사실적인 컴퓨터 애니메이션 구현을 위한 증분형 영상 기반 운동 렌더링 기법 (Incremental Image-Based Motion Rendering Technique for Implementation of Realistic Computer Animation)

  • 사실적인 컴퓨터 애니메이션 제작 시 종종 모션 캡쳐 기술이 사용된다. 모션 캡쳐 기술은 대상체의 운동을 측정해서 모델링한 운동 렌더링 결과를 그래픽스로 표현한다. 본 논문에서는 카메라를 사용해서 얻어진 2차원 영상 정보로부터 대상체의 3차원 운동을 측정하여 모델링하는 영상 기반 운동 렌더링 문제를 다룬다. 기존의 영상 기반 운동 렌더링 알고리즘은 계산량이 너무 많거나 정확도가 떨어지는 등의 단점이 있었다. 첫 번 째 단점은 장편 애니메이션 제작시 제작 시간이 너무 길어서 문제가 되고, 두 번 째 단점은 사실적인 애니메이션 구현시 사실감이 저하되는 문제를 야기 시킨다. 이와 같은 기존 방식의 단점을 보완하기 위하여 본 논문에서는 계산량이 적고 정확도가 높은 영상 기반 운동 렌더링 알고리즘을 제안한다. 본 논문에서 제안한 방식에서는 계산량이 적은 증분형 운동렌더링 알고리즘을 최적제어 이론의 시각에서 분석하여 정확도를 향상시키도록 개조한다. 본 방식을 광학식 모션 캡쳐 기술에 적용할 경우 표시자(marker)의 부착 없이도 모션 캡쳐가 가능하다는 부가적 인 장점 또한 얻을 수 있다.

말라리아 매개 모기 Anopheles stephensi에서 트랜스포존 piggyBac을 이용한 Pax6 발현 (Transposon piggyBac mediated Ipax6 Expression in Malaria Vector Anopheles stephensi)

  • Pax6는 진화적으로 잘 보존된 homeobox유전자 그룹의 하나로 배 발생기 동안 시공간적으로 제한되어 발현된다. 이 실험은 말라리아 매개모기인 Anopheles stephemi에서의 Pax6 발현을 서로 다른 분자환경 조건에서 조사해 보기 위해 트랜스포존의 하나인 piggyBac과 Pax6에 결합하는 3xp3-EGFP를 사용한 생식세포 형질전환 방법을 사용하였다. 4개의 형질 전환 계열이 만들어졌고 형질전환율은 6.7%였으며, 도입 유전자는 여러 세대에 걸쳐 안정적으로 발현되었다. 4계열은 3가지의 공간적 발현 형태를 보였으며 이는 트랜스포존 삽입 위치에 따른 enhancing혹은 silencing의 결과로 예상된다. 이 결과를 통해 트랜스포존 piggyBac을 사용한 형질전환 시스템은 일반적인 보고자 유전자 발현 실험에서 다양한 형태의 공간적 발현 결과를 유도하는 매우 효율적인 방법으로 사용될 수 있으리라 예상된다

Association of Single Nucleotide Polymorphisms in Exon 6 Region of BMPRIB Gene with Litter Size Traits in Sheep

  • The objective of this study was to investigate polymorphisms of BMPRIB (bone morphogenetic protein type IB receptor) gene and its effect on litter size traits in sheep. Three populations including 101 Small Tailed Han sheep, 79 Poll Dorset and 81 hybrids (Poll Dorset${\times}$Small Tailed Han sheep) were used to detect the polymorphisms in exon 6 region of sheep BMPRIB gene. A fragment of approximately 190bp was amplified by one pair of primers, the polymorphism was revealed from the analysis of three populations by the technique of PCR -SSCP, and a mutation from A to G at 746 of the coding region was confirmed by sequencing in several individual. Statistical results indicated the distribution of allele B (with a A$\longrightarrow$G mutation) and A (without mutation) or genotype AA, AB and BB frequencies differed in three populations. BB genotype (44.55%) and B allele (66.34%) frequencies of Small Tailed Han sheep were higher than those of the others. Analysis of variance showed that the polymorphism of BMPRIB gene was associated with positive effect on litter size traits. The means of genotype BB and AB were about 1.04 and 0.74 more than genotype AA for litter size (p<0.05). Analysis of BMPRIB genotype effects on litter size in three populations indicates the existence of genotype BB or B allele increases the litter size. It suggested that the polymorphism in exon 6 (at 746 in the coding region) of sheep BMPRIB gene may be used as a marker for early selection of prolificacy in sheep.

Expression of $Ca^{2+}$-activated $K^+$ Channels and Their Role in Proliferation of Rat Cardiac Fibroblasts

  • Cardiac fibroblasts constitute one of the largest cell populations in the heart, and contribute to structural, biochemical, mechanical and electrical properties of the myocardium. Nonetheless, their cardiac functions, especially electrophysiological properties, have often been disregarded in studies. $Ca^{2+}$-activated $K^+\;(K_{Ca})$ channels can control $Ca^{2+}$ influx as well as a number of $Ca^{2+}$-dependent physiological processes. We, therefore, attempted to identify and characterize $K_{Ca}$ channels in rat Cardiac fibroblasts. First, we showed that the cells cultured from the rat ventricle were cardiac fibroblasts by immunostaining for discoidin domain receptor 2 (DDR-2), a specific fibroblast marker. Secondly, we detected the expression of various $K_{Ca}$ channels by reverse transcription polymerase chain reaction (RT-PCR), and found all three family members of $K_{Ca}$ channels, including large conductance $K_{Ca}$ (BK-${\alpha}1-\;and\;-{\beta}1{\sim}4$subunits), intermediate conductance $K_{Ca}$ (IK), and small conductance $K_{Ca}$ (SK$1{\sim}4$ subunits) channels. Thirdly, we recorded BK, IK, and SK channels by whole cell mode patch clamp technique using their specific blockers. Finally, we performed cell proliferation assay to evaluate the effects of the channels on cell proliferation, and found that the inhibition of IK channel increased the cell proliferation. These results showed the existence of BK, IK, and SK channels in rat ventricular fibroblasts and involvement of IK channel in cell proliferation.

Expression of Kainate Glutamate Receptors in Type II Cells in Taste Buds of Rats

  • Glutamate-induced cobalt uptake reveals non-NMDA glutamate receptors (GluRs) in rat taste bud cells. Previous studies suggest that glutamate-induced cobalt uptake in taste cells occurs mainly via kainate type GluRs. Cobaltstained cells were immunoreactive against GluR6 and KA1 subunits of GluRs. However, the functions of those type of receptors are not known yet. It is important question which types of taste cells are cobalt-stained when stimulated by glutamate and whether they express these kinds of GluRs. Circumvallate and foliate papilla of Sprague-Dawley rats (45-60 days old) were used. A cobalt-staining technique combined with immunohistochemistry against specific markers for taste bud cell types, such as blood group H antigen (BGH), $\alpha$-gustducin (Gus), or neural cell adhesion molecule (NCAM) was employed. We also performed double labeling of GluR6 or KA1 subunits of GluR with each specific marker for taste bud cell types. Lots of cobaltstained taste bud cells expressed Gus-like immunoreactivity, and subsets of the cobalt stained cells appeared NCAM- or BGH-like immunoreactivity. Stimulation with 1 mM glutamate significantly increased the number of cobaltstained cells in Gus-like immunoreactive cells, but not in NCAM- or BGH-like immunoreactive cells. In the double labeling experiments, GluR6 and KA1 subunits of GluRs were mainly expressed with Gus. These results suggest that kainate glutamate receptors preferentially expressed in type II taste bud cells in rat.

Comparative Study of Carcinoembryonic Antigen Tumor Marker in Stomach and Colon Cancer Patients in Khyber Pakhtunkhwa

  • Background: Due to the increase in morbidity and mortality rate, cancer has become an alarming threat to the human population worldwide. Since cancer is a progressive disorder, timely diagnosis would be helpful to prevent/stop cancer from progressing to severe stage. In Khyber Pakhtunkhwa, Pakistan, most of the time, tumors are diagnosed with endoscopy and biopsy; therefore rare studies exist regarding the diagnosis of gastrointestinal (GIT) carcinomas based on tumor markers, especially CEA. Objectives: This study made a comparative analysis of CEA in admitted hospitalized stomach and colon cancer patients diagnosed as GIT with biopsy. Materials and Methods: In this study, a total of 66 cases were included. The level of CEA was determined in the blood of these patients using ELISA technique. Results: Out of 66 patients, the level of CEA was high in 59.1% of the total, 60.7% in colon cancer patients and 57.9 % in stomach cancer patients. Moreover, the incidence of colorectal and stomach cancer was greater in males as compared to females. Patients were more of the age group of 40-60 and the level of CEA was comparatively higher in patients (51.5%) with histology which was moderately differentiated, than patients with well differentiated and poorly differentiated tumor histology. Conclusions: CEA level was high in more than 50% of the total patients. Moreover, CEA exhibited higher sensitivity for colon than stomach cancer.

Na+/K+-ATPase Alpha Subunit in the Monogonont Rotifer, Brachionus koreanus: Molecular Cloning and Response to Different Salinity

  • $Na^+/K^+$-ATPase is a membrane protein and plays a key role in osmotic regulation in living organisms. In the present study, a cDNA sequence encoding the $Na^+/K^+$-ATPase alpha subunit from the monogonont rotifer, Brachionus koreanus was cloned by rapid amplification of cDNA ends technique. To investigate the role of this enzyme in osmotic stress, enzymatic activities of $Na^+/K^+$-ATPase were measured after exposure to different salinities for 48 h. The full-length Bk $Na^+/K^+$-ATPase cDNA was 3069 bp-long, encoding a 1022-amino acid polypeptide. Bk $Na^+/K^+$-ATPase possesses eight membrane spanning regions and five conserved domains. Phylogenetic analysis showed that Bk $Na^+/K^+$-ATPase had high identity with those of other species, and was closely clustered with other Brachionus sp. These findings indicate that this protein was conserved both structurally and functionally. B. koreanus $Na^+/K^+$-ATPase activity was stimulated in both hyposaline (6 psu) and hypersaline (32 psu) conditions, suggesting that this protein may play a role in osmoregulation. This study would provide better understanding of the physiology of B. koreanus and this enzyme may be useful as a molecular marker for evaluation of osmotic stress in aquatic environment.

A Gene-Tagging System for Monitoring of Xanthomonas Species

  • A novel chromosomal gene tagging technique using a specific fragment of the fatty acid desaturase-like open reading frame (des-like ORF) from the tox-argK gene cluster of Pseudomonas syringae pv. phaseolicola was developed to identify Xanthomonas spp.released into the environment as biocontrol agents. X. campestris pv. convolvuli FB-635, a pathogen of Convolvulus arvensis L., (bindweed), was chosen as the organism in which to develop and test the system. A 0.52 kb DES fragment amplified from P. syringae pv. phaseolicola C-199 was inserted into pGX15, a cosmid clone containing a 10.3 kb Eco RI-HindIII fragment derived from the xanthomonadin biosynthetic gene cluster contained in plasmid pIG102, to create a pigG::DES insertion. The 10.8 kb EcoRI-BamHI fragment carrying the pigG:: DES insertion was cloned into pLAFR3 to generate pLXP22. pLXP22 was then conjugated into X. campestris pv. convolvuli FB-635 and the pigG::DES insertion integrated into the bacterial chromosome by marker exchange. Rifampicin resistant, tetracycline sensitive, starch hydrolyzing, white colonies were used to differentiate the marked strain from yellow pigmented wild-type ones. PCR primers specific for the unique DES fragment were used for direct detection of the marked strain. Result showed the marked strain could be detected at very low levels even in the presence of high levels of other closely related or competitive bacteria. This PCR-based DES-tagging system provides a rapid and specific tool for directly monitoring the dispersal and persistence of Xanthomonas spp.released into the environment.

Relationship between Center of Pressure and Local Stability of the Lower Joints during Walking in the Elderly Women

  • Objective: The purpose of this study was to determine the relationship between center of pressure (CoP) and local stability of the lower joints, which was calculated based on approximate entropy (ApEn) during walking in elderly women. Method: Eighteen elderly women were recruited (age: $66.4{\pm}1.2yrs$; mass: $55.4{\pm}8.3kg$; height: $1.56{\pm}0.04m$) for this study. Before collecting data, reflective marker triads composed of 3 non-collinear spheres were attached to the lateral surface of the thigh and shank near the mid-segment to measure motion of the thigh and shank segments. To measure foot motion, reflective markers were placed on the shoe at the heel, head of the fifth metatarsal, and lateral malleolus, and were also placed on the right anterior-superior iliac spine, left anterior-superior iliac spine, and sacrum to observe pelvic motion. During treadmill walking, kinematic data were recorded using 6 infrared cameras (Oqus 300, Qualisys, Sweden) with a 100 Hz sampling frequency and kinetic data were collected from a treadmill (Instrumented Treadmill, Bertec, USA) for 20 strides. From kinematic data, 3D angles of the lower extremity's joint were calculated using Cardan technique and then ApEn were computed for their angles to evaluate local stability. Range of CoP was determined from the kinetic data. Pearson product-moment and Spearman rank correlation coefficient were applied to find relationship between CoP and ApEn. The level of significance was determined at p<.05. Results: There was a negative linear correlation between CoP and ApEn of hip joint adduction-abduction motion (p<.05), but ApEn of other joint motion did not affect the CoP. Conclusion: It was conjectured that ApEn, local stability index, for adduction/abduction of the hip joint during walking could be useful as a fall predictor.

상호작용 확장을 위한 상황적 UX 기반의 스마트 증강현실 시스템 설계 및 구현 (Design and Implementation of the Smart AR System based on Contextual UX for Expansion of the Interaction)

  • IT융합기술은 점차적으로 인간 중심적, 저탄소 녹색성장을 모토로 하여 진화하고 있다. 증강현실 기술이란 현실 세계의 감각을 유지하면서 가상의 객체를 혼합할 수 있는 융합콘텐츠 기술이다. 시스템과 사용자 사이의 상호작용이 가능하고, 다양한 응용을 할 수 있으며, 확장성이 매우 높다. 본 논문에서는 기존의 상호작용에 대한 범위를 확장하기 위해 상황적 UX 기반의 스마트 증강현실 시스템을 제안하고자 한다. 실세계 환경 변화에 반응하는 가상 객체를 생성할 수 있는 시스템으로써 사용자는 본인의 개인적인 취향을 직접 가상 객체에 적용할 수 있다. 또한 다수의 가상 객체를 한꺼번에 영상 정합할 수 있으며, 조명의 밝기에 따라 낮과 밤에 해당하는 애니메이션을 보여주기도 한다. 이와 같이 복합적인 사용성을 갖는 시스템을 통해 사용자의 만족도를 높이고, 증강현실 콘텐츠에 대한 지속적인 사용을 유도할 수 있도록 한다.
