10종의 한약재 MeOH 추출물과 EtOH 추출물의 E. coli에 대한 항균력을 조사한 결과 소목과 오배자의 MeOH 추출물과 EtOH 추출물은 5 mg/ml 수준에서 항균활성을 나타내었으며 목단피와 황금등은 10 mg/ml 이상에서 항균성을 보였다. 최소저해농도는 MeOH 추출물에서는 $1.4{\sim}8mg/ml$, EtOH 추출물에서는 $1.2{\sim}12mg/ml$ 농도에서 항균활성을 보였으며 분획층 중 EtOAc층에서 성장억제효과가 가장 높게 나타났다. 항균력이 우수한 것으로 확인된 소목과 오배자 추출물의 미생물 증식억제 효과를 조사하기 위해 증식배지에 각각의 추출물을 0, 100, 300 및 500 ppm의 농도로 첨가하여 균주의 증식을 조사 한 결과 배양 후 24시간에 소목 추출물 무첨가구의 $OD_{620}$값이 0.6인 반면 300ppm 이상의 추출물 첨가 시 $0.02{\sim}0.1$정도로 균증식이 현저히 억제되었고, 오배자 추출물 무첨가구의 $OD_{620}$값이 0.5인 반면 500ppm 이상의 추출물 첨가 시 $0.02{\sim}0.2$정도로 균증식이 현저히 억제되었다. 이상의 결과로 볼 때 질병을 일으키는 E. coli의 감염을 예방하거나 또는 치료할 수 있는 한약재 추출물의 개발 가능성을 확인할 수 있었다.

This study was aimed to evaluate the in vitro effects of medicinal herb extracts (MHEs) on ruminal fermentation characteristics and the inhibition of protozoa to reduce methane production in the rumen. A fistulated Hanwoo was used as a donor of rumen fluid. The MHEs (T1, Veratrum patulum; T2, Iris ensata var. spontanea; T3, Arisaema ringens; T4, Carduus crispus; T5, Pueraria thunbergiana) were added to the in vitro fermentation bottles containing the rumen fluid and medium. Total volatile fatty acid (tVFA), total gas production, gas profiles, and the ruminal microbe communities were measured. The tVFA concentration was increased or decreased as compared to the control, and there was a significant (p<0.05) difference after 24 h incubation. pH and ruminal disappearance of dry matter did not show significant difference. As the in vitro ruminal fermentation progressed, total gas production in added MHEs was increased, while the methane production was decreased compared to the control. In particular, Arisaema ringens extract led to decrease methane production by more than 43%. In addition, the result of real-time polymerase chain reaction indicted that the protozoa population in all added MHEs decreased more than that of the control. In conclusion, the results of this study indicated that MHEs could have properties that decrease ruminal methanogenesis by inhibiting protozoa species and might be promising feed additives for ruminants.

Aralia cordata (A. cordata), which belongs to Araliaceae, is a perennial herb widely distributed in East Asia. We evaluated the anti-inflammatory effect of stems (AC-S), roots (AC-R) and leaves (AC-L) extracted with 100% methanol of A. cordata and elucidated the potential signaling pathway in LPS-stimulated RAW264.7 cells. The AC-L showed a strong anti-inflammatory activity through inhibition of NO production. AC-L dose-dependently inhibited NO production by suppressing iNOS, COX-2 and $IL-{\beta}$ expression in LPS-stimulated RAW264.7 cells. AC-L inhibited the degradation and phosphorylation of $I{\kappa}B-{\alpha}$, which donated to the inhibition of p65 nuclear accumulation and $NF-{\kappa}B$ activation. Furthermore, AC-L suppressed the phosphorylation of ERK1/2 and p38. These results suggested that AC-L may utilize anti-inflammatory activity by blocking $NF-{\kappa}B$ and MAPK signaling pathway and indicated that the AC-L can be used as a natural anti-inflammatory drugs.
As an attempt to develop new functional health beverage by using medicinal herb, Artemisia capillaris, we investigated the effect of scopoletin in Artemisia capillaris on vessel and regional cerebral blood flow (rCBF) of rats ingesting health drink prepared with Artemisia capillaris extracts and various ingredients. Artemisia capillaris extract decreased the vessel contraction and increased rCBF significantly. The extracts were grouped by heat temperature and mixed ratio and tested their respective characteristics. Then each condition was combined and produced the most effective one. The drink produced consisted of Artemisia capillaris extract 42%, honey 9.8%, citric acid 0.035%, cyclodextrin 1.47% and water. Brix, pH and acidity of the product were 9.2, 4.4 and 0.04%, respectively, This drink scored to have highest level on overall acceptance by the sensory evaluation. The above results showed that development of such functional beverage using Artemisia capillaris can be used as a functional material improving blood circulation in beverage industry.
The methanol extracts of 340 kinds of Korean medicinal herbs were examined in vitro for the inhibitory effect on the acetylcholinesterase (AChE, E.C. 3.1.1.7) from electric eel using acetylthiocholine as a substrate. Among tested, the extracts of Coptidis Rhizoma, Phellodendri Cortex, Evodiae Fructus, Myrrha, Arecae Semen and Piperis nigri Fructus were found to exhibit a significant inhibition upon the acetylcholinesterase in a dose dependent manner, respectively.
The water extracts of 82 Korean medicinal herbs were examined for the binding affinity on the recombinant human muscarinic acetylcholine receptor subtype 1 $(mAChR-M_1)$ produced from the CHO (Chinese Hamster Ovary) cell line. Of those tested, the extracts of Coptidis Rhizoma, Phellodendri Cortex, Hedyotis Herba and of Terminariae Fructus were found to exhibit a significant competition with $[^3H]$ N-methyl-scopolamine for the specific binding to $mAChR-M_1$ in a dose dependent manner, respectively.
The water extracts of 82 Korean medicinal herbs were prepared and were examined for the binding affinity on the glycine binding site of NMDA (N-methyl-D-aspartate) receptor prepared by the synaptic membranes from the forebrains of male Sprague-Dawley rats. Among the tested, the extracts of Dioscoreae Rhizoma, Hoveniae Semen cum Fructus, Astragali Radix, Armeniacae Semen, Huttuynia cordata Herba, Acanthopanacis Cortex, Aurantii nobilis Pericarpium, Phellinus linteus, Amomi Fructus, Artemisiae capillaris Herba, Polyporus, Agastachis Herba and of Galli Stomachichum Corium were found to exhibit significant competitions with $[^3H]-MDL$ 105,519 for the glycine specific binding site of NMDA receptor in a dose dependent manner, respectively.
Herbal medicine is the use of medicinal plants for prevention and treatment of diseases: it ranges from traditional and popular medicines of every country to the use of standardized and tritated herbal extracts. Generally cultural rootedness enduring and widespread use in a traditional medical system may indicate safety, but also efficacy of treatments, especially in herbal medicine where tradition is almost completely based on remedies containing active principles at very low and ultra low concentrations or relying on magical-energetic principles. The efficacy and safety assessments of medicines, whether modern or herbal, invariably encounter challenges or problems during the course of pre-clinical and clinical research. Some of the challenges in evidence-based herbal medicinal research are unique, and the researcher must be cognizant of them in order to safeguard the quality of the data obtained. Key challenges are: the quality of raw materials; appropriateness of biological/pharmacological activity assessment methodology, and data interpretation; standardization methodology; pharmacokinetics and bioavailability of active constituents and metabolites; clinical dosage formulation/production; and clinical study designs and outcome measures.
These experiments were performed to investigate the safety of the three medicinal herbs- Curcuma longa Linne, Paeonia japonica Miyabe, Scutellaria baikalensis George-irradiated with gamma rays in respect of genotoxicity. The methanol-soluble and water-soluble fractions of the methanol-water extracts of the 10 kGy gamma-irradiated herbs were examined in two short-term in vitro tests : (1) Salmonella typhimurium reversion assay (Ames test) in strain TA 98, TA 100 and TA 012 (2) Micronucleus test in cultured Chinese hamster ovary (CHO) cells. No mutagenicity was detected in the assays with or without metabolic activation. From these results, the safety of the herbs irradiated with gamma rays at practical doses could be revealed in further tests of genotoxicity in vivo, chronic and reproductive toxiceity.
Objectives: The aim of this study was to investigate deodorizing effectsof medicinal herbs (Mume Fructus, Eriobotryae Folium, Acanthopanacis Cortex, Angelicae Dahuricae Radix) for development of a gargle solution. Methods: 1. The antimicrobial effects of medicinal herbs were evaluated with the minimal bactericidal concentration (MBC) and the change of the number of viable cells in the herb extracts(1%) for 48 hrs against P. gingivalis 2561 and Pr. intermedia ATCC 25611. 2. Deodorizing activity of each herb and Garglin $Mint^{(R)}$gainst methyl mercaptan were analyzed by gas chromatography (GC). 3. We used the malodor modeling of the salivary sediment system with a Halimeter. 4. In the preliminary clinical study, the baseline concentration of VSC in the oral cavity of each subject was measured by Halimeter. Subjects would gargle for 30 seconds with cysteine. After 4 minutes subjects would gargle for 30 seconds with Garglin and herb extracts (2%). Subsequently, concentration of VSC were measured at 0, 4, 8, 12 and 20 minutes. Results: 1. MBC of Mume Fructusfor P. gingivalis 2561 was determined to be <1% and MBCs of Eriobotryae Folium for P. gingivalis 2561 and Pr. intermedia ATCC 25611 were determined to be <2% and <1%, respectively. Mume Fructus (1%) completely suppressed the P. gingivalis cell viability from 5 hrs and Eriobotryae Folium (1%) completely suppressed the Pr. intermedia cell viability from 48 hrs. 2. In GC analysis, deodorizing activities were 91.54% with Mume Fructus, 87.97% with Eriobotryae Folium, 100% with Acanthopanacis Cortex, 72.36% with Angelicae Dahuricae Radix and 40.54% with Garglin $Mint^{(R)}$. 3. In malodor modeling of the salivary sediment system, each of the medicinal herbs had significantly inhibitory effect on malodor formation (p<0.05). 4. In the preliminary clinical study, the concentration of VSC of the herb groups was significantly lower than of the control group, but not in Garglin $Mint^{(R)}$. Conclusions: Mume Fructus, Eriobotryae Folium, Acanthopanacis Cortex and Angelicae Dahuricae Radixhave deodorizing activities and potential as an effective mouthwash against oral malodor.
