  • Two new bisbenzylisoquinoline alkaloids tiliacosine (1) and tiliasine (2) were isolated from the leaves of Tiliacora racemosa Colebr.. The structures of these alkaloids were established on the basis of spectral evidence and by the correlation of their $^1H-NMR$ spectral data with those of the congeners N-methyltiliamosine (3), tiliamosine (4) and tiliacorine (5).

  • Botulinum toxins are neurotoxic modular proteins composed of a heavy chain and a light chain connected by a disulfide bond and are produced by Clostridium botulinum. Although lethally toxic, botulinum toxin in low doses is clinically effective in numerous medical conditions, including muscle spasticity, strabismus, hyperactive urinary bladder, excessive sweating, and migraine. Globally, several companies are now producing products containing botulinum toxin for medical and cosmetic purposes, including the reduction of facial wrinkles. To test the efficacy and toxicity of botulinum toxin, animal tests have been solely and widely used, resulting in the inevitable sacrifice of hundreds of animals. Hence, alternative methods are urgently required to replace animals in botulinum toxin testing. Here, the various alternative methods developed to test the toxicity and efficacy of botulinum toxins have been briefly reviewed and future perspectives have been detailed.

  • Microbial metabolism studies of yangonin (1), a major styryl lactone from Piper methysticum, have resulted in the production of three hydroxylated metabolites (2-4). The chemical structures of these compounds were elucidated to be 4-methoxy-6-(12-hydroxystyryl)-2-pyrone (2),4-methoxy-6-(11,12-dihydroxystyryl)-2-pyrone (3),and 4,12-dimethoxy-6-(7,8-dihydroxy-7,8-dihydrostyryl)-2-pyrone (4) on the basis of the chemical and spectroscopic analyses. The compounds 3 and 4 are reported herein as microbial metabolites of yangonin for the first time.

  • Livestock excretes are frequently used in organic farming to produce vegetables and crops. However, zoonotic parasite eggs can be contaminated into such products and people can be infected with parasites of animal origin. Sporadic zoonotic infections with Ascaris suum have been reported even in developed countries, such as North America, Denmark, and the United Kingdom. Ascaris eggs are resistant to most adverse environmental conditions, and pigs and humans become infected by ingestion of fecally excreted eggs through contaminated food, water, or soil. This article discusses the resistant nature of ascarid eggs against harsh environment, and conditions of kimchi to be safe from consuming viable A. suum eggs.

  • A virtual human which can evaluate Kansei such as comfort, pain, etc. when the virtual human uses some product is developed. In this paper, method of the evaluation of Kansei by the virtual human is presented. The body of our virtual human is modeled as an uniform non-linear elastic one with a skeleton. The deformation of the body on the contact with some product is simulated using a FEM analysis, and by using of the simulated results (load distribution, strain, etc.) on the contact surface the Kansei is predicted. As examples of the application, comfort of buttocks on seating and pain of arm on hanging of bag are shown. This virtual human can apply for the design of virtual products and also the simulation of medical care.

The Inhibitory Effect of Gut Microbiota and Its Metabolites on Colorectal Cancer

  • Colorectal cancer (CRC) is regarded as one of the most common and deadly forms of cancer. Gut microbiota is vital to retain and promote several functions of intestinal. Although previous researches have shown that some gut microbiota have the abilities to inhibit tumorigenesis and prevent cancer from progressing, they have not yet clearly identified associative mechanisms. This review not only concentrates on the antitumor effects of metabolites produced by gut microbiota, for example, SCFA, ferrichrome, urolithins, equol and conjugated linoleic acids, but also the molecules which constituted the bacterial cell wall have the antitumor effect in the host, including lipopolysaccharide, lipoteichoic acid, β-glucans and peptidoglycan. The aim of our review is to develop a possible therapeutic method, which use the products of gut microbiota metabolism or gut microbiota constituents to help treat or prevent colorectal cancer.

  • Doxorubicin as a chemotherapeutic drug is widely used for the treatment of patients with cancer. However, clinical use of this drug is hampered by its cardiotoxicity, which is manifested as electrocardiographic abnormalities, arrhythmias, irreversible degenerative cardiomyopathy and congestive heart failure. The precise mechanisms underlying the cardiotoxicity of doxorubicin are not clear, but impairment of calcium homeostasis, generation of iron complexes, production of oxygen radicals, mitochondrial dysfunction and cell membrane damage have been suggested as potential etiologic factors. Compounds that can neutralize the toxic effect of doxorubicin on cardiac cells without reducing the drug's antitumor activity are needed. In recent years, numerous studies have shown that herbal medicines and bioactive phytochemicals can serve as effective add-on therapies to reduce the cardiotoxic effects of doxorubicin. This review describes different phytochemicals and herbal products that have been shown to counterbalance doxorubicin-induced cardiotoxicity.

  • Emerge of nanotechnology impacts all aspects of humans' life. One of important aspects of the nanotechnology and nanoparticles (NPs) is in the food production industry. The safety of such foods is not well recognized and producing safe foods using nanoparticles involves delicate experiments. In this study, we aim to incorporate intelligent computer simulation in predicting safety degree of nanofoods. In this regard, the safety concerns on the nano-foods are addressed considering cytotoxicity levels in metal oxides nanoparticles using adaptive neuro-fuzzy inference system (ANFIS) and response surface method (RSM). Three descriptors including chemical bond length, lattice energy and enthalpy of formation gaseous cation of 15 selected NPs are examined to find their influence on the cytotoxicity of NPs. The most effective descriptor is selected using RSM method and dependency of the toxicity of these NPs on the descriptors are presented in 2D and 3D graphs obtained using ANFIS technique. A comprehensive parameters study is conducted to observe effects of different descriptors on cytotoxicity of NPs. The results indicated that combinations of descriptors have the most effects on the cytotoxicity.

Hot water extract of Loliolus beka attenuates methylglyoxal-induced advanced glycation end products formation in human umbilical vein endothelial cells

  • Over production of methylglyoxal (MGO) a highly reactive dicarbonyl compound, has been associated in progressive diabetes with vascular complication. Therefore, we investigated whether hot water extract of Loliolus beka meat (LBM-HWE) presents a preserve effect against MGO-induced cellular damage in human umbilical vein endothelial cells (HUVECs). The LBM-HWE extract showed to inhibit MGO-induced cytotoxicity. Additionally, the LBM-HWE reduced mRNA expression of pro-inflammatory cytokines, and reduced MGO-induced advanced glycation end product (AGEs) formation. Furthermore, LBM-HWE induced glyoxalase-1 mRNA expression and reduced MGO-induced carbonyl protein formation in HUVECs. The results implicate that LBM-HWE has protective ability against MGO-induced HUVECs toxicity by preventing AGEs formation. In conclusion, LBM-HWE could be used as a potential treatment material for the prevention of vascular complications of diabetes.

  • Fluorine compounds have attracted interest of scientists for immense applications in medicinal chemistry and pharmaceuticals. Recently, photoredox catalysts, both organic-based and metal-based compounds, have been employed in organic synthetic methodology to achieve desirable products due to facile operation and mild reaction condition. Various protocols to prepare fluorination adducts in the presence of photoredox catalysts have been developed from several starting materials with formation of radical scaffolds. In this review, we describe recent advances in the fluorination using light.
