사업장 내 사무실의 $PM_{2.5}$ 노출 평가 (Exposure Assessment of $PM_{2.5}$ in Manufacturing Industry Office Buildings)

  • Objectives: This study was conducted in order to evaluate $PM_{2.5}$ concentrations at 20 offices connected to the manufacturing industry from the beginning of September to the end of November 2012. Methods: A total of 20 samples were collected from 20 office buildings. Each $PM_{2.5}$ sample was collected by a 37 mm PTFE filter attached to a Personal Environment Monitor. Results: The geometric mean concentrations of $PM_{2.5}$ in the offices was $23.47{\mu}g/m^3$, and the mean $PM_{2.5}$ concentrations measured in smoking offices were much higher than those of measured in non-smoking offices($24.83{\mu}g/m^3$ and $21.55{\mu}g/m^3$, respectively). $PM_{2.5}$ was revealed to be higher in small offices($39.52{\mu}g/m^3$) than in medium or large offices($22.69{\mu}g/m^3$ and $11.04{\mu}g/m^3$, respectively). The mean $PM_{2.5}$ concentration of offices located on the 1st floor was higher than that of those on the 2nd floor, and those of offices located in the workplace were higher than those out of the workplace. The multiple regression model showed that concentration of $PM_{2.5}$ was positively associated with the method of ventilation. Conclusions: Smoking, ventilation method, location, and inflow of outdoor particulate matter are the most important factors for office $PM_{2.5}$ concentrations.

참다래 꼭지썩음병을 일으키는 Diaporthe actinidiae을 억제하는 길항성 Bacillus sp. #72의 분리 및 동정 (Screening and Identification of Antifungal Bacillus sp. #72 against the Pathogenic Stem-end Rot of Kiwi Fruit)

  • This study was conducted to select and screen for an antifungal bacterial strain showing pathogen inhibitory activity against Diaporthe actinidiae, which causes stem-end rot in kiwi fruit. Four bacterial strains were isolated which strongly inhibit Diaporthe actinidiae from among two hundred and fifty bacterial strains screened from the soil where kiwi fruit were grown. By co-culturing bacterial strain #72 and the pathogen causing the stem-end rot of kiwi fruit, bacterial strain #72 showed 81.0% antifungal activity against Diaporthe actinidiae. Bacterial strain #72 was identified to be from the genus Bacillus sp. based on morphological and biochemical characterization. The bacterialization of culture broth for Bacillus sp. #72 which was sterilized at $121^{\circ}C$ for 15 minutes and than purified by $0.45{\mu}m$ membrane filter showed almost all of the antagonistic activity against Diaporthe actinidiae. We have also confirmed that in vitro treatment of Bacillus sp. #72 cultured in SD+B+P(sugar 5%, soy sauce 3%, beef extract 0.2%, peptone 0.2%) medium efficiently inhibited the growth of Diaporthe actinidiae responsible for stem-end rot in kiwi fruit.

Effects on amplification of strong ground motion due to deep soils

  • Many seismically vulnerable regions in India and worldwide are located on deep soil deposits which extend to several hundred meters of depth. It has been well recognized that the earthquake shaking is altered by geological conditions at the location of building. As seismic waves propagates through uppermost layers of soil and rock, these layers serve as filter and they can increase the duration and amplitude of earthquake motion within narrow frequency bands. The amplification of these waves is largely controlled by mechanical properties of these layers, which are function of their stiffness and damping. Stiffness and damping are further influenced by soil type and thickness. In the current study, an attempt has been made to study the seismic site response of deep soils. Three hypothetical homogeneous soil models (e.g., soft soil, medium soil and hard soil) lying on bedrock are considered. Depth of half space is varied from 30 m to 2,000 m in this study. Controlled synthetic motions are used as input base motion. One dimensional equivalent linear ground response analyses are carried out using a computer package DEEPSOIL. Conventional approach of analysing up to 30 m depth has been found to be inadequate for deep soil sites. PGA values are observed to be higher for deeper soil profiles as compared to shallow soil profiles indicating that deeper soil profiles are more prone to liquefaction and other related seismic hazards under earthquake ground shaking. The study recommends to deal the deeper soil sections more carefully for estimating the amplification factors for seismic hazard assessment at the surface.

Extracting Gold from Pyrite Roster Cinder by Ultra-Fine-Grinding/Resin-in-Pulp

  • A new method to extract gold from pyrite roster cinder, which combines ultra-fine-grinding with resin-in-pulp, has been studied in this paper. Compared with traditional leaching technology, it can short leaching time, avoid complex filter process, lower sodium cyanide consumption and increase gold recovery by 35%. During leaching, aluminium oxide ball was used as stirred medium, hydrogen peroxide as leaching aid and sodium hexametaphosphate as grinding aid. With the high efficiency and chemistry effect of ultra-fine-grinding, the leaching process was developed and the gold leaching rate may reach 88%. With AM-2 Б resin as abosorber and sulfocarbamide (TU) as eluent, gold was recovered from cyanide pulp by resin-in-pulp. AM-2 Б resin has good adsorbability in cyanide solution(pH=10). It was easy to elude gold from the loaded resin with 0.1㏖/L cholhydric acid and 1㏖/L sulfocabamide. The effect of contact time, temperature and acidity etc. on the gold absorption had been examined with static methods. The results showed that the adsorption and desorption of gold could both reach over 98%. The effects of flow rate of solution on dynamic adsorption and elution of gold had been examined with dynamic methods. Breakthrough curve and elution curve had been drawn in this paper. A mild condition was determined through a number of experiments: leaching time 2 hours, liquid solid ratio 4:1, sodium cyanide 3kg/t, hydrogen peroxide 0.05%, sodium hexametaphosphate 0.05%; adsorption time 30 minutes, temperature 10-3$0^{\circ}C$, resin($m\ell$) solid(g) ratio 1:10, eluent resin ratio 10-20:1, velocity of eluent $1.5m\ell$/min. Under the mild condition, the gold recovery may reach 85%.

Preparation of high Purity manganese oxide by Pyrolysis of solution extracted from ferromanganese dust in AOD process

  • The high purity manganese oxides were made from the dust, generated in AOD process that produces a medium-low carbon ferromanganese and collected in the bag filter. Manganese oxide content in the dust was about 90%, and its phase was confirmed as Mn₃O₄. In the extraction of manganese, because of remaining amorphous MnO₂, the dust was reduced to MnO by roasting with charcoal. The pulp density of the reduced dust can control pH of the solution more than 4 and then Fe ion is precipitated to a ferric hydroxide. Because a ferric hydroxide co precipitates with Si ion etc, Fe, Si ion was removed f개m the solution. Heating made water to be volatized and nitrates was left in reactor Then nitrates were a liquid state and stirring was possible. Among the nitrates in reactor, only the manganese nitrate which have the lowest pyrolysis temperature pyrolyzed into β-MnO₂powder and NO₂(g) at the temperature less than 200℃. When the pyrolysis of manganese nitrate has been completed about 90%, injection of water stopped the pyrolysis. Nitrates of impurity dissolved and the spherical high purity β-MnO₂powders were obtained by filtering and washing. Mn₂O₃or Mn₃O₄ powder could be manufactured from β-MnO₂powder by controlling the heating temperature. Lastly, a manufactured manganese oxide particle has 99.97% purity.

  • As traffic in city-centre around the world continues to increase, so levels of atmospheric pollutants continue to rise. High concentrations of NOx can have negative effects on human health, and we must find new ways to reduce their levels in the air we breathe. Nitrogen oxide gas (NOx), consisting of nitrogen monoxide (NO) and nitrogen dioxide $(NO_2)$ produced using $O_3$ oxidation, at a low concentration corresponding to that on roads as a result of exhaust from automobiles, was carried out to evaluate the removal characteristics of NOx through a laboratory-scale biofilter packed with soil as a packing material. A mixture media (yellow soil (30%): soil (40%): compost (10%): a used briquet (20%)) was applied. After about 1day of operation, the removal efficiency for $NO_2$ in all experiments with a constant condition ($25^{\circ}C$ and water humidity (60%)) was over 98%. The retention times of the section between phase I and phase II for formation and reduction of $NO_3$ NO and $NO_2$ on the initial $NO_3$ concentration was 50min $(O_3:195\;ppb),\;55min\;(O_3:925\;ppb),\;65min\;(O_3:1743\;ppb),\;70min\;(O_3:2616\;ppb),\;75min\;(O_3:3500\;ppb)$, respectively The soil biofilter system is a unique technology that purifies urban air by utilizing the natural processes that take place in the soil. Although some of the processes are quite complex, they can broadly be summarized as adsorption onto soil particles, dissolution into soil pore water, and biochemical.

Ionized Fe Objects in UWIFE survey and IGRINS

  • The UKIRT Wide-field Infrared survey for Fe+(UWIFE) is an unbiased survey of the first Galactic quadrant, with narrow-band filter centered on $1.644{\mu}m$. This survey covers $7^{\circ}$ < l < $62^{\circ}$ and |b| < $1.5^{\circ}$, where active interaction of stars and interstellar medium is expected. With median seeing of 0.8 arcsec, 5 - sigma detection limit of 18.7 mag and surface brightness limit of $8.1{\times}10^{-20}W\;m^{-2}arcsec^{-2}$, this survey gives an opportunity to statistically study Galactic [Fe II] - emitting sources for the first time. In order to identify Ionized Fe Objects (IFOs) in survey area systematically, we conducted visual inspection and automatic detection simultaneously. Total of ~300 extended IFOs are identified, most of them are found out to be part of supernova remnants (SNRs), young stellar objects, HII regions and planetary nebulae. The majority of IFOs are new discoveries which reveal shocked structures in high-extinction region. Spatial distribution of IFOs suggest that they trace Galactic structure. As a part of spectroscopic follow-up, we observed SNR candidate IFO J183740.829-061452.41 with IGRINS (Immersion Grating Infrared Spectrograph, Yuk+2010), mounted on 2.7m Harlan Smith telescope. This unknown arc-like, 6'-long IFO is coincident with inner part of radio continuum loop G25.8+0.2, which has been known as HII region. However, interior of this radio shell is filled with diffuse soft X-ray emission, and possible association of hard X-ray pulsar / pulsar wind nebula makes the nature of the IFO unclear. The H and K-band 2D spectrum shows shock-ionized [Fe II] filaments, which is apart from photoionized HII filaments. In this presentation we present basic statistics of newly identified IFOs, as well as the follow-up study of IFO J183740.829-061452.41.

Effects of LCFA on the Gas Production, Cellulose Digestion and Cellulase Activities by the Rumen Anaerobic Fungus, Neocallimastix frontalis RE1

  • Responses of the rumen fungus, Neocallimastix frontalis RE1, to long chain fatty acid (LCFA) were evaluated by measuring gas production, filter paper (FP) cellulose digestion and polysaccharidase enzyme activities. LCFA (stearic acid, $C_{18:0}$; oleic acid, $C_{18:1}$; linoleic acid, $C_{18:2}$ and linolenic acid, $C_{18:3}$) were emulsitied by ultrasonication under anaerobic condition, and added to the medium. When N frontalis RE1 was grown in culture with stearic, oleic and linoleic acid, the cumulative gas production, gas pool size, FP cellulose digestion and enzymes activities significantly (p<0.05) increased at some incubation times(especially, exponential phases of fungal growth, 48~120 h of incubation) relative to that for control cultures. However, the addition of linolenic acid strongly inhibited all of the investigated parameters up to 120 h incubation, but not after 168 and 216 h of incubation. These results indicated that stearic, oleic and linoleic acids tended to have great stimulatory effects on fungal cellulolysis, whereas linolenic acid caused a significant (p<0.05) inhibitory effects on the cellulolysis by the rumen fungus. These results are the first report of the effect of LCFAs on the ruminal fungi. Further research is needed to identify the mode of action of LCFAs on fungal strains and to verify whether or not ruminal fungi have ability to hydrate unsaturated LCFAs to saturated FAs. There was high correlation between cumulative in vitro gas production and fungal growth (94.78%), FP cellulose degradation (96.34%), CMCase activity(90.86%) or xylanase activity (87.67%). Thus measuring of cumulative gas production could be a useful tool for evaluating fungal growth and/or enzyme production by ruminal fungi.

LC-MS/MS를 이용하여 먼지 속의 NNK (4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone) 정량 분석법 개발 (Development of a method for the determination of 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone in dust using liquld chromatography tandem mass spectrometry)

  • 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK), a tobacco specific nitrosamine found only in tobacco products. The ability to monitor biomarker concentrations is very important in understanding environmental tobacco smoke (ETS). In this study, an efficient and sensitive method for the analysis of NNK in dust was developed and validated using liquid chromatography tandem mass spectrometry. Dust was collected with filter paper soaked in methanol. The standard solution and dust sample were diluted with 100 mM ammonium acetate and extracted using dichloromethane. Our calibration curves ranged from 25 to $10^4pg/mL$. Excellent linearity was obtained with correlation coefficient values between 0.9996 and 1.0000. The limit of detection (LOD) was 5 pg/mL ($S/N{\geq}3$) and the retention time was 10 min. The limit of quantification (LOQ) was 25 pg/mL, and the acceptance criteria was the rate of 98-103% (80-120% at levels up to $3{\times}LOQ$). The coefficient of variations (CV) was 2.8%. Accuracies determined from dust samples spiked with four different levels of NNK racurves ranged that from 25 to 104 pg/mL. Excellent linearity was obtained between 92.1% and 114%. The precision of the method was acceptable (5% of CV). The recovery rates of the whole analytical procedure at low, medium, and high levels were 105.7-116.5% for NNK. The carry-over effects during LC-MS/MS analysis were not observed for NNK. This manuscript summarizes the scientific evidence on the use of markers to measure ETS.

생물방제능과 식물성장촉진능을 동시에 가지는 Bacillus licheniformis K11의 non-siderophore 항진균 물질 및 cellulase의 생산조건 확인 (Confirmation of Non-Siderophore Antifugal Substance and Cellulase from Bacillus lichemiformis Kll Containing Antagonistic Ability and Plant Growth Promoting Activity)

  • Auxin, siderophore, 그리고 cellulase를 동시에 생산하는 생물방제균주 B. licheniformis Kll을 식물병원성진균을 대상으로 균사 성장억제능을 확인결과 6종의 식물병원성 진균에서 균사체 성장억제능을 확인하였으며, 그 중에서 토마토 시들음병을 유발하는 F. oxysporum(KACC 40037)에 가장 강력한 억제능을 나타내었다. 그리고 본 균주가 생산하는 항진균성 siderophore이외에 세포벽이 cellulose로 구성된 P. capsici의 cell wall을 분해하는 cellulase를 생산하는 것을 추가적으로 확인하였다. 뿐만아니라 B. licheniformis Kll은 nutrient broth(pH 8.0), $30{\circ}C$에서 96시간 배양시 토마토 시들음병에 대한 항진균 활성이 가장 높았고, 이는 cellulase의 활성과 sideropore의 최대 생산조건과는 상이하였다. 또한 탄소원과 질소원으로 starch와 urea를 각각 첨가시 항진균성 활성이 가장 높았고, 이 역시는 cellulase의 활성과 항진균성 siderophore의 최대 생산조건과 일치하지 않았다. 그리하여 본균주 B. licheniformis Kll은 식물성장촉진 물질은 auxin, 항진균성 siderophore와 cellulase를 생산함과 동시에 또 다른 강력한 항진균성 물질을 생산하는 것을 추가로 확인하였다.
