Ameliorative Effect of Pu-erh Tea on DSS-induced Colitis through Regulation of NF-κB Activation in Mice

  • Ulcerative colitis (UC), chronic inflammatory bowel disease, is characterized by severe inflammation in the colon. Tea is one of the most popular beverages consumed worldwide. Pu-erh tea, a unique Chinese tea produced by microbial activities, possesses a broad range of health-promoting effects, including anti-aging, anti-Alzheimer's disease, antioxidation and anti-obesity. However, the inhibitory effect of Pu-erh tea on intestinal inflammation and the underlying mechanism remain unclear. The present study was designed to evaluate the regulatory effect of Pu-erh tea extract (PTE) on dextran sulfate sodium (DSS)-induced colitis clinical signs by analyzing the weight loss and colon length in mice. The inhibitory effects of PTE on inflammatory mediators, such as interleukin (IL)-6 and tumor necrosis factor (TNF)-α, and the activation of nuclear factor-κB (NF-κB) were also determined in DSS-treated colitis tissue. We observed that PTE treatment significantly inhibited the DSS-induced clinical symptoms of weight loss, decrease,in colon length, and colon tissue damage in mice. Moreover, PTE attenuated the DSS-induced levels of IL-6 and TNF-α in colon tissue. We also demonstrated the anti-inflammatory mechanism of PTE by suppressing the activation of NF-κB in DSS-treated colon tissues. Collectively, the findings provide experimental evidence that PTE may be effective in preventing and treatment of intestinal inflammatory disorders, including UC.

Undaria pinnatifida Inhibits the Mast Cell-Mediated Inflammatory Response via NF-κB/Caspase-1 Suppression

  • Marine sources as potential treatment options for various diseases have been a subject of growing interest. However, information on the anti-inflammatory mechanism employed by Undaria pinnatifida (UP) remains limited. The present study was conducted to investigate the mechanisms of UP on the mast cell-mediated inflammatory response. To determine the pharmacological mechanism of UP in inflammatory reaction, we evaluated the effects of UP on interleukin (IL)-6, IL-8 and tumor necrosis factor (TNF)-α production and nuclear factor-κB (NF-κB) and caspase-1 activation in calcium ionophore A23187 plus phorbol 12-myristate 13-acetate-stimulated human mast cells-1 (HMC-1). The results showed that UP suppressed IL-6, IL-8 and TNF-α production in a dose-dependent manner. Moreover, UP significantly attenuated NF-kB/caspase-1 activation in stimulated HMC-1. Collectively, these findings provide experimental evidence that UP may be a useful candidate for the inflammation-related diseases treatment.

An Interactive Multi-Factor User Authentication Framework in Cloud Computing

  • Identity and access management in cloud computing is one of the leading significant issues that require various security countermeasures to preserve user privacy. An authentication mechanism is a leading solution to authenticate and verify the identities of cloud users while accessing cloud applications. Building a secured and flexible authentication mechanism in a cloud computing platform is challenging. Authentication techniques can be combined with other security techniques such as intrusion detection systems to maintain a verifiable layer of security. In this paper, we provide an interactive, flexible, and reliable multi-factor authentication mechanisms that are primarily based on a proposed Authentication Method Selector (AMS) technique. The basic idea of AMS is to rely on the user's previous authentication information and user behavior which can be embedded with additional authentication methods according to the organization's requirements. In AMS, the administrator has the ability to add the appropriate authentication method based on the requirements of the organization. Based on these requirements, the administrator will activate and initialize the authentication method that has been added to the authentication pool. An intrusion detection component has been added to apply the users' location and users' default web browser feature. The AMS and intrusion detection components provide a security enhancement to increase the accuracy and efficiency of cloud user identity verification.

Transcription Factor EB-Mediated Lysosomal Function Regulation for Determining Stem Cell Fate under Metabolic Stress

  • Stem cells require high amounts of energy to replicate their genome and organelles and differentiate into numerous cell types. Therefore, metabolic stress has a major impact on stem cell fate determination, including self-renewal, quiescence, and differentiation. Lysosomes are catabolic organelles that influence stem cell function and fate by regulating the degradation of intracellular components and maintaining cellular homeostasis in response to metabolic stress. Lysosomal functions altered by metabolic stress are tightly regulated by the transcription factor EB (TFEB) and TFE3, critical regulators of lysosomal gene expression. Therefore, understanding the regulatory mechanism of TFEB-mediated lysosomal function may provide some insight into stem cell fate determination under metabolic stress. In this review, we summarize the molecular mechanism of TFEB/TFE3 in modulating stem cell lysosomal function and then elucidate the role of TFEB/TFE3-mediated transcriptional activity in the determination of stem cell fate under metabolic stress.

  • It is well known that osteoblasts and osteoc1asts playa key role in bone metabolism. They involve in osteoformation or bone destruction which are ragulated by various factors such as thyroid hormone, parathyroid hormone, estrogen, growth factor and cytokine. Recently, it is demonstrated that oxidative stress is one of pathological factors in bone metabolism, but it is left unknown about mechanism between oxidative stress and bone metabolism.(omitted)

Inferring candidate regulatory networks in human breast cancer cells

  • Human cell regulatory mechanism is one of suspicious problems among biologists. Here we tried to uncover the human breast cancer cell regulatory mechanism from gene expression data (Marc J. Van de vijver, et. al., 2002) using a module network algorithm which is suggested by Segal, et. al.(2003) Finally, we derived a module network which consists of 50 modules and 10 tree depths. Moreover, to validate this candidate network, we applied a GO enrichment test and known transcription factor-target relationships from Transfac(R) (V. Matys, et. al, 2006) and HPRD database (Peri, S. et al., 2003).

MiT Family Transcriptional Factors in Immune Cell Functions

  • The microphthalmia-associated transcription factor family (MiT family) proteins are evolutionarily conserved transcription factors that perform many essential biological functions. In mammals, the MiT family consists of MITF (microphthalmia-associated transcription factor or melanocyte-inducing transcription factor), TFEB (transcription factor EB), TFE3 (transcription factor E3), and TFEC (transcription factor EC). These transcriptional factors belong to the basic helix-loop-helix-leucine zipper (bHLH-LZ) transcription factor family and bind the E-box DNA motifs in the promoter regions of target genes to enhance transcription. The best studied functions of MiT proteins include lysosome biogenesis and autophagy induction. In addition, they modulate cellular metabolism, mitochondria dynamics, and various stress responses. The control of nuclear localization via phosphorylation and dephosphorylation serves as the primary regulatory mechanism for MiT family proteins, and several kinases and phosphatases have been identified to directly determine the transcriptional activities of MiT proteins. In different immune cell types, each MiT family member is shown to play distinct or redundant roles and we expect that there is far more to learn about their functions and regulatory mechanisms in host defense and inflammatory responses.

제품에서 Fun감성이 유발되는 요인의 분석방법에 관한 연구 (Research on Analytical Method of fun Generating Factor of Product)

  • 제품디자인에서 Fun이라는 주제를 표현하기 위해 많은 디자이너들이 심리학적 측면에서의 접근방법을 사용하고 있다. 그렇다면 심리학에서 이야기 하는 Fun발생 메커니즘은 제품디자인에 접목하기에 많은 보완이 필요하다고 본다. 현재 심리학에서는 Fun이라는 감성을 유발시키기 위한 메커니즘으로 어떤 상황을 직면하였을 때 심적 정체를 겪은 후, 어떠한 실마리에 의한 정체의 해소를 겪게 될 때 Fun을 느끼게 된다고 이야기 한다. 심리학적 측면에서 Fun을 발생시키는 메커니즘의 도출은 그 접근 방식 자체에서 인지적인 측면의 Fun만을 이야기 하는 경향이 있다. 이러한 성향은 조형을 가지고 있는 제품디자인을 이야기 할 경우, 제품에서 중요한 역할을 가지는 지각적인 측면의 재미를 충분히 설명하기에는 역부족인면이 있다. 본 연구에서는 심리학에서 다루어지는 인지적 재미요인 이외에 지각적인 측면의 Fun을 제품디자인에서 같이 논의하는 것이 좀더 타당한 접근방법이라는 가설을 세웠다. 이를 위해 지각적 측면의 Fun을 이해하기 위해 키치라는 문화현상 중에 키치제품의 유희성이라는 측면을 도입하기로 한다. 키치제품의 유머러스한 형태를 도출하는 방법을 이야기 하고 있으며 이러한 방법들이 지각적인 측면에서도 같이 적용될 수 있는 것이다. 이러한 가설을 통하여, 내면의 기능적 특성인 인지적 요소와 표면적 고유특성인 지각적인 요소가 모두 거론되어야 실질적인 방법으로 사용될 수 있으리라 생각된다.

  • 현재의 인터넷망에서 채택하고 있는 최선노력 서비스만으로는 실시간 응용 서비스들이 요구하는 서비스 품질 (QoS: Quality of Service) 보장을 만족시키는 것이 어렵다. 따라서 세계적으로 Q0S를 제공하기 위한 방법에 대한 연구가 진행되고 있다. 그 중에서 본 논문에서는 차등 서비스(Diffserv: Differentiated Service)를 이용한 QoS 보장 기법에 대하여 기술하였다. 먼저 차등 서비스의 성능이 어떻게 트래픽 모델에 의하여 영향을 받는지 분석하였다. 이를 위하여, 랜덤, 버스티, 자기유사 입력 트래픽 모델링 방법에 대하여 연구를 수행하였다. 그리고 OPNET 시뮬레이터를 설계 및 구현하고 다양한 입력 파리미터에 대하여 모의실험과 성능평가를 수행하였다. 성능분석 결과 모든 환경에서 DiffServ 기능을 이용하여 EF(Expedited Forwarding) 클래스에 대하여 QoS 보장이 가능하고, EF와 BE(Best Effort) 클래스의 서비스 분리가 가능함을 확인할 수 있었다. WFQ(Weighted Fair Queueing)에서 EF와 BE 클래스 사이에 주어진 자원의 분배(즉, Weight factor)에 따른 성능 변화와 역동적 행위에 대하여도 분석하였다.

A Study on the Structural Causal Relationship of Marketing Stimulating Factors on Online Impulse Purchase through Consumer Value: Focusing on the Mediating Effect of Consumer Value

  • Purpose: With the introduction of Web 2.0, the advent of smartphones, and the recent outbreak of COVID-19 worldwide, interest in online shopping has soared. In an online shopping environment, the desire for impulse buying increases compared to offline shopping. Research design, data and methodology: In this study, three marketing stimulating factors such as product factor, price factor, and facilitating factor were selected which affect impulse buying for clothing and accessory products in the Korean online shopping environment. The mechanism of causal relationship among them, and the role customer value between marketing stimulating factor and impulse buying was analyzed. Results: The analysis results are as follows. First, the product factor had a significant positive effect on consumer value and impulse buying. Second, the price factor also had a significant positive effect on consumer value and impulse buying. Conclusions: Although the facilitating factor had no significant influence on consumer value and impulse buying, the indirect effect through consumer value was positively significant. Consumer value had a positive mediating effect on impulse purchase in the order of price factor, product factor, and promotion factor.
