Interaction Between Barbiturate and Membrane Components

  • Intermolecular interaction between barbiturates and membrane components such as phospholipid and cholesterol were investigated on $^1$H-NMR spectra and infrared spectra. According to previous reports, barbiturates interacted with phospholipid through intermolecular hydrogen bonds. We also investigated thi observation using dipalmitoyl-phosphatidylcholine (DPPC) as phospholipid in deuterochloroform, and characterized quantitatively. Also, the observed drug could interact with cholesterol which is one of the major components of biomembranes through hydrogen bonds. It was the carbonyl groups of barbiturate and the hydroxyl group of cholesterol that formed hydrogen bond complex. In addition to spectroscopic studies, we investigated the direct effect of phenobarbital on lipid multibilayer vesicles, whose compositions were varied, by calorimetric method. Phenobarbital caused a reduction in the temperature of phase transition of vesicles. These studies may provided a basis for interpreting the mode of action of barbiturates.

대두사포닌이 침투된 인공 인지질 생체유사막의 열에 의한 상변화에 관한 연구 (The Thermotropic Phase Behaviors of Artificial Phospholipid Liposomes Incorporated with Soyasaponin)

  • 인공 생체유사막을 인지질인 dimyristoyl phosphatidylcholine (DMPC)로 제조하여 인지질 막에 미치는 대두사포닌의 영향을 시차열량 분석계로 연구하였다. 대두사포닌이 침투되기 전, 후의 인지질막의 상그림을 얻어 상 전이시 엔탈피의 변화와 협동단위수를 상법에 따라 계산하였다. 대두사포닌이 침투된 인지질 막의 상그림은 순수 인지질 막의 상 그림에 비해 넓적하게 변하였으며 특이하게 상전이 온도를 낮추었다. 이것은 인지질막의 이중층에 침투된 대두사포닌이 인지질 이중층의 협동단위를 감소시킨 것으로 추정된다. 이러한 결과로 보아 대두사포닌은 인지질 막 이중층의 유동성에 중요한 영향을 미친다고 사료된다.

Radish phospholipid hydroperoxide glutathione peroxidase provides protection against hydroperoxide-mediated injury in mouse 3T3 fibroblasts

  • Overexpression of phospholipid hydroperoxide glutathione peroxidase (PHGPx) genes has been reported to play an important role in protecting host cells from oxidative injury in several model systems. A radish phospholipid hydroperoxide glutathione peroxidase (RsPHGPx) known to have high catalytic activity was applied to mouse 3T3 fibroblasts to determine the protective effects of PHGPx against oxidative injury triggered by hydroperoxides such as hydrogen peroxide ($H_2O_2$), tert-butyl hydroperoxide (t-BHP) and phosphatidylcholine hydroperoxide (PCOOH). We observed that preincubation of cells with RsPHGPx significantly increased cell viability, reduced levels of malondialdehyde (MDA), inhibited generation of reactive oxygen species (ROS), and maintained natural cell shapes after treatment with $H_2O_2$, t-BHP or PCOOH, indicating that the exogenous RsPHGPx can act as an effective hydroperoxide-scavenger and may also protect target cells from oxidative damage. These results suggest the possibility for use of RsPHGPx as a therapeutic protectant.

Interactions of Membrane and PMAP-23 Studied by $^{31}P$ solid-state NMR Spectroscopy

  • [ $^{31}P$ ] powder pattern spectra were measured to investigate the aspects of the interaction between the MLV (Multilamellar vesicle) and PMAP-23, a membrane of cathelicidin family and then CSAs(chemical shift anisotropy) were calculated to indentify the extent of perturbation of phospholipid mobility by the peptides. We found that acidic phospholipid interacts strongly with PMAP-23, and the analogues which modified to increase the amphipathic property showed that larger change of CSA. The analogue which introduced positive charge showed the same effects with amphipathic property.

Determination of Saturated and Unsaturated Fatty Acids Amount in Leukocyte Membranes from Subjects Fed with Solid and Fluid Oils

  • Modifications in dietary fatty acid intake might lead to a modification in membrane phospholipid fatty acid composition. The purpose of this study was to investigate relationship between different type of oil consumption and leukocyte membrane phospholipid composition. This study was carried out in subjects utilizing butter (n = 15), margarine (n = 15), fluid oil (n = 15) and mixed types of oils (n = 15) in total 60 subjects. Leukocytes were separated from total blood by dextran sedimentation method. Membrane lipids and proteins were isolated following the cell disruption. Fatty acids of membrane phospholipids were isolated by hydrolysation with phospholipase B under ultrasonic dismembranator. Free fatty acids were identified with gas chromatography at chloroform phase. The results obtained were compared with data obtained by chromatograms of the standards. Results more prominent values of arachidic, dihomo-$\gamma$-linolenic and palmitoleic acids were found in butter-or mixed oil-user groups; eicosadienoic, eicosamonoenoic, dihomo-$\gamma$-linolenic and behenic acids in fluid oil; heptanoic, valeric, eicosadienoic and linolenic acids in margarine groups. The fatty acid composition of mixed oil was similar to butter, while other two oils were so different. From this study, it was concluded that the type of oil consumption might have an influence on phospholipid components of plasma membranes.

우유 지방구막의 고밀도 표피에 결합된 지질의 조성 (Composition of Lipids Associated with Dense Coat-enriched Fractions of Bovine Milk Fat Globule Membrane)

  • 우유 지방구막의 고밀도 표피에 결합된 지질의 조성을 분석하기 위하여 지방구막의 고밀도 표피부분을 여러 농도의 비이온성 세제 Triton X-100으로 처리하였고 세제에 용해되지 않는 물질, 즉 지방과 결합된 성분을 분석하였다. 유지방구막의 단백질, 인지질, 콜레스테롤과 ganglioside의 양은 Triton X-100의 농도가 증가함에 따라 감소하였다. 불용성 표피물질로서 butyrophilin(band 12), xanthine oxidase (band 3)와 band 16이 SDS-polyacrylamide gel을 이용한 전기영동에서 나타났다. Phosphatidyl ethanolamine, phosphatidyl choline, phosphatidyl serine, phosphatidyl inositol 및 spingomyeline 함량은 처리되지 않은 원래의 막의 것과 큰 차이가 있어 표피물질의 주요 인지질로 규명되었다. 전체 지질에서 지방산은 myristate, palmitate, stearate(주요 포화지방산), oleate, linoleate(주요 불포화지방산)이었다. 단백질에 결합된 콜레스테롤은 다른 성분에서보다 단백질에 더 견고하게 부착되어 있었다. Ganglioside의 함량은 Triton X-100의 농도가 증가함에 따라 비례 감소하였다.

Characterization of ATPase Activity of Free and Immobilized Chromatophore Membrane Vesicles of Rhodobacter sphaeroides

  • The intracytoplasmic membrane of Rhodobacter sphaeroides readily vesiculates when cells are lysed. The resulting chromatophore membrane vesicle (CMV) contains the photosynthetic machineries to synthesize ATP by ATPase. The light-dependent ATPase activity of CMV was lowered in the presence of $O_2$, but the activity increased to the level observed under anaerobic condition when the reaction mixture was supplemented with ascorbic acid (${\geq}0.5mM$). Cell lysis in the presence of biotinyl cap phospholipid (bcp) resulted in the incorporation of bcp into the membrane to form biotinylated CMV (bCMV), which binds to streptavidin resin at a ratio of approximately $24{\mu}g$ bacteriochlorophyll a/ml resin. The ATPase activity of CMV was not affected by biotinylation, but approximately 30% of the activity was lost by immobilization to resin. Interestingly, the remaining 70% of ATPase activity stayed constant during 7-day storage at $4^{\circ}C$. On the contrary, the ATPase activity of bCMV without immobilization gradually decreased to approximately 40% of the initial level in the same comparison. Thus, the ATPase activity of CMV is sustainable after immobilization, and the immobilized bCMV can be used repeatedly as an ATP generator.

Effect of Lipid Peroxidation on the Fluidity of Erythrocyte Ghost and Phospholipid Liposomal Membranes

  • The effects of lipid peroxidation on the fluidity of the lipid bilayers of the human erythrocyte ghosts and egg-lecithin phospholipid liposomes have been studied. For the measurements of the peroxidation extent and the fluidity of the membranes, the thiobarbituric acid-reactive substances and the fluorescence depolarization of 1, 6-diphynyl-1, 3, 5-hexatriene labelled into the membrane were employed, respectively. The lipid peroxidation was performed in hypoxanthine/xanthine oxidase/ferrous ion, and hydrogen peroxide/ferrous ion systems. The results of these experiments show that both of the xanthine oxidase and hydrogen peroxide systems effectively. The lipid peroxidation decreased the fluidity of the membranes, especially at the very early stage of the peroxidation reaction. The decrease in the fluidity of membrane by the lipid peroxidation has been ascribed to the alteration of the polyunsaturated acyl chains of lipids and cross linkages among the membrane components. However, under drastic condition of lipid peroxidation, tdhe fluidity of the membrane rather increased possibly due to the deterioration of the membrane integrity by the peroxidation. Morphological change of the erythrocyte on peroxidation has also been observed.

Effects of Brazilin on Lipid and Phosphatidyl Fatty Acid Composition of Erythrocyte Membrane in Streptozotocin-induced Diabetic Rats

  • In diabetes, the abnormal increase of the membrane cholesterol/phospholipid ratio (C/PL) is consdered to be the main reason for the decreased membrane fluidity, which then results in impaired erythrocyte deformability and subsequent microcirculatory disturbances. In this study, we examined the effects of brazilin on lipid and phosphatidyl fatty acid composition of erythrocyte membranes in streptozotocin induced diabetic rats. Treatment of brazilin (10mg/kg or 100 mg/kg for 2 weeks, i.p) altered and cholesterol contents in diabetic erythrocyte membranes. The C/PL ratio of brazilin treated groups decreased compared with that of diabetic control group while no change was observed in normal erythrocytes. In streptozotocin induced diabetic rats, alterations in phosphatidyl fatty acid compositioin of erythrocyte membranes were observed and brazilin could reverse these alterations. Arachidonic acid level reumed to a normal level while linoleic acid level remained unchanged by the treatment of brazilin. The results suggest that brazilin might increase erythrocyte membrane fluidity which plays a key role inregulating erythrocyte deformability, thereby it could exert positive effects on microdiculatory disturbances.

  • Apo, holo, native-type of lactoferrin showed a similar pattern of circular dichroism(CD) spectra. Lactoferrin appeared to interact more easily with negatively charged liposome than neutral liposome. Holo-type of lactoferrin showed the highest degree of leakage, whereas apo-type of lactoferrin showed the lowest level of leakage. Holo-type lactoferrin could more easily interact with phospholipid liposomes than apo-type lactoferrin, when used as an artificial membrane.
