수산물통조림의 살균조건에 관한 연구(1) 홍합 보일드 통조림 및 홍합 훈제 기름담금 통조림 (Evaluation of Thermal Processes for Canned Marine Products (1) Canned Boiled Sea-mussel in Brine and Canned Smoked Sea-mussel in Oil)

  • 수산물통조림의 살균조건을 검토하기 위한 연구의 일환으로 홍합 보일드 통조림(평1호부) 및 홍합 훈제 기름담금 통조림(각 3호 B관)에 대하여 업계에서 적용하고 있는 살균조건하에서 열전달치를 측정하고 이로부터 $F_0$값을 산출하여 검토하였다. 홍합 보일드 통조림 (평 1 호부)의 $F_0$ 값은 25.33이었고, 홍합 훈제 기름담금 통조림(각3호 B부)의 $P_0$값은 13.84로, 모두 Clostridium botulinum A 형균의 치사치인 $F_0=4.03$을 충족시키고도 남는 높은 살균강도였다. 특히, 홍합 보일드 통조림(평 1 호관)은 홍합 훈제 기를담금 통조림(각 3호 B관)에 비하여 훨씬 강한 살균강도로 처리하고 있음을 알 수 있었다. 또한, 동일제품에 있어서 살균온도 및 살균시간이 변동하는데 따라서 $F_0$값이 어떻게 변화하는가를 알기 위하여 살균온도별로 $F_0$값에 따른 B값을 산출하여 그 nomograh를 작성하였다.

독도 연안 암반에 서식하는 초대형 저서동물의 수직분포와 산업종의 현존량 추정 (Vertical Distribution of Mega-invertebrate and Calculation to the Stock Assessment of Commercial Species Inhibiting Shallow Hard-bottom in Dokdo, Korea)

  • Distribution and stock assessment of mega-benthos living in the shallow hard bottom around Dokdo were studied in July,2000. Depth and topographical conditions have affected to the densities and biomass of benthic animals. In shallow area, less than 10 m depth, turbo shell Batillus cornutus, mussel, Mytilus corusucs were dominated and showed distinct patterns in vertical distribution. On the other hand, the area over 10m depth, it showed diverse pattern depending on topography. Turbo shell, mussel and sea cucumber Stichopus japonicus were dominated in terms of fishery resources, but abalones were rarely sampled. Stock assessment were estimated to be 6.54 M/T, 3.89 M/T and 8.92 M/T, respectively. Some parts of coastal hard bottom around Dokdo, such as the area between Dongdo and Seodo, seemed to play an Important role as nursery ground. Therefore, it is necessary to the environmental monitoring for coastal fishery managements aspects.

Composition and Structure of Macrofouling Communities on Ocean-going Ships in the Far East Sea Basin

  • Species composition and community structure of the fouling found on the hulls of 28 ships traveling through 6 main shipping routes (SR)in the Far East Sea Basin were analyzed using statistical methods. Samples obtained during 1976-1990 expeditions of the Institute of Marine Biology were used for the analyses. These samples were taken from the ships anchored in the harbor by SCUBA diving and in dry-docks of the Vladivostok ship-repairing yard. Similar composition of the fouling communities occurred on the ships travelling the same SR. In five cases, fouling was dominated by different Cirripedia communities. And, in one case, a community of the mussel Mytilus trossulus was found. In most cases the results of the factor analyses showed extremely low level of the relationships among different animals and algal species in fouling communities. Each ocean-going ship had an original structure of the fouling. Spatially disconnected animal associations of tropical and boreal origin may simultaneously coexist at the same ship. This paper testified to the originality of the zone of anthropogenic substrata as a benthos concentrator in the pelagic regions of the world ocean. The fouling from different zones showed that each zone possesses peculiar features and regularities of the composition and relationships between organisms dwelling here.

Studies for Reestabilishment of Approval Toxin Amount in Paralytic Shellfish Poison-Infested Shellfish 1. Toxicity Change in Paralytic Shellfish Poison-Infested Blue mussel, Mytilus edulis and Oyster, Crassostrea gigas during Boiling and Canning Processes

  • The studies on the detoxification of paralytic shellfish poison (PSP)-infested blue mussels, Mytilus edulis and oyster, Crassostrea gigas were performed for using of available processing resource. Toxic blue mussel and oysters from Nampo in Masan Bay, Hachong in Koje Bay and Woepori in Koje were used for experimental samples. The toxicity of low toxic blue mussel $(A,\;84{\mu}g/100g;\;B,\;166{\mu}g/100g;\;C,\;295{\mu}g/l00g;\;D,\;557{\mu}g/100g)$ and oyster $(740{\mu}g/100g)$ were reduced below the regulation limit of PSP $(80{\mu}g/100g)$ or undetected level by mouse bioassay after boiling at $98^{\circ}C$ for 10 min and retorting at $115^{\circ}C$ for 70 min, while the toxicity of high toxic blue mussel $(E,\;8,760{\mu}g/100g)$ remained beyond the regulation limit after boiling and retorting at same condition. These results suggested that the regulation limit of PSP could be level up from $(80{\mu}g/100g)$ to about $160{\mu}g/100g$.

  • 1976년과 1997년 남해안의 거제도 외포리에서 채취한 진주담치와 굴의 독력 덴 독소성분을 비교 조사하였다. 독성은 진주담치가 굴에 비하여 약 10배 정도 높았으며(1996년, 진주담치, 8,670 $\mu\textrm{g}$, 굴 860$\mu\textrm{g}$; 1997년, 진주담치 5,657 $\mu\textrm{g}$, 굴 531$\mu\textrm{g}$/100g), 독화기간도 진주담치가 굴에 비하여 길었다. 두 종류의 시료 모두 독소 주성분은 Cl 및 C2 (20~65%)와 gonyautoxin 1, 2, 3, 4 (38~78%)이었다. 그리고 독화초기에는 11$\beta$-epimer toxin(C2, GTX4)의 비율이 25~56mo1e%(1996년)와 25~80mo1e% (1997년)로 11$\alpha$-epimer toxin(Cl, GTX2)의 비율도다 높았다. 그러나 독화기간이 지남에 따라 11$\alpha$-epimer toxin의 비율이 41~57mo1e%(1996년)와 25~56mo1e%(1997년)로 11$\beta$-epimer toxin의 비율보다 높게 나타났다. 이와 같은 독소성분 조성의 변화는 패류내에서 독소가 대사되기 때문인 것으로 추측된다.

설사성 패류독의 LC-MS/MS에 의한 분석 (Detection of Diarrhetic Shellfish Poisons by LC-MS/MS)

  • LC-MS/MS법에 의하여 설사성 패류독소 7종(okadaic acid, dinophysistoxin-1, -3, pectebotoxin-1, -2, -6, yessotoxin)의 동시 분석 조건을 확립하였고, 2006년 3월부터 9월까지 매월 5종의 이매패(굴, 진주담치, 반지락, 가리비, 개조개, 6월부터 9월까지는 가리비 대신 피조개)를 정기적으로 통영시장에서 구입하여 분석하였으며, 기존의 마우스 검사법과 비교하였다. 마우스 검사법으로는 총 35종 시료 중 3월의 굴과 진주담치에서 0.05${\sim}$0.1 MU/g의 독성을 나타내었으나, 이미 알려진 설사성 패류독소는 전혀 검출되지 않아 다른 성분에 의한 것으로 판단되었다. 한편, LC-MS/MS법에 의하여서는 6월 진주담치에서 dinophysistoxin-1(DTX1)이 0.05 ${\mu}g/g$ 검출되었으며, 8월의 모든 시료에서 okadaic acid(OA)가 0.01${\sim}$0.02 ${\mu}g/g$ 검출되었다. 이들은 외국의 기준치[0.05 MU/g(일본), OAs(OA+DTX1) 0.16 ${\mu}g/g$(EU)]보다 매우 낮은 함량으로 식품 위생상 안전하였다.

  • Mussels have been commonly used in bioassay for quality assessments of environment. Moreover, several standard protocols for the developmental bioassay of bivalves have been proposed. In this study, the EC50 of polycyclic aromatic hydrocarbons (PAHs) was determined using mussel, Mytilus galloprovincialis embryonic developmental bioassay. To determine the sensitivity of M. galloprovincialis, their fertilized eggs were exposed to five PAHs (2-metylnaphthalene, fluorene, dibenzothiophene, phenanthrene, pyrene). The EC50 of 2-metylnaphthalene, fluorene, dibenzothiophene, phenanthrene, and pyrene were 232, 273, 67.9, 43.2, and $33.1{\mu}g/L$, respectively. The overall sensitivity of M. galloprovincialis in the present developmental bioassay was similar to or more sensitive than that of other marine organisms commonly used in aquatic bioassays. The results of this study could be provide with fundamental data of setting standard for protection of marine life and or can use prediction the aquatic toxicity of PAHs.

유전독성물질로 오염된 해양생물의 생물검정법으로서 comet assay 이용 (Use of comet assay as a bioassay in marine organisms exposed to genotoxicants)

  • Using single cell gel electrophoresis, DNA single strand breaks were determined in various marine organisms. DNA damage on fish blood cells was detected to know whether there was a difference between Incheon, Pohang, Masan, and Tongyeong as a control site. Tongyeong showed the lowest DNA damage among the study areas. Mussels, transplanted to Masan Bay for one month, revealed high DNA damage at sites with high economical activity. In two weeks exposure of polychaete to Incheon sediments, higher DNA damage was detected in the sediment adjacent to Incheon harbor than open sea. These results suggested that the marine organism from the polluted area revealed a relatively high DNA damage. In addition, these areas might be contaminated with genotoxic compounds and comet assay was useful as a bioassay to detect DNA damage in marine organisms.
