Tae-Um-Jo-wee-Tang (TUJWT) and Do-Dam-Tang (DDT) have been used as an anti-obesity herbal medicine but their effect and mechanism of action have not been studied. We investigated the effects of TUJWT and DDT on energy and glucose homeostasis using Sprague Dawley female rats with diet-induced obesity. The mechanisms of action of TUJWT and DDT were studied whether they had anti-obesity effects. Rats fed a high-fat diet were divided into 3 groups: rats in each group received 2 g water extracts of modified TUJWT and DDT, or 2 g cellulose per kg body weight (a negative control) on a daily basis. A further group was fed a low-fat diet as a positive control. We found that DDT significantly decreased body weight and body fat (mesenteric fat and retroperitoneal fat) more than the control. This decrease was due to the reduction in energy intake but no changes of energy expenditure. However, DDT increased fat oxidation as a major energy source than the control. In addition, modified TUJWT and DDT improved glucose tolerance without changing serum insulin levels during an oral glucose tolerance test. In conclusion, DDT have a better anti-obesity effect than TUJWT by decreasing energy intake in female rats with diet-induced obesity. It also improves glucose tolerance.
The protective effect of'ischemic preconditioning'on ischemid-reperfusion injury of heart has been reported in various animal species. but without known mechAnism in detail, In An attempt to investigate the cardioprotective mechanism of ischemic preconditioning, we examined the effects of nitric oxide(UO) synthesis in preconditioned heart of rat The isolated hearts perfused by Langendorfr's method were ex- posed to 30min global ischemia followed by 30min reperfusion with oxygenated Krebs-Henseleit(K-H) sol- ution. Ischemic preconditioning was performed with three episodes of Sm n ischemia and Smin repeyfusion before the induction of prolong ischemia(30min)-reperfusion(30min). Ischemic preconditioning prevented the depression of cardiac function(left ventricular pressure .K heart rate) observed in the ischemia- reperfusion hearts and reduced the release of lactate dehydrogenase during the reperfusion period. On electromicroscopic pictures, myocardial ultrastructures wore relatively well preserved in isthemic preconditioned hearts. The failure of the WO synthesis inhibitor to reduce t e effect of ischemic preconditioning may be related to be species specific in that NO may allot be the trigger for ischemic preconditioning in rats.
The East China Sea and the Yellow Sea are abundant in nutritions because of river inflows and are important as the nursery and spawning grounds of demersal and pelagic fishes. The remarkable thermal front between the Yellow Sea Bottom Cold Water and the Tsushima Warm Current is formed in this region, and the fluctuation of this front may affect the variation of the yellow croaker fishing ground. To investigate the mechanism of the yellow croaker fishing ground, the distribution ana seasonal change of the fishing ground are examined by using catch of stow net fishery (Fisheries Research and Development Agency, 1970-1979) and the water temperature data (Japan Hydrographic Association, 1978). The main fishing ground of yellow croaker was nine sea areas (rectangle of 30' latitude by 30' longitude) located at 40-150 nautical miles west and southwest of Jeju Island, the area of which occupies no more than $11\%$ of all fishing grounds, and it appeared that about $70\%$ of total catch of ten years was concentrated in this area. The main fishing periods were from March to May and September to October. The coefficients of variation of the catch for the main fishing ground were from 0.8 to 2.1 and the condition of all fishing grounds was generally unstable. The mean CPUE was 27kg/haul at the main fishing ground, while it was the largest on boundary area of the Yellow Sea Bottom Cold Water. It was found that the seasonal movement of fishing ground is related to the expansion and reduction of the Yellow Sea Bottom Cold Water ($10^{\circ}C$).
Target herbal ingredient (THI) is an extract made from two herbs, Scutellariae Radix and Platycodi Radix. It has been developed as a treatment for metabolic diseases such as hyperlipidemia, atherosclerosis, and hypertension. One component of these two herbs has been reported to have anti-inflammatory, anti-hyperlipidemic, and anti-obesity activities. However, there have been no reports about the effects of the mixed extract of these two herbs on metabolic diseases. In this study, we investigated the metabolic effects of THI using a diet-induced obesity (DIO) mouse model. High-fat diet (HFD) mice were orally administered daily with 250 mg/kg of THI. After 10 weeks of treatment, the THI-administered HFD mice showed reduction of body weights and epididymal white adipose tissue weights as well as improved glucose tolerance. In addition, the level of total cholesterol in the serum was markedly reduced. To elucidate the molecular mechanism of the metabolic effects of THI in vitro, 3T3-L1 cells were treated with THI, after which the mRNA levels of adipogenic transcription factors, including C/$EBP{\alpha}$ and $PPAR{\gamma}$, were measured. The results show that the expression of these two transcription factors was down regulated by THI in a dose-dependent manner. We also examined the combinatorial effects of THI and swimming exercise on metabolic status. THI administration simultaneously accompanied by swimming exercise had a synergistic effect on serum cholesterol levels. These findings suggest that THI could be developed as a supplement for improving metabolic status.

PURPOSES : The objective of this study is to evaluate the effect of size and depth of cavities on the pavement failure using the full-scale accelerated pavement testing. METHODS : A full-scale testbed was constructed by installing the artificial cavities at a depth of 0.3 m and 0.7 m from the pavement surface for accelerated pavement testing. The cavities were made of ice with a dimension of 0.5 m*0.5m*0.3m, and the thickness of asphalt and base layer were 0.2 m and 0.3 m, respectively. The ground penetrating radar and endoscope testing were conducted to determine the shape and location of cavities. The falling weight deflectometer testing was also performed on the cavity and intact sections to estimate the difference of structural capacity between the two sections. A wheel loading of 80 kN was applied on the pavement section with a speed of 10 km/h in accelerated pavement testing. The permanent deformation was measured periodically at a given number of repetitions. The correlation between the depth and size of cavities and pavement failure was investigated using the accelerated pavement testing results. RESULTS : It is found from FWD testing that the center deflection of cavity section is 10% greater than that of the intact section, indicating the 25% reduction of modulus in subbase layer due to the occurrence of the cavity. The measured permanent deformation of the intact section is approximately 10 mm at 90,000 load repetitions. However, for a cavity section of 0.7 m depth, a permanent deformation of 30 mm was measured at 90,000 load repetitions, which is three times greater than that of the intact section. At cavity section of 0.3 m, the permanent deformation reached up to approximately 90 mm and an elliptical hole occurred at pavement surface after testing. CONCLUSIONS : This study is aimed at determining the pavement failure mechanism due to the occurrence of cavities under the pavement using accelerated pavement testing. In the future, the accelerated pavement testing will be conducted at a pavement section with different depths and sizes of cavities. Test results will be utilized to establish the criteria of risk in road collapse based on the various conditions.
Alzheimer's disease (AD) is the most common cause of dementia in the elderly, and its pathology is characterized by the presence of numerous numbers of senile plaques and neurofibrillary tangles. Several genetic and transgenic studies have indicated that excess amount of $\beta$-amyloid protein (A$\beta$) is produced by mutations of $\beta$TEX>$\beta$-amyloid precursor protein and causes learning impairment. Moreover, $A\beta$ has a toxic effect on cultured nerve cells. To prepare AD model animals, we have examined continuous (2 weeks) infusion of $A\beta$ into the cerebral ventricle of rats. Continuous infusion of $A\beta$ induces learning impairment in water maze and passive avoidance tasks, and decreases choline acetyltransferase activity in the frontal cortex and hippocampus. Immunohistochemical analysis revealed diffuse depositions of $A\beta$ in the cerebral cortex and hippocampus around the ventricle. Furthermore, the nicotine-evoked release of acetylcholine and dopamine in the frontal cortex/hippocampus and striatum, respectively, is decreased in the $A\beta$-infused group. Perfusion of nicotine (50 $\mu\textrm{M}$) reduced the amplitude of electrically evoked population spikes in the CA1 pyramidal cells of the control group, but not in those of the $A\beta$-infused group, suggesting the impairment of nicotinic signaling in the $A\beta$-infused group. In fact, Kd, but not Bmax, values for [$^3H$] cytisine binding in the hippocampus significantly increased in the $A\beta$-infused rats. suggesting the decrease in affinity of nicotinic acetylcholine receptors. Long-term potentiation (LTP) induced by tetanic stimulations in CA1 pyramidal cells, which is thought to be an essential mechanism underlying learning and memory, was readily observed in the control group, whereas it was impaired in the $A\beta$-infused group. Taken together, these results suggest that $A\beta$ infusion impairs the signal transduction mechanisms via nicotinic acetylcholine receptors. This dysfunction may be responsible, at least in part, for the impairment of LTP induction and may lead to learning and memory impairment. We also found the reduction of glutathione- and Mn-superoxide dismutase-like immunoreactivity in the brains of $A\beta$-infused rats. Administration of antioxidants or nootropics alleviated learning and memory impairment induced by $A\beta$ infusion. We believe that investigation of currently available transgenic and non-transgenic animal models for AD will help to clarify the pathogenic mechanisms and allow assessment of new therapeutic strategies.
Photocatalytic degradation of bisphenol A (BPA) in aqueous solution was investigated using $TiO_2$ nanoparticles (Degussa P25) in this study. After a 3 hr photocatalytic reaction (${\lambda}=365nm$ and $I=3mW\;cm^{-2}$, $[TiO_2]=2.0g\;L^{-1}$), 98% of BPA ($1.0{\times}10^{-5}M$) was degraded and 89% of the total organic carbon was removed. In addition, BPA degradation by photolytic, hydrolytic and adsorption reactions was found to be 2%, 5% and 13%, respectively. The reaction rate of BPA degradation by photocatalysis decreased with increasing concentration of methanol that is used as a hydroxyl radical scavenger. This indicates that the reaction between BPA and hydroxyl radical was the key mechanism of BPA degradation. The pseudo-first-order reaction rate constant for this reaction was determined to be $7.94{\times}10^{-4}min^{-1}$, and the time for 90% BPA removal was found to be 25 min. In addition, acute toxicity testing using Daphnia magna neonates (< 24 h old) was carried out to evaluate the reduction of BPA toxicity. Acute toxicity (48 hr) to D. magna was decreased from 2.93 TU (toxic unit) to non-toxic after photocatalytic degradation of BPA for 3 hr. This suggests that there was no formation of toxic degradation products from BPA photocatalysis.

Three-dimensional (3D) numerical analyses have been performed to study the behaviour of single piles and grouped piles to adjacent tunnelling in the lateral direction of the pile. In the numerical analyses, the interaction between the tunnel, the pile and the soil next to the piles and shear transfer mechanism have been analysed allowing soil slip at the pile-soil interface by using interface elements. The study includes the shear stresses at the soil next to the pile, the axial force distributions on the pile and the pile settlement. It has been found that existing elastic solutions may not accurately estimate the pile behaviour since several key issues are excluded. Due to changes in the shear transfer between the pile and the soil next to the pile with tunnel advancement, the shear stresses and axial force distributions along the pile change drastically. Downward shear stress develops above the tunnel springline while upward shear stress is mobilised below the tunnel springline, resulting in a compressive force on the pile. In addition, mobilisation of shear strength at the pile-soil interface was found to be a key factor governing pile-soil-tunnelling interaction. It has been found that grouped piles are less influenced by the tunnelling than the single pile in terms of the axial pile forces. The reduction of apparent allowable pile capacity due to pile settlement resulted from the tunnelling seemed to be insignificant.

Diazinon is a phosphorothiate insecticide widely used in the world including Korea. This study investigates the feasibility of photolysis and photocatalysis processes for the degradation of diazinon in water. Both photolysis and photocatalysis reactiosn were effective in degrdading diazinon, however lower TOC removals were achieved. In case of photocatalysis, approximately 40% of nitrogen from diazinon was recovered as NO$_3^-$, and less than 5% of phosphorus as PO$_4{^{3-}}$. However, the sulfur in diazinon molecule was completely recovered to SO$_4{^{2-}}$ from photocatalysis reaction, and the recovery from photolysis was 50%, indicating that P=S bond easily breaks first during photolysis and photocatalysis. The poor recoveries of ionic byproducts and TOC from photolysis and photocatalysis indicate the presence of other organic intermediates during reactions. The formation of organic intermediates were identified during reactions using GC/MS and LC/MS/MS, and the main degradation products were diazoxon, and 2-isopropyl-4-methyl-6-hydroxypyrimidine (IMP), respectively. Finally, the acute 48-hr toxicity test using Daphnia magna were employed to measure the toxicity reduction during photocatalysis of degradation. The results showed that the toxicity increased until 180 min of the photocatalysis reaction (from EC$_{50}$ (%) of 69.6 to 13.2%), however, acute toxicity completely disappeared (>100%) after 360 min. The toxicity results showed that the intermediates from photocatalysis such as diazoxon were more toxic than diazinon itself, however these intermediates can be degraded or mineralized with further reaction.

Ischemic preconditioning (IPC) is known to protect the heart against ischemia/reperfusion (IR)-induced injuries, and regional differences in the mitochondrial antioxidant state during IR or IPC may promote the death or survival of viable and infarcted cardiac tissues under oxidative stress. To date, however, the interplay between the mitochondrial antioxidant enzyme system and the level of reactive oxygen species (ROS) in the body has not yet been resolved. In the present study, we examined the effects of IR- and IPC-induced oxidative stresses on mitochondrial function in viable and infarcted cardiac tissues. Our results showed that the mitochondria from viable areas in the IR-induced group were swollen and fused, whereas those in the infarcted area were heavily damaged. IPC protected the mitochondria, thus reducing cardiac injury. We also found that the activity of the mitochondrial antioxidant enzyme system, which includes manganese superoxide dismutase (Mn-SOD), was enhanced in the viable areas compared to the infarcted areas in proportion with decreasing levels of ROS and mitochondrial DNA (mtDNA) damage. These changes were also present between the IPC and IR groups. Regional differences in Mn-SOD expression were shown to be related to a reduction in mtDNA damage as well as to the release of mitochondrial cytochrome c (Cyt c). To the best of our knowledge, this might be the first study to explore the regional mitochondrial changes during IPC. The present findings are expected to help elucidate the molecular mechanism involved in IPC and helpful in the development of new clinical strategies against ischemic heart disease.
