Production of PMA-induced MMP-2 and MMP-9 in the HT-1080 Fibrosarcoma Cell Line is Inhibited by Corydalis heterocarpa via the MAPK-related Pathway (PMA로 자극된 HT-1080 세포에서 염주괴불주머니 추출물의 MAPK 경로를 통한 MMP-2, MMP-9 발현 억제 효과)

  • Matrix metalloproteinase (MMP) enzymes are responsible for the degradation and formation of the extracellular matrix (ECM), and overproduction of MMPs is observed in several diseases, such as cancer and asthma, that progress with metastatic characteristics. Natural products, especially phytochemicals, have been an important source of MMP inhibitors with reduced side effects. Although the majority of phytochemicals inhibit the enzymatic activity of MMPs, some suppress MMP production. In this context, the current study evaluated the potential of Corydalis heterocarpa, a halophyte with reported bioactivities, to inhibit MMP expression in PMA-stimulated HT-1080 cells. A crude C. heterocarpa extract was shown to decrease the mRNA and protein expression of MMP-2 and MMP-9 while increasing the endogenous MMP inhibitors TIMP-1 and TIMP-2 which regulate MMP expression in healthy tissues. In addition, our results show that the inhibitory effects of C. heterocarpa might occur through suppression of the phosphorylation of MAPK signaling, the upstream activator of MMP overexpression. In conclusion, C. heterocarpa is a potential source of antimetastatic compounds that might serve as lead molecules to develop novel MMP inhibitors.

Ameliorative Effect of Persicaria Poliata Etract through the Rgulation of AP-1, PI3K/Akt and MAPK Sgnaling Mlecules in UVB-Iradiated HaCaT Clls (HaCaT 세포에서 며느리 배꼽 추출물의 AP-1, PI3K/Akt 및 MAPK 활성 조절을 통한 광손상 억제 효과)

  • Purpose : Skin is the primary barrier to protect the body from various exogenous factors. Among them, UVB exposure can cause the induction of not only excessive inflammatory responses but also the degradation of extracellular matrix (ECM), including collagen and elastin. This study tried to investigate the ameliorative effect of Persicaria perfoliata ethanol extract (PPEE) on UVB-irradiated photodamage through the regulation of activator protein (AP)-1, phosphoinositide 3-kinase (PI3K)/Akt, and mitogen-activated protein kinase (MAPK) signaling molecules in HaCaT cells. Methods : The cytotoxicity of PPEE on HaCaT cells was evaluated by the WST-1 assay. The 80 mJ/cm2 of UVB (312 nm) was irradiated on HaCaT cells to induce the photodamage. Western blot analysis was conducted to investigate the protein expression levels of cyclooxygenase (COX)-2, matrix metalloproteinase (MMP)-9, and heme oxygenase (HO)-1 for ameliorative status by PPEE treatment in UVB-exposed HaCaT cells. In addition, the activated status of the inflammatory transcription factor, AP-1, as well as upstream signaling molecules, PI3K/Akt, and MAPK, were also evaluated by Western blot analysis. Results : Any cytotoxic effect was not induced at the concentration up to 200 ㎍/ml by PPEE treatment. Protein expression levels of COX-2 and MMP-9 were significantly down- and up-regulated by PPEE treatment. The inflammatory transcription factor AP-1, stimulated by UVB irradiation, was also significantly attenuated by PPEE treatment. The phosphorylated status of PI3K/Akt and MAPK were mitigated by PPEE treatment in UVB-exposed HaCaT cells. Moreover, PPEE treatment potently accelerated the expression of HO-1 and its transcription factor, nuclear factor-erythroid 2-related factor (Nrf)2, which is known for its anti-inflammatory activity. Conclusion : Consequently, PPEE treatment significantly regulated COX-2 and MMP-9 expressions in UVB-irradiated HaCaT cells. The inflammatory transcription factor AP-1, along with upstream signaling molecules PI3K/Akt and MAPKs, were also attenuated by PPEE treatment in UVB-exposed HaCaT cells. Additionally, PPEE treatment exaggerated HO-1 expression and Nrf2 activation, which might have contributed to the anti-inflammatory activity of PPEE. These results indicate that PPEE could be a candidate for attenuating UVB-induced photodamage in human skin.

Anti-aging Effect of Akebia quinata Decaisne Ethanol Extract (으름덩굴 에탄올 추출물의 항노화 효과)

  • Skin aging progresses due to external factors such as ultraviolet rays and infections. These factors cause skin fibroblasts to secrete proteolytic enzymes, matrix metalloproteinases (MMPs). MMPs induce the degradation of collagen located in the extracellular matrix, directly influencing aging. The stems of Akebia quinata Decaisne have been reported to have antioxidant and anti-inflammatory effects. However, the anti-aging effect of Akebia quinata Decaisne stem ethanol extract (AQSEE) is not known. Therefore, we studied the TNF-α-induced MMP-1 inhibitory effect in human fibroblasts. When the cell viability of AQSEE was confirmed through MTT asaay, it showed no toxicity up to 400 ㎍/mL. The inhibition of MMP-1 mRNA and protein secretion was confirmed through RT-qPCR and ELISA, and results showed a significant decrease at concentrations of 100, 200, 400 ㎍/mL. We also confirmed by Western blotting that phosphorylation of MAPKs signaling pathway and transcription factors was reduced. As a result, phosphorylation of p38, c-Jun, p65 was significantly decreased at all concentrations. DPPH and ABTS assays were performed to confirm the radical scavenging ability of AQSEE, and the results showed a significant decrease at all concentrations. The results of this study confirmed the MMP-1 inhibitory effect and radical scavenging ability, which suggests that it can be used as an anti-aging substance.

Inhibitory Effect on Kaempferia Parviflora Ethanol Extract of IL-1β Induced Inflammation and MMP Expression in CHON-001 Cells (흑생강 추출물의 CHON-001 세포에서의 IL-1β로 유도된 염증과 MMPs 발현)

  • The potential therapeutic effects of Kaempferia Parviflora ethanol extract (KPE) on osteoarthritis were investigated using the human chondrocyte cell line (CHON-001) to explore its application in functional foods. The CHON-001 cells were pre-treated with either 1 ㎍/ml or 5 ㎍/ml of KPE before exposure to 10 ng/ml of IL-1β to induce osteoarthritis. Results showed that KPE treatment significantly suppressed IL-1β-induced TNF-α production by 66% and 50% at concentrations of 1 ㎍/ml and 5 ㎍/ml KPE, respectively. In addition, COX-2 protein expression was reduced by 26% and 34% compared to control levels. The preservation of chondrocyte-specific matrix proteins, aggrecan, and collagen type II, was notable, with aggrecan and mRNA levels maintained by 5% and 8%, and collagen II levels preserved by 62% and 47% at the same KPE concentrations. This preservation is likely due to the reduced expression of MMP1 and MMP13, enzymes responsible for matrix protein degradation. Overall, the current results suggest that KPE may protect chondrocytes from IL-1β-induced osteoarthritis by suppressing TNF-α production and COX-2 expression while preserving critical matrix proteins like aggrecan and collagen II by suppressing the expressions of their degrading enzymes (MMP-1 and MMP-13). Therefore, KPE holds promise as a candidate for developing functional foods aimed at reducing osteoarthritis.

Proteinases and their Inhibitors in Cartilage and Synovial Fluid Acquired from a Canine Osteoarthritic Model (개 퇴행성 관절염 모델을 이용한 연골과 활액 내 단백질 분해 효소와 억제제의 작용 연구)

  • Chondrocytes and synovial fluid derived markers are used to monitor for osteoarthritis(OA). Specific inhibitors, known as tissue inhibitors of metalloproteinases(TIMP), regulate the proteolytic activity of matrix metalloproteinases(MMP). This study investigated whether MMP and TIMP levels were altered in synovial fluid and cartilage following the experimental induction of OA in canines. Twenty mature beagle dogs underwent a unilateral surgical transection of the cranial cruciate ligament and the medial collateral ligament as well as a medial meniscectomy. Matrix metalloproteinase-2 and MMP-9 levels were assayed using Western blot and TIMP-2 levels were measured with enzyme-linked immunosorbent assays four weeks after OA induction. Increased MMP-2 expression was observed in chondrocytes isolated from cartilage following OA induction, but MMP-9 expression decreased. Matrix metalloproteinase-2 and MMP-9 levels in synovial fluid from the OA induced joint significantly increased compared to those of the sham group. Tissue inhibitors of metalloproteinase-2 concentrations were higher in chondrocytes from the OA cartilage, yet TIMP-2 remained lower in the synovial fluid of OA. This suggests the elevated release of MMP-9 over MMP-2 into the synovial fluid following the cartilage degradation-related death of chondrocytes after OA. Osteoarthritis can be further deteriorated by increased MMP activity in the synovial fluid because TIMP-2 exist low concentration into the extracellular matrix. As a result, MMP activity, particularly MMP-9 activity, can be useful as a biomarker in diagnosing and monitoring the early stages of canine OA.

  • Schisandrae Fructus (SF), the fruit of Schisandra chinensis (Turcz.) Baill., is widely used in traditional medicine for the treatment of a number of chronic diseases. SF extracts have been recently reported to attenuate the inflammatory responses in SW1353 human chondrocyte cells in in vitro and monosodium iodoacetate (MIA)-induced cartilage degradation in in vivo osteoarthritis (OA) models. However, their protective and therapeutic potentials against OA in primary culture chondrocytes and animal models remain unclear. Therefore, we investigated the effects of the ethanol extract of SF on the activity of matrix metalloproteinases (MMPs), biomarkers for diagnosis of OA, on interleukin $(IL)-1{\beta}-induced$ primary cultured rat cartilage chondrocytes and MIA-induced osteoarthritis in a rat model. Our data indicated that SF treatment significantly reduced the mRNA expression and enzyme activity of MMP-1, -3 and -13 in $IL-1{\beta}-induced$ primary cultured rat cartilage chondrocytes. The chondro-protective effects of SF were then analyzed in a rat OA model using a single intra-articular injection of MIA in the right knee joint. According to our results, the elevated levels of MMP-1 and -3 were markedly ameliorated by SF administration. Collectively, these findings indicate that SF could be a candidate for the treatment of OA.

Evaluation of the Impact on Manufacturing Temperature and Time in the Production Process of Bio-composites (바이오복합재료 제조 공정시 제조온도 및 시간에 의한 영향 평가)

  • The main objective of this research was conducted to evaluate the impacts on the thermoplastic polymer which is a matrix polymer and the rice husk flour (RHF) which is a reinforcing filler relative to the manufacturing temperature and time when bio-composites were manufactured. In order to evaluate the impacts on the rice husk flour relative to the manufacturing temperature, the rice husk flour was persevered for 10 minutes to 2 hours period at $220^{\circ}C$ temperature which was then added with the polypropylene (PP) and low-density polyethylene (LDPE) to complete the manufacturing process of the bio-composites and measure the corresponding mechanical properties. As preserving time increased at $220^{\circ}C$, the tensile and impact strength were decreased due to the thermal degradation of the main components within the rice husk flour. The thermogravimetric analysis (TGA) was used to measure weight loss caused by the actual manufacturing temperature and the result was that the thermoplastic polymer had not scarcely occurred weight change, but there had been increasing rate of weight loss relative to time for the rice husk flour and the bio-composites under the consistent temperature of $220^{\circ}C$ for 2 hour time period. Therefore, the proper manufacturing temperature and time settings are significantly important features in order to prevent the reduction of mechanical properties which were induced throughout the manufacturing process under the high manufacturing temperature.

A Study on Phase-Multiplexed Volume Hologram using Spatial Light Modulator (공간광변조기를 이용한 위상다중화 체적 홀로그램에 관한 연구)

  • For an effective phase multiplexing in a volume holographic system, four types of phase code, pseudo random code(PSC), Hadamard matrix(HAM), pure random code(PRC), equivalent random code(ERC), used as reference beams are generated. In case of $32{\times}32$ address beam, a phase error with 0%, 5%, 10%, 15%, 20%, and 25% error rate, is purposely added to the real phase values in order to consider the practical SLM's nonlinear characteristics of phase modulation in computer simulation. Crosstalks and SNRs(signal-to-ratio) are comparatively analyzed for these phase codes by the auto-correlation and cross-correlation. PSC has the lowest cross-correlation mean value of 0.067 among four types of phase code, which means the SNR of the pseudo random phase code is higher than other phase codes. Also, the standard deviation of the pseudo random phase code indicating the degree of recalled data degradation is the lowest value of 0.0113. In order to analyze the affect by variation of pixel size, simulation is carried out by same method for the case of $32{\times}32$, $64{\times}64$, $128{\times}128$, $256{\times}256$ address beams.

Influence of Strain-Hardening Cement Composite's Tensile Properties on the Seismic Performance of Infill Walls (변형경화형 시멘트 복합체의 인장성능에 따른 끼움벽의 내진성능)

  • This paper describes experimental results on the seismic performance of SHCC (strain-hardening cement composite) infill wall for improving damage tolerance capacity of non-ductile frame. To investigate the effect of tensile strain capacity and cracking behavior of SHCC materials on the shear behavior of SHCC infill wall, three infill walls were fabricated and tested under cyclic loading. The test parameter in this study is a type of cement composites; concrete and SHCCs. The two types of SHCC materials were prepared for infill walls. In order to induce crack damages into the mid-span of the infill wall, each infill wall had two 100-mm-deep-notches on both sides. Test results indicated that SHCC infill walls showed superior crack control capacities and much larger drift ratios at the peak loads than RC (reinforced concrete) infill wall, as expected. In particular, due to the bridging actions of the reinforcing fibers, SHCC matrix used in this study would delay the stiffness degradation of infill wall after the first inclined cracking. Moreover, from the damage classes based on the cracks' maximum width in the infill walls, it was observed that PIW-SHD specimen possessed nearly threefold seismic capacities compared to PIW-SLD specimen. Also, from the results on the strain of diagonal reinforcements, it can be concluded that the SHCC matrix would resist a part of tensile stresses transferred along steel rebar in the infill wall.

Improved Direction of Arrival Estimation Based on Coprime Array and Propagator Method by Noise Power Spectral Density Estimation (잡음 파워 스펙트럼 밀도 추정을 이용한 서로소 배열과 프로퍼게이터 기법 기반의 향상된 도래각 추정 기법)

  • We propose an improved direction of arrival (DoA) estimation algorithm based on co-prime array and propagator method. The propagator method with co-prime array does not require singular value decomposition (SVD) requiring much less computational complexity but exhibiting somewhat worse performance in comparison with MUSIC based on co-prime array. We notice that one cause of the performance degradation was in the avoidance of the usage of the diagonal elements of the signal autocorrelation matrix that contains the noise power spectral density. So we propose an algorithm with the diagonal elements of the signal autocorrelation matrix based on the fact that the noise power spectral density can be estimated using noise observation over a long period of time. We observe, through simulations, that the proposed scheme in this paper improves the performance, with 4 times more computational requirement, by signal-to-noise ratio of 1.5dB and by DoA resolution of $0.7^{\circ}$ at the detection probability of 95% compared with the previously introduced co-prime array propagator scheme, resulting in performance much closer to that of co-prime array-based MUSIC scheme.
