A Linear Beacon System Featuring an Internal Deoxyguanine Quencher Allows Highly Selective Detection of Single Base Mismatches

  • The fluorescence intensity of a single-stranded oligonucleotide containing a fluorene-labeled deoxyuridine $(U^{Fl})$ unit increases by only 1.5-fold upon formation of its perfectly matched duplex. To increase the fluorescence signal during hybridization, we positioned a quencher strand containing a deoxyguanine (dG) nucleobase, functioning as an internal quencher, opposite to the $U^{Fl}$ unit to reduce the intrinsic fluorescence upon hybridization with a probe. From an investigation of the optimal length of the quencher strand and the effect of the neighboring base sequence, we found that a short strand (five-nucleotide) containing all natural nucleotides and dG as an internal quencher was effective at reducing the intrinsic fluorescence of a linear beacon; it also exhibited high total discrimination factors for the formation of perfectly matched and single base-mismatched duplexes. Such assays that function based on clear changes in fluorescence in response to single-base nucleotide mutations would be useful tools for accelerating diagnoses related to various diseases.

Enantiomeric Resolution of α-Amino Acid Derivatives on Two Diastereomeric Chiral Stationary Phases Based on Chiral Crown Ethers Incorporating Two Different Chiral Units

  • Two diastereomeric chiral stationary phases (CSPs) were applied to the liquid chromatographic resolution of various racemic ${\alpha}$-amino methyl esters, ${\alpha}$-amino N,N-diethylamides and ${\alpha}$-amino N-propylamides. The CSP incorporating (R)-3,3'-diphenyl-1,1'-binaphtyl and (R,R)-tartaric acid unit as chiral barriers did not show any chiral recognition. In contrast, the CSP incorporating (R)-3,3'-diphenyl-1,1'-binaphtyl and (S,S)-tartaric acid unit as chiral barriers was found to show excellent chiral recognition especially for the two enantiomers of ${\alpha}$-amino N-propylamides. Some of ${\alpha}$-amino methyl esters and ${\alpha}$-amino N,N-diethylamides were also resolved on the CSP incorporating (R)-3,3'-diphenyl-1,1'-binaphtyl and (S,S)-tartaric acid unit. From these results it was concluded that the two chiral units composing the diastereomeric CSPs can show "matched" or "mismatched" effect on the chiral recognition according to their absolute stereochemistry.

Affective Priming Effect on Cognitive Processes Reflected by Event-related Potentials (ERP로 확인되는 인지정보 처리에 대한 정서 점화효과)

  • This study was conducted to investigate whether Stroop-related cognitive task will be affected according to the preceding affective valence factored by matchedness in response time(RT) and whether facial recognition will be indexed by specific event-related potentials(ERPs) signature in normal person as in patients suffering from affective disorder. ERPs primed by subliminal(30ms) facial stimuli were recorded when presented with four pairs of affect(positive or negative) and cognitive task(matched or mismatched) to get ERP effects(N2 and P300) in terms of its amplitude and peak latency variations. Behavioral response analysis based on RTs confirmed that subliminal affective stimuli primed the target processing in all affective condition except for the neutral stimulus. Additional results for the ERPs performed in the negative affect with mismatched condition reached significance of emotional-face specificity named N2 showing more amplitude and delayed peak latency compared to the positive counterpart. Furthermore the condition shows more positive amplitude and earlier peak latency of P300 effect denoting cognitive closure than the corresponding positive affect condition. These results are suggested to reflect that negative affect stimulus in subliminal level is automatically inhibited such that this effect had influence on accelerating detection of the affect and facilitating response allowing adequate reallocation of attentional resources. The functional and cognitive significance with these findings was implied in terms of subliminal effect and affect-related recognition modulating the cognitive tasks.

Optical transition dynamics in ZnO/ZnMgO multiple quantum well structures with different well widths grown on ZnO substrates

  • ZnO is a promising material for the application of high efficiency light emitting diodes with short wavelength region for its large bandgap energy of 3.37 eV which is similar to GaN (3.39 eV) at room temperature. The large exciton binding energy of 60 meV in ZnO provide provides higher efficiency of emission for optoelectronic device applications. Several ZnO/ZnMgO multiple quantum well (MQW) structures have been grown on various substrates such as sapphire, GaN, Si, and so on. However, the achievement of high quality ZnO/ZnMgO MQW structures has been somehow limited by the use of lattice-mismatched substrates. Therefore, we propose the optical properties of ZnO/ZnMgO multiple quantum well (MQW) structures with different well widths grown on lattice-matched ZnO substrates by molecular beam epitaxy. Photoluminescence (PL) spectra show MQW emissions at 3.387 and 3.369 eV for the ZnO/ZnMgO MQW samples with well widths of 2 and 5 nm, respectively, due to the quantum confinement effect. Time-resolved PL results show an efficient photo-generated carrier transfer from the barrier to the MQWs, which leads to an increased intensity ratio of the well to barrier emissions for the ZnO/ZnMgO MQW sample with the wider width. From the power-dependent PL spectra, we observed no PL peak shift of MQW emission in both samples, indicating a negligible built-in electric field effect in the ZnO/$Zn_{0.9}Mg_{0.1}O$ MQWs grown on lattice-matched ZnO substrates.

Emission Detection of Mercuric Ions in Aqueous Media Based-on Dehybridization of DNA Duplexes

  • To quantify the presence of mercuric ions in aqueous solution, double-stranded DNA (dsDNA) of poly(dT) was employed using a light switch compound, $Ru(phen)_2(dppz)^{2+}$ (1) which is reported to intercalate into dsDNA of a right-handed B-form. Addition of mercuric ions induced the dehybridization of poly(dT)${\cdot}$poly(dA) duplexes to form a hairpin structure of poly(dT) at room temperature and the metal-to-ligand charge transfer emission derived from the intercalation of 1 was reduced due to the dehybridization of dsDNA. As the concentration of $Hg^{2+}$ was increased, the emission of 1 progressively decreased. This label-free emission method had a detection limit of 0.2 nM. Other metal ions, such as $K^+$, $Ag^+$, $Ca^{2+}$, $Mg^{2+}$, $Zn^{2+}$, $Mn^{2+}$, $Co^{2+}$, $Ni^{2+}$, $Cu^{2+}$, $Cd^{2+}$, $Cr^{3+}$, $Fe^{3+}$, had no significant effect on reducing emission. This emission method can differentiate matched and mismatched poly(dT) sequences based on the emission intensity of dsDNA.

Impact of social relationships on self-related information processing and emotional experiences (사회적 관계가 개인의 정보처리와 정서경험에 미치는 효과)

  • Do social situations have an impact on an individual's information processing and emotional experiences? Two studies were conducted to investigate relationships between self-reference effects, emotional experiences and social information processing. Study 1 examined whether biases favoring self-related stimuli could occur automatically. Participants had to judge whether sequential geometric shape-label pairs matched or mismatched. The results showed that self-related stimuli are more rapidly processed than friends/others-related stimuli. In Study 2, the participants had to recall items which were presented with different instructions (either chosen by a friend or by the computer). Here we explored whether the self-reference effect is reduced in a social learning condition. When comparing the social learning condition (seated in pairs) with the nonsocial learning condition (seated alone), the participants recalled more self-related words in the nonsocial learning condition than in the social learning condition. Importantly, the automatic self-reference effect disappeared in the social learning condition. More friends-related words were recalled in the social condition than self-related words. In addition, while tasting chocolates, the participants judged them to be more likeable in the social condition than in the nonsocial condition. These results implicated that social processing can be useful for reducing the automatic self-reference effects and shared experiences are perceived more intensely than unshared experiences.

Induced Prism by the Categories of Spectacle Frames (안경테 종류에 따른 유발프리즘)

  • Purpose: One of the critical aspects on dispensing glasses is to match the center of pupils to the optical center of lenses as the mismatched glasses are able to induce uncomfortable effects called prism which has been known to induce phoria, a main cause for asthenopia in many cases. Therefore, we investigated the induced prism occurred by mismatching centers between the center of pupils and the optical center of lenses. Methods: In this study, total 103 subjects were examined whether the center of pupils and the optical center of lenses are matched in horizontal and vertical directions, and then, the data was categorized into 4 groups based on the structural components of glasses. Total amount of prism was compared to show the effect of the glasses frame on the prism induction, and the value of measured prism was compared with the German RAL-RG 915 regulations. Results: The results in respect to the horizontal component showed that the induced prism was not found in 10.7% of total subjects. 73.8% of total subjects were influenced by induced prism, the range of prism was in a tolerance level. However, the 15.5% of total subjects seemed to be influenced by prism which is out of the criterion of tolerance. In case of vertical component, 23.3% of total subjects showed no effects of prism while early adopting glasses, 54.4% of total subjects showed a little prism effect within the criterion of tolerance, and 22.3% of total subjects showed the prism effect out of the tolerance range. This data indicates that group A and B that are less likely adjustable by fitting induce more prism than group C and D in horizontal and vertical components. Conclusions: In higher refractive error condition, it was found that aligning the optical center of lenses with the center of pupils by pre-fitting of glasses frame minimized prism induction in horizontal and vertical components, which ameliorates ocular fatigue. Therefore, appropriate optometric dispensing through fitting by opticians and precise design about monocular PD and monocular Oh are necessary to improve visual perception.

Allogeneic Hematopoietic Stem Cell Transplantation in Juvenile Myelomonocytic Leukemia (연소형 골수단구성 백혈병에서의 동종 조혈 모세포 이식)

  • Purpose : The purpose of this study was to evaluate the outcome of children with juvenile myelomonocytic leukemia(JMML) treated with allogeneic hematopoietic stem cell transplantation(allo-HSCT). Methods : Eleven JMML patients aged 8-39 months underwent allo-HSCT. The sources of grafts were unrelated donors(n=7), HLA-matched siblings(n=3) and an HLA 1-antigen mismatched familial donor. All patients had received chemotherapy ${\pm}13$-cis-retinoic acid(CRA) before transplant, and CRA was used, posttransplant, in six patients. Results : Only three patients were in complete remission(CR) at the time of transplantation. Initial chimeric status revealed complete donor chimerism(CC) in five patients, mixed chimerism(MC) in five and autologous recovery(AR) in one. One patient with MC having persistent splenomegaly eventually turned to CC and CR after rapid tapering of cyclosporine, combined with daily use of CRA. An AR case relapsed shortly after transplant but was rescued with second, unrelated cord blood transplantation. Ultimately, six patients are alive, event-free, with a median follow-up of 15.5 months posttransplant. All three deaths occurred in patients who failed to achieve CC, leading to disease progression. Conclusion : We suggest that graft-versus-leukemia effect play an important role and CRA a possible role in posttransplant leukemic involution in JMML. In patients whose leukemic burden is still high with MC after transplant, early tapering of immunosuppressants and introduction of CRA might provide a chance of a cure for some patients.
