Effects of Aqueous Extracts from Twelve Herbs on ${\alpha}$-melanocyte Stimulating Hormone-induced Melanogenesis in B16F10 Mouse Melanoma Cell (한약재 12종의 열수추출물이 ${\alpha}$-melanocyte stimulating hormone에 의해 유도된 B16F10 흑색종 세포의 멜라닌형성에 미치는 영향)

  • We investigated the effects of aqueous extracts from twelve medical herbs on ${\alpha}$-melanocyte stimulating hormone (${\alpha}$-MSH)-induced melanogenesis in B16F10 mouse melanoma cell. The cells were incubated with ${\alpha}$-MSH and aqueous extracts 5 days and 2 days and then analysed melanin amount and tyrosinase activities, respectively. Nine aqueous extract of herbs examined at 1 mg/m${\ell}$ level decreased melanin synthesis in B16F10 cell, especially in Agastache rugosa, Leonurus siviricus and Murus bombycis. The significant decrease of released extracellular melanin were also observed in treated cells with aqueous extract from Leonurus siviricus, Murus bombycis and Ledebouriella seseioides. The ${\alpha}$-MSH-induced activation of tyrosinase was inhibited in cells treated with aqueous extract from Cuscutae semen, Angelica tenuissima and Agastache rugosa. These results suggest that herbs inhibiting melanogenesis through tyrosinase activity may apply to develop whitening drugs and cosmetics.

Stability of W/O Nanoemulsions with Low Viscosity Prepared by PIC Method (PIC 방법으로 제조된 저점도 W/O 나노에멀젼의 안정성)

  • In this study, water-in-oil (W/O) nanoemulsions of water/Span 80-Nikkol BL 25/oil system were prepared by the PIC method at elevated temperature. This method allows the formation of finely dispersed W/O nanoemulsions with low viscosity in this system. However, macroemulsions rather than nanoemulsions were prepared by PIC method at room temperature. As a result of the significant change of interfacial tension with temperature, the emulsion droplet size decreases from $2{\mu}m$ to 100 nm with the increase in temperature from $30^{\circ}C$ to $80^{\circ}C$. The droplet size of nanoemulsions prepared at $80^{\circ}C$ was in the range of 50 ~ 200 nm and the internal phase content could reach as high as 15 wt%. The most stable nanoemulsion was formed in the vicinity of 7.0 of optimum HLB of the emulsifier mixture. The obtained nanoemulsions were stable without obvious change in droplet size in one month. This study provides valuable information for optimizing the formation of W/O nanoemulsions with low viscosity. These results suggest that W/O nanoemulsions of low viscosity could be useful for cosmetics with soft feeling.

A Review on the Health Risks Associated with the Use of Products Containing Benzalkonium Chloride (BKC), Focusing on Humidifier Disinfectant Products (염화벤잘코늄(Benzalkonium chloride) 살균제의 용도 및 건강 위험에 대한 고찰 - 가습기 살균제를 중심으로)

  • Background: Benzalkonium chloride (BKC) has been extensively used as a preservative in industrial products and in hygiene, medical, and cosmetic applications. Humidifier disinfectant (HD) products containing BKC have been used in South Korea. Objectives: This study was aimed to review types of products containing BKC, to summarize the regulations in the US, EU, Japan, and South Korea, and to review the health effects associated with the use of HD. Methods: We reviewed and summarized documents which were searched through PubMed and Google Scholar with the key words: BKC and asthma/contact dermatitis, humidifier disinfectant-associated lung injury (HDLI), and more. Results: Regulations in most countries including South Korea do not allow its use as disinfectants in spray-type of products for medicine, cosmetics, and household products. Two types of HD containing BKC (800~1,270 ppm) were marketed in South Korea from 1996~2003. Health effects reported from people who used products containing BKC were allergic contact dermatitis, erythema, and respiratory disease, including asthma. Two people who responded as HD users containing BKC only were confirmed to have developed asthma. HD-associated lung injury (HDLI) was reported by consumers who used both HD containing polyhexamethylene guanidine (PHMG) and HD containing BKC. Conclusions: In conclusion, the use of BKC as a biocide has to be controlled considering the route and pattern of exposure. Products containing BKC as preservatives were reviewed with exposure routes and sites in the human body such as skin, eyes, and the respiratory tract. HD containing BKC was clinically evaluated to be associated with asthma.

  • This study analyzed intra-regional disparities by using the concentration index of industrial organization theory. And the range of study is 11 cities in Chungcheongbuk-do province. This study aims to clarify the characteristics of the intra-regional disparities by using concentration ratio(CR) and Herfindahl-Hirschman Index. After the review of previous studies, we intend to study the intra-regional disparities using these two indexes. Target is the bio-health industry that all povinces in South Korea want to develop competitively. In addition the analysis was conducted by dividing the bio-health industry into four detailed fields(pharmaceuticals, medical devices, health functional foods, and cosmetics). These fields are based on the results of the city and county competitiveness-enhancing industry. The research results derived the intra-regional disparities index based on sales, the number of workers, and R&D expenses by detailed field. As a result it was confirmed that the strengths according to the detailed fields differed by region. It can be used as basic data in establishing differentiated industrial development strategies for each region in the future.

Antioxidant Effect of Hibiscus Extract (히비스커스 추출물의 항산화 효과)

  • Due to COVID-19, modern people have come to prefer natural substances as anxiety due to harmful environments and various stimuli has increased. Therefore, in order to find out the appropriateness of hibiscus, which is recognized as a non-toxic plant in traditional medicine, as a natural cosmetic material, the antioxidant effects (polyphenol, flavonoid, DPPH, ABTS) of hibiscus extract were studied, and the following results were obtained. First, the total polyphenol of hibiscus was found to be 433 ㎍/mL when the HSE was 100%. Second, the total flavonoids showed high antioxidant capacity at 488 ㎍/mL in 100% of the HSE. Third, the DPPH radical scavenging ability was found to be 94.04% in the undiluted HES and 89.54% in the diluted HSE 20%. Fourth, the ABTS radical scavenging ability was 98.95% in 100% of the HSE stock solution and 94.84% in the diluted HSE 20%, respectively, showing a high scavenging ability of more than 90%. As a result of these studies, it is thought that the hibiscus extract can be used as an antioxidant raw material for natural cosmetics in the future.

The Correlativity of Ulva lactuca Fractions, LPS, Enzymatic Activity and the Evaluation of Water Fraction (갈파래 분획, LPS, 효소활성의 상관성과 수층분획의 가치평가)

  • Lipopolysaccharide(LPS) was posttreated after the 14 day-pretreatment of Ulva lactuca fractions(ULF), and their correlativity to enzymatic activity alteration was investigated in the liver of rats. ULF was intraperitoneally administered into rats at dose of $1m{\ell}/kg$ of 100 mg/kg concentration for 14 days. On the day 15, $1m{\ell}/kg$ of LPS was injected. The corelativity was examined by measuring the changed values of superoxide dismutase(SOD) in mitochondrial fraction and catalase(CAT), glutathione peroxidase(GPx) in liver homogenate. The results showed that LPS treatment decreased the high values of SOD, CAT, GPx to the low values, but ULF pretreatment increased the low values of SOD, CAT, GPx to the high values. It was suggested that ULF, LPS and antioxidative enzymes like SOD, CAT, GPx had the corelativity of the high-low-high pattern and that the ULF pretreatment played the proper preventive role in the protection against the LPS treatment-induced enzymatic inactivity in the water fraction.

Antioxidant and Anti-inflammatory Effects of Extracts from the Flowers of Weigela subsessilis on RAW 264.7 Macrophages (RAW 264.7 대식세포에 미치는 병꽃나무 꽃 추출물의 항산화 및 항염증 효과)

  • This study investigated the antioxidant and anti-inflammatory activity of ethanol extract from the flowers of Weigela subsessilis (WS-E) in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. The total polyphenol and flavonoid content was 719.19±0.04 μg tannic acid equivalents/ml and 644.87±0.02 μg quercetin equivalents/ml, respectively. The antioxidant activities of WS-E were measured by 1,1-diphenyl-2-picrylhydrazyl (DPPH) and superoxide anion radical scavenging activity. The antioxidant activities of WS-E increased markedly, in a dose-dependent manner. To screen for anti-inflammatory agents, the inhibitory effects of WS-E on the production of proinflammatory cytokines in the LPS-stimulated RAW 264.7 macrophages was examined. WS-E had no effect on cell viability at a concentration of 100 μg/ml. Nitric oxide (NO) and interleukin (IL)-6 production were inhibited in a dose-dependent manner (p<0.05). WS-E had no effect on the production of tumor necrosis factor (TNF)-α at a concentration of 0.16–20 μg/ml but induced TNF-α at a concentration of 100 μg/ml. Inducible nitric oxide synthase (iNOS) expression was also inhibited at lower concentrations (p<0.05). In addition, WS-E reduced the activation of nuclear factor (NF)-κB by inhibition of inhibitoy (I) κB phosphorylation in RAW 264.7 macrophages upon stimulation with LPS (100 ng/ml) for 24 h but not that of mitogen-activated protein kinase (MAPK). These results suggest that WS-E may be a useful antioxidant and anti-inflammatory agent in functional cosmetics.

Human Studies on Functional Foods: How They Are Regulated (기능성식품 인체적용시험 제도 비교)

  • Along with the steady growth of health functional food (HFF) markets, research evaluating the human effects of HFF has been expanding. In this study, we investigated the regulatory and management system of human study on HFF in the USA, Japan and UK, and the Korean domestic regulations on HHF, medicines, medical devices, cosmetics and biotechnology in order to improve the domestic management system. In these four countries, institutional review board (IRB) or research ethics committee (REC) approvals are required for on human study of HHF, but regulatory and management systems differ from country to country. In the USA, human studies on HFF for structure/function claims do not require the FDA's prior approval but clinical trials of the disease treatment effects of HHF require prior approval from the FDA. In the USA, IRBs are managed by the Department of Health and Human Services (DHHS) rather than the FDA, and IRBs in those institutions which would execute the clinical trials requiring prior approval from the FDA or human studies funded by the USA federal government are required to be registered on the DHHS. In the UK, although the government does not require prior approval of human study, authorized RECs managed by the National Research Ethics Service (NRES) and other independent RECs review the human study. In Japan, human study for HFF must conform with "Ethical guidelines for epidemiological research" and IRB registration has not been required. In Korean domestic regulations, the responsibilities, compositions, functions and operations of IRBs on medicines, medical devices and biotechnology are legally specified, but not those of IRB on HHF. These foreign statuses for the management of human study on HFF and comparisons with Korean regulations are expected to be used as basic data to improve the domestic legal system.

Stable Liquid Paraffin-in-Water Nanoemulsions Prepared by Phase Inversion Composition Method (조성 상전이 방법으로 제조된 안정한 액상 파라핀-물 나노에멀젼)

  • Oil-in-water nanoemulsions were prepared in the system of water/Span 80-Tween 80/long-chain paraffin oil via the PIC (phase inversion composition) method. With the increase of preparation temperature from $30^{\circ}C$ to $80^{\circ}C$, the diameter of emulsion droplets decreased from 120 nm to 40 nm, proving the formation of nanoemulsions. By varying the HLB (hydrophilic lipophilic balance) of mixed surfactants, we found that there was an optimum HLB around 12.0 ~ 13.0 corresponding to the minimum droplet size. The viscosity of nanoemulsions clearly increased with droplet volume fraction, f, but the droplet size slightly increased. Significantly, at ${\phi}{\leq}0.3$, the size distribution of nanoemulsions kept constant more than 2 months. These results proved that the viscous paraffin oil can hardly be dispersed by the PIC method at $30^{\circ}C$, but the increase in preparation temperature makes it possible for producing monodisperse nanoemulsions. Once the nanoemulsion is produced, the stability against Ostwald ripening is outstanding due to the extremely low solubility of the liquid paraffin oil in the continuous phase. The highly stable nanoemulsions are of great importance in cosmetic applications.

Antioxidant and Antibacterial Effects of Korean Isodon japonicus H. (한국산 연명초(延命草)(Isodon japonicus Hara)의 항산화, 항균효과)

  • Biological activities and application of Isodon japonicus H. were investigated. In the physiological activities, the electron donating ability (EDA) was 66.3% in 100 ppm and SOD-like activity was as high as 85.0% in 1,000 ppm with gradual increase. As for the inhibitory effect of xanthine oxidase, it was 70.0% in 1,000 ppm and as low as 40.0% in 500 ppm; also, as for the inhibitory effect of tyrosinase, it was as low as 20.5% below 1,000 ppm. The tyrosinase inhibition effect related to skin whitening function showed 30.0% at 1,000 ppm level or below, indicating a relatively low effect. As for the result of measuring the lipid oxidation, all the concentrations of medical ion treatments showed anti-acidification ability; also, as for the metal ion blocking effects against the lipid oxidation promoting factors $(Fe^{2+}\;and\;Cu^{2+})$, $Fe^{2+}$ was better than $Cu^{2+}$ and all concentrations of medical ion treatments was 60.0% in 100 ppm. Also, the clear zone against various bacteria at 0.5 and 1.0 mg/disc was clearly shown. When it was applied into a normal skin-softener, it was safe, showing its potential as a natural material of cosmetics.
