linear relationship between them at different protein quality and level. The affinity parameter showed that arginine, tryptophan and isoleucine were more efficient in the stimulation ofbody protein synthesis. Lysine and phenylalanine+tyrosine were less efficient. The oxidation parameter showed that histidine, pheyalanine+tyrosine were less efficient. The oxidation parameter showed that histidine, phenyalanine+tyrosine, and methionine+cystine were oxidized in larger magnitude than lysine and threonine. The oxidation parameter of most amino acids increased with the increase of protein intake beyond the requirement level, but not any more at highest protein intake level. Finally it was found that the improvement of feed protein quality at the lower or required level of protein intake increase protein deposition through a parallel increase of protein synthesis and breakdown.

A survey of research papers on the health benefits of kimchi and kimchi lactic acid bacteria (김치 및 김치 유래 유산균의 건강 기능성에 대한 연구 동향 조사)

  • Purpose: This review article provides an overview of the trends of research papers on the health benefits of kimchi and kimchi lactic acid bacteria published from 1995 to 2017. Methods: All publications from 1995 to 2017 regarding kimchi and kimchi lactic acid bacteria were collected, reviewed, and classified. This review article covers the publications of the health benefits of kimchi and kimchi lactic acid bacteria on experimental, clinical trials, and epidemiology studies. Results: The number of publications on kimchi over the period were 590: 385 publications in Korean and 205 publications in English. The number of publications on the health benefits of kimchi and kimchi lactic acid bacteria were 95 in Korean and 54 in English. The number of publications on kimchi and kimchi lactic acid bacteria were 84 and 38, respectively, in the experimental models. Ten research papers on kimchi in clinical trials and 7 publications in epidemiology were found. Kimchi or kimchi lactic acid bacteria had protective effects against oxidative stress, mutagenicity, toxicity, cancer, dyslipidemia, hypertension, immunity, and inflammation in in vitro, cellular, and in vivo animal models. Moreover, kimchi had effects on the serum lipids, intestinal microbiota, iron status, obesity, and metabolic parameters in human clinical trials. In epidemiology, kimchi had effects on hypertension, asthma, atopic dermatitis, rhinitis, cholesterol levels, and free radicals. Conclusion: This review focused on the publications regarding the health benefits of kimchi and kimchi lactic acid bacteria, suggesting the future directions of studies about kimchi and kimchi lactic acid bacteria by producing a database for an evaluation of the health benefits of kimchi.

Efficacy and Safety of Deflazacort in Korean Children with Nephrotic Syndrome (신증후군 환아에 대한 Deflazacort의 효과와 안전성)

  • Deflazacort, an oxazoline derivative of prednisolone, has been claimed to have anti-inflammatory effects with fewer side effects compared to prednisone. The objectives of the study were to evaluate efficacy and safety of deflazacort in children with nephrotic syndrome. Eligible Patients were the children with nephrotic syndrome who were treated with deflazacort from October. 1994 to April. 1999. Nephrotic syndrome was defined as having albumin level of less than 2.5 mg/dL and 24-hour urinary protein excretion of greater than $40\;mg/m^2/hr$. The primary parameters evaluating the efficacy of deflazacort were response rate, time to respond and relapse frequency. The safety profiles were the impact on children's growth, calcium sparing effect, glucose metabolism, lipid profile and adverse drug reactions. As results, total of 60 children were evaluated (47 boys, 13 girls). Response rate was $95\%$ (57/60) for initial and late responders. Median time to respond was 12 days (range 7-110 days) and median relapse frequency was one time (range 0-6). Weight/height ratio increased from $22.05\pm3.47\;to\23.20\pm3.44\;kg/m$ (p<0.001) and plasma calcium level, from $7.55\pm3.86\;to\;9.98\pm3.77\;mg/dL$ after treatment (p<0.001). Change of fasting glucose level was not statistically significant $(91.92\pm3.53\;vs.\;98.19\pm4.78\;mg/dL,\;p=0.072)$, while change of total cholesterol was significant $(362.3\pm12.0\;vs\;251.4\pm11.5\;mg/dL$, p<0.001). In conclusion, patients on deflazacort showed similar efficacy in treatment of nephrotic syndrome as reported for prednisone with less impact on growth inhibition and metabolic side effects of hyperglycemia and hyperlipidemia.

Effects of Combination Therapy with Anti-Obesity Herbal Medicine Including Ephedra Herba and Lorcaserin in Obese Patients: Two Case Reports (마황이 포함된 한방비만 처방과 Lorcaserin 병용 투여 후 호전된 비만 환자 치험 2예)

  • Although there has been many studies about the co-administration of herb formula and western medicine, there is none about obesity. The aim of this study was to report the effects of co-administration of herb formula containing ephedra sinica and lorcaserin on two obese patients for about 2~4 months. During the treatment, we checked body weight and body composition, and collected blood samples to evaluate liver, kidney, thyroid function and lipid profile. This treatment not only decreased body weight but also improved body composition and lipid profile in both patients. There was no hepatic or renal toxicity, nor any serious side effects of treatment. This study demonstrated that co-administration of herb formula containing ephedra sinica and lorcaserin is a safe and effective therapy to lose weight and improve metabolic parameters for obese patients. Further large-scale clinical trials are needed to evaluate the anti-obesity effect of the combination of herbal and western medicines.

Development of a Redox Dye-Based Rapid Colorimetric Assay for the Quantitation of Viability/Mortality of Pine Wilt Nematode

  • Control of pine wilt disease, which is caused by pine wilt nematode Bursaphelenchus xylophilus, is heavily dependent on the use of chemicals such as abamectin. Although such chemicals are highly effective, demands for alternatives that are derived preferentially from natural sources, are increasing out of environmental concerns. One of the challenges to discovery of alternative control agents is lack of fast and efficient screening method that can be used in a high-throughput manner. Here we described the development of colorimetric assay for the rapid and accurate screening of candidate nematicidal compounds/biologics targeting B. xylophilus. Contrary to the conventional method, which relies on laborious visual inspection and counting of nematode population under microscope, our method utilizes a redox dye that changes its color in response to metabolic activity of nematode population in a given sample. In this work, we optimized parameters of our colorimetric assay including number of nematodes and amount of redox dye, and tested applicability of our assay for screening of chemicals and biologics. We demonstrated that our colorimetric assay can be applied to rapid and accurate quantification of nematode viability/mortality in a nematode population treated with candidate chemicals/biologics. Application of our method would facilitate high-throughput endeavors aiming at finding environment-friendly control agents for deadly disease of pine trees.

Plasma Adropin as a Potential Marker Predicting Obesity and Obesity-associated Cancer in Korean Patients With Type 2 Diabetes Mellitus

  • Background: Type 2 diabetes mellitus (T2DM) and cancer are serious health problems worldwide, and their prevalences have been on the rise in recent years. It has been reported that adropin plays an important role in the development of T2DM, oxidative stress, inflammation, and obesity. However, there is limited information available on T2DM from human studies, especially for the Korean population. In this study, we aimed to investigate the correlation between adropin levels and obesity of Korean T2DM patients. Methods: Thirty-six T2DM patients were recruited for this study. The participants were further classified into female (n = 12) and male (n = 24). Their body composition, metabolic parameters, inflammatory factors, and oxidative stress were measured. Results: The severity of obesity is more manifested in male than in female. Plasma triglyceride (TG) and high-sensitivity C-reactive protein (hs-CRP) levels of male were significantly higher than female. The plasma adropin and adiponectin level of female was significantly higher than male. The body weight, body mass index (BMI), body fat mass were negatively correlated with the plasma adropin level in female, whereas adropin has positive correlation with adiponectin in female. The hs-CRP was negatively correlated with the plasma adropin level in female and male. malondialdehyde, reactive oxidative species, and $TNF-{\alpha}$ was not significantly correlated with adropin in patients with T2DM. Conclusions: These findings suggest that adropin may be more used as a biomarker for predicting the risk of obesity and inflammation in Korean patients with T2DM, especially women.
