Evaluation of Ethanol Production Activity by Engineered Saccharomyces cerevisiae Fermenting Cellobiose through the Phosphorolytic Pathway in Simultaneous Saccharification and Fermentation of Cellulose

  • In simultaneous saccharification and fermentation (SSF) for production of cellulosic biofuels, engineered Saccharomyces cerevisiae capable of fermenting cellobiose has provided several benefits, such as lower enzyme costs and faster fermentation rate compared with wild-type S. cerevisiae fermenting glucose. In this study, the effects of an alternative intracellular cellobiose utilization pathway-a phosphorolytic pathway based on a mutant cellodextrin transporter (CDT-1 (F213L)) and cellobiose phosphorylase (SdCBP)-was investigated by comparing with a hydrolytic pathway based on the same transporter and an intracellular ${\beta}$-glucosidase (GH1-1) for their SSF performances under various conditions. Whereas the phosphorolytic and hydrolytic cellobiose-fermenting S. cerevisiae strains performed similarly under the anoxic SSF conditions, the hydrolytic S. cerevisiae performed slightly better than the phosphorolytic S. cerevisiae under the microaerobic SSF conditions. Nonetheless, the phosphorolytic S. cerevisiae expressing the mutant CDT-1 showed better ethanol production than the glucose-fermenting S. cerevisiae with an extracellular ${\beta}$-glucosidase, regardless of SSF conditions. These results clearly prove that introduction of the intracellular cellobiose metabolic pathway into yeast can be effective on cellulosic ethanol production in SSF. They also demonstrate that enhancement of cellobiose transport activity in engineered yeast is the most important factor affecting the efficiency of SSF of cellulose.

  • The effects of dietary cellulose to utilize the intake nutrients in the albino rats were investigated by the two consecutive experiments in this study. The author wanted to observe the effects of the age of animals and different level of dietary cellulose, high and low in the first experiment and the effects of dietary cellulose from three different sources, rice bran, vegetable and pure-cellulose at high level in the second experiment. The results were summarized as follows. (1) The obvious effect of dietary cellulose caused to increase feces weight. Consequently apparent digestibilities of intake nutrients in the diet decreased. It was noteworthy that fecal glucose excretion was significantly affected by cellulose intake. Since three nutrients, protein, fat and carbohydrate serve as energy sources in the body, this result can be interpreted to be lowered in the efficiency of energy utilization out of intake from the diet. (2) The nutrients loss due to dietary cellulose resulted in reduction of skeletal muscle and bone weights which lead eventually to affect in reduction of body weight. (3) The effects of dietary cellulose showed differently according to the amount of cellulose ingested, dietary nutrients composition, the kinds of dietary cellulose supplemented and the age of experimental animals. When the intake diet was proportionately imbalanced in three different energy sources, carbohydrate, fat and protein, the cellulose intake was adversely affected in the utilization of the nutrients in the body. Cellulose from rice bran decreased body weight gain, cellulose from vegetable maintained medium level in body weight and pure-cellulose increased body weight. Growing rats showed more sensitive effects of cellulose tolerance in the body than grown rats in every concerns of this experiment.

Wastewater-based epidemiology for the management of community lifestyle and health: An unexplored value of water infrastructure (하수기반역학을 이용한 커뮤니티 생활상 및 건강 관리: 물 인프라의 새로운 가치)

  • Traditional wastewater research mainly focuses on 1) estimating the amount of waste entering sewage treatment facilities, 2) evaluating the treatment efficiency of sewage facilities, 3) investigating the role of sewage treatment effluent as a point source, and 4) designing and managing sewage treatment facilities. However, since wastewater contains a variety of chemical and biological substances due to the discharge of human excreta and material used for daily living into it, the collective constituents of wastewater are likely a reflection of a community's status. Wastewater-based epidemiology (WBE), an emerging and promising field of study that involves the analysis of substances in wastewater, can be applied to monitor the state of a defined community. WBE provides opportunities for exploiting indicators in wastewater to fulfill various objectives. The data analyzed under WBE are those pertaining to selected natural and anthropogenic substances in wastewater that are a result of the discharge of metabolic excreta, illicit or legal drugs, and infectious pathogens into the wastewater. This paper reviews recent progress in WBE and addresses current challenges in the field. It primarily discusses several representative applications including the investigation of drug consumption across different communities and the management of community disease and health. Finally, it summarizes established indicators for WBE.

  • $^{18}F$-fludeoxyglucose PET (FDG-PET) can help finding an abnormal metabolic activity in brain. In this study, we evaluated an efficiency of volume- and cortical surface-based analysis which were used to determine whether standardized uptake value ratio (SUVR) of FDG-PET was different among Alzheimer's disease (AD), mild cognitive impairment (MCI), and healthy control (HC). Each PET image was rigidly co-registered to the corresponding magnetic resonance imaging (MRI) using mutual information. All voxels of the co-registered PET images were divided by the mean FDG uptake of the cerebellum cortex which was thresholded by partial volume effect (>0.9). Also, the SUVR value of each vertex was linearly interpolated from volumetric SUVR image which was thresholded by gray matter partial volume effect (>0.1). Lobar mean values were calculated from both volume- and cortical surface-based SUVRs. Statistical analysis was conducted to compare two measures for AD, MCI and HC groups. Even though the results of volume (SUVR_vol) and cortical surface-based SUVR (SUVR_surf) analysis were not significantly different from each other, the latter would be better for detecting group differences in SUVR of PET.

Effects of Engineered Saccharomyces cerevisiae Fermenting Cellobiose through Low-Energy-Consuming Phosphorolytic Pathway in Simultaneous Saccharification and Fermentation

  • Until recently, four types of cellobiose-fermenting Saccharomyces cerevisiae strains have been developed by introduction of a cellobiose metabolic pathway based on either intracellular β-glucosidase (GH1-1) or cellobiose phosphorylase (CBP), along with either an energy-consuming active cellodextrin transporter (CDT-1) or a non-energy-consuming passive cellodextrin facilitator (CDT-2). In this study, the ethanol production performance of two cellobiose-fermenting S. cerevisiae strains expressing mutant CDT-2 (N306I) with GH1-1 or CBP were compared with two cellobiose-fermenting S. cerevisiae strains expressing mutant CDT-1 (F213L) with GH1-1 or CBP in the simultaneous saccharification and fermentation (SSF) of cellulose under various conditions. It was found that, regardless of the SSF conditions, the phosphorolytic cellobiose-fermenting S. cerevisiae expressing mutant CDT-2 with CBP showed the best ethanol production among the four strains. In addition, during SSF contaminated by lactic acid bacteria, the phosphorolytic cellobiose-fermenting S. cerevisiae expressing mutant CDT-2 with CBP showed the highest ethanol production and the lowest lactate formation compared with those of other strains, such as the hydrolytic cellobiose-fermenting S. cerevisiae expressing mutant CDT-1 with GH1-1, and the glucose-fermenting S. cerevisiae with extracellular β-glucosidase. These results suggest that the cellobiose-fermenting yeast strain exhibiting low energy consumption can enhance the efficiency of the SSF of cellulosic biomass.

Nitrogen metabolism and mammary gland amino acid utilization in lactating dairy cows with different residual feed intake

  • Objective: This study was conducted to enhance our understanding of nitrogen (N) metabolism and mammary amino acid (AA) utilization in lactating cows with divergent phenotypes of residual feed intake (RFI). Methods: Fifty-three multiparous mid-lactation Holstein dairy cows were selected for RFI measurements over a 50-d experimental period. The 26 cows with the most extreme RFI values were classified into the high RFI (n = 13) and low RFI (n = 13) groups, respectively, for analysis of N metabolism and AA utilization. Results: Compared with the high RFI cows, the low RFI animals had lower dry matter intake (p<0.01) with no difference observed in milk yield between the two groups (p>0.10). However, higher ratios of milk yield to dry matter intake (p<0.01) were found in the low RFI cows than in the high RFI cows. The low RFI cows had significant greater ratios of milk protein to metabolizable protein (p = 0.02) and milk protein to crude protein intake than the high RFI cows (p = 0.01). The arterial concentration and mammary uptake of essential AA (p<0.10), branched-chain AA (p<0.10), and total AA (p<0.10) tended to be lower in the low RFI cows. Additionally, the low RFI cows tended to have a lower ratio of AA uptake to milk output for essential AA (p = 0.08), branched-chain AA (p = 0.07) and total AA (p = 0.09) than the high RFI cows. Conclusion: In summary, both utilization of metabolizable protein for milk protein and mammary AA utilization are more efficient in cows with lower RFI than in the high RFI cows. Our results provide new insight into the protein metabolic processes (related to N and AA) involved in feed efficiency.

The Protective Effects of Dioscoreae Rhizoma on the Exposure to UVA of MEF cells (산약의 Mouse embryonic fibroblast cell에 대한 자외선 손상 방어효과)

  • Purpose: This study was to determine the protective effects of Dioscoreae Rhizoma on the Mouse Embrio Fibroblast (MEF) cells exposed to the ultraviolet rays(UVA). Methods: The samples were assigned randomly to five groups; control group without any treatments, UVA group exposed only to UVA, DR group exposed only to the Dioscoreae Rhizoma, UVA-DR group exposed to UVA before being treated with the Dioscoreae Rhizoma, and DR-UVA group treated with the Dioscoreae Rhizoma before being exposed to UVA. The survival rate of cells, metabolic rate of cells, transformation of nucleus within cells, alteration of cell cycle, effects on the apoptosis, the change of the amount of protein related to cell cycle were measured in order to determine the cell protective effects of the Dioscoreae Rhizoma on each group. Results: 1. DR-UVA group has more cell protective effects compared to the UVA group in all experiments, indicating that the Dioscoreae Rhizoma protects skin from UVA physically and chemically. 2. UVA-DR group shows more efficiency compared to UVA group in rapid recovery of damaged cell and leading highly damaged cells to apoptosis, preventing the expression of abnormal cells. Conclusions: Dioscoreae Rhizoma has effects of protecting MEF cells from UVA, of recovering cells damaged by UVA, and of prohibiting the expression of abnormal cells.

Species Specificity Evaluation for Wastewater Treatment Application of Alkaliphilic Microalgae Arthrospira platensis (호염기성 미세조류 Arthrospira platensis의 폐수처리 적용을 위한 종특이성 평가)

  • Since the efficiency of wastewater treatment using microalgae differs depending on the metabolic characteristics of the species, it is important to understand the characteristics of target algae prior to the application in wastewater treatment. In this study, for the application of Arthrospira platensis to wastewater treatment, which is a filamentous alkaliphilic cyanobacteria, basic species specificity was identified and the possibility of application to wastewater treatment was investigated. As a result of the species specificity investigation, the specific growth rate between pH 7.0 and 11.0 showed the highest value near pH 9 at 0.25/day. The reason for the relatively low growth(0.08/day) at pH 11 was thought to be the CA(carbonic anhydrase) enzyme that is involved in carbon fixation during photosynthesis has the highest activity at pH 8.0 to 9.0, and at pH 11, CA activity was relatively low. In addition, A. platensis showed optimal growth at 400 PPFD(photosynthetic photon flux density) and 30℃, and this means that cyanobacteria such as A. platensis have a larger number of PS-I(photosystem I) than that of PS-II(photosystem II). It was speculated that it was because higher light intensity and temperature were required to sufficiently generate electrons to transfer to PS-I. Regarding the applicability of A. platensis, it was suggested that if a system using the synergistic effect of co-culture of A. platensis and bacteria was developed, a more efficient system would be possible. And different from single cocci, filamentous A. platensis expected to have a positive impact on harvesting, which is very important in the latter part of the wastewater treatment process.

Effects of ambient temperature and dietary glycerol addition on growth performance, blood parameters and immune cell populations of Korean cattle steers

  • Objective: This study was performed to evaluate whether ambient temperature and dietary glycerol addition affect growth performance, and blood metabolic and immunological parameters, in beef cattle. Methods: Twenty Korean cattle steers ($405.1{\pm}7.11kg$ of body weight [BW], $14.2{\pm}0.15$ months of age) were divided into a conventional control diet group (n = 10) and a 2% glycerol- added group (n = 10). Steers were fed 1.6% BW of a concentrate diet and 0.75% BW of a timothy hay diet for 8 weeks (4 weeks from July 28th to August 26th and 4 weeks from August 27th to September 26th). Blood was collected four times on July 28th, August 11th, August 27th, and September 26th. Results: The maximum indoor ambient temperature-humidity index in August (75.8) was higher (p<0.001) than that in September (70.0), and in August was within the mild heat stress (HS) category range previously reported for dairy cattle. The average daily gain (ADG; p = 0.03) and feed efficiency (p<0.001) were higher in hotter August than in September. Glycerol addition did not affect ADG and feed efficiency. Neither month nor glycerol addition affected blood concentrations of cortisol, triglyceride, or non-esterified fatty acid. Blood concentrations of cholesterol, low-density lipoprotein, high-density lipoprotein, glucose, and albumin were lower (p<0.05) on August 27th than on September 26 th, and blood phosphorus, calcium and magnesium concentrations were also lower on August 27th than on September 27th. Glycerol addition did not affect these blood parameters. Percentages of $CD4^+$ T cells and $CD8^+$ T cells were higher (p<0.05) on July 28th than on August 27th and September 26th. The blood $CD8^+$ T cell population was lower in the glycerol supplemented-group compared to the control group on July 28th and August 27th. Conclusion: Korean cattle may not be significantly affected by mild HS, considering that growth performance of cattle was better in hotter conditions, although some changes in blood metabolic and mineral parameters were observed.

A study on verification of exercise intensity and efficiency in Nurida-ball exercise equipment (누리다 볼 운동기구의 운동 강도와 운동 효율성 검증연구)

  • The aim of this study was to evaluate the physiological responses following acute and chronic Nurida-ball exercise to measure the relative exercise intensity and health-related fitness in middle-aged men. In study 1, eight middle-aged men carried out acute Nurida-ball exercise for 30 min. The physiological responses were normalized and compared with a graded treadmill exercise test (GXT) to evaluate the relative exercise intensity. In study 2, twelve middle-aged men were divided into two groups: Nurida-ball exercise group (NB, n=6) and control group (CON, n=6). NB group performed the Nurida-ball exercise for 8 weeks (30min/day, 3 days/week). And then, we measured the body composition, cardiovascular factors, blood lipid factors, and muscle damage markers to confirm the exercise efficiency. In study 1, the related exercise intensity of acute Nurida-ball is the range of GXT-HRpeak (55~85%) and $GXT-VO_2peak$(23~61%). The change in body fat-related factors was significantly lower in the NB compared to the CON group. Moreover, TC and LDL were decreased whereas BMC was increased in the NB group after Nurida-ball exercise. The change in VEpeak and performance time of GXT were significantly greater in the NB compared to the CON group. The performance time of GXT was increased in the NB group after Nurida-ball exercise. Our results indicated that Nurida-ball exercise would be effective equipment for the improvement of health-related fitness. Especially, Nurida-ball exercise can affect body composition and cardiovascular function, which might alleviate metabolic syndrome-related diseases in middle-aged men.
