Transition Metal Induces Apoptosis in MC3T3E1 Osteoblast: Evidence of Free Radical Release

  • Transition metal ions including $Se^{2+},\;Cd^{2+},\;Hg^{2+}\;or\;Mn^{2+}$ have been thought to disturb the bone metabolism directly. However, the mechanism for the bone lesion is unknown. In this study, we demonstrated that MC3T3E1 osteoblasts, exposed to various transition metal ions; selenium, cadmium, mercury or manganese, generated massive amounts of reactive oxygen species (ROS). The released ROS were completely quenched by free radical scavengers-N-acetyl cysteine (NAC), reduced glutathione (GSH), or superoxide dismutase (SOD). First, we have observed that selenium $(10\;{\mu}M),$ cadmium $(100\;{\mu}M),$ mercury $(100\;{\mu}M)$ or manganese (1 mM) treatment induced apoptotic phenomena like DNA fragmentation, chromatin condensation and caspase-3-like cysteine protease activation in MC3T3E1 osteoblasts. Concomitant treatment of antioxidant; N-acetyl-L-cysteine (NAC), reduced-form glutathione (GSH), or superoxide dismutase (SOD), prevented apoptosis induced by each of the transition metal ions. Catalase or dimethylsulfoxide (DMSO) has less potent inhibitory effect on the apoptosis, compared with NAC, GSH or SOD. In line with the results, nitroblue tetrazolium (NBT) stain shows that each of the transition metals is a potent source of free radicals in MC3T3E1 osteoblast. Our data show that oxidative damage is associated with the induction of apoptosis in MC3T3E1 osteoblasts following $Se^{2+},\;Cd^{2+},\;Hg^{2+}\;or\;Mn^{2+}$ treatment.

  • Mordant Red 19(MR19) is reduced at mercury electrode at -0.67 V vs. Ag/AgCl with two electrons per molecule in pH 9.2 buffer by differential pulse polarography and linear sweep voltammetry. The peak potential is dependent on the pH of solution. The exhaustive electrolysis, however, gives 4 electrons per molecule because of the disproportionation of the unstable hydrazo intermediate. The electrochemical reduction of lanthanide-MR19 complexes is observed at more cathodic potential than that of free ligand. The difference in peak potentials between complex and free ligand varies from 75 mV for $La^{3+}$ to 165 mV for $Tb^{3+}$ and increases with increasing the atomic number of lanthanide. The electrochemical reduction of lanthanide complexes with MR19 is due to the reduction of ligand itself, and it can be potentially useful as an indirect method for the determination of lanthanides. The shape of i-E curves and the scan rate dependence indicates the presence of adsorption and the adsorption was confirmed by potential double-step chronocoulometry and the effect of standing time. Also the surface excess of the adsorbed species and diffusion coefficients are determined. The composition of the complex is determined to be 1 : 2 by spectrophotometric and electrochemical methods.

Effect of $Li_2O$ and $Na_2O$ addition on the microstructure and humidity sensitivity of $V_ 2O_5$-doped $TiO_2$ ($V_2O_5$를 도판트한 $TiO_2$의 미세구조와 감습에 미치는 알카리 옥사이드$ M_2O(M=Li,Na)$의 영향)

  • In this paper, the effect of alkaline oxide addition such as Li$_{2}$O and Na$_{2}$O on the microstructure and humidity sensitivity of V$_{2}$O$_{5}$(2 mol%)-doped TiO$_{2}$(98 mol%) was investigated as a function of amount (0, 1, 2, 5, 10 mol%) of Li$_{2}$O and Na$_{2}$O additives. The pores in the alkaline free sample were distributed mostly in the range between 0.16 and 1.0.mu.m in diameter and its porosity was 23.29%. Li$_{2}$O caused grain overgrowth and reduced the porosity with a narrow distribution of the pore size, leading to poor humidity sensitivity. Na$_{2}$O helped to enlarge the distribution of the pore size through the formation of small soluble phases. The pore sizes of the sample containing Na$_{2}$O 2mol% were distributed mostly in the range between 1.0 and 2.5.mu.m in diameter and its porosity and intrusion volume of mercury were 31.13 % and 0.1155 mL/g respectively, which consequently improved the humidity sensing characteristics such as the sensitivity and temperature-stability. Especially, the addition of 2mol% of Na$_{2}$O improved the humidity-sensing characteristics such as sensitivity and linearity in the whole range between 30 and 90 %RH (Percentage Relative Humidity)y)

  • Mercury-free lamp, external electrode fluorescent lamp (EEFL) which includes the xenon gas, is now going on the research for the replacement of mercury lamp. The densities of excited xenon atom in the $1s_4$ resonance state and the $1s_5$ metastable state are investigated in the EEFL by a laser absorption spectroscopy under various gas pressures. We have measured the absorption signals for both $1s_4$ resonance and the $1s_5$ metastable state in the EEFL by varying the discharge currents for a given pressure. This basic absorption characteristic is very important for improvement of the VUV luminous efficiency of the EEFL as well as plasma display panel.

  • 탄소나노튜브를 이용한 이미터 전계방출 면광원용 안정기를 제안한다. 안정기는 고전압 직류전압 부분과 양극의 펄스를 생성하는 부분으로 구성되어 있다. 탄소나노튜브를 이용한 전계방출 램프는 3가지의 전극 (애노드, 게이트, 캐소드)으로 구성되어 있는데, 애노드와 게이트 사이에는 고전압 직류가 공급되고 게이트와 캐소드 사이에는 양극의 펄스가 공급된다. 램프 및 안정기를 보호하기 위하여 과전류, 과전압, 과온도에 대한 보호 기능을 추가하였고, 실험을 통하여 제안된 방식이 탄소나노튜브 램프 구동을 위한 적합함을 검증하였다.

  • 최근에 환경친화적인 관점에서 많은 형태의 무수은을 이용한 형광램프가 연구되어오고 있다. 그 중에서도 제논을 이용한 형광램프는 수은을 대체하는 방전기체의 조건을 만족하고 있다. 따라서 제논을 이용한 면광원의 구동 시 지금의 수은이 들어간 광원의 구동에 있어서 안정기요건이 차이가 있으므로, 본 논문에서 제논을 이용한 면광원을 위한 안정기를 제안한다. 제안된 면광원 안정기는 크게 AC입력 측의 PFC (power factor correction)부분과 면광원을 구동하는 인버터 부분으로 구성되어있다. 특히, 인버터 부분에서는 제논 면광원의 특성에 대응하기 위해서 정전력회로, 아크 방지 기동회로 등이 포함되어 있다.

  • In this paper, parameters of electron temperature and density for the mercury-free lighting-source were measured to diagnosis and analyze in Xe based inductively coupled plasma(ICP). In results at several dependences of 20~100 mTorr Xenon pressure, 50~200W RF power and horizontal distribution were especially mentioned. When Xe pressure was 20mTorr and RF power was 200W, the electron temperature and density were respectively 3.58eV and $3.56{\times}10^{12}cm^{-3}$. The key parameters of Xe based ICP depended on Xe pressure more than RF power that could be verified. A high electron temperature and low electron density with a suitable Xe pressure are indispensible parameters for Xe based ICP lighting-source.

  • The ammonium salt of Nitrosophenylhydroxylamine "called cupferron", is a well-known analytical reagent, which precipitates a great number of metal ions in acid medium. The electrode reduction velocities of cupferron at droping mercury electrode in various supporting electrolytes were measured by Delahay method. The free energy of activation and transfer coefficient for a given electrode reaction are computed from the corresponding current-voltage curve. The number of electrons involved in the rate determining step of the reduction of cupferron is calculated for solutions of various pH.

  • Malfunction of Klippon relay in Cheju-Haenam HVDC system has been caused by the inflow of high voltage arc. In this paper, we have studied on the theoretical examination and data analysis of Klippon relay, and the countermeasures against the problems were suggested according to their causes. Grounding problem in Klippon relay is removed by one-point earth connection and by modification of grounding circuit. The effects of over current surge was removed by attaching blocking diodes by series In Klippon relay to remove surge input in Klippon relay's mercury contact. The problem of Induced overvoltage by 86re1ay excitation coil, was removed by attaching Free-wheeling diode in parallel with the excitation coil of Lock-out relay.
