Kinetics of Horseradish Peroxidase-Catalyzed Nitration of Phenol in a Biphasic System

  • The use of peroxidase in the nitration of phenols is gaining interest as compared with traditional chemical reactions. We investigated the kinetic characteristics of phenol nitration catalyzed by horseradish peroxidase (HRP) in an aqueous-organic biphasic system using n-butanol as the organic solvent and ${NO_2}^-$ and $H_2O_2$ as substrates. The reaction rate was mainly controlled by the reaction kinetics in the aqueous phase when appropriate agitation was used to enhance mass transfer in the biphasic system. The initial velocity of the reaction increased with increasing HRP concentration. Additionally, an increase in the substrate concentrations of phenol (0-2 mM in organic phase) or $H_2O_2$ (0-0.1 mM in aqueous phase) enhanced the nitration efficiency catalyzed by HRP. In contrast, high concentrations of organic solvent decreased the kinetic parameter $V_{max}/K_m$. No inhibition of enzyme activity was observed when the concentrations of phenol and $H_2O_2$ were at or below 10 mM and 0.1 mM, respectively. On the basis of the peroxidase catalytic mechanism, a double-substrate ping-pong kinetic model was established. The kinetic parameters were ${K_m}^{H_2O_2}=1.09mM$, ${K_m}^{PhOH}=9.45mM$, and $V_{max}=0.196mM/min$. The proposed model was well fit to the data obtained from additional independent experiments under the suggested optimal synthesis conditions. The kinetic model developed in this paper lays a foundation for further comprehensive study of enzymatic nitration kinetics.

Suitable Dietary Protein/Lipid Ratio for Hybrid, Female Red Sea Bream Pagrus major and Male Black Sea Bream Acanthopagrus schlegeli in the Juvenile Stage, Compared with Red Sea Bream

  • To determine a suitable dietary protein/lipid (CP/CL) ratio in the early juvenile stages of hybrid porgy ($F_1$), female red sea bream (RSB) ${\times}$ male black sea bream, five diets with various CP/CL ratios-60/7, 55/12, 51/17, 46/23, and 41/28-were prepared and provided to juveniles in triplicate. At the smaller juvenile stage, $F_1$, weighing 0.32 g, a significantly higher specific growth rate (SGR) and feed efficiency (FE) were seen with 60/7 and 55/12 diets. However, in RSB weighing 0.26 g, SGR and FE were higher with the 60/7 diet than the other diets at $21^{\circ}C$. At the larger juvenile stage, $F_1$, weighing 3.7 g, there was no significant difference in SGR or FE among the diets, but RSB weighing 4.0 g fed 60/7, 55/12, and 51/17 diets had higher SGR and FE than 46/23 and 41/28 diets at $24^{\circ}C$. Moreover, survival and apparent nutrient retention of $F_1$ at both stages were significantly higher than those in RSB. These results indicate that both $F_1$ and RSB weighing ca. 0.3 g require a higher dietary CP/CL than those weighing ca. 4 g. Additionally, $F_1$ in both trials showed the suitability of a lower dietary CP/CL than RSB, indicating that mass production of $F_1$ juveniles will be more economical than RSB.

  • With its technical development, digital printing is being universally introduced to the mass production of clothing factories. At the same time, many fashion platforms have been made for customers' participation using digital printing, and a tool is provided in platforms for customers to make designs. However, there is no sufficient solution in the production stage for automatically converting a customer's design into a file before printing other than designating a square area for the pattern designed by the customer. That is, if 30 different designs come in from customers for one shirt, designers have to do the work of reproducing the design on the clothing pattern in the same location and in the same angle, and this work requires a great deal of manpower. Therefore, it is necessary to develop a technology which can let the customer make the design and, at the same time, reflect it in the clothing pattern. This is defined in relation to the existing clothing pattern with digital printing. This study yields a clothing pattern for digital printing which reflects a customer's design in real time by matching the diagram area where a customer designs on a given clothing model and the area where a standard pattern reflects the customer's actual design information. Designers can substitute the complex mapping operation of programmers with a simple area-matching operation. As there is no limit to clothing designs, the variousfashion design creations of designers and the diverse customizing demands of customers can be satisfied at low cost with high efficiency. This is not restricted to T-shirts or eco-bags but can be applied to all woven wear, including men's, women's, and children's clothing, except knitwear.

Modeling of 3D Monte Carlo Ion Implantation in the Ultra-Low Energy for the Fabrication of Giga-Bit Devices (기가 비트급 소자 제작을 위한 3차원 몬테카를로 극 저 에너지 이온 주입 모델링)

  • A rigorous modeling of ultra-low energy implantation is becoming increasingly more important as devices shrink to deep submicron dimensions. In this paper, we have developed an efficient three-dimensional Monte Carlo ion implantation model based on a modified Binary Collision Approximation(BCA). To this purpose, the modified electronic stopping model and the multi-body collision model have been taken into account in this simulator. The dopant and damage profiles show very good agreement with SIMS(Secondary Ion Mass Spectroscopy) data and RBS(Rutherford Backscattering Spectroscopy) data, respectively. Moreover, the ion distribution replica method has been implemented into the model to get a computational efficiency in a 3D simulation, and we have calculated the 3D Monte Carlo simulation into the topographically complex structure.

A Survey of the Implication of the Cancer Screening Program in Health District Centers (보건소의 국가 암 검진사업 실태)

  • Purpose: To evaluate every step of the cancer screening program. Method: 146 of 233 health centers participated in this study. Data were collected by mailing questionnaires between December 2002 and January 2003. The response rate was 65.5%. Result: The government cancer screening program was directed by a variety of departments of the health centers. 41.1% of persons in charges were nurses. 41.3% of the health centers received a list of the appropriate person from the National Health Insurance Cooperation within one or two months. 26.1% of health centers received ??? after five month from when the program started. All the health centers advertised their services, and most of them used mail and mass media 56.4%, used other institute's materials, and 72.2% of them used the government's materials. 76.7% of the heath centers recommended secondary health screening to the persons who had positive results at the first screening. 71.6% of the health centers ascertained the patients with cancer who were diagnosed at secondary screenings. 67.9% of the health centers had registered home-based cancer patients at the health center. 137 out of 146 health centers had a local institute for cancer screening; an average of 4.9 institutes had cancer screening. 80.1% of the persons in charge of the programs felt they needed this program, but 80.8% of them thought the program should be modified. Conclusion: Organized and standardized programs are needed to promote the efficiency of National Cancer Screening Program.

Purification and biological activity of recombinant human bone morphogenetic protein-2 produced by E. coli expression system (E. coli 발현 시스템에 의해 생산된 recombinant human bone morphogenetic protein-2의 정제와 생물학적 활성)

  • Purpose: Bone morphogenetic protein-2(BMP-2) has been shown to possess significant osteoinducitve potential. There have been attempts to overcome a limitation of mass production, and economical efficiency of BMP. The aim of this study was to produce recombinant human BMP-2(rhBMP-2) from E. coli in a large scale and evaluate its biological activity. Materials and Methods: The E.coli strain BL21(DE3) was used as a host for rhBMP-2 production. Dimerized rhBMP-2 was purified by affinity chromatography using Heparin column. To determine the physicochemical properties of the rhBMP-2 expressed in E. coli, we examined the HPLC profile and performed Western blot analysis. The effect of the purified rhBMP-2 dimer on osteoblast differentiation was examined by alkaline phosphatase (ALP) activity and representing morphological change using C2C12 cell. Results: E. coli was genetically engineered to produce rhBMP-2 in a non-active aggregated form. We have established a method which involves refolding and purifying a folded rhBMP-2 dimer from non-active aggregates. The purified rhBMP-2 homodimer was characterized by SDS-PAGE as molecular weight of about 28kDa and eluted at 34% acetonitrile, 13.27 min(retention time) in the HPLC profile and detected at Western blot. The purified rhBMP-2 dimer stimulated ALP activity and induced the transformation from myogenic differentiation to osteogenic differentiation. Conclusion: rhBMP-2 was produced in E. coli using genetic engineering. The purified rhBMP-2 dimer stimulated ALP activity and induced the osteogenic differentiation of C2C12 cells.

Natural Occurring Substances as Corrosion Inhibitors for Tin inSodium Bicarbonate Solutions (중탄산소듐 용액에서 납의 부식방지제인 자연산물질들)

  • The inhibitive action of the aqueous extract of lawsonia, licorice root and carob toward the corrosion of tin electrode in 0.1 M $NaHCO_3$ solutions was investigated using galvanostatic polarization measurements. It was found that the corrosion rate decreases in the presence of these extracts indicating the inhibiting of these compounds. The inhibition efficiency increases with increasing extract concentration. The inhibition action of these extracts was explained in view of adsorption of its compounds onto the tin surface, making a barrier to mass and charge transfer. The adsorption of these extracts on the tin surface was found to be a spontaneous process and follow Freundlich adsorption isotherm. It is also found that these extracts provide a good protection to tin against pitting corrosion in chloride containing solution using potentiodynamic anodic polarization technique.

Applicability of Solidified Soil as a Filling Materials in the Drilling of the Bored-precast Pile (매입말뚝 시공시 현장토를 활용한 고화처리 충전재의 현장 적용성 평가)

  • The use of filling material based on cement paste is inefficient at field construction because it needs a lot of the charging mass. In addition, it has environmental problem according to the large amount of cement use because its strength is also larger than criterion. The excavated soil with stabilizer can be used as the filling materials when the bored pile is constructed. Therefore, this paper describes field application of solidified soil for economical efficiency and environment-friendly. The static axial load tests and the load-transfer measurements were performed to examine the axial resistant behavior of the piles. As results, the flowability, segregation and bleeding, and bond strength of filling materials was a good performance than that of the existing cement paste. But the skin friction of pile by PDA was slightly decreased than that of the existing cement paste. However, as pile filling materials, and in terms of economics and environment, the applicability of filling material is considered very effective.

  • Korea Institute of Civil Engineering and Building Technology has constructed a large scale Dust Thermal Vacuum Chamber to simulate extreme lunar terrestrial environments and to study the Moon as an outposts for space development and exploration. Although a large amount of KLS-1 (Korean Lunar Simulant-1) is required for research, its massive production is practically difficult. This paper describes semi-automatic manufacturing system for massive production of KLS-1 in detail, which is seven times more efficient than manual production. In addition, to increase the similarity with lunar regolith, hydrogen reduction reaction using ilmenite which is one of the minerals was also conducted to simulate nanophase Fe(0) which is the unique property of lunar regolith. As a result, it was found that np-Fe(0) was formed at a temperature of 700℃ or higher, and increased in proportion to the temperature until 900℃.

IEEE 802.11s based Wireless Mesh Networks for Smart Grid (스마트 그리드를 위한 IEEE 802.11s 기반 무선 메쉬 네트워크)

  • We present the concept of applying Wireless Mesh Networking (WMN) technology into Smart Grid, which is recently rising as a potential technology in various areas thanks to its advantages such as low installation costs, high scalability, and high flexibility. Smart Grid is an intelligent, next-generation electrical power network that can maximize energy efficiency by monitoring utility information in real-time and controlling the flow of electricity with IT communications technology converged to the existing power grid. WMNs must be designed for Smart Grid communication systems considering not only the high level of reliability, QoS support and mass-data treatment but also the properties of the traditional power grid. In addition, it is essential to design techniques based on international standards to support interoperability and scalability. In this paper, we evaluate the performance of IEEE 802.11s based Smart Grid Mesh Networks by conducting preliminary simulation studies with the ns-3 simulator. We also outline some challenging issues that should be reviewed when considering WMNs as the candidate for Smart Grid communication infrastructure.
