The Improvement of the Ionization on Micro Mass Spectrometer using Carbon Nanotube Emitter (탄소나노튜브 방출원을 통한 초소형 질량분석기의 이온화 향상)

  • Recently, mass spectrometers are widely used for in-situ chemical analysis. It has rapid response and high sensitivity. In this paper, we present the fabrication and test of a cold cathode emitter for micro mass spectrometer using CNTs(Carbon nano tubes). The CNTs have good mechanical, electrical and chemical characteristics. So they have a long life time and strong robustness. The micro mass spectrometer is composed of the glass substrate and the silicon substrate. The glass substrate is constructed by electrodes for TOF(Time-of-flight) which analyze an ion with mass to charge ratio as ion separator. The silicon substrate is highly doped wafer which is patterned for gate electrode and then 100 11m dry etching to grow the CNTs as the electron emitter. The CNTs are grown by HFCVD(Hot filament chemical vapor deposition) with sputtering the catalyst. We successfully attained to grow the CNTs and to test the characteristics.

  • In this paper, we are proposing a robust tool which is capable of measuring the size and elemental composition of submicron particles from twenty to several hundreds nanometers at the same time, i.e., named Single Particle Mass Spectrometer (SPMS). The home-made SPMS employs a laser ablation/multi-photon ionization method to tear a nanoparticle into the constituent elemental ions. One thing different from the conventional Aerosol Time-of-Flight Mass Spectrometer (ATOFMS) is the power of the ionization laser. Much strong laser used in this work makes it possible to generate elemental ions rather than molecular ions from a nanoparticle. Also the use of high power laser may guarantee a complete ionization of a particle, which was confirmed by the existence of multiple charged ions. If a particle is evaporated/ionized completely and detected through electric field-free TOF tube without any loss, we can extract the original particle volume from the measured total ion numbers. Collecting a number of particles mass spectra, we get a database of size and elemental composition of nanoparticles, with which we may take a took into any kinds of chemical reaction occurring at nanoscale. Several issues related to size estimation by SPMS will be discussed.

A Study on Particle Diffusion to Develop Faraday Cup Array of Particle Beam Mass Spectrometer System (Faraday cup array 개발을 위한 Particle Beam Mass Spectrometer 시스템 내에서의 입자 확산 연구)

  • The Faraday cup electrode of different size has been developed and evaluated to investigate the diffusion effect of particles by Brownian motion in a particle beam mass spectrometer(PBMS). Particles which focused and accelerated by aerodynamic lens are charged to saturation in an electron beam, and then deflected electrostatically into a Faraday cup detector for measurement of the particle current. The concentration of particles is converted from currents detected by Faraday cup. Measurements of particle current as a function of deflection voltage are combined with measured relationships between particle velocity and diameter, charge and diameter, and mass and diameter, to determine the particle size distribution. The particle currents were measured using 5, 10, 20, 40 mm sized Faraday cup that can be move to one direction by motion shaft. The current difference for each sizes as a function of position was compared to figure out diffusion effect during transport. Polystyrene latex(PSL) 100, 200 nm sized standard particles were used for evaluation. The measurement using 5 mm sized Faraday cup has the highest resolution in a diffusion distance and the smaller particles had widely diffused.

Improvement of protein identification performance by reinterpreting the precursor ion mass tolerance of mass spectrum (질량스펙트럼의 펩타이드 분자량 오차범위 재해석에 의한 단백질 동정의 성능 향상)

  • In proteomics research, proteins are digested into peptides by an enzyme and in mass spectrometer, these peptides break into fragment ions to generate tandem mass spectra. The tandem mass spectral data obtained from the mass spectrometer consists of the molecular weights of the precursor ion and fragment ions. The precursor ion mass of tandem mass spectrum is the first value that is fetched to sort the candidate peptides in the database search. We look far the peptide sequences whose molecular weight matches with precursor ion mass of the mass spectrum. Then, we choose one peptide sequence that shows the best match with fragment ions information. The precursor ion mass of the tandem mass spectrum is compared with that of the digested peptides of protein database within the mass tolerance that is assigned by users according to the mass spectrometer accuracy. In this study, we used reversed sequence database method to analyze the molecular weight distribution of precursor ions of the tandem mass spectra obtained by the FT LTQ mass spectrometer for human plasma sample. By reinterpreting the precursor ion mass distribution, we could compute the experimental accuracy and we suggested a method to improve the protein identification performance.

  • Recently, we reported that significant ion loss occurred prior to its detection in the conventional single particles mass spectrometry and more seriously the loss is ion-kinetic-energy-dependent. These lead to significant error in the measured chemical composition of nanoparticles. Here we attempted to design a novel ion optics that is capable of 100% detection of ions generated from single nanoparticle. Using a commercial software SIMION, we simulated the trajectories of ions launched at different speeds inside the previous single particle mass spectrometer We tested how affect changes in shape of repelling plate, adding Einzel lens, substitution of tube electrode between extraction and acceleration grids. As a results, we could find a best design by assembling the trials in the present condition.

Sll0396 regulates transcription of the phycocyanin genes in Synechocystis sp. PCC 6803

  • An olive-green mutant was generated in Synechocystis sp. strain PCC 6803 by inactivation of the sll0396 gene. Whole-cell absorption spectra of the mutant revealed the missing of phycocyanin peak. An investigation of the low-temperature fluorescence emission spectra revealed that the $sll0396{\Omega}$ mutant has a reduced amount of phycocyanin. Western blot analysis showed that the mutant contained less phycocyanin ${\beta}$- and ${\alpha}$-subunits and lacked the 30- and 32-kDa linker polypeptides, and northern blot analysis revealed that the transcription of the 1.4-kb cpcBA gene encoding the phycocyanin ${\beta}$- and ${\alpha}$-subunits was lower in the mutant. The Sll0396 protein has a DNA-binding motif and shares homology with known response regulators. Our results indicate that Sll0396 plays a regulatory role in the transcription of the phycocyanin genes during phycobilisome synthesis.

  • Potential of mass spectrometer-based electronic nose to discriminate habitat of Angelicae gigantis radix was evaluated using 24 and 86 Korean and non-Korean samples, respectively. Loading plot(s) of principal component analysis of data measured through this system revealed difference between Korean samples (probability; 100%) and non-Korean ones (probability; 95.3%), suggesting this technique could be used as efficient method to differentiate habitat of A. gigantis radix.

  • In this paper, we present the fabrication of a thermoelectron emitter for a micro mass spectrometer. The micro mass spectrometer consists of a silicon substrate with a thermoelecton emitter and glass substrate with an acceleration electrode and a repeller electrode. The hot-filament is fabricated by electroplating. We designed two shapes of thermoelectron emitter and compared the emission current.

A High-Lateral Resolution MALDI Microprobe Imaging Mass Spectrometer Utilizing an Aspherical Singlet Lens

  • We report the construction of a MALDI imaging mass spectrometer equipped with a specially designed laser focusing lens, a compact aspherical singlet lens, that obtains a high-lateral imaging resolution in the microprobe mode. The lens is specially designed to focus the ionization laser (${\lambda}$ = 355 nm) down to a $1{\mu}m$ diameter with a long working distance of 34.5 mm. With the lens being perpendicular to the sample surface and sharing the optical axis with the ion path, the imaging mass spectrometer achieved an imaging resolution of as good as $5{\mu}m$ along with a high detection sensitivity of 100 fmol for peptides. The mass resolution was about 900 (m/${\Delta}m$) in the linear TOF mode. The high-resolution capability of this instrument will provide a new research opportunity for label-free imaging studies of various samples including tissues and biochips, even for the study at a single cell level in the future.

  • This paper proposes methods for improving mass spectral resolution for a gas chromatograph mass spectrometer. The slope signs of the 1st and 2nd fitting functions for the ion signal block of each mass index are obtained, and the unnecessary element signals in the ion signal block are removed. The spectrum can be obtained by obtaining the second-order fitting function of the reconstructed ion signal block using only the effective ion signals. In addition, the resolution of the mass spectrum can be improved by correcting the error caused by the shift of the spectral peak position. To verify the performance of the proposed methods, computer simulations were performed using the actual ion signals obtained from the GC-MS system under development. Simulation results show that the proposed method is valid.
